Growth potential (delta) is defined as the difference between the population of a microorganism at the end of shelf-life of specific food and its initial population. The determination of 6 of Salmonella and Listeria monocytogenes in RTE vegetables can be very useful to determine likely threats to food safety. However, little is known on the behavior of these microorganisms in several RTE vegetables. Therefore, the aim of this study was to determine the delta of both pathogens in nine different types of RTE vegetables (escarole, collard green, spinach, watercress, arugula, grated carrot, green salad, and mix for yakisoba) stored at refrigeration (7 degrees C) and abuse temperature (15 degrees C). The population of aerobic microorganisms and lactic acid bacteria, including those showing antimicrobial activity has been also determined. Results indicated that L monocytogenes was able to grow (delta >= 0.5 log(10)) in more storage conditions and vegetables than Salmonella. Both microorganisms were inhibited in carrots, although a more pronounced effect has been observed against L monocytogenes. The highest 5 values were obtained when the RTE vegetables were stored 15 degrees C/6 days in collard greens (delta=3.3) and arugula (delta=3.2) (L monocytogenes) and arugula (delta=4.1) and escarole (delta=2.8) (Salmonella). In most vegetables and storage conditions studied, the counts of total aerobic microorganisms raised significantly independent of the temperature of storage (p<0.05). Counts of lactic acid bacteria were higher in vegetables partially or fully stored at abuse temperature with recovery of isolates showing antimicrobial activity. In conclusion, the results of this study show that Salmonella and L monocytogenes may grow and reach high populations in RTE vegetables depending on storage conditions and the definition of effective intervention strategies are needed to control their growth in these products. The hydroponic cultivation of vegetables has increased markedly in recent years, however, little is known about its chemical composition, which is of extreme importance in view of changing food habits of a considerable fraction of the population seeking food quality. For this reason, cultivation of watercress, chicory, rocket and lettuce American (Lucy Brown) and smooth (cv. Regina) were grown in hydroponics, NFT system, aiming to evaluate the yield, nitrate content and chemical composition. The experiment was conducted in a greenhouse at the Universidade Federal de Santa Catarina, Florianopolis city in the Santa Catarina State (Brazil), from may to july 2004 under a completely randomized design with four replications. The spacing used was 25 x 25 cm for the crops of watercress, chicory and lettuce (cv. Lucy Brown and cv. Regina) and 5 x 5 cm to the rocket. The traits were: number of leaves, nitrate content and chemical composition of vegetables. The lettuce cv. Regina and chicory had higher number of leaves per plant. The watercress had lower water content and higher dry mass of shoots. The largest increase in fresh weight was obtained in chicory, lettuce, cv. Regina. Higher levels of lipids, protein, ash, carbohydrates, calories, fiber and nitrate were obtained from the watercress. The rocket had lower values for the variables fresh and dries the whole plant, shoot and root and leaf number per plant. All cultures showed good visual appearance, low calorie and nitrate levels suitable for human consumption.

The aim of this study was to investigate the effects of the use of chlorine or ozone as sanitizing agents in the water of chicken immersion chilling, using the residual levels usually applied in Brazil (1.5 ppm), comparing the effects of these treatments on the microbiological, physicochemical, and sensory characteristics of carcasses. Chicken carcasses were chilled in water (4 degrees C) with similar residual levels of ozone and chlorine until reaching temperatures below 7 degrees C (around 45 min). The stability of carcasses was assessed during 15 days of storage at 2 +/- 1 degrees C. Microbiological, surface color (L*, a*, b* parameters), pH value, lipid oxidation (thiobarbituric acid reactive substances index), and sensory evaluation (on a 9-point hedonic scale for odor and appearance) analyses were carried out. The presence of Salmonella was not detected, coagulase-positive staphylococci counts were below 10(2) CFU/ml of rinse fluid, and Escherichia coil and total coliform counts were below 10(5) CFU/ml of rinse fluid until the end of the storage period for both treatments. Psychrotrophic microorganism counts did not differ (P > 0.05) between chlorine and ozone treatments, and both values were near 10(9) CFU/ml of rinse fluid after 15 days at 4 +/- 1 degrees C. pH values did not differ between treatments (P > 0.05) or during the storage period (P > 0.05). In addition, neither chlorine nor ozone treatment showed differences (P > 0.05) in the lipid oxidation of carcasses; however, the thiobarbituric acid reactive substances index of both treatments increased (P <= 0.05) during the storage period, reaching values of approximately 0.68 mg of malonaldehyde per kg. Samples from both treatments did not differ (P > 0.05) in their acceptance scores for odor and overall appearance, but in the evaluation of color, ozone showed an acceptance score significantly higher (P <= 0.05) than that for the chlorine treatment. In general, under the conditions tested, ozone showed results similar to the results for chlorine in the disinfection of chicken carcasses in the immersion chilling, which may indicate its use as a substitute for chlorine in poultry slaughterhouses.

