The transcription factor REST is a key suppressor of neuronal genes in non-neuronal tissues. REST has been shown to suppress pro-neuronal microRNAs in neural progenitors indicating that REST-mediated neurogenic suppression may act in part via microRNAs. We used neural differentiation of Rest-null mouse ESC to identify dozens of microRNAs regulated by REST during neural development. One of the identified microRNAs, miR-375, was upregulated during human spinal motor neuron development. We found that miR-375 facilitates spinal motor neurogenesis by targeting the cyclin kinase CCND2 and the transcription factor PAX6. Additionally, miR-375 inhibits the tumor suppressor p53 and protects neurons from apoptosis in response to DNA damage. Interestingly, motor neurons derived from a spinal muscular atrophy patient displayed depressed miR-375 expression and elevated p53 protein levels. Importantly, SMA motor neurons were significantly more susceptible to DNA damage induced apoptosis suggesting that miR-375 may play a protective role in motor neurons.

Amyotrophic lateral sclerosis (ALS) is an incurable neuromuscular disease that leads to a profound loss of life quality and premature death. Around 10% of the cases are inherited and ALS8 is an autosomal dominant form of familial ALS caused by mutations in the vamp-associated protein B/C (VAPB) gene. The VAPB protein is involved in many cellular processes and it likely contributes to the pathogenesis of other forms of ALS besides ALS8. A number of successful drug tests in ALS animal models could not be translated to humans underscoring the need for novel approaches. The induced pluripotent stem cells (iPSC) technology brings new hope, since it can be used to model and investigate diseases in vitro. Here we present an additional tool to study ALS based on ALS8-iPSC. Fibroblasts from ALS8 patients and their non-carrier siblings were successfully reprogrammed to a pluripotent state and differentiated into motor neurons. We show for the first time that VAPB protein levels are reduced in ALS8-derived motor neurons but, in contrast to over-expression systems, cytoplasmic aggregates could not be identified. Our results suggest that optimal levels of VAPB may play a central role in the pathogenesis of ALS8, in agreement with the observed reduction of VAPB in sporadic ALS.

Motor cortex stimulation (MCS) has been used to treat patients with neuropathic pain resistant to other therapeutic approaches; however, the mechanisms of pain control by MCS are still not clearly understood. We have demonstrated that MCS increases the nociceptive threshold of naive conscious rats, with opioid participation. In the present study, the effect of transdural MCS on neuropathic pain in rats subjected to chronic constriction injury of the sciatic nerve was investigated. In addition, the pattern of neuronal activation, evaluated by Fos and Zif268 immunolabel, was performed in the spinal cord and brain sites associated with the modulation of persistent pain. MCS reversed the mechanical hyperalgesia and allodynia induced by peripheral neuropathy. After stimulation, Fos immunoreactivity (Fos-IR) decreased in the dorsal horn of the spinal cord and in the ventral posterior lateral and medial nuclei of the thalamus, when compared to animals with neuropathic pain. Furthermore, the MCS increased the Fos-IR in the periaqueductal gray, the anterior cingulate cortex and the central and basolateral amygdaloid nuclei. Zif268 results were similar to those obtained for Fos, although no changes were observed for Zif268 in the anterior cingulate cortex and the central amygdaloid nucleus after MCS. The present findings suggest that MCS reverts neuropathic pain phenomena in rats, mimicking the effect observed in humans, through activation of the limbic and descending pain inhibitory systems. Further investigation of the mechanisms involved in this effect may contribute to the improvement of the clinical treatment of persistent pain. Physical exercise is known to enhance brain function in several aspects. We evaluated the acute effects of a moderate forced exercise protocol on synaptic proteins, namely synapsin 1 (SYN) and synaptophysin (SYP), and structural proteins (neurofilaments, NFs) in rat brain regions related to motor function and often affected by neurodegenerative disorders. Immunohistochemistry, Western blotting and real-time PCR were used to analyze the expression of those proteins after 3, 7 and 15 days of exercise (EX3, EX7 and EX15). In the cerebellum, increase of SYN was observed at EX7 and EX15 and of NF68 at EX3. In the substantia nigra, increases of protein levels were observed for NF68 and NF160 at EX3. In the striatum, there was an increase of SYN at EX3 and EX7, of SYP at EX7 and of NF68 at EX3. In the cortex, decreased levels of NF68 and NF160 were observed at EX3, followed by an increase of NF68 at EX15. In the reticular formation, all NF proteins were increased at EX15. The mRNA data for each time-point and region also revealed significant exercise-related changes of SYN, SYP and NF expression. These results suggest that moderate physical exercise modulates synaptic and structural proteins in motor brain areas, which may play an important role in the exercise-dependent brain plasticity. Este estudo demonstrou os impactos do E.C.R. ¿ Resposta Eficiente ao Consumidor na cadeia de suprimentos de uma empresa de varejo, precisamente do grupo Sendas, empresa esta que busca na logística um diferencial competitivo para sobreviver neste mercado acompanhando as mudanças e as melhorias operacionais no fluxo de produtos e das operações, causadas pela evolução tecnológica que o varejo vem sofrendo nos últimos anos. Para tanto foram realizadas pesquisas bibliográfica e documental, de forma a embasar conceitualmente e com informações relevantes a caracterização da empresa inserindo a mesma neste contexto de cadeia de suprimentos e foco no cliente, estabelecendo uma relação entre a empresa pesquisada e a aplicação da tecnologia existente. Quanto aos aspectos relacionados aos ganhos de eficiência, houve uma preocupação com a implantação das ferramentas do E.C.R. como um fator determinante para a perpetuação do negócio. Os ganhos de eficiência permitiram a empresa este diferencial competitivo na disputa pelo mercado, certamente outros aspectos como atendimento são relevantes para garantir a eficiência da cadeia, portanto, avaliamos as interfaces entre os participantes da cadeia (fabricantes, fornecedores), grupo Sendas (varejista) e consumidores. Assim, concluímos este estudo mapeando pontos fortes e fracos desta relação, identificando as barreiras a serem vencidas e algumas recomendações.

