The influence of nitric-perchloric, aqua regia, dry ashing and microwave digestion methods, in combination with 100 and 200 mg of sample, on the characterization and recovery of nutrients in samples of sludge sewage, poultry, swine, quail and bovine manures, organic compost, organic substrate and humic material were studied. Nitric-perchloric digestion with 200 mg samples recovered the higher nutrient contents. The nitric-perchloric method recovered also low levels of K. Dry ashing caused S volatilization and microwave digestion produced dark color extracts and this impaired S determination. Aqua regia recovered the lowest contents of nutrients in the organic residues evaluated.

This review attempts to provide an updated overview of the Quick, Easy, Cheap, Effective, Ruged and Safe (QuEChERS) multiresidue extraction method, that involves initial extraction in acetonitrile, an extraction/partition step after the addition of salt, and a cleanup step utilizing dispersive solid phase extraction. QuEChERS method is nowadays the most applied extraction method for the determination of pesticide residues in food samples, providing acceptable recoveries for acidic, neutral and basic pesticides. Several applications for various food matrices (fruits, vegetables, cereals and others) in combination with chromatographic mass spectrometry analysis were presented.

Samplings of atmospheric particulate matter (PM) were carried out between the months of March and April of 2007, simultaneously in two areas of Londrina, an urban (Historical Museum) and other rural (Farm School-UEL). PM was collected using the cascade impactor consisting of four impaction stages (0.25 to 10 μm). The results indicated that the fine fraction (PM2.5) represented a significant portion of the mass of PM10 (70 and 67% in the urban and rural places, respectively). Cl-, NO3- and SO4(2-) were determined by ion chromatography and the size distribution is presented. Natural and anthropogenic sources were suggested to the ionic components in the fine and coarse mode of PM.

The determination of acetic acid in vinegar adulterated sample using simultaneous potentiometric and condutometric titrations was used as an example of integrated experiment in instrumental analysis. An Excel® spreadsheet, which allows the entry of simultaneous data and the construction of the superimposed experimental curves (condutometric, potentiometric, first and second derivative potentiometric curve and, distribution diagrama of the acetic species as function of pH), was used as powerful tool to discuss the fundamental concepts involved in each technique and choose the best of them to quantify, without mutual interference, H3CCOOH and HCl in vinegar adulterated sample.

Conventional sample holder cells used to the electric characterization of ceramics at high temperature consists of an alumina tube and platinum wires and plates using a complex design. The high cost materials used in the conventional sampler holder cell were replaced by stainless steel and conventional ceramics. The sample holder was validated by characterizing yttria-stabilized-zirconia in a temperature range of 25 to 700 ºC. The results do not present variations, discontinuity or unusual noise in the electric signals. Several samples were characterized without maintenance, which demonstrates that the sample holder is electric and mechanic adequate to be used to electrical characterization of ceramics up to 700 ºC.

The restricted availability of water sources suitable for consumption and high costs for obtaining potable water has caused an increase of the conscience concerning the use. Thus, there is a high demand for "environmentally safe methods" which are according to the principles of Green Chemistry. Moreover, these methods should be able to provide reliable results for the analysis of water quality for various pollutants, such as phenol. In this work, greener alternatives for sample preparation for phenol determination in aqueous matrices are presented, which include: liquid phase microextraction, solid phase microextraction, flow analysis, cloud point extraction and aqueous two-phase systems.

The variability in aflatoxin B1 concentration among peanut subsamples of 4 sample preparation procedures was evaluated. For each procedure, 18 samples were prepared involving dry comminuting/homogenization or dry roughly comminuting followed by the preparation of a aqueous slurry. Ten analytical samples were withdrawn from each sample/procedure and analyzed by thin layer chromatography. The coefficient of variation (CV%) among each set of 10 analytical samples was assumed to be associated with the sample preparation procedure. The procedure that made use of a subsample mill and preparation of a subsample slurry, showed lower variability (CV%) among the analytical subsamples.

The phytochemical investigation of a sample of propolis from Passa Quatro-Minas Gerais State, Brazil, where nine constituents were isolated: a mixture of α- and β-amyrin, lupeol, a mixture of flavonols ramnocitrin and eupalitin, acacetin, 3-prenyl-4-hydroxycinnamic acid, 3,5-diprenyl-4-hydroxycinnamic acid the new compound, the (E)-3-[4-(3-phenylpropanoiloxy)]-3,5-diprenil-cinnamic acid. The structures of the isolated compounds were characterized by 1D- and 2D-NMR experiments, MS and IR spectrometry, and comparison with data described in the literature.

The determination of pesticide residues in water samples by Liquid Chromatography require sample preparation for extraction and enrichment of the analytes with the minimization of interferences to achieve adequate detection limits. The Solid Phase Extraction (SPE), Solid Phase Microextraction (SPME), Stir Bar Sorptive Extraction (SBSE) and Dispersive Liquid-Liquid Microextraction (DLLME) techniques have been widely used for extraction of pesticides in water. In this review, the principles of these sample preparation techniques associated with the analysis by Liquid Chromatography with Diode Array Detection (LC-DAD) or Mass Spectrometry (LC-MS) are described and an overview of several applications were presented and discussed.

