Oggetto di questa tesi è lo studio parametrico del comportamento di paratie mediante modellazione numerica agli elementi finiti. Si è anche analizzato l’aspetto normativo relativo ad esse ed in particolare si è applicato il D.M. 2008 ad una paratia sottolineando le innovazioni nella progettazione geotecnica. Le paratie sono opere di sostegno flessibili interagenti con i fronti di scavo e costituite da una porzione fuori terra, soggetta a condizioni di spinta attiva, e una porzione interrata che contrappone la resistenza passiva del terreno. Le opere di sostegno flessibile si dividono in due categorie: diaframmi e palancole che differiscono molto fra loro sia per il materiale con cui sono realizzate, sia per la tecnica di messa in opera, sia per la geometria, ma hanno in comune il meccanismo di funzionamento. Sono stati illustrati i metodi di calcolo per lo studio delle paratie: metodi dell’equilibrio limite a rottura, metodi a molle e metodi agli elementi finiti. I metodi agli elementi finiti negli ultimi anni hanno avuto maggior successo grazie alla possibilità di definire il modello costitutivo sforzi-deformazioni, le condizioni al contorno e le condizioni iniziali. Esistono numerosi programmi di calcolo che si basano sul metodo agli elementi finiti, tra i quali è da annoverare il programma Plaxis che viene molto usato grazie alla presenza di vari modelli costitutivi in grado di simulare il comportamento del terreno. Tra i legami costitutivi presenti nel programma Plaxis, sono stati presi in esame il modello Mohr-Coulomb, l’Hardening model e il Soft Soil Creep model e sono poi stati applicati per eseguire uno studio parametrico del comportamento delle paratie. Si è voluto comprendere la sensibilità dei modelli al variare dei parametri inseriti nel programma. In particolare si è posta attenzione alla modellazione dei terreni in base ai diversi modelli costitutivi studiando il loro effetto sui risultati ottenuti. Si è inoltre eseguito un confronto tra il programma Plaxis e il programma Paratie. Quest’ultimo è un programma di calcolo che prevede la modellazione del terreno con molle elasto-plastiche incrudenti. Il lavoro di tesi viene illustrato in questo volume come segue: nel capitolo 1 si analizzano le principali caratteristiche e tipologie di paratie mettendo in evidenza la complessità dell’analisi di queste opere e i principali metodi di calcolo usati nella progettazione, nel capitolo 2 si è illustrato lo studio delle paratie secondo le prescrizioni del D.M. 2008. Per comprendere l’argomento è stato necessario illustrare i principi base di questo decreto e le caratteristiche generali del capitolo 6 dedicato alla progettazione geotecnica. Le paratie sono poi state studiate in condizioni sismiche secondo quanto previsto dal D.M. 2008 partendo dall’analisi degli aspetti innovativi nella progettazione sismica per poi illustrare i metodi pseudo-statici utilizzati nello studio delle paratie soggette ad azione sismica. Nel capitolo 3 si sono presi in esame i due programmi di calcolo maggiormente utilizzati per la modellazione delle paratie: Paratie e Plaxis. Dopo aver illustrato i due programmi per comprenderne le potenzialità, i limiti e le differenze, si sono analizzati i principali modelli costituitivi implementati nel programma Plaxis, infine si sono mostrati alcuni studi di modellazione presenti in letteratura. Si è posta molta attenzione ai modelli costitutivi, in particolare a quelli di Mohr-Coulomb, Hardening Soil model e Soft Soil Creep model capaci di rappresentare il comportamento dei diversi tipi di terreno. Nel capitolo 4, con il programma Plaxis si è eseguita un’analisi parametrica di due modelli di paratie al fine di comprendere come i parametri del terreno, della paratia e dei modelli costitutivi condizionino i risultati. Anche in questo caso si è posta molta attenzione alla modellazione del terreno, aspetto che in questa tesi ha assunto notevole importanza. Dato che nella progettazione delle paratie è spesso utilizzato il programma Paratie (Ceas) si è provveduto ad eseguire un confronto tra i risultati ottenuti con questo programma con quelli ricavati con il programma Plaxis. E’ stata inoltra eseguita la verifica della paratia secondo le prescrizioni del D.M. 2008 dal momento che tale decreto è cogente.

