The present invention discloses the use of the bacterial cellulose membrane in ligament, tendon and synovial capsule reconstructions according to the methods described in the technical description of the invention. Said material could be used in the reconstruction of ligaments and tendons (the knee cruciate ligaments, patellar ligament, Achilles tendon, quadriceps tendon, etc.) and the synovial capsule.

The purpose of this work was to validate isolation methods for sheep mesenchymal stem cells (MSC) from different sources and to explore the hypothesis that MSC exhibit markers of the same phenotype independent from tissue source. Cells derived from ovine bone marrow, synovial membrane and adipose tissue were characterized using the following markers: CD44, CD45, CD11b and MHC-I. The isolated MSC were cultivated, went through osteogenic, chondrogenic and adipogenic differentiation, and were characterized by flow cytometry using mouse anti-ovine CD44, CD45 and MHC-I monoclonal antibody (mAb), and mouse anti-bovine CD11b mAb. Ovine MSC from all three sources differentiated under chondorgenic, osteogenic and adipogenic conditions. Also, MSC from the three tissues were found to express CD44 and MHC-I but lack of CD11b and CD45. The results obtained revealed that our isolation methods for the different tissues tested are valid and that MSC from the three sources studied have same immunophenotic characteristics. Background: Soft tissue sarcomas (STSs) are a group of neoplasms, which, despite current therapeutic advances, still confer a poor outcome to half of the patients. As other solid tumors, STSs exhibit high glucose consumption rates, associated with worse prognosis and therapeutic response. As highly glycolytic tumors, we hypothesized that sarcomas should present an increased expression of lactate transporters (MCTs).Methods: Immunohistochemical expression of MCT1, MCT2, MCT4 and CD147 was assessed in a series of 86 STSs and the expression profiles were associated with patients' clinical-pathological parameters.Results: MCT1, MCT4 and CD147 were mainly observed in the plasma membrane of cancer cells (around 60% for MCTs and 40% for CD147), while MCT2 was conspicuously found in the cytoplasm (94.2%). Importantly, we observed MCT1 nuclear expression (32.6%). MCT1 and MCT4, alone or co-expressed with CD147 in the plasma membrane, were associated with poor prognostic variables including high tumor grade, disease progression and shorter overall survival. Conversely, we found MCT1 nuclear expression to be associated with low grade tumors and longer overall survival.Conclusions: The present work represents the first report of MCTs characterization in STSs. We showed the original finding of MCT1 expression in the nucleus. Importantly, opposite biological roles should be behind the dual sub-cellular localization of MCT1, as plasma membrane expression of MCT1 is associated with worse patients' prognosis, while nuclear expression is associated with better prognosis.

Aim: Injury of tendons contained within a synovial environment, such as joint, bursa or tendon sheath, frequently fails to heal and releases matrix proteins into the synovial fluid, driving inflammation. This study investigated the effectiveness of cells to seal tendon surfaces and provoke matrix synthesis as a possible effective injectable therapy. Materials & methods: Equine flexor tendon explants were cultured overnight in suspensions of bone marrow and synovium-derived mesenchymal stems cells and, as controls, two sources of fibroblasts, derived from tendon and skin, which adhered to the explants. Release of the most abundant tendon extracellular matrix proteins into the media was assayed, along with specific matrix proteins synthesis by real-time PCR. Results: Release of extracellular matrix proteins was influenced by the coating cell type. Fibroblasts from skin and tendon appeared less capable of preventing the release of matrix proteins than mesenchymal stems cells. Conclusion: The source of cell is an important consideration for cell therapy.

