Nutrient sensitive insulin-like peptides (ILPs) have profound effects on invertebrate metabolism, nutrient storage, fertility and aging. Many insects transcribe ILPs in specialized neurosecretory cells at changing levels correlated with life history. However, the major site of insect metabolism and nutrient storage is not the brain, but rather the fat body, where functions of ILP expression are rarely studied and poorly understood. Fat body is analogous to mammalian liver and adipose tissue, with nutrient stores that often correlate with behavior. We used the honey bee (Apis mellifera), an insect with complex behavior, to test whether ILP genes in fat body respond to experimentally induced changes of behavioral physiology. Honey bee fat body influences endocrine state and behavior by secreting the yolk protein precursor vitellogenin (Vg), which suppresses lipophilic juvenile hormone and social foraging behavior. In a two-factorial experiment, we used RNA interference (RNAi)-mediated vg gene knockdown and amino acid nutrient enrichment of hemolymph (blood) to perturb this regulatory module. We document factor-specific changes in fat body ilp1 and ilp2 mRNA, the bee`s ILP-encoding genes, and confirm that our protocol affects social behavior. We show that ilp1 and ilp2 are regulated independently and differently and diverge in their specific expression-localization between fat body oenocyte and trophocyte cells. Insect ilp functions may be better understood by broadening research to account for expression in fat body and not only brain.

We used high-resolution SNP genotyping to identify regions of genomic gain and loss in the genomes of 212 medulloblastomas, malignant pediatric brain tumors. We found focal amplifications of 15 known oncogenes and focal deletions of 20 known tumor suppressor genes (TSG), most not previously implicated in medulloblastoma. Notably, we identified previously unknown amplifications and homozygous deletions, including recurrent, mutually exclusive, highly focal genetic events in genes targeting histone lysine methylation, particularly that of histone 3, lysine 9 (H3K9). Post-translational modification of histone proteins is critical for regulation of gene expression, can participate in determination of stem cell fates and has been implicated in carcinogenesis. Consistent with our genetic data, restoration of expression of genes controlling H3K9 methylation greatly diminishes proliferation of medulloblastoma in vitro. Copy number aberrations of genes with critical roles in writing, reading, removing and blocking the state of histone lysine methylation, particularly at H3K9, suggest that defective control of the histone code contributes to the pathogenesis of medulloblastoma.

Background and Objectives: This study evaluated the hybrid layer (HL) morphology created by three adhesive systems (AS) on dentin surfaces treated with Er:YAG laser using two irradiation parameters. Study Design: Occlusal flat dentin surfaces of 36 human third molars were assigned into nine groups (n = 4) according to the following ASs: one bottle etch&rinse Single Bond Plus (3M ESPE), two-step Clearfil Protect Bond (Kuraray), and all-in-one S3 Bond (Kuraray) self-etching, which were labeled with rhodamine B or fluorescein isothiocyanate dextran and were applied to dentin surfaces that were irradiated with Er:YAG laser at either 120 (38.7 J/cm(2)) or 200 mJ/pulse (64.5 J/cm(2)), or were applied to untreated dentin surfaces (control group). The ASs were light-activated following MI and the bonded surfaces were restored with resin composite Z250 (3M ESPE). After 24 hours of storage in vegetable oil, the restored teeth were vertically, serially sectioned into 1-mm thick slabs, which had the adhesive interfaces analyzed with confocal laser microscope (CLSM-LSM 510 Meta). CLSM images were recorded in the fluorescent mode from three different regions along each bonded interface. Results: Non-uniform HL was created on laser-irradiated dentin surfaces regardless of laser irradiation protocol for all AS, while regular and uniform HL was observed in the control groups. ""Stretch mark""-like red lines were found within the HL as a result of resin infiltration into dentin microfissures, which were predominantly observed in 200 mJ/pulse groups regardless of AS. Poor resin infiltration into peritubular dentin was observed in most regions of adhesive interfaces created by all ASs on laser-irradiated dentin, resulting in thin resin tags with neither funnel-shaped morphology nor lateral resin projections. Conclusion: Laser irradiation of dentin surfaces at 120 or 200 mJ/pulse resulted in morphological changes in HL and resin tags for all ASs evaluated in the study. Lasers Surg. Med. 42:662-670, 2010. Objective. This study evaluated the degree of conversion (DC), maximum rate of cure (R(p)(max)), and polymerization stress (PS) developed by an experimental dental composite subjected to different irradiant energies (3,6,12, 24, or 48J/cm(2)) under constant irradiance (500 mw/cm(2)). Methods. DC and R(p)(max) were monitored for 10 min on the bottom surface of 2-mm thick disks and on 150-mu m thick films (representing the top of the specimen) using ATR-FTIR. PS was monitored for 10 min in 2-mm thick disks bonded to two glass rods (O = 5 mm) attached to a universal testing machine. One-way ANOVA/Tukey tests were used and differences in DC and R(p)(max) between top and bottom surfaces were examined using Student`s t-test. Statistical testing was performed at a pre-set alpha of 0.05. Results. For a given surface, DC showed differences among all groups, except at the top between 24 and 48 J/cm(2). R(p)(max) was similar among all groups at the same surface and statistically higher at the top surface. PS also showed significant differences among all groups. Data for 48 J/cm(2) were not obtained due to specimen failure at the glass/composite interface. Significance. Objective: Verify the influence of radiant exposure (H) on composite degree of conversion (DC) and mechanical properties. Methods: Composite was photoactivated with 3, 6, 12, 24, or 48 J/cm(2). Properties were measured after 48-h dry storage at room temperature. DC was determined on the flat surfaces of 6 mm x 2 mm disk-shaped specimens using FTIR. Flexural strength (FS) and modulus (FM) were accessed by three-point bending. Knoop microhardness number (KHN) was measured on fragments of FS specimens. Data were analyzed by one-way ANOVA/Tukey test, Student`s t-test, and regression analysis. Results: DC/top between 6 and 12 J/cm(2) and between 24 and 48 J/cm(2) were not statistically different. No differences between DC/top and bottom were detected. DC/bottom, FM, and KHN/top showed significant differences among all H levels. FS did not vary between 12 and 24 J/cm(2) and between 24 and 48 J/cm(2). KHN/bottom at 3 and 6 J/cm(2) was similar. KHN between top and bottom was different up to 12 J/cm(2). Regression analyses having H as independent variable showed a plateau region above 24 J/cm(2). KHN increased exponentially (top) or linearly (bottom) with DC. FS and FM increased almost linearly with DC/bottom up to 55% conversion. Conclusions: DC and mechanical properties increased with radiant exposure. Variables leveled off at high H levels. This study has a vast analysis, studying almost all the pre-electoral polls published or issued in Portugal in the month previous to each of the elections, since 1991 until the last one that took place in February 2005. The accuracy measures I used were adapted from the study carried out by Frederick Mosteller in the report to the Committee on Analysis of Pre-election Polls, regarding the USA elections of 1948.