Vegetables are widely consumed in Brazil and exported to several countries. This study was performed to evaluate the phenolic content and antioxidant activity of vegetables commonly consumed in Brazil using five different methods, namely DPPH and ABTS free radical, beta-carotene bleaching, reduction of Fe3+ (FRAP), oxidative stability in Rancimat, and the chemical composition using gas chromatography-mass spectrometry (GC-MS). The content of phenolic compounds ranged from 1.2 mg GA/g (carrot) to 16.9 mg GA/g (lettuce). Vegetables presenting the highest antioxidant activity were lettuce (77.2 mu mol Trolox/g DPPH center dot; 447.1 mu mol F2+/g FRAP), turmeric (118.6 mu mol Trolox/g ABTS(center dot+); 92.8% beta-carotene), watercress and broccoli (protective factor 1.29-Rancimat method). Artichoke, spinach, broccoli, and asparagus also showed considerable antioxidant activity. The most frequent phenolic compounds identified by GC-MS were ferulic, caffeic, p-coumaric, 2-dihydroxybenzoic, 2,5-dihydroxybenzoic acids, and quercetin. We observed antioxidant activity in several vegetables and our results point out their importance in the diet.

O presente trabalho teve como objetivos determinar os teores de nutrientes e de nitrato e realizar análise sensorial de alface cultivada em sistema hidropônico sob água salina. O experimento foi conduzido em ambiente protegido no período de dezembro de 2007 a janeiro de 2008, em Piracicaba (SP). O delineamento experimental foi de blocos ao acaso, sendo estudados cinco níveis de salinidade da água de irrigação, utilizando-se NaCl (Condutividade elétrica da água (CEa): 0,42, 1,53, 3,52, 5,55 e 7,43 dS m-1) em duas cultivares de alface (Verônica e Pira Roxa), em esquema fatorial. Foram avaliados a análise sensorial aplicando escala hedônica e através dessa, o que os provadores mais apreciaram e o que menos gostaram de cada amostra, intenção de compra e consumo, teor de nitrato, absorção de nutrientes pela folhas e determinação dos nutrientes presentes na solução nutritiva. Para o atributo sabor a cultivar Verônica recebeu as melhores notas no nível de salinidade 1,53 e 5,55 dS m-1. Para a cultivar Pira Roxa a melhor nota foi atribuída para a alface produzida na água não salina. Para os demais atributos não houve diferença significativa nos diferentes níveis de salinidade. Com relação à intenção de compra, constatou-se que as alfaces cv. Verônica e Pira Roxa apresentaram boa aceitação de mercado. Os menores níveis de nitrato (1960 mg kg-1 e 2620 mg kg-1 de massa de matéria fresca), da Verônica e Pira Roxa, respectivamente, foram relacionados à condutividade elétrica de 0,42 dS m-1, aumentando a salinidade da água para 7,43 dS m-1, o teor foliar de nitrato aumentou para 2500 mg kg-1 e 3420 mg kg-1 para as cultivares Verônica e Pira Roxa. Como o tempo de exposição da alface à salinidade em sistema hidropônico foi curto, em torno de 25 dias, não se verificou sintomas de deficiência nutricional e toda alface foi classificada como apropriada para consumo humano.

O vigor da semente tem sido avaliado por meio de testes fisiológicos, bioquímicos e de tolerância a estresse. Mais recentemente, com o uso da análise computadorizada de imagens de plântulas, podem ser obtidas informações objetivas, em período relativamente curto, com menor interferência humana. O objetivo nesta pesquisa foi identificar testes eficientes para avaliação do vigor de sementes de alface, com ênfase para a análise de imagens digitais de plântulas. Sementes de 12 lotes (seis da cultivar Vanda e seis da Vera) foram avaliadas por meio de testes de germinação, germinação a baixa temperatura (15ºC), emergência de plântulas e envelhecimento acelerado tradicional e com solução salina (NaCl 40%) saturada, a 41ºC/48 h e 41ºC/72 h. Utilizou-se, também, o sistema de análise computadorizada de imagens de plântulas (SVIS®), desenvolvido pela Ohio State University (EUA), que forneceu dados referentes ao índice de vigor, uniformidade e crescimento de plântulas. Os testes foram realizados a cada três meses, durante 12 meses de armazenamento, sob condições ambiente. Com base nos resultados obtidos, concluiu-se que os testes de envelhecimento acelerado com uso de solução saturada de NaCl, a 41ºC por 48 e 72 horas e de velocidade de germinação, e os parâmetros obtidos com o software SVIS, índice de crescimento de plântulas e comprimento da raiz primária, são eficientes na avaliação do vigor de lotes de sementes de alface.