Objective: To assess 3D morphological variations and local and systemic biomarker profiles in subjects with a diagnosis of temporomandibular joint osteoarthritis (TMJ OA).Design: Twenty-eight patients with long-term TMJ OA (39.9 +/- 16 years), 12 patients at initial diagnosis of OA (47.4 +/- 16.1 years), and 12 healthy controls (41.8 +/- 12.2 years) were recruited. All patients were female and had cone beam CT scans taken. TMJ arthrocentesis and venipuncture were performed on 12 OA and 12 age-matched healthy controls. Serum and synovial fluid levels of 50 biomarkers of arthritic inflammation were quantified by protein microarrays. Shape Analysis MANCOVA tested statistical correlations between biomarker levels and variations in condylar morphology.Results: Compared with healthy controls, the OA average condyle was significantly smaller in all dimensions except its anterior surface, with areas indicative of bone resorption along the articular surface, particularly in the lateral pole. Synovial fluid levels of ANG, GDF15, TIMP-1, CXCL16, MMP-3 and MMP-7 were significantly correlated with bone apposition of the condylar anterior surface. Serum levels of ENA-78, MMP-3, PAI-1, VE-Cadherin, VEGF, GM-CSF, TGF beta b1, IFN gamma g, TNF alpha a, IL-1 alpha a, and IL-6 were significantly correlated with flattening of the lateral pole. Expression levels of ANG were significantly correlated with the articular morphology in healthy controls.Conclusions: Bone resorption at the articular surface, particularly at the lateral pole was statistically significant at initial diagnosis of TMJ OA. Synovial fluid levels of ANG, GDF15, TIMP-1, CXCL16, MMP-3 and MMP-7 were correlated with bone apposition. Serum levels of ENA-78, MMP-3, PAI-1, VE-Cadherin, VEGF, GM-CSF, TGF beta 1, IFN gamma, TNF alpha, IL-1 alpha, and IL-6 were correlated with bone resorption. The distal sesamoid bone, also known as navicular bone, is located inside the hoof, palmar (thoracic limbs) or plantar (pelvic limbs) to distal interphalangeal joint. Its extremities are fixed by collateral medial and lateral sesamoidean ligament and distal impar sesamoidean ligament. Navicular disease diagnostic is made through a thorough clinical exam, nerve blocks and imaging exams. Even though imaging exams are not conclusive, they are essential to evaluate the extension of soft tissue lesions. Radiographic projections used to evaluate navicular bone are lateromedial (LM); dorsoproximal-palmaro/plantarodistal oblíqua (D30Pr-PaDiO); dorsoproximal-palmaro/plantarodistal oblíqua (D60Pr-PaDiO) e a palmaro/plantaroproximal-palmaro/plantarodistal oblíqua (PaPr-PaDiO). This exam allows to identify number and shape alterations of synovial invaginations on the distal (foramem nutricio) in the distal margin of distal sesamoid, osteophytes, enthesophytes and periarticular lesions. There are four ultrasonographic accesses described in literature to evaluate podotroclear apparatus, they are: palmar or plantar distal do the pastern, through the heel bulbs, through coronary band and transcuneal. These images allow a beeter the evaluation of soft tissue next to the distal sesamoid, because it is more sensitive than radiographic exam to evaluate acute lesions in soft tissues and perioesteum

A busca pela identificação de fatores que possam apontar o diagnóstico, a resposta terapêutica e a sobrevida dos pacientes portadores de sarcoma ósseos tem sido incessante. Presumir a agressividade tumoral, capacidade de invasão tecidual, probabilidade de recidiva, propensão ao desenvolvimento de metástases e resposta ao tratamento, poderá ser valioso expediente na escolha da proposta terapêutica. Embora fatores ambientais e dietéticos contribuam para a etiologia do câncer, as neoplasias se originam de um processo de múltiplos passos envolvendo alterações de genes e seleção clonal da progênie variante. Estas mutações ocorrem em classes de genes reguladores da proliferação celular como os oncogenes, genes supressores de tumor, fatores de crescimento, vias de sinalização e genes de reparo de DNA. Os conhecimentos sobre a biologia tumoral melhoraram o entendimento sobre os múltiplos aspectos da carcinogênese. No entanto, embora as perspectivas permaneçam, até agora, há poucos benefícios para prevenção, diagnóstico, tratamento e seguimento dos pacientes com sarcomas ósseos. Este trabalho teve por objetivo detectar e descrever alterações cromossômicas consistentes e recorrentes, através da utilização de análises com a citogenética