Vertebrate hematopoietic stem cells are derived from vental mesoderm, which is postulated to migrate to both extra- and intraembryonic positions during gastrula and neurula stages. Extraembryonic migration has previously been documented, but the origin and migration of intraembryonic hematopoietic cells have not been visualized. The zebrafish and most other teleosts do not form yolk sac blood islands during early embryogenesis, but instead hematopoiesis occurs solely in a dorsal location known as the intermediate cell mass (IM) or Oellacher. In this report, we have isolated cDNAs encoding zebrafish homologs of the hematopoietic transcription factors GATA-1 and GATA-2 and have used these markers to determine that the IM is formed from mesodermal cells in a posterior-lateral position on the yolk syncytial layer of the gastrula yolk sac. Surprisingly, cells of the IM then migrate anteriorly through most of the body length prior to the onset of active circulation and exit onto the yolk sac. These findings support a hypothesis in which the hematopoietic program of vertebrates is established by variations in homologous migration pathways of extra- and intraembryonic progenitors.

We investigated the relationship between the fusion selectivity of the envelope glycoprotein (env) and the tropism of different human immunodeficiency virus type 1 (HIV-1) isolates for CD4+ human T-cell lines vs. primary macrophages. Recombinant vaccinia viruses were prepared encoding the envs from several well-characterized HIV-1 isolates with distinct cytotropisms. Cells expressing the recombinant envs were mixed with various CD4+ partner cell types; cell fusion was monitored by a quantitative reporter gene assay and by syncytia formation. With CD4+ continuous cell lines as partners (T-cell lines, HeLa cells expressing recombinant CD4), efficient fusion occurred with the envs from T-cell line-tropic isolates (IIIB, LAV, SF2, and RF) but not with the envs from macrophage-tropic isolates (JR-FL, SF162, ADA, and Ba-L). The opposite selectivity pattern was observed with primary macrophages as cell partners; stronger fusion occurred with the envs from the macrophage-tropic than from the T-cell line-tropic isolates. All the envs showed fusion activity with peripheral blood mononuclear cells as partners, consistent with the ability of this cell population to support replication of all the corresponding HIV-1 isolates. These fusion selectivities were maintained irrespective of the cell type used to express env, thereby excluding a role for differential host cell modification. We conclude that the intrinsic fusion selectivity of env plays a major role in the tropism of a HIV-1 isolate for infection of CD4+ T-cell lines vs. primary macrophages, presumably by determining the selectivity of virus entry and cell fusion.

In addition to their well-recognized hepatotropism, all hepatitis B viruses (HBVs) display marked species specificity, growing poorly or not at all in species other than those closely related to their natural hosts. We have examined the molecular basis for this narrow host range, using duck HBV (DHBV) and heron HBV (HHBV) as a model system. HHBV virions will not infect ducks in vivo and infect cultured duck hepatocytes extremely inefficiently in vitro. Mutant HHBV genomes lacking all viral envelope proteins (HHBV env-) can be complemented in trans with DHBV envelope proteins; the resulting pseudotyped virions can efficiently infect duck hepatocytes. Further complementation analysis reveals that of the two viral surface proteins (L and S), it is the L protein that determines host range. Pseudotyping of HHBV env- with DHBV/HHBV chimeric envelope proteins reveals that replacement of as few as 69 amino acids of the pre-S domain of the HHBV L protein by their DHBV counterparts is sufficient to permit infection of duck hepatocytes. These studies indicate that the species-specificity of hepadnaviral infection is determined at the level of virus entry and is governed by the pre-S domain of the viral L protein.

Research has shown that more than half of attempted recovery efforts fail, producing a ‘double deviation’ effect. Surprisingly, these double deviation effects have received little attention in marketing literature. This paper examines what happens after these critical encounters, which behavior or set of behaviors the customers are prone to follow and how customers’ perceptions of the firm’s recovery efforts influence these behaviors. For the analysis of choice of the type of response (complaining, exit, complaining and exit, and no-switching), we estimate multinomial Logit models with random coefficients (RCL). The results of our study show that magnitude of service failure, explanations, apologies, perceived justice, angry and frustration felt by the customer, and satisfaction with service recovery have a significant effect on customers’ choice of the type of response. Implications from the findings are offered.