A reduced cancer risk associated with fruit and vegetable phytochemicals initially dictated chemopreventive approaches focused on specific green variety consumption or even single nutrient supplementations. However, these strategies not only failed to provide any health benefits but gave rise to detrimental effects. In parallel, public-health chemoprevention programmes were developed in the USA and Europe to increase whole vegetable consumption. Among these, the National Cancer Institute (NCI) sponsored plan “5 to 9 a day for a better health” was one of the most popular. This campaign promoted wide food choice through the consumption of at least 5 to 9 servings a day of colourful fruits and vegetables. In this study the effects of the diet suggested by NCI on transcription, translation and catalytic activity of both xenobiotic metabolizing (XME) and antioxidant enzymes were studied in the animal model. In fact, the boost of both antioxidant defences and “good” phase-II together with down-regulation of “bad” phase-I XMEs is still considered one of the most widely-used strategies of cancer control. Six male Sprague Dawley rats for each treatment group were used. According to the Italian Society of Human Nutrition, a serving of fruit, vegetables and leafy greens corresponds to 150, 250 and 50 g, respectively, in a 70 kg man. Proportionally, rats received one or five servings of lyophilized onion, tomato, peach, black grape or lettuce – for white, red, yellow, violet or green diet, respectively - or five servings of each green (“5 a day” diet) by oral gavage daily for 10 consecutive days. Liver subcellular fractions were tested for various cytochrome P450 (CYP) linked-monooxygenases, phase-II supported XMEs such as glutathione S-transferase (GST) and UDP-glucuronosyl transferase (UDPGT) as well as for some antioxidant enzymes. Hepatic transcriptional and translational effects were evaluated by reverse transcription-polymerase chain reaction (RT-PCR) and Western blot analysis, respectively. dROMs test was used to measure plasmatic oxidative stress. Routine haematochemical parameters were also monitored. While the five servings administration didn’t significantly vary XME catalytic activity, the lower dose caused a complex pattern of CYP inactivation with lettuce exerting particularly strong effects (a loss of up to 43% and 45% for CYP content and CYP2B1/2-linked XME, respectively; P<0.01). “5 a day” supplementation produced the most pronounced modulations (a loss of up to 60% for CYP2E1-linked XME and a reduction of CYP content of 54%; P<0.01). Testosterone hydroxylase activity confirmed these results. RT-PCR and Western blot analysis revealed that the “5 a day” diet XMEs inactivations were a result of both a transcriptional and a translational effect while lettuce didn’t exert such effects. All administrations brought out none or fewer modulation of phase-II supported XMEs. Apart from “5 a day” supplementation and the single serving of lettuce, which strongly induced DT- diaphorase (an increase of up to 141 and 171%, respectively; P<0.01), antioxidant enzymes were not significantly changed. RT-PCR analysis confirmed DT-diaphorase induction brought about by the administration of both “5 a day” diet and a single serving of lettuce. Furthermore, it unmasked a similar result for heme-oxygenase. dROMs test provided insight into a condition of high systemic oxidative stress as a consequence of animal diet supplementation with “5 a day” diet and a single serving of lettuce (an increase of up to 600% and 900%, respectively; P<0.01). Haematochemical parameters were mildly affected by such dietary manipulations. According to the classical chemopreventive theory, these results could be of particular relevance. In fact, even if antioxidant enzymes were only mildly affected, the phase-I inactivating ability of these vegetables would be a worthy strategy to cancer control. However, the recorded systemic considerable amount of reactive oxygen species and the complexity of these enzymes and their functions suggest caution in the widespread use of vegan/vegetarian diets as human chemopreventive strategies. In fact, recent literature rather suggests that only diets rich in fruits and vegetables and poor in certain types of fat, together with moderate caloric intake, could be associated with reduced cancer risk.