A presente pesquisa possui um enfoque no processo de internacionalização das empresas brasileiras, um fenômeno que vem ocorrendo de forma lenta e gradual ao longo dos últimos 25 anos, com grande ênfase nos anos 1990, quando o Brasil abriu sua economia ao capital externo, proporcionando maiores investimentos em novos produtos, novas tecnologias, e a adoção por parte das nossas organizações de novas formas e práticas de gestão empresarial, as quais já faziam parte há um bom tempo das organizações multinacionais pelo mundo afora, que sempre utilizaram essas melhores formas de gestão empresarial em seu estado da arte. Este atraso do Brasil em relação ao mundo desenvolvido, em que o País se fechou como uma verdadeira "ostra", refletiu-se em prejuízos imensuráveis ao desenvolvimento social, político e econômico, resultando em atrasos tecnológicos e educacionais e na consequente falta de pessoal qualificado em face de um mundo globalizado, no qual se mostram imprescindíveis as inovações tecnológicas e a capacitação humana. Nesse contexto, o processo de internacionalização de empresas, especificamente o segmento do cooperativismo de agronegócio no estado do Paraná, ganha um impulso significativo, quando as cooperativas, a partir da década de 1980, buscam sua inserção internacional no mundo globalizado através de um aumento gradativo em suas exportações, inicialmente somente com produtos in natura e, posteriormente, com a introdução de novas tecnologias, equipamentos, pesquisas, modernas formas de gestão e educação continuada de pessoal, possibilitando às cooperativas, e de forma geral ao setor cooperativista paranaense, um grande salto financeiro e um aumento expressivo na composição do PIB do Paraná, representando nos dias atuais aproximadamente 40% do total das exportações do estado. Dessa forma, este trabalho procura contribuir com a academia e com as organizações cooperativistas do estado do Paraná no sentido de buscar as razões para esse importante e irreversível processo de internacionalização, que trouxe, e ainda traz, vantagens financeiras, tecnológicas e dinamismo à economia local com os seus diversos atores, e que transforma essas organizações paranaenses e brasileiras em grandes players globais nos negócios internacionais.

A presente Tese tem por objetivo propor um sistema de inovação permanente para a área do Agronegócio do setor da Horticultura da Região Serrana do Estado do Rio de Janeiro. Este trabalho parte da suposição de que existe um potencial não explorado de inovação pelo trabalho atualizável mediante um modelo referencial composto por um conjunto de indutores denominados de INDILABs – Indutores da Inovação Laboral: a tecnologia, a gestão, o processo de cognição e a atitude e ação. A Tecnologia promove mudanças econômicas, políticas e culturais e influencia o trabalho, a vida e o processo de cognição. A Gestão promove a personalização dos indivíduos e desenvolver a criatividade. O Processo de Cognição contribui para articular e harmonizar os sistemas sociais e deve ser centrado nos valores éticos e humanos para que haja equilíbrio entre o ser e o seu valor de uso, enquanto que a Atitude e Ação buscam consolidar os movimentos mais criativos, autônomos, humanos, prazerosos e desafiadores nos espaços coletivos. Discute-se a essencialidade da capacidade de inovação para a continuidade dos sistemas produtivos, sendo considerada uma condição fundamental para a sobrevivência dos negócios, dos empregos, do trabalho e da renda. Relaciona-se o Estado com a inovação, afirmando que além de apoiar o desenvolvimento de atividades econômicas, dos indivíduos e da sociedade, ele deve assegurar a continuidade dos sistemas produtivos pela capacidade de inovação por meio e mediante o trabalho. Utilizando procedimentos metodológicos baseados no estruturalismo, buscou-se a estrutura subjacente às práticas dos trabalhos dos agricultores familiares e suas relações com o sistema de apoio ao desenvolvimento da inovação. A coleta de dados para a pesquisa foi realizada com base em entrevistas individuais e em grupos, formais e informais, através de seminário específico e da observação do contexto e exame de documentos. O modelo referencial de inovação proposto visa fundamentar a formulação de políticas públicas nas seguintes áreas: a) Desenvolvimento do empreendedorismo; b) Formação e Capacitação do agricultor e de mão de obra; c) Promover a cooperação e inter-relacionamento institucional; d) Comunicação e difusão; e) Apoio a Comercialização; e f) Atuar em programas de diversificação da Produção.