The wheel - rail contact analysis plays a fundamental role in the multibody modeling of railway vehicles. A good contact model must provide an accurate description of the global contact phenomena (contact forces and torques, number and position of the contact points) and of the local contact phenomena (position and shape of the contact patch, stresses and displacements). The model has also to assure high numerical efficiency (in order to be implemented directly online within multibody models) and a good compatibility with commercial multibody software (Simpack Rail, Adams Rail). The wheel - rail contact problem has been discussed by several authors and many models can be found in the literature. The contact models can be subdivided into two different categories: the global models and the local (or differential) models. Currently, as regards the global models, the main approaches to the problem are the so - called rigid contact formulation and the semi – elastic contact description. The rigid approach considers the wheel and the rail as rigid bodies. The contact is imposed by means of constraint equations and the contact points are detected during the dynamic simulation by solving the nonlinear algebraic differential equations associated to the constrained multibody system. Indentation between the bodies is not permitted and the normal contact forces are calculated through the Lagrange multipliers. Finally the Hertz’s and the Kalker’s theories allow to evaluate the shape of the contact patch and the tangential forces respectively. Also the semi - elastic approach considers the wheel and the rail as rigid bodies. However in this case no kinematic constraints are imposed and the indentation between the bodies is permitted. The contact points are detected by means of approximated procedures (based on look - up tables and simplifying hypotheses on the problem geometry). The normal contact forces are calculated as a function of the indentation while, as in the rigid approach, the Hertz’s and the Kalker’s theories allow to evaluate the shape of the contact patch and the tangential forces. Both the described multibody approaches are computationally very efficient but their generality and accuracy turn out to be often insufficient because the physical hypotheses behind these theories are too restrictive and, in many circumstances, unverified. In order to obtain a complete description of the contact phenomena, local (or differential) contact models are needed. In other words wheel and rail have to be considered elastic bodies governed by the Navier’s equations and the contact has to be described by suitable analytical contact conditions. The contact between elastic bodies has been widely studied in literature both in the general case and in the rolling case. Many procedures based on variational inequalities, FEM techniques and convex optimization have been developed. This kind of approach assures high generality and accuracy but still needs very large computational costs and memory consumption. Due to the high computational load and memory consumption, referring to the current state of the art, the integration between multibody and differential modeling is almost absent in literature especially in the railway field. However this integration is very important because only the differential modeling allows an accurate analysis of the contact problem (in terms of contact forces and torques, position and shape of the contact patch, stresses and displacements) while the multibody modeling is the standard in the study of the railway dynamics. In this thesis some innovative wheel – rail contact models developed during the Ph. D. activity will be described. Concerning the global models, two new models belonging to the semi – elastic approach will be presented; the models satisfy the following specifics: 1) the models have to be 3D and to consider all the six relative degrees of freedom between wheel and rail 2) the models have to consider generic railway tracks and generic wheel and rail profiles 3) the models have to assure a general and accurate handling of the multiple contact without simplifying hypotheses on the problem geometry; in particular the models have to evaluate the number and the position of the contact points and, for each point, the contact forces and torques 4) the models have to be implementable directly online within the multibody models without look - up tables 5) the models have to assure computation times comparable with those of commercial multibody software (Simpack Rail, Adams Rail) and compatible with RT and HIL applications 6) the models have to be compatible with commercial multibody software (Simpack Rail, Adams Rail). The most innovative aspect of the new global contact models regards the detection of the contact points. In particular both the models aim to reduce the algebraic problem dimension by means of suitable analytical techniques. This kind of reduction allows to obtain an high numerical efficiency that makes possible the online implementation of the new procedure and the achievement of performance comparable with those of commercial multibody software. At the same time the analytical approach assures high accuracy and generality. Concerning the local (or differential) contact models, one new model satisfying the following specifics will be presented: 1) the model has to be 3D and to consider all the six relative degrees of freedom between wheel and rail 2) the model has to consider generic railway tracks and generic wheel and rail profiles 3) the model has to assure a general and accurate handling of the multiple contact without simplifying hypotheses on the problem geometry; in particular the model has to able to calculate both the global contact variables (contact forces and torques) and the local contact variables (position and shape of the contact patch, stresses and displacements) 4) the model has to be implementable directly online within the multibody models 5) the model has to assure high numerical efficiency and a reduced memory consumption in order to achieve a good integration between multibody and differential modeling (the base for the local contact models) 6) the model has to be compatible with commercial multibody software (Simpack Rail, Adams Rail). In this case the most innovative aspects of the new local contact model regard the contact modeling (by means of suitable analytical conditions) and the implementation of the numerical algorithms needed to solve the discrete problem arising from the discretization of the original continuum problem. Moreover, during the development of the local model, the achievement of a good compromise between accuracy and efficiency turned out to be very important to obtain a good integration between multibody and differential modeling. At this point the contact models has been inserted within a 3D multibody model of a railway vehicle to obtain a complete model of the wagon. The railway vehicle chosen as benchmark is the Manchester Wagon the physical and geometrical characteristics of which are easily available in the literature. The model of the whole railway vehicle (multibody model and contact model) has been implemented in the Matlab/Simulink environment. The multibody model has been implemented in SimMechanics, a Matlab toolbox specifically designed for multibody dynamics, while, as regards the contact models, the CS – functions have been used; this particular Matlab architecture allows to efficiently connect the Matlab/Simulink and the C/C++ environment. The 3D multibody model of the same vehicle (this time equipped with a standard contact model based on the semi - elastic approach) has been then implemented also in Simpack Rail, a commercial multibody software for railway vehicles widely tested and validated. Finally numerical simulations of the vehicle dynamics have been carried out on many different railway tracks with the aim of evaluating the performances of the whole model. The comparison between the results obtained by the Matlab/ Simulink model and those obtained by the Simpack Rail model has allowed an accurate and reliable validation of the new contact models. In conclusion to this brief introduction to my Ph. D. thesis, we would like to thank Trenitalia and the Regione Toscana for the support provided during all the Ph. D. activity. Moreover we would also like to thank the INTEC GmbH, the society the develops the software Simpack Rail, with which we are currently working together to develop innovative toolboxes specifically designed for the wheel rail contact analysis.