Für die Etablierung einer Transformationsmethode züchterisch relevanter Sorten von Osteospermum ecklonis (Kapmargerite) wurde zunächst ein geeignetes Protokoll für die Regeneration adventiver Sprosse aus vegetativem Gewebe entwickelt. Anschließend wurden Transformationen von Markergenen durch Kokultur mit Agrobacterium tumefaciens durchgeführt. Hierzu wurden Konstrukte verwendet, die das Gen für ß-D-Glucuronidase (GUS) enthielten und deren Expression in transgenen Pflanzen histochemisch nachgewiesen werden konnte. Kanamycinresistenz erwies sich als geeigneter Selektionsmarker für die Transformation. Es konnten von verschiedenen O. ecklonis Sorten GUS-transgene, nicht-chimäre Pflanzen regeneriert werden.Zur Erzeugung transgener Pflanzen mit dem Ziel der Resistenz gegen LMV (lettuce mosaic potyvirus, Salat Mosaik Virus) wurden drei Konstrukte verwendet. Das erste enthält die kodierende Sequenz der Virusproteine VPg, Pro und 6K2. Durch PCR-Mutation wurde die Proteinase-Schnittstelle zwischen 6K2 und VPg zerstört, sowie Start- und Stopcodon eingeführt. Die anderen LMV-abgeleiteten Konstrukte enthalten nicht translatierbare Fragmente des coat protein Gens in sense und antisense Orientierung.Außerdem wurde O. ecklonis noch mit dem Gen des mutmaßlichen Transkriptionsfaktor SPL3 aus Arabidopsis thaliana unter der Kontrolle eines konstitutiven Promotors transformiert. SPL3 ist an der Regulierung der Blüteninduktion in A. thaliana beteiligt.Regenerierte O. ecklonis wurden durch PCR mit konstruktspezifischen Primern auf Anwesenheit des Transgens und Kontamination durch A. tumefaciens überprüft.

The demand of minimally processed fruits and vegetables has increased in the last years. However, their intrinsic characteristics may favor the growth of pathogens and spoilage microbiota. The negative effects on human health reported for some traditional chemical sanitizers have justified the search for substitutes to guarantee food safety and quality. In this work we have evaluate the potential of some essential oils and their components to improve the safety and the shelf life of Lamb’s lettuce (Valerianella locusta) and apples (Golden delicious). Moreover, the effects of selected lactic acid bacteria alone or in combination with essential oils or their components, on the shelf-life and safety as well as organoleptic properties of minimally processed products, were evaluated. Since the lack of knowledge of microbial cell targets of essential oils represent one of the most important limit to the use of these molecules at industrial level, another aim of this thesis was the study of the action mechanisms of essential oils and their components. The results obtained showed the beneficial effects of the natural antimicrobials as well as the selected lactic acid bacteria on minimally processed fruit and vegetable safety and shelf-life, without detrimental effects on the quality parameters. The beneficial effects obtained by the use of the selected biocontrol agents were further increased combining them with selected natural antimicrobials. The natural antimicrobial employed induced noticeable modifications of membrane fatty acid profiles and volatile compounds produced by microbial cells during the growth. The modification of the expression in genes involved in fatty acid biosynthesis suggesting that the cytoplasmic membrane of microbial cells is one of the major cellular target of essential oils and their components. The comprehension of microbial stress response mechanisms can contribute to the scaling up of natural antimicrobials and bio-control agents at industrial level.