The effects of serum and brain calcium concentration on rat behavior were tested by maintaining animals on either distilled water (N = 60) or water containing 1% calcium gluconate (N = 60) for 3 days. Animals that were maintained on high calcium drinking water presented increased serum calcium levels (control = 10.12 ± 0.46 vs calcium treated = 11.62 ± 0.51 µg/dl). Increase of brain calcium levels was not statistically significant. In the behavioral experiments each rat was used for only one test. Rats that were maintained on high calcium drinking water showed increased open-field behavior of ambulation (20.68%) and rearing (64.57%). on the hole-board, calcium-supplemented animals showed increased head-dip (67%) and head-dipping (126%), suggesting increased ambulatory and exploratory behavior. The time of social interaction was normal in animals maintained on drinking water containing added calcium. Rats supplemented with calcium and submitted to elevated plus-maze tests showed a normal status of anxiety and elevated locomotor activity. We conclude that elevated levels of calcium enhance motor and exploratory behavior of rats without inducing other behavioral alterations. These data suggest the need for a more detailed analysis of several current proposals for the use of calcium therapy in humans, for example in altered blood pressure states, bone mineral metabolism disorders in the elderly, hypocalcemic states, and athletic activities.

This work describes the study and the implementation of the speed control for a three-phase induction motor of 1,1 kW and 4 poles using the neural rotor flux estimation. The vector speed control operates together with the winding currents controller of the stator phasis. The neural flux estimation applied to the vector speed controls has the objective of compensating the parameter dependences of the conventional estimators in relation to the parameter machine s variations due to the temperature increases or due to the rotor magnetic saturation. The implemented control system allows a direct comparison between the respective responses of the speed controls to the machine oriented by the neural rotor flux estimator in relation to the conventional flux estimator. All the system control is executed by a program developed in the ANSI C language. The main DSP recources used by the system are, respectively, the Analog/Digital channels converters, the PWM outputs and the parallel and RS-232 serial interfaces, which are responsible, respectively, by the DSP programming and the data capture through the supervisory system

The aim of this Study was to compare the learning process of a highly complex ballet skill following demonstrations of point light and video models 16 participants divided into point light and video groups (ns = 8) performed 160 trials of a pirouette equally distributed in blocks of 20 trials alternating periods of demonstration and practice with a retention test a day later Measures of head and trunk oscillation coordination d1 parity from the model and movement time difference showed similarities between video and point light groups ballet experts evaluations indicated superiority of performance in the video over the point light group Results are discussed in terms of the task requirements of dissociation between head and trunk rotations focusing on the hypothesis of sufficiency and higher relevance of information contained in biological motion models applied to learning of complex motor skills

Background: Cerebral palsy (CP) presents changes in posture and movement as a core characteristic, which requires therapeutic monitoring during the habilitation or rehabilitation of children. Besides clinical treatment, it is fundamental that professionals use systems of evaluation to quantify the difficulties presented to the individual and assist in the organization of a therapeutic program. The aim of this study was to quantitatively verify the performance of children with spastic di-paresia type CP.Methods: The Pediatric Evaluation of Disability Inventory (PEDI) and Gross Motor Function Classification System (GMFM) tests were used and classification made through the GMFCS in the assessment of 7 patients with CP, 4 females and 3 males, average age of 9 years old.Results: According to GMFCS scales, 17% (n=1) were level II and 83% (n=6) were level III. The PEDI test and 88 GMFM items were used in the area of mobility. We observed that there was high correlation between mobility and gross motor function with Pearson's correlation coefficient =0.929) showing the likely impact of these areas in the functional skills and the quality of life of these patients.Conclusion: We suggest the impact of the limitation of the areas in functional skills and quality of life of these patients.