In Gesteinen, die bei schwach- bis mittelgradiger Metamorphose deformiert wurden, sind Mikrostrukturen, wie z.B. undulöse Auslöschung, Subkörner und kristallographische Regelungen, typisch. Sie werden i.a. als das Resultat eines kristallplastischen Deformationsprozesses interpretiert. Das häufige Auftreten dieser Strukturen führte zu der Annahme, daß kristallplastische Deformationsprozesse zu den dominierenden duktilen Deformationsmechanismen während der Erdkrustendeformation gehören.Die vorliegende Arbeit beschäftigt sich mit Experimenten, bei denen die zeitabhängige kata-klastische Deformation polykristalliner, kompaktierter Aggregatproben des sehr gut löslichen, elastisch/spröden Salzes Natriumchlorat (NaClO3) untersucht wurde. Dieses Salz wurde als Analogmaterial gesteinsbildender Minerale wie Quarz und Feldspat ausgewählt. Es wurde nun untersucht, ob und in welcher Weise spröde Deformationsmechanismen in Anwesenheit von Wasser scheinbar kristall-plastische Mikrostrukturen hervorrufen können.Aus den Versuchen ergibt sich nun die Folgerung, daß scheinbar kristallplastisch erzeugte Mikrostrukturen wie Subkörner auch durch spröde zeitunabhängige Risse und subkritische (langsame) Kataklase entstehen können. Diese Erkenntnisse sind wichtig für das rheologische Verhalten der Erdkruste und somit auch für Modellierungen der Erdkrustendeformation. Ihre Betrachtung und Übertragung auf die Deformationsstrukturen der natürlich deformierten kontinentalen, oberkrustalen Gesteine der Erde würde die Interpretation der Mikrostrukturen in diesen Bereichen stark ändern. Dies hätte eine wesentliche Änderung auf den Verlauf der Gesteinsfestigkeitskurven in den Spannungsprofilen der Erdoberkruste zur Folge.

In this thesis is studied the long-term behaviour of steel reinforced slabs paying particular attention to the effects due to shrinkage and creep. Despite the universal popularity of using this kind of slabs for simply construction floors, the major world codes focus their attention in a design based on the ultimate limit state, restraining the exercise limit state to a simply verification after the design. For Australia, on the contrary, this is not true. In fact, since this country is not subjected to seismic effects, the main concern is related to the long-term behaviour of the structure. Even if there are a lot of studies about long-term effects of shrinkage and creep, up to date, there are not so many studies concerning the behaviour of slabs with a cracked cross section and how shrinkage and creep influence it. For this reason, a series of ten full scale reinforced slabs was prepared and monitored under laboratory conditions to investigate this behaviour. A wide range of situations is studied in order to cover as many cases as possible, as for example the use of a fog room able to reproduce an environment of 100% humidity. The results show how there is a huge difference in terms of deflections between the case of slabs which are subjected to both shrinkage and creep effects soon after the partial cracking of the cross section, and the case of slabs which have already experienced shrinkage effects for several weeks, when the section has not still cracked, and creep effects only after the cracking.

Experimental study on the long-term deformations of the fibre reinforced concrete. Steel and macro-synthetic fibers were used to evaluate the shrinkage, creep, mid-span deflection, cracking and rupture analysis of three different types of samples. At the end the main topics of ACI guidelines were analyzed in order to perform an overview of design.

