Aeromonas hydrophila flagella glycosylation: involvement of a lipid carrier.

Polar flagellin proteins from Aeromonas hydrophila strain AH-3 (serotype O34) were found to be O-glycosylated with a heterogeneous glycan. Mutants unable to produce WecP or Gne enzymes showed altered motility, and the study of their polar flagellin glycosylation showed that the patterns of glycosylation differed from that observed with wild type polar flagellin. This suggested the involvement of a lipid carrier in glycosylation. A gene coding for an enzyme linking sugar to a lipid carrier was identified in strain AH-3 (WecX) and subsequent mutation abolished completely motility, flagella production by EM, and flagellin glycosylation. This is the first report of a lipid carrier involved in flagella O-glycosylation. A molecular model has been proposed. The results obtained suggested that the N-acetylhexosamines are N-acetylgalactosamines and that the heptasaccharide is completely independent of the O34-antigen lipopolysaccharide. Furthermore, by comparing the mutants with differing degrees of polar flagellin glycosylation, we established their importance in A. hydrophila flagella formation and motility.

Role of Aeromonas hydrophila flagella glycosylation in adhesion to Hep-2 cells, biofilm formation and immune stimulation.

Polar flagellin proteins from Aeromonas hydrophila strain AH-3 (serotype O34) were found to be O-glycosylated with a heterogeneous heptasaccharide glycan. Two mutants with altered (light and strong) polar flagella glycosylation still able to produce flagella were previously obtained, as well as mutants lacking the O34-antigen lipopolysaccharide (LPS) but with unaltered polar flagella glycosylation. We compared these mutants, altogether with the wild type strain, in different studies to conclude that polar flagella glycosylation is extremely important for A. hydrophila adhesion to Hep-2 cells and biofilm formation. Furthermore, the polar flagella glycosylation is an important factor for the immune stimulation of IL-8 production via toll receptor 5 (TLR5).

Developing a new service business model for an industrial supplier

Project business companies are moving towards solution offering in order to avoid discontinuity of project business and to gain other advantages. An option to implement solution business could be BOOT (Build-Own-Operate-Transfer) business model where a company is given the responsibilities to design, finance, build, own, operate and maintain for example production facilities of a client. The contract is made for 10-30 years, the client pays the solution during this period of time and after contract termination the facilities are transferred to the ownership of the client. The purpose of this study was to provide knowledge about BOOT business model for the company in question and its employees and to create a settlement of the advantages, disadvantages and risks of it. Furthermore, one of the main objectives was to create a description of the network needed to run a BOOT project. The objectives were met through a literature study and an explorative case study with appropriate interviews. Based on this study, the company should be able to evaluate the applicability of BOOT business model to their business environment better.

The grammar of psychosis

This thesis is concerned with the philosophical grammar of certain psychiatric concepts, which play a central role in delineating the field of psychiatric work. The concepts studied are ‘psychosis’, ‘delusion’, ‘person’, ‘understanding’ and ‘incomprehensibility’. The purpose of this conceptual analysis is to provide a more perspicuous view of the logic of these concepts, how psychiatric work is constituted in relation to them, and what this tells us about the relationships between the conceptual and the empirical in psychiatric concepts. The method used in the thesis is indebted primarily to Ludwig Wittgenstein’s conception of philosophy, where we are urged to look at language uses in relation to practices in order to obtain a clearer overview of practices of interest; this will enable us to resolve the conceptual problems related to these practices. This questioning takes as its starting point the concept of psychosis, a central psychiatric concept during the twentieth century. The conceptual analysis of ‘psychosis’ shows that the concept is logically dependent on the concepts of ‘understanding’ and ‘person’. Following the lead found in this analysis, the logic of person-concepts in psychiatric discourse is analysed by a detailed textual analysis of a psychiatric journal article. The main finding is the ambiguous uses of ‘person’, enabling a specifically psychiatric form of concern in human affairs. The grammar of ‘understanding’ is then tackled from the opposite end, by exploring the logic of the concept of ‘incomprehensibility’. First, by studying the DSM-IV definition of delusion it is shown that its ambiguities boil down to the question of whether psychiatric practice is better accounted for in terms of the grammar of ‘incorrectness’ or ‘incomprehensibility’. Second, the grammar of ‘incomprehensibility’ is further focused on by introducing the distinction between positive and negative conceptions of ‘incomprehensibility’. The main finding is that this distinction has wide-ranging implications for our understanding of psychiatric concepts. Finally, some of the findings gained in these studies are ‘put into practice’ in studying the more practical question of the conceptual and ethical problems associated with the concept of ‘prodromal symptom of schizophrenia’ and the agenda of early detection and intervention in schizophrenia more generally.