Different catalysts, based on heteropolyacids supported on activated carbon fibers, have been prepared for palmitic acid esterification reaction. The influence of the catalyst (heteropolyacid) and the support on the catalytic activity have been analyzed. The results prove that an adequate combination of both is required to achieve the most suitable catalysts. Regarding to the heteropolyacid, phosphomolybdic acid seems to be the most suitable appropriate taking into account its lowest leaching. About the support, it must show an optimum microporosity, which must be wide enough to allow the entrance and exit of the reagents and products but not too wide in order to avoid the leaching of the catalyst. In addition, both decreasing of the catalytic activity and its recovery over several cycles have been analyzed.

College Book 6 is often referred to as the Hollis book, reflecting its contents. It was created following an April 4, 1726 Corporation vote that "Mr. Treasurer procure a Book, into which shall be transmitted, and a Register kept of, Mr Hollis's Rules, orders, Gifts & Bounties past & to come; together with ye Names & age, & Charecter of his Scholars, ye time of their Entry and Dismission; and also all ye Votes of ye Overseers & Corporation from time to time relating to ye said orders, Bounties & Scholars of the said Mr Hollis." Entries are primarily in Benjamin Wadsworth's hand and record donations from Thomas Hollis and his descendants, with transcriptions of related Corporation minutes. They also provide detailed information about the allocation of Hollis funds and scholarships, and the rules governing the Hollis Professorship of Divinity (established in 1721) and the Hollis Professorship of Mathematics and Natural Philosophy (established in 1727). The volume also contains inventories of books in the official library of the Hollis Professor of Divinity and two inventories – created in 1779 and in 1790 – of the mathematical and philosophical apparatus purchased with Hollis funds. Many entries related to the purchase of scientific instruments and supplies include the cost in sterling of each item. Also included are entries related to financial accounts and expenditures, as well as copies of letters from Nathaniel Hollis.

The EU began railway reform in earnest around the turn of the century. Two ‘railway packages’ have meanwhile been adopted amounting to a series of directives and a third package has been proposed. A range of complementary initiatives has been undertaken or is underway. This BEEP Briefing inspects the main economic aspects of EU rail reform. After highlighting the dramatic loss of market share of rail since the 1960s, the case for reform is argued to rest on three arguments: the need for greater competitiveness of rail, promoting the (market driven) diversion of road haulage to rail as a step towards sustainable mobility in Europe, and an end to the disproportional claims on public budgets of Member States. The core of the paper deals respectively with market failures in rail and in the internal market for rail services; the complex economic issues underlying vertical separation (unbundling) and pricing options; and the methods, potential and problems of introducing competition in rail freight and in passenger services. Market failures in the rail sector are several (natural monopoly, economies of density, safety and asymmetries of information), exacerbated by no less than 7 technical and legal barriers precluding the practical operation of an internal rail market. The EU choice to opt for vertical unbundling (with benefits similar in nature as in other network industries e.g. preventing opaque cross-subsidisation and greater cost revelation) risks the emergence of considerable coordination costs. The adoption of marginal cost pricing is problematic on economic grounds (drawbacks include arbitrary cost allocation rules in the presence of large economies of scope and relatively large common costs; a non-optimal incentive system, holding back the growth of freight services; possibly anti-competitive effects of two-part tariffs). Without further detailed harmonisation, it may also lead to many different systems in Member States, causing even greater distortions. Insofar as freight could develop into a competitive market, a combination of Ramsey pricing (given the incentive for service providers to keep market share) and price ceilings based on stand-alone costs might be superior in terms of competition, market growth and regulatory oversight. The incipient cooperative approach for path coordination and allocation is welcome but likely to be seriously insufficient. The arguments to introduce competition, notably in freight, are valuable and many e.g. optimal cross-border services, quality differentiation as well as general quality improvement, larger scale for cost recovery and a decrease of rent seeking. Nevertheless, it is not correct to argue for the introduction of competition in rail tout court. It depends on the size of the market and on removing a host of barriers; it requires careful PSO definition and costing; also, coordination failures ought to be pre-empted. On the other hand, reform and competition cannot and should not be assessed in a static perspective. Conduct and cost structures will change with reform. Infrastructure and investment in technology are known to generate enormous potential for cost savings, especially when coupled with the EU interoperability programme. All this dynamism may well help to induce entry and further enlarge the (net) welfare gains from EU railway reform. The paper ends with a few pointers for the way forward in EU rail reform.