Il basso tasso d'usura e l'alta resistenza meccanica dell'UHMWPE reticolato e additivato ha posto l'attenzione verso l'uso di teste femorali con diametri maggiori per diminuire il rischio d'impingement e dislocazioni. Questo richiede l'utilizzo d'inserti acetabolari più sottili di quelli attualmente in commercio. In quest'ottica è necessario porre particolare attenzione alla resistenza meccanica d'inserti più sottili, e all'efficacia della vitamina E nel combattere l'effetto dell'ossidazione che si manifesta in seguito al processo di reticolazione. Lo scopo del lavoro è quindi di studiare un inserto più sottile di quelli attualmente in commercio per verificarne le performance. Tale studio è svolto su una serie di taglie (compreso inserto prodotto ad-hoc con spessore di 3,6 mm) con spessore di 5,6 mm e di 3,6 mm dalle quali viene isolato il worst-case tramite analisi FEM. Con prove sperimentali è testata la resistenza meccanica del worst-case, e sono monitorate le deformazioni subite e l'ossidazione del campione. Dagli studi FEM è risultato che le tensioni sono mediamente le stesse in tutti i campioni, anche se si sono registrate tensioni leggermente superiori nella taglia intermedia. A differenza delle attese la taglia in cui si sono riscontrate le tensioni massime è la F (non è l'inserto che ha diametro inferiore). A seguito della messa a punto del modello FEM si è identificato un valore d'attrito inferiore a quello atteso. In letteratura i valori d'attrito coppa-inserto sono più grandi del valore che si è identificato tramite simulazioni FEM. Sulla base dei risultati FEM è isolato il worst-case che viene quindi sottoposto a un test dinamico con 6 milioni di cicli atto a valutarne le performance. Gli inserti di spessore ridotto non hanno riportato alcun danno visibile, e la loro integrità strutturale non è stata modificata. Le considerazioni preliminari sono confermate dalla verifica al tastatore meccanico e dall'analisi chimica, dalle quale non si sono evidenziate particolari problematiche. Infatti, da queste verifiche si rileva che l'effetto del creep nella prova accelerata è pressoché trascurabile, e non si riscontrano variazioni dimensionali rilevanti. Anche dall'analisi chimica dei campioni non si evidenzia ossidazione. I valori d'ossidazione dell'inserto testato sono analoghi a quelli del campione non testato, anche quando viene confrontato con l'inserto in UHMWPE vergine si evidenzia un'ossidazione di molto superiore. Questo prova che la vitamina E inibisce i radicali liberi che quindi non causano l'ossidazione con susseguente fallimento dell'inserto. Dai risultati si vede che i campioni non subiscono danni rilevanti, le deformazioni elastiche monitorate nel test dinamico sono pressoché nulle, come gli effetti del creep misurati analizzando i dati ottenuti al tastatore meccanico. Grazie alla presenza della vitamina E non si ha ossidazione, quella rilevata è vicina a zero ed è da imputare alla lavorazione meccanica. Secondo tali considerazioni è possibile affermare che la riduzione dello spessore degli inserti da 5,6 mm a 3,6 mm non ha conseguenze critiche sul loro comportamento, e non comporta un fallimento del dispositivo.

The production, segregation and migration of melt and aqueous fluids (henceforth called liquid) plays an important role for the transport of mass and energy within the mantle and the crust of the Earth. Many properties of large-scale liquid migration processes such as the permeability of a rock matrix or the initial segregation of newly formed liquid from the host-rock depends on the grain-scale distribution and behaviour of liquid. Although the general mechanisms of liquid distribution at the grain-scale are well understood, the influence of possibly important modifying processes such as static recrystallization, deformation, and chemical disequilibrium on the liquid distribution is not well constrained. For this thesis analogue experiments were used that allowed to investigate the interplay of these different mechanisms in-situ. In high-temperature environments where melts are produced, the grain-scale distribution in “equilibrium” is fully determined by the liquid fraction and the ratio between the solid-solid and the solid-liquid surface energy. The latter is commonly expressed as the dihedral or wetting angle between two grains and the liquid phase (Chapter 2). The interplay of this “equilibrium” liquid distribution with ongoing surface energy driven recrystallization is investigated in Chapter 4 and 5 with experiments using norcamphor plus ethanol liquid. Ethanol in contact with norcamphor forms a wetting angle of about 25°, which is similar to reported angles of rock-forming minerals in contact with silicate melt. The experiments in Chapter 4 show that previously reported disequilibrium features such as trapped liquid lenses, fully-wetted grain boundaries, and large liquid pockets can be explained by the interplay of the liquid with ongoing recrystallization. Closer inspection of dihedral angles in Chapter 5 reveals that the wetting angles are themselves modified by grain coarsening. Ongoing recrystallization constantly moves liquid-filled triple junctions, thereby altering the wetting angles dynamically as a function of the triple junction velocity. A polycrystalline aggregate will therefore always display a range of equilibrium and dynamic wetting angles at raised temperature, rather than a single wetting angle as previously thought. For the deformation experiments partially molten KNO3–LiNO3 experiments were used in addition to norcamphor–ethanol experiments (Chapter 6). Three deformation regimes were observed. At a high bulk liquid fraction >10 vol.% the aggregate deformed by compaction and granular flow. At a “moderate” liquid fraction, the aggregate deformed mainly by grain boundary sliding (GBS) that was localized into conjugate shear zones. At a low liquid fraction, the grains of the aggregate formed a supporting framework that deformed internally by crystal plastic deformation or diffusion creep. Liquid segregation was most efficient during framework deformation, while GBS lead to slow liquid segregation or even liquid dispersion in the deforming areas.