A method based on pyrohydrolysis was proposed for cement sample preparation and further chloride determination by spectrophotometry using flow injection analysis. Analytical parameters were evaluated and, under the selected conditions, the calibration curve was linear in the range of 0.2 to 10.0 µg mL-1 with r2 = 0.998. The limit of detection was5 µg g-1 of chloride and the relative standard deviation was less than 7%. The proposed pyrohydrolysis method is relatively simple and can be used for sample preparation for further spectrophotometric determination of low concentrations of chloride in cement.

This paper describes the development of methods in micro-scale for the determination of K, Mg, Na and Zn in meat by atomic spectrometry techniques. The limits of detection (LOD) for K and Na by microdigestion were 0.18 and 0.20 mg g-1, respectively whereas LOD for Mg and Zn by microsolubilization with TMAH were 2.40 and 18.4 µg g-1, respectively. The RSD values were lower than 6.0% and the CRMs analyzed showed values with 95% agreement. The proposed methods are simple, fast and use small amounts of sample (around 10 mg) yet do not require special equipment for sample preparation.

Para o controle integrado de doenças, o tratamento químico das sementes de milho (Zea mays) torna-se indispensável, sendo a sua eficácia e influenciada por vários fatores, entre eles o tamanho das sementes. O objetivo do trabalho foi avaliar a relação do tamanho da semente e doses do fungicida thiabendazole no controle de Stenocarpella maydis, considerando-se diferentes tamanhos de sementes e doses do fungicida. As sementes classificadas nas peneiras 19, 22 e 24 foram inoculadas com S. maydis e tratadas com o fungicida thiabendazole em três doses, sendo avaliadas a incidência do patógeno e desempenho das sementes por meio de testes de qualidade fisiológica. Constatou-se com o tratamento fungicida, aumento na germinação, maior peso de plântulas e redução de tombamento provocado por S. maydis, havendo comportamento diferenciado do tratamento químico em relação ao tamanho de sementes e doses do produto.

Eletrocatalisadores de Pt/C são preparados por diferentes métodos para a reação de redução de oxigênio em célula a combustível alimentada com H2/O2. Este trabalho mostra a caracterização física e eletroquímica mediante DRX, MET, VC e CP dos catalisadores de Pt/C preparados por diferentes métodos. Os resultados mostraram que a atividade catalítica está correlacionada com a morfologia e o diâmetro médio das partículas de Pt suportadas em carbono de alta área superficial.

Os cultivos de alface e couve-chinesa podem ter a produção reduzida devido à ocorrência da podridão-mole, causada por Pectobacterium carotovorum subsp. carotovorum. O objetivo deste estudo foi determinar os tamanhos ideais das amostras para quantificação da incidência dessa doença em levantamentos no campo. Foram realizadas amostragens da incidência da podridão-mole em oito áreas de plantio de alface e cinco de couve-chinesa, situadas nos principais municípios produtores do Estado de Pernambuco. Considerando os resultados obtidos e um erro aceitável de 20%, em futuros levantamentos da incidência da podridão-mole em alface recomenda-se a amostragem de 32 parcelas de 4,5 m²/ha e 20 plantas/parcela, enquanto em couve-chinesa a amostragem de 22 parcelas de 10,5 m²/ha e 20 plantas/parcela. Para ambas as culturas não houve correlação significativa (P=0,05) entre os níveis de incidência da doença e os tamanhos das amostras.

Nos trabalhos em comunidades florestais, tradicionalmente são estudadas apenas a composição e a estrutura do componente arbóreo, relegando o estrato herbáceo-arbustivo ao esquecimento ou ao segundo plano. Os objetivos deste trabalho foram descrever a estrutura fitossociológica desse estrato para entender suas relações sinecológicas e, por fim, estudar a distribuição dos indivíduos pelas classes de tamanho para inferir sobre fatores e processos determinantes da organização florestal da Mata da Silvicultura. Para o estudo da fitossociologia da área foram utilizados os parâmetros de abundância obtidos a partir de 100 m² de amostra subdividida em parcelas de 1 m². A estrutura fitossociológica horizontal considerou todos os indivíduos com CAP menor que 10 cm ou com altura maior que 20 cm. Os aspectos dinâmicos foram avaliados por meio da distribuição de tamanhos individuais expressos pelos diâmetros à altura do solo em cada população amostrada. Foram amostrados 1.193 indivíduos de 109 espécies, pertencentes a 41 famílias botânicas, resultando em um índice de diversidade de Shannon (H') de 3,38 e equabilidade (J') de 0,72, valores considerados altos para a heterogeneidade do estrato herbáceo-arbustivo. As espécies mais importantes (VI) foram Piper lucaeanum, Psychotria conjugens, Olyra micrantha, Psychotria sessilis, Siparuna guianensis, Bambusa tuldoides, Ottonia leptostachya, Aparisthmium cordatum e Psychotria hastisepala. As famílias mais importantes (VI) foram Rubiaceae, Piperaceae, Poaceae, Monimiaceae, Leguminosae (Mimosoideae), Myrtaceae, Euphorbiaceae, Meliaceae, Lauraceae e Flacourtiaceae. Pela análise de distribuição de tamanhos foi levantada a hipótese de existirem dois grupos de espécies, segundo a estratégia que possuem de habitar o estrato herbáceo-arbustivo da Mata da Silvicultura. Um dos grupos seria formado pelas espécies que investem preferencialmente recursos energéticos no sistema caulinar e o outro, pelas espécies que investem recursos energéticos preferencialmente no sistema fotossintético.