clássica. Também descreveu o envolvimento dessas alterações com o prognóstico em sarcomas ósseos primários e secundários, na tentativa de contribuir para a realização de estratégias mais eficazes para melhorar as taxas de sobrevida e beneficiar maior número de pacientes portadores de sarcomas ósseos. O Herpesvírus associado ao sarcoma de Kaposi (KSHV), ou Herpesvírus Humano tipo 8 (HHV-8), é o agente etiológico do sarcoma de Kaposi (SK), uma neoplasia maligna vascular. O ciclo biológico do KSHV apresenta duas fases, denominadas ciclo latente e ciclo lítico (ou produtivo). O ciclo latente é marcado pela expressão de um número reduzido de genes virais, com destaque para LANA e vFLIP. No ciclo lítico ocorre a replicação do genoma viral e a produção de novas partículas virais infecciosas; dentre seus principais produtos destacam-se as proteínas Rta, vGPCR e K1. LANA, vFLIP, vGPCR e K1 apresentam propriedades oncogênicas relatadas na literatura, enquanto Rta têm papel importante na regulação da transição entre os ciclos lítico e latente do KSHV. O KSHV é requerido para o desenvolvimento do SK. No entanto, a infecção pelo vírus não é suficiente para o desenvolvimento da doença. Por outro lado, sabe-se que o HIV é um co-fator importante, que favorece o desenvolvimento dessa neoplasia. Sugere-se que a proteína tat do HIV-1 amplifica a infectividade do KSHV, hiper-regulando a expressão de diferentes genes herpesvirais e colaborando para o crescimento e sobrevivência de células endoteliais que compõem as lesões do SK. A fim de contribuir para um melhor entendimento dos efeitos da proteína tat do HIV-1 em células infectadas pelo KSHV, o presente trabalho descreveu eventuais alterações na expressão dos genes codificadores de vFLIP, LANA, vGPCR, Rta e K1 em células endoteliais de veia umbilical humana imortalizada pela telomerase e infectada pelo KSHV a longo prazo (TIVE-LTCs) expostas à proteína tat do HIV-1 produzida por células linfóides T (CLTs) em co-cultivo. Células Jurkat contendo ou não vetor da proteína tat do HIV-1 foram utilizadas como CLTs e co-cultivadas com TIVE-LTCs por 48, 72 e 96 horas. One of the main causes of incapacity in athletes, be they human or equines, is the occurrence of intra-articular lesions. The equines are each time more required in his athletic performance, resulting in intense stress to the structures that composes the locomotor device. The leading cause of human and equine athlete’s functional incapacity is the intra-articular disorders. One of the greatest advances in sports medicine was the development of arthroscopy as a minimal invasive intra-articular surgery. The defining characteristic of diagnostic or surgical arthroscopy is featured by minimal tissue damage and broad inspection of internal structures inside the joint associated with low morbidity and complications. The advantages of surgical arthroscopy over traditional surgery are well known: limited hospitalization, early return to competition, lower risks of post-operative joint rigidity, magnification of inspected structures, joint lavage associated or not with removal of potentially dangerous substances. Arthroscopy cannot replace conventional methods and must not do so; however, the intrinsic limitations of conventional diagnostic techniques, such as radiology and synovial fluid analysis, must be kept in mind, particularly in evaluating damage to cartilage and the synovial membrane. This study gathered some of the most relevant researches already undertaken regarding the Temporomandibular Joint, presenting descriptions of its anatomy and dynamics, and relating them to the main aspects of joint dysfunctions. Literature recognizes that the Temporomandibular Joint, responsible for lower jaw movements, comprehends the skull base and the jaw bone. Its anatomy and dynamics are of great importance in order to understand the functions of Stomatognathic System as well as to study Temporomandibular Joint Dysfunction. The elements that compose this synovial joint and the bilateral characteristic confer eminent importance to the ATM before the skeleton head, the ability to perform broad movements and high complexity in its operation. It is accepted that the overload of one anatomical structure from the TMJ or other components of the Stomatognathic System leads to Temporomandibular Joint Dysfunctions. These disorders have different etiologies, ranging from malocclusion to emotional stress, and various forms of treatment, which are related in this study