This thesis presents a new method to explore the local mechanical properties such as bending modulus or surface tension of artificial and native pore-spanning membranes. Therefore the elastic response of a free-standing membrane to a local indentation by the means of atomic force microscopy is measured. Starting point are highly hexagonal ordered pores in alumina produced by electrochemical anodization of planar aluminium. The homogeneous pore radius can by tailored in the range of 10 nm up to 200 nm, but radius of 33 nm, 90 nm and 200 nm turned out to be best suited for investigation of the mechanical properties of pore-spanning native or artificial membranes. In this work artificial membrane systems consisting of DODAB as a bilayer in gel phase or DOTAP as a fluide membrane are spreaded by vesicle absorption on hexagonal structured pores after chemisorption of a 3-mercaptopropionic acid monolayer. Centrally indenting these nanodrums with an atomic force microscope tip yields force-indentation curves, which are quantitatively analyzed by solving the corresponding shape equations of continuum curvature elasticity. Since the measured response depends in a known way on the system geometry (pore size, tip radius) and on material parameters (bending modulus, lateral tension, adhesion), this opens the possibility to monitor local elastic properties of lipid membranes in a well-controlled setting. Additionally the locally distributed mechanical properties of pore-spanning artificial membranes are compared to those of native pore-spanning membranes. Therefore the basal membrane of MDCK II cells was prepared on porous alumina assays and their mechanical properties were analyzed by means of atomic force microscopy. Finally the elastic behavior such as the Young modulus of living MDCK II cells under various osmotic pressures is investigated. By changing the osmolarity in the extracellular region of MDCK II cells a volume change is induced according to hydration and dehydration of the cells, respectively. This volume change induces also a change in the elastic behavior of the cell, which is quantified by the means of force spectroscopy.

The research work was aimed at studying, with a deterministic approach, the relationships between the rock’s texture and its mechanical properties determined at the laboratory scale. The experimentation was performed on a monomineralic crystalline rock, varying in texture, i.e. grains shape. Multi-scale analysis has been adopted to determine the elasto-mechanical properties of the crystals composing the rock and its strength and deformability at the macro-scale. This let us to understand how the structural variability of the investigated rock affects its macromechanical behaviour. Investigations have been performed on three different scales: nano-scale (order of nm), micro-scale (tens of m) and macro-scale (cm). Innovative techniques for rock mechanics, i.e. Depth Sensing Indentation (DSI), have been applied, in order to determine the elasto-mechanical properties of the calcite grains. These techniques have also allowed to study the influence of grain boundaries on the mechanical response of calcite grains by varying the indents’ sizes and to quantify the effect of the applied load on the hardness and elastic modulus of the grain (indentation size effect, ISE). The secondary effects of static indentation Berkovich, Vickers and Knoop were analyzed by SEM, and some considerations on the rock’s brittle behaviour and the effect of microcracks can be made.