A figura do iluminador (lighting designer) surge nesta dissertação como ponto de partida para um esboço de estudo acerca do autor do desenho de luz no espectáculo teatral em Portugal. Baseámo-nos num dos mais conceituados iluminadores (lighting designer) portugueses, Orlando Worm, para pesquisarmos o que o próprio trouxe aos novos designers de luz que continuam a percorrer um longo e, por vezes, solitário caminho nesta arte criativa. Considerámos para tal tecnologias criadas entre as décadas de 30 a 70 por alguns dos maiores nomes ligados à matéria: Stanley McCandless, Frederick Bentham, Richard Pilbrow e Francis Reid. Estes autores servem-nos como ponto de partida para depois indagarmos se a partir da década de 80 se utiliza algum destes métodos. Ao reflectir acerca destas tecnologias e da própria experiência de Orlando Worm, pretende-se concluir que o trabalho de desenho de luz é, de facto, um trabalho criativo e artístico, na medida em que a figura do iluminador (lighting designer) tem, ou deve ter, um contributo activo no processo teatral.

A comunicação apresentada reporta-se a um projeto em curso, que se propõe desenvolver uma análise alargada sobre os conteúdos e os propósitos dos jornais internos no nosso país nos dias de hoje. A finalidade desta comunicação que integra o colóquio de Relações Públicas subordinado ao tema “O Jornal de Empresa ao Serviço da Comunicação”, passa por identificar pela positiva certas caraterísticas que contribuem para o sucesso dos jornais internos (o que fazer, os do’s) e, por contraponto, o que evitar se se pretende alcançar a excelência nas publicações empresariais (o que não fazer, ou seja, os dont’s). O ponto de partida foi a reflexão sobre um conjunto de indicadores que constituem o fundamento da avaliação do júri que preside ao concurso do Grande Prémio da Associação Portuguesa de Comunicação da Empresa (APCE), nomeadamente no que se refere à categoria de “Publicação Interna”. Esta iniciativa foi criada em 1995 para distinguir a excelência na estratégia da comunicação organizacional, estimulando, reconhecendo e divulgando as iniciativas dos profissionais desta área. A ideia e a motivação para este projeto surgem a propósito da realização da exposição “Imprensa Empresarial em Portugal – 145 anos de Jornais de Empresa”, organizada pelo Dr. João Moreira dos Santos, para assinalar os 100 sobre a publicação, em 1915 nos EUA, da primeira obra dedicada aos jornais de empresa, pela mão de George Frederick Wilson, e ao mesmo tempo registar os 20 anos passados sobre a publicação da sua obra “Imprensa Empresarial – Da Informação à Comunicação”. No âmbito desta exposição comemorativa, realizada na Biblioteca Nacional, é dirigido um convite ao Grupo de Trabalho em Comunicação Organizacional e Institucional da SOPCOM (GT COI) e à Associação Portuguesa de Comunicação de Empresa (APCE) para a realização de um colóquio, que em simultâneo aproximasse o passado e o futuro da imprensa empresarial em Portugal e reunisse em debate académicos e responsáveis por publicações internas no mundo empresarial, com o objetivo de promover uma reflecção sobre o percurso, as caraterísticas, as limitações e as potencialidades, a função/utilidade do Jornal de Empresa.

Sem medir o tempo não teria feito sentido falar-se, como tanto se falou na indústiia desde finais do século passado, pelo menos', em organização racional, ou científica, do trabalho. Foi empunhando o cronômetio que Frederick Winslow Taylor (1856-1915) observou as práticas de trabalho, experimentou e estabeleceu seqüências "racionais". Contando tempos, demonstrou a eficácia quantitativa da segmentação das tarefas e depuração dos gestos técnicos até ao estritamente necessário, com exclusão de todo o "desperdício" gestual susceptível de gerar lentidão ou atraso (Left-anc, 1975: 330-31; SainsauUeu, 1985: 374-76; Freire, 1993: 63-67, por exemplo).
