For the first time horseradish peroxidase (HRP) immobilized on the surface of active carbon powder modified at the surface of a glassy carbon electrode has been shown to undergo a direct quasi-reversible electrochemical reaction. Its formal potential, E-o/, is -0.363 V in phosphate buffer solution (pH 6.8) at a scan rate of 100 mV/s and is almost independent of the scan rate in the range of 50-700 mV/s. The dependence of E-o/ on the pH of the buffer solution indicated that the conversion of HRP-Fe(III)/HRP-Fe(II) is a one-electron-transfer reaction process coupled with one-proton-transfer. The experimental results also demonstrated that the immobilized HRP retained its bioelectrocatalytic activity to the reduction of H2O2. Furthermore, the HRP adsorbed oil the surface of the active carbon powder can be stored at 4 degreesC for several months without any loss of the enzyme activity. The method presented for immobilizing HRP can be easily extended to immobilize and obtain the direct electrochemistry of other enzymes.

The partitioning behavior of four amino acids, cysteine, phenylalanine, methionine, and lysine in 15 aqueous two-phase systems (ATPSs) with different polyethylene glycol (PEG) molecular weights and phosphate buffers has been studied in the present paper. The phase diagrams of the systems are investigated together with the effect of the PEG molecular weight and pH of the phosphate solutions. The composition of these systems and some parameters such as density and refractive index are determined. The influences of salts in ATPSs, side chain structure of the amino acids, pH of ATPSs, and the PEG molecular weight on the distribution ratios of the amino acids have been studied. This work is useful for the purification of amino acids and the separation of some proteins whose main surface exposed amino acid residues are these four amino acids, respectively.

The target DNA was immobilized successfully on gold colloid particles associated with a cysteamine monolayer on gold electrode surface. Self-assembly of colloidal An onto a cysteamine modified gold electrode can enlarge the electrode surface area and enhance greatly the amount of immobilized single stranded DNA (ssDNA). The electrontransfer processes of [Fe(CN)(6)](4)-/[Fe(CN)(6)](3-) on the gold surface were blocked due to the procedures of the target DNA immobilization, which was investigated by impedance spectroscopy. Then single stranded target DNA immobilized on the gold electrode hybridized with the silver nanoparticle-oligonucleotide DNA probe, followed by the release of the silver metal atoms anchored on the hybrids by oxidative metal dissolution, and the indirect determination of the released solubilized Ag-1 ions by anodic stripping voltammetry (ASV) at a carbon fiber microelectrode. The results show that this method has good correlation for DNA detection in the range of 10-800 pmol/1 and allows the detection level as low as 5 pmol/1 of the target oligonucleotides.

A novel sensitive electrochemical immunoassay with colloidal gold as the antibody labeling tag and subsequent signal amplification by silver enhancement is described. Colloidal gold was treated by a light-sensitive silver enhancement system which made silver deposit on the surface of colloidal gold(form Au/Ag core-shell structure), followed by the release of the metallic silver atoms anchored on the antibody by oxidative dissolution of them in an acidic solution and the indirect determination of the dissolved Ag+ ions by anodic stripping voltammetry(ASV) at a carbon fiber microelectrode. The electrochemical signal is directly proportional to the amount of analyte(goat IgG) in the standard or a sample. The method was evaluated by means of a noncompetitive heterogeneous immunoassay of immunoglobulin G(IgG) with a concentration as low as 0.2 ng/ mL. The high performance of the method is related to the sensitive ASV determination of silver(I) at a carbon fiber microelectrode and to the release of a large number of Ag+ ions from each silver shell anchored on the analyte(goat IgG).

A novel phosphor Sr2CeO4 was synthesized by the citrate-gel method. The results of XRD show that the temperature at which the crystallization starting is lowered and the soak time for complete crystallization is decreased. The values of pH of the precursor and the ratio( R) between the citrate and cation ions have an effect on the crystallization process. The host can transfer its exciting energy to rare earth ion Eu3+. The doped compound emits strong white light when the concentration of the doped Eu3+ is low. When that of Eu3+ is increased, it emits strong red light. The fluorescence from the higher excited states can be observed because the multiphonon relaxation probability between Eu3+ ions is low.

The abzyme (Se-6E8) with a higher thyroxine deiodinase activity was prepared by modifying the serine residues of monoclonal antibody (6E8)with phenylmethanesulfonyl fluoride and sodium hydrogen selenide, and the 6E8 against O-methyl-T-4, which is a kind of thyroxine derivatives and was taken as a hapten for the first time. Two bands were found corresponding to the 5.5 kD heavy chain and the 2.7 kD light chain respectively by SDS-PAGE. The characteristics of dissociation constants, pH, and temperature were also studied. The results show that the activity of Se-6E8 is 2 010 U/mumol protein, and the proper temperature and pH of the catalytic reactions is 57 degreesC and 8.2 respectively.