A broad variety of solid state NMR techniques were used to investigate the chain dynamics in several polyethylene (PE) samples, including ultrahigh molecular weight PEs (UHMW-PEs) and low molecular weight PEs (LMW-PEs). Via changing the processing history, i.e. melt/solution crystallization and drawing processes, these samples gain different morphologies, leading to different molecular dynamics. Due to the long chain nature, the molecular dynamics of polyethylene can be distinguished in local fluctuation and long range motion. With the help of NMR these different kinds of molecular dynamics can be monitored separately. In this work the local chain dynamics in non-crystalline regions of polyethylene samples was investigated via measuring 1H-13C heteronuclear dipolar coupling and 13C chemical shift anisotropy (CSA). By analyzing the motionally averaged 1H-13C heteronuclear dipolar coupling and 13C CSA, the information about the local anisotropy and geometry of motion was obtained. Taking advantage of the big difference of the 13C T1 relaxation time in crystalline and non-crystalline regions of PEs, the 1D 13C MAS exchange experiment was used to investigate the cooperative chain motion between these regions. The different chain organizations in non-crystalline regions were used to explain the relationship between the local fluctuation and the long range motion of the samples. In a simple manner the cooperative chain motion between crystalline and non-crystalline regions of PE results in the experimentally observed diffusive behavior of PE chain. The morphological influences on the diffusion motion have been discussed. The morphological factors include lamellar thickness, chain organization in non-crystalline regions and chain entanglements. Thermodynamics of the diffusion motion in melt and solution crystallized UHMW-PEs is discussed, revealing entropy-controlled features of the chain diffusion in PE. This thermodynamic consideration explains the counterintuitive relationship between the local fluctuation and the long range motion of the samples. Using the chain diffusion coefficient, the rates of jump motion in crystals of the melt crystallized PE have been calculated. A concept of "effective" jump motion has been proposed to explain the difference between the values derived from the chain diffusion coefficients and those in literatures. The observations of this thesis give a clear demonstration of the strong relationship between the sample morphology and chain dynamics. The sample morphologies governed by the processing history lead to different spatial constraints for the molecular chains, leading to different features of the local and long range chain dynamics. The knowledge of the morphological influence on the microscopic chain motion has many implications in our understanding of the alpha-relaxation process in PE and the related phenomena such as crystal thickening, drawability of PE, the easy creep of PE fiber, etc.

Membranen spielen eine essentielle Rolle bei vielen wichtigen zellulären Prozessen. Sie ermöglichen die Erzeugung von chemischen Gradienten zwischen dem Zellinneren und der Umgebung. Die Zellmembran übernimmt wesentliche Aufgaben bei der intra- und extrazellulären Signalweiterleitung und der Adhäsion an Oberflächen. Durch Prozesse wie Endozytose und Exozytose werden Stoffe in oder aus der Zelle transportiert, eingehüllt in Vesikel, welche aus der Zellmembran geformt werden. Zusätzlich bietet sie auch Schutz für das Zellinnere. Der Hauptbestandteil einer Zellmembran ist die Lipiddoppelschicht, eine zweidimensionale fluide Matrix mit einer heterogenen Zusammensetzung aus unterschiedlichen Lipiden. In dieser Matrix befinden sich weitere Bausteine, wie z.B. Proteine. An der Innenseite der Zelle ist die Membran über Ankerproteine an das Zytoskelett gekoppelt. Dieses Polymernetzwerk erhöht unter anderem die Stabilität, beeinflusst die Form der Zelle und übernimmt Funktionenrnbei der Zellbewegung. Zellmembranen sind keine homogenen Strukturen, je nach Funktion sind unterschiedliche Lipide und Proteine in mikrsokopischen Domänen angereichert.Um die grundlegenden mechanischen Eigenschaften der Zellmembran zu verstehen wurde im Rahmen dieser Arbeit das Modellsystem der porenüberspannenden Membranen verwendet.Die Entwicklung der porenüberspannenden Membranen ermöglicht die Untersuchung von mechanischen Eigenschaften von Membranen im mikro- bis nanoskopischen Bereich mit rasterkraftmikroskopischen Methoden. Hierbei bestimmen Porosität und Porengröße des Substrates die räumliche Auflösung, mit