Background. The prevalence of obesity and overweight children has been an ongoing health epidemic in the US for the last several decades. The problem has consistently worsened and has disproportionately been the most prevalent among low socioeconomic status (SES) populations. Food availability in the home has been suggested to be a potential factor related to overweight and obesity, as availability is likely associated with intake. Food availability of low SES preschool aged children has not been well examined. The purpose of this study was to explore the food environment of the Harris County Department of Education (HCDE) Head Start population, and describe reported frequency of intake of particular food groups. The effect of food availability on reported intake was also examined.^ Methods. This was a cross-sectional study of secondary data analysis. Data obtained from 17 HCDE Head Start Centers was analyzed using PASW 18 Statistical Software. Demographic analyses included population, age, gender, race, parent occupation, type of home, and language spoken in the home. Descriptive statistics included reported availability of foods in the home as well as frequency of intake.^ Regression analysis examined the relationship of availability of foods on intake. The food categories included were: dark leafy green and orange vegetables, other vegetables, fruits, soda, salty snacks, and sweet snacks. For both vegetable categories reported intake of fresh, frozen, and canned vegetables were included. For the fruit category, intake of fresh, frozen, canned, and dried fruits were reported.^ Results. Results showed that 90-95% of parents reported having vegetables and fruits available in the home. However, the only significant relationship between availability and intake was for fresh fruit and dried fruit. No associations were seen among the vegetable groups. Other vegetables (bell peppers, eggplant, tomatoes, onions, iceberg lettuce, asparagus) that were frozen, approached significance for availability on intake, however once adjusted for confounders the relationship was no longer present. Among soda, salty snacks, and sweet snacks the only significant relationship was seen for soda availability and intake. Salty snacks and sweet snacks presence in the home was not a predictor of increased frequency of intake.^ Conclusions. This research supported the hypothesis that availability of foods has an impact on intake for fresh fruits, dried fruits and soda. No associations were seen for vegetables, salty snacks and sweet snacks. Additionally, most of the parents reported having fruits and vegetables in the home, but reported intakes were not meeting the Dietary Guidelines for Americans recommendations. Strengths of the study included the large sample size taken from numerous HCDE Head Start Centers. Limitations included questionable reliability of participant’s responses, ability to generalize to other populations, and the use of secondary data rather than prospectively collected data.^

Para la cuantificación de nitratos hay numerosas técnicas y no existe entre los analistas unanimidad en la selección de la más adecuada. Por tal motivo, se compararon cuatro métodos para la determinación de nitratos en muestras vegetales con el fin de evaluar la correlación entre los mismos y establecer pautas para su utilización. Se utilizaron 690 muestras de lechuga (Lactuca sativa L. var. capitata), pertenecientes a los tipos arrepollado y mantecoso, recolectadas a lo largo de un año en el Mercado Cooperativo de Guaymallén (Mendoza, Argentina). Según los tenores de nitratos encontrados en la población estudiada se efectuó un sub-muestreo aleatorio estratificado proporcional para lograr un número de muestras que representaran la variabilidad del total de la población. Se utilizaron cuatro métodos para la determinación de nitratos: 1. destilación por arrastre con vapor, considerado como método de referencia 2. colorimetría por nitración con ácido salicílico 3. colorimetría modificada 4. potenciometría con electrodo selectivo Se probaron diferentes modelos de regresión entre el método de referencia y los otros tres, siendo el lineal el que mejor se ajustó en todos los casos. Los métodos estudiados tuvieron comportamiento semejante. La mayor correlación (r2 = 93 %) se observó entre la destilación por arrastre con vapor y la potenciometría; no obstante, los restantes también presentaron alta correlación. Consecuentemente, la elección del procedimiento analítico dependerá principalmente del número de muestras a analizar, del tiempo requerido por el análisis y del costo del mismo.

En la provincia de La Pampa son escasos los datos sobre relevamiento de plagas y de especies benéficas en lechuga, por ello resulta necesario conocer e investigar los organismos perjudiciales y sus enemigos naturales y las condiciones predisponentes al incremento de sus poblaciones. Los objetivos de este trabajo fueron determinar las especies fitófagas para dos variedades de lechuga, en dos sistemas de producción, e identificar las especies benéficas que puedan actuar como controladores biológicos. El estudio se realizó en la huerta de la Facultad de Agronomía de la UNLPam, con muestreos semanales, desde julio hasta octubre de 2004, a campo con riego complementario y bajo cubierta en macrotúnel con riego por goteo, con las variedades de lechuga Grand Rapid y Gallega de Invierno. En cada variedad y en cada sistema se revisaron 6 plantas al azar, registrándose: especie, número de individuos y su estado de desarrollo. Se comparó mediante ANOVA y test de Tukey (a = 0,05). Se detectó la presencia de dos especies de pulgones: Myzus persicae Sulzer (Homoptera: Aphididae) y Nasonovia ribisnigri (Mosley) (primera cita de N. ribisnigri para la provincia de La Pampa). El mayor número de pulgones se concentró en el cultivo protegido bajo cubierta. Se registraron dos enemigos naturales de los pulgones: un hongo entomopatógeno y un parasitoide: Aphidius ervi (Hymenoptera: Braconidae). También se encontraron, en bajas densidades, larvas de Naupactus leucoloma Boheman (Coleoptera: Curculionidae) que se alimentaban de las hojas.