The behavior of arachidic acid on the surface of YCI3 aqueous subphase was studied by LB and Brewster angle microscopy techniques. The results showed that the pre-compressing time and the pH of the subphase played an important role in the forming of the monolayer. The monolayer on the subphase surface was irreversible. If the monolayer was compressed into wrinkles, the monolayer could not become uniform again. The optimum transferring conditions were selected and the ordered yttrium arachidate multilayer with a long spacing of 4.96 nm and a tilt angle of 28.5degrees of the three alkyl chains from the surface normal was fabricated and characterized.

The general and synchronous spectra of phospholipase A(2) (PLA(2)) isolated from Chinese agkistrodon blomhoffii Ussurensis snake venom were studied. The chromophores of PLA(2) were mainly contributed by tyrosine and tryptophane residues when the intervals between the excitation wavelength and the emssion waveleagth (Delta lambda) were 20nm and 75nm, respectively. The pH of buffers could change the fluorescence intensities of PLA(2) by changing the charge distribution of its amino acid chain. Ca2+ can not only increase the emission fluorescence intensity of PLA(2) but also improve the reaction rate of PLA(2) with its corresponding substrate DPPC.

Electrocatalytic reduction of O-2 and H2O2 at the glass carbon electrode modified with microperoxidase-11 immobilized with Nafion film has been studied by means of cyclic voltammetry and rotating disk electrode techniques. The modified electrode shows high catalytic activity toward the reduction of both O-2 and H2O2. The rate constants of Oz and H2O2 reduction at the modified electrode have been measured and compared. It is found that O-2 undergoes a four-electron reduction at the modified electrode and the catalytic activity for the reduction of O-2 is dependent on the pH of the solutions.

The p-toluene sulfonic acid (MA) in phenol matrix was separated and determined by capillary electrophoresis with ultraviolet detector. the effect of the concentration and pH of the buffer on separation was investigated. Cinnamic acid has been chosen as the internal standard from four compounds, the calibration curves of PTSA in 50 mg/L phenol matrix were obtained with and without the internal standard. The linear range was from 1.25 to 12.5 mg/L and the correlation coefficient was 0.9999 for both curves. The limit of detection of PISA was 0.75 mg/L at 3 times of SIN. Finally, the concentration of PTSA in four synthesized samples was determined with method of standard additions, and the effect of matrix was discussed. The values of MA in these samples were 1.01, 0.94, 1.56 and 0.00 mg/L respectively.

Non-stoichiometric mixed-valent molybdenum(VI, V) oxide film was grown on carbon substrates by the electrodeposition method. Responses of the prepared molybdenum oxide thin films to potential and to different solution acidities were studied by cyclic voltammetry, and the corresponding morphological changes of the film were monitored by atomic force microscopy (AFM). AFM images of the molybdenum oxide film show that the characteristic domed structure on the film surface increased during the transition from the oxidized state to the reduced state without signification change in the KMS surface roughness value. Furthermore, AFM studies show that the solution acidity has great effect on the morphology of the films, and the films undergo a homogenizing process with increasing pH of the solutions. Binary blends of polyamide 1010/poly(propylene) and polyamide 1010 (PA1010)/poly(propylene)-graft-(glycidyl methacrylate) (PP-g-GMA) were prepared. The epoxy groups in PP-g-GMA react with the amino end-groups in PA1010, thus a PA1010-graft-PP copolymer is formed and acts as a compatibilizer between PA1010 and PP-g-GMA. The reaction was confirmed by electron spectroscopy for chemical analysis (ESCA) and attenuated total reflection (ATR)-FTIR spectroscopic analysis, and also evaluated by the stability of the suspension obtained by dissolving the blends in formic acid and by the morphologies of the blends.

Hydrotalcite-like compounds (HTLcs): (CuMAlCO3)-Al-II-HTLcs, where M-II=Co2+, Ni2+, Cu2+, Zn2+ and Fe2+, were synthesized by coprecipitation and characterized with XRD and IR. The catalysis of these HTLcs was studied in the phenol hydroxylation by H2O2 in liquid phase; then the effects of the ratio of Cu/Al, reaction temperature, solvent and pH of medium were investigated. It has been found that the uncalcined HTLcs have higher activities than those of calcined samples in this reaction. The catalyst CuAlCO3-HTLcs having Cu/Al=3 efficiently oxidized phenol and gave high yields of the corresponding diphenols in appropriate reaction conditions. A tentative reaction mechanism is also proposed. (C) 1998 Elsevier Science B.V.