welcher die mechanischen Parameter untersucht werdenrnkönnen. Porenüberspannende Lipiddoppelschichten und Zellmembranen auf neuartigen porösen Siliziumsubstraten mit Porenradien von 225 nm bis 600 nm und Porositäten bis zu 30% wurden untersucht. Es wird ein Weg zu einer umfassenden theoretischen Modellierung der lokalen Indentationsexperimente und der Bestimmung der dominierenden energetischen Beiträge in der Mechanik von porenüberspannenden Membranen aufgezeigt. Porenüberspannende Membranen zeigen eine linear ansteigende Kraft mit zunehmender Indentationstiefe. Durch Untersuchung verschiedener Oberflächen, Porengrößen und Membranen unterschiedlicher Zusammensetzung war es für freistehende Lipiddoppelschichten möglich, den Einfluss der Oberflächeneigenschaften und Geometrie des Substrates, sowie der Membranphase und des Lösungsmittels auf die mechanischen Eigenschaften zu bestimmen. Es ist möglich, die experimentellen Daten mit einem theoretischen Modell zu beschreiben. Hierbei werden Parameter wie die laterale Spannung und das Biegemodul der Membran bestimmt. In Abhängigkeit der Substrateigenschaften wurden für freitragende Lipiddoppelschichten laterale Spannungen von 150 μN/m bis zu 31 mN/m gefunden für Biegemodulde zwischen 10^(−19) J bis 10^(−18) J. Durch Kraft-Indentations-Experimente an porenüberspannenden Zellmembranen wurde ein Vergleich zwischen dem Modell der freistehenden Lipiddoppelschichten und nativen Membranen herbeigeführt. Die lateralen Spannungen für native freitragende Membranen wurden zu 50 μN/m bestimmt. Weiterhin konnte der Einfluss des Zytoskeletts und der extrazellulä-rnren Matrix auf die mechanischen Eigenschaften bestimmt und innerhalb eines basolateralen Zellmembranfragments kartiert werden, wobei die Periodizität und der Porendurchmesser des Substrates das räumliche Auflösungsvermögen bestimmen. Durch Fixierung der freistehenden Zellmembran wurde das Biegemodul der Membran um bis zu einem Faktor 10 erhöht. Diese Arbeit zeigt wie lokal aufgelöste, mechanische Eigenschaften mittels des Modellsystems der porenüberspannenden Membranen gemessen und quantifiziert werden können. Weiterhin werden die dominierenden energetischen Einflüsse diskutiert, und eine Vergleichbarkeit zurnnatürlichen Membranen hergestellt.rn

As the elastic response of cell membranes to mechanical stimuli plays a key role in various cellular processes, novel biophysical strategies to quantify the elasticity of native membranes under physiological conditions at a nanometer scale are gaining interest. In order to investigate the elastic response of apical membranes, elasticity maps of native membrane sheets, isolated from MDCK II (Madine Darby Canine kidney strain II) epithelial cells, were recorded by local indentation with an Atomic Force Microscope (AFM). To exclude the underlying substrate effect on membrane indentation, a highly ordered gold coated porous array with a pore diameter of 1.2 μm was used to support apical membranes. Overlays of fluorescence and AFM images show that intact apical membrane sheets are attached to poly-D-lysine coated porous substrate. Force indentation measurements reveal an extremely soft elastic membrane response if it is indented at the center of the pore in comparison to a hard repulsion on the adjacent rim used to define the exact contact point. A linear dependency of force versus indentation (-dF/dh) up to 100 nm penetration depth enabled us to define an apparent membrane spring constant (kapp) as the slope of a linear fit with a stiffness value of for native apical membrane in PBS. A correlation between fluorescence intensity and kapp is also reported. Time dependent hysteresis observed with native membranes is explained by a viscoelastic solid model of a spring connected to a Kelvin-Voight solid with a time constant of 0.04 s. No hysteresis was reported with chemically fixated membranes. A combined linear and non linear elastic response is suggested to relate the experimental data of force indentation curves to the elastic modulus and the membrane thickness. Membrane bending is the dominant contributor to linear elastic indentation at low loads, whereas stretching is the dominant contributor for non linear elastic response at higher loads. The membrane elastic response was controlled either by stiffening with chemical fixatives or by softening with F-actin disrupters. Overall, the presented setup is ideally suitable to study the interactions of the apical membrane with the underlying cytoskeleton by means of force indentation elasticity maps combined with fluorescence imaging.