Konstvetenskap - konsthistoria

En större samling konstvetenskaplig litteratur finns vid huvudbiblioteket och av dem handlar några tusen volymer om konsthistoria. Vid Humanistiska biblioteket finns dessutom en stor samling konsthistorisk litteratur i anslutning till ämnet Konstvetenskap Bokdonationer både från Sverige och från Finland kompletterar det som biblioteket erhållit genom sin friexemplarsrätt efter 1918. Bland donationer från Sverige kan nämnas bankdirektör L.A. Odencranz konsthistoriska samling och från Finland kan nämnas några samlingar ur sterbhus så som efter målarinnan Sigrid Granfelt, häradshövding J.V.Brummer och systrarna Annie och Berta Edelfelt. Återvinning av konsthistorisk litteratur tryckt före 1980 kan göras genom manuell sökning i huvudbibliotekets systematiska kortkatalog med signum IVd1. Retroaktiv inmatning i databasen Alma gör att all konstvetenskaplig litteratur tryckt mellan 1830 och 1979 med tiden blir sökbar i Alma med sökfunktionen Signum och söktermen HB IVd. Samlingen med signum HB IVa finns vid huvudbiblioteket och innehåller litteraturvetenskaplig litteratur till och med år 1999. Litteraturvetenskaplig litteratur finns dessutom vid Humanistiska biblioteket i anslutning till ämnet Litteraturvetenskap. Litteraturvetenskaplig litteratur har ingått i talrika donationer till akademibiblioteket. Språkvetenskaplig litteratur finns också vid Humanistiska biblioteket. Språkvetenskaplig litteratur har ingått i talrika bokdonationer till akademibiblioteket, som exempel kan nämnas bokdonationer av professor Julio Reuter, fru Aino Åberg, kamrer Magnus Åberg, docent Väinö Nordström, professor Hugo Pipping, lektor Ludvig Granit, lektor John Schlüter och FM Karl Kekoni samt J.V. Brummer. Friexemplaren som erhållits från 1919 samt inköp har kompletterat samlingen. Den språkvetenskapliga litteraturen vid huvudbiblioteket söks i Alma med sökfunktionen Signum och söktermen HB III. En del litteratur tryckt före 1980 skall sökas manuellt i den systematiska kortkatalogen, men retroaktiv inmatning i Alma av litteratur tryckt mellan 1830 och 1979 pågår. Statskunskap och politik

Samlingen med signum HB VIc1 finns vid huvudbiblioteket och innehåller litteratur om statskunskap och politik till och med år 1999. Liknande litteratur finns i anslutning till ämnet Statskunskap. Litteratur om statskunskap och politik har ingått i bokdonationer såväl från hemlandet som från Sverige. Några donatorer av speciellt stora samlingar statsvetenskaplig litteratur kan nämnas, J.V. Brummer, J.A. Nordman, Rafael Erich, Ernst Estlander, Erik Ehrström och Tor von Wright. Politisk litteratur har donerats av Edvard Öhrnberg, Märta Björkenheim, Rafael Erich, Ernst Estlander, Hjalmar Crohn, Erik Ehrström, Carl von Bonsdorff, Jon Hartman, Allan Wallenius, Nils Källström och Hjalmar J. Samlingen teologisk litteratur med signum TF finns vid Teologiska biblioteket. Vid huvudbiblioteket finns teologisk litteratur under signum HB IIc1. Många tusen volymer teologisk litteratur har genom åren donerats till akademibiblioteket och donationer görs allt fortfarande. Bland tidigare donatorer kan som exempel nämnas prostinnan Hortense Grönstrand, häradshövding J.V. Brummer, vicehäradshövding Jon Hartman samt prostarna Konrad Ahlman, Karl Anders Troberg, Viktor Roslin, Theodor von Pfaler, Werner Wirén och C.H. Ekblom.