Regulators and competition authorities often prevent firms with significant market power or dominant firms from practicing price discrimination. The goal of such an asymmetric no- discrimination constraint is to encourage entry and serve consumers’ interests. This constraint prohibits the firm with significant market power to practice both behaviour-based price discrimination within the competitive segment and third-degree price discrimination across the monopolistic and competitive segments. We find that this constraint hinders entry and reduces welfare when the monopolistic segment is small.

Confronted by the current refugee crisis, most Member States are turning inwards. But migration will continue to rise in the future. Given that migration is an unstoppable trend, the EU has everything to win from turning this crisis into an opportunity for its own citizens and economy, for the refugees and migrants it hosts and for their countries of origin. The manner in which the EU addresses this challenge will truly prove if it can live up to its founding principles of human dignity, solidarity, freedom, democracy and equality. This policy brief summarises European measures taken in the last few months and proposes four key actions to create a well-framed European migration policy: effectively implementing the principle of solidarity and fair-sharing of responsibility between Member States; creating more legal entry and integration channels; addressing the root causes of migration; and broadcasting a constructive and positive narrative on migration.

Introdução: A Educação Inclusiva surge como um modelo de escola que fomenta o ingresso e permanência de todos os alunos em um único sistema de ensino. Particularmente, no que compete à educação física inclusiva, esta é a Educação Física Adaptada, aplicada em condições especiais, visando uma população especial que necessita de estímulos particulares de desenvolvimento motor e funcional. Objetivo geral: Compreender as práticas de ensino de Educação Física Inclusiva em escolas públicas de Maceió. Como objetivos específicos pretende-se saber o que pensam os professores e gestores pedagógicos sobre a educação inclusiva e Educação Física Inclusiva; Conhecer as estratégias e estruturas escolares para a sua implementação; Identificar a preparação dos professores para desenvolver educação física inclusiva; Saber se reconhecem benefícios na sua implementação; Descrever o apoio fornecido pela escola e município. Métodos: Estudo qualitativo de abordagem fenomenológica, realizado com uma amostra constituída por quatro professores de educação física e quatro gestores pedagógicos com idades entre 33 e 50 anos, que integravam o quadro ativo das escolas públicas do município de Maceió, Brasil, no ano de 2014. O instrumento de colheita de dados utilizado foi a entrevista semiestruturada, recorrendo-se ao método de análise de conteúdo. Resultados: Em linhas gerais os professores concordam com a escola inclusiva e apontam benefícios psicossociais e a nível do convívio social, contudo referem não possuir preparação adequada para trabalhar com alunos que têm necessidades educacionais especiais (NEE), assumindo alguns investimentos na formação contínua e na adaptação nos conteúdos e nas metodologias de ensino. A falta de material apropriado para lecionar e a situação estrutural precária que as escolas oferecem aos alunos são outros aspectos negativos que enfrentam, além deles não participarem da elaboração dos documentos legais da escola. Os gestores apontaram como principais dificuldades, a falta de apoio da secretaria municipal de educação (SEMED) para com os profissionais que trabalham diretamente com esses alunos, a falta de capacitação na área da inclusão e consequentemente o ensino fornecido por eles. Os profissionais acreditam que essa disciplina tem um papel importante no processo de inclusão dos alunos com NEE, pois contribui para a socialização, reforça a autoestima e a qualidade de vida, além de ajudar no desenvolvimento cognitivo, afetivo e psicomotor. Conclusões: O processo de inclusão ainda está caminhando a passos lentos, e particularmente nas aulas de educação física. São poucos os alunos com NEE que frequentam as escolas regulares e, menos ainda, os que participam das aulas de educação física. Entendemos que são necessárias mudanças para que a inclusão nas aulas de educação física possa realmente favorecer os alunos com NEE. Enfatizamos contudo, que a educação física não é a base para a inclusão escolar, porém, um bom acolhimento e uma boa qualidade de ensino podem ter efeitos significantes na vida desses estudantes.