The understanding of the coupling between superconducting YBa2Cu3O7 (YBCO) layers decoupled by non superconducting PrBa2Cu3O7 (PBCO) layers in c-axis oriented superlattices was the aim of this thesis. For this purpose two conceptually different kind of transport experiments have been performed. rnrnIn the first type of transport experiments the current is flowing parallel to the layers. Here the coupling is probed indirectly using magnetic vortex lines, which are penetrating the superlattice. Movement of the vortex segments in neighbouring YBCO layers is more or less coherent depending on the thickness of both the superconducting and non superconducting layers. This in-plane transport was measured either by sending an external current through bridges patterned in the superlattice or by an induced internal current. rnThe vortex-creep activation energy U was determined by analysis of the in-plane resistive transition in an external magnetic field B oriented along the c-axis. The activation energies for two series of superlattices were investigated. In one series the thickness of the YBCO layers was constant (nY=4 unit cells) and the number of the PBCO unit cells was varied, while in the other the number of PBCO layers was constant (nP=4) and nY varied. The correlation length of the vortex system was determined to be 80 nm along the c-axis direction. It was found that even a single PBCO unit cell in a superlattice effectively cuts the flux lines into shorter weakly coupled segments, and the coupling of the vortex systems in neighbouring layers is negligible already for a thickness of four unit cells of the PBCO layers. A characteristic variation of the activation energy for the two series of superlattices was found, where U0 is proportional to the YBCO thickness. A change in the variation of U0 with the current I in the specimen was observed, which can be explained in terms of a crossover in the vortex creep process, generated by the transport current. At low I values the dislocations mediated (plastic) vortex creep leads to thermally assisted flux-flow behaviour, whereas at high current the dc transport measurements are dominated by elastic (collective) creep.rnThe analysis of standard dc magnetization relaxation data obtained for a series superlattices revealed the occurrence of a crossover from elastic (collective) vortex creep at low temperature to plastic vortex creep at high T. The crossover is generated by the T dependent macroscopic currents induced in the sample. The existence of this creep crossover suggests that, compared with the well known Maley technique, the use of the normalized vortex creep activation energy is a better solution for the determination of vortex creep parameters.rnrnThe second type of transport experiments was to measure directly a possible Josephson coupling between superconducting CuO2 double planes in the superlattices by investigation of the transport properties perpendicular to the superconducting planes. Here three different experiments have been performed. The first one was to pattern mesa structures photolithographically as in previous works. The second used three-dimensional nanostructures cut by a focused ion beam. For the these two experiments insufficient patterning capabilities prevented an observation of the Josephson effect in the current voltage curves. rnA third experiment used a-axis and (110) oriented YBCO films, where in-plane patterning can in principle be sufficient to measure transport perpendicular to the superconducting planes. Therefore the deposition of films with this unusual growth orientation was optimized and investigated. The structural and microstructural evolution of c-axis to a-axis orientation was monitored using x-ray diffraction, scanning electron microscopy and magnetization measurements. Films with full a-axis alignment parallel to the substrate normal could be achieved on (100)SrTiO3. Due to the symmetry of the substrate the c-axis direction in-plane is twofold. Transferring the deposition conditions to films grown on (110)SrTiO3 allowed the growth of (110) oriented YBCO films with a unique in-plane c-axis orientation. While these films were of high quality by crystallographic and macroscopic visual inspection, electron microscopy revealed a coherent crack pattern on a nanoscale. Therefore the actual current path in the sample was not determined by the macroscopic patterning which prohibited investigations of the in-plane anisotropy in this case.rn

Bone is continually being removed and replaced through the actions of basic multicellular units (BMU). This constant upkeep is necessary to remove microdamage formed naturally due to fatigue and thus maintain the integrity of the bone. The repair process in bone is targeted, meaning that a BMU travels directly to the site of damage and repairs it. It is still unclear how targeted remodelling is stimulated and directed but it is highly likely that osteocytes play a role. A number of theories have been advanced to explain the microcrack osteocyte interaction but no complete mechanism has been demonstrated. Osteocytes are connected to each other by dendritic processes. The “scissors model" proposed that the rupture of these processes where they cross microcracks signals the degree of damage and the urgency of the necessary repair. In its original form it was proposed that under applied compressive loading, microcrack faces will be pressed together and undergo relative shear movement. If this movement is greater than the width of an osteocyte process, then the process will be cut in a “scissors like" motion, releasing RANKL, a cytokine known to be essential in the formation of osteoclasts from pre-osteoclasts. The main aim of this thesis was to investigate this theoretical model with a specific focus on microscopy and finite element modelling. Previous studies had proved that cyclic stress was necessary for osteocyte process rupture to occur. This was a divergence from the original “scissors model" which had proposed that the cutting of cell material occurred in one single action. The present thesis is the first study to show fatigue failure in cellular processes spanning naturally occurring cracks and it's the first study to estimate the cyclic strain range and relate it to the number of cycles to failure, for any type of cell. Rupture due to shear movement was ruled out as microcrack closing never occurred, as a result of plastic deformation of the bone. Fatigue failure was found to occur due to cyclic tensile stress in the locality of the damage. The strain range necessary for osteocyte process rupture was quantified. It was found that the lower the process strain range the greater the number of cycles to cell process failure. FEM modelling allowed to predict stress in the vicinity of an osteocyte process and to analyse its interaction with the bone surrounding it: simulations revealed evident creep effects in bone during cyclic loading. This thesis confirms and dismisses aspects of the “scissors model". The observations support the model as a viable mechanism of microcrack detection by the osteocyte network, albeit in a slightly modified form where cyclic loading is necessary and the method of rupture is fatigue failure due to cyclic tensile motion. An in depth study was performed focusing on microscopy analysis of naturally occurring cracks in bone and FEM simulation analysis of an osteocyte process spanning a microcrack in bone under cyclic load.
