Introduction The progression of human papillomavirus (HPV) infection in the anogenital tract has been associated with the involvement of cells with regulatory properties. Evidence has shown that glucocorticoid-induced tumor necrosis factor receptor (GITR) is an important surface molecule for the characterization of these cells and proposes that GITR ligand may constitute a rational treatment for many cancer types. We aimed to detect the presence of GITR and CD25 in cervical stroma cells with and without pathological changes or HPV infection to better understand the immune response in the infected tissue microenvironment. Methods We subjected 49 paraffin-embedded cervical tissue samples to HPV DNA detection and histopathological analysis, and subsequently immunohistochemistry to detect GITR and CD25 in lymphocytes. Results We observed that 76.9% of all samples with high GITR expression were HPV-positive regardless of histopathological findings. High GITR expression (77.8%) was predominant in samples with ≥1,000 RLU/PCB. Of the HPV-positive samples negative for intraepithelial lesion and malignancy, 62.5% had high GITR expression. High GITR expression was observed in both carcinoma and high-grade squamous intraepithelial lesion (HSIL) samples (p = 0.16). CD25 was present in great quantities in all samples. Conclusions The predominance of high GITR expression in samples with high viral load that were classified as HSIL and carcinoma suggests that GITR+ cells can exhibit regulatory properties and may contribute to the progression of HPV-induced cervical neoplasia, emphasizing the importance of GITR as a potential target for immune therapy of cervical cancer and as a disease evolution biomarker.

RESUMO - Objetivo: Caraterizar e analisar o acesso dos utentes inscritos no Agrupamento de Centros de Saúde Lisboa Central ao serviço de urgência do hospital de referência (Hospital de S. José). Metodologia: O presente estudo é do tipo observacional, descritivo, transversal e exploratório. A análise da informação incidiu sobre a base de dados cedida pelo CHLC que continha os registos de todas as admissões do ano de 2014 referentes ao Serviço de Urgência Polivalente do CHLC, foram analisados 81928 episódios e 15042 episódios referentes a utentes frequentadores. Resultados: A proximidade ao SU é um fator que potencia a procura de cuidados hospitalares urgentes, no entanto, o facto de o utente não possuir médico de família não se encontra relacionado com a procura do serviço de urgência. Metades das admissões são consideradas “pouco urgentes” (pulseira verde), não existindo uma variação significativa entre as unidades funcionais e o tipo de modelo das mesmas (UCSP ou USF) no que respeita à procura das urgências por parte dos seus utentes. Conclui-se ainda que o horário de funcionamento dos CSP encontra-se de acordo com as necessidades dos utentes considerando que a maior procura de cuidados urgentes ocorre durante o horário de funcionamento dos centros de saúde. Conclusões: A procura de cuidados hospitalares urgentes por parte dos utentes do ACES Lisboa Central é significativa e maioritariamente injustificada do ponto de vista clínico, pelo que revela-se necessário repensar nas estratégias ao nível dos CSP para responder às efetivas necessidades em saúde.

OBJECTIVE: To analyze the correlation of risk factors to the occurrence of urinary tract infection in full-term newborn infants. PATIENTS AND METHODS: Retrospective study (1997) including full-term infants having a positive urine culture by bag specimen. Urine collection was based on: fever, weight loss > 10% of birth weight, nonspecific symptoms (feeding intolerance, failure to thrive, hypoactivity, debilitate suction, irritability), or renal and urinary tract malformations. In these cases, another urine culture by suprapubic bladder aspiration was collected to confirm the diagnosis. To compare and validate the risk factors in each group, the selected cases were divided into two groups: Group I - positive urine culture by bag specimen collection and negative urine culture by suprapubic aspiration, and Group II - positive urine culture by bag specimen collection and positive urine culture by suprapubic aspiration . RESULTS: Sixty one infants were studied, Group I, n = 42 (68.9%) and Group II, n = 19 (31.1%). The selected risk factors (associated infectious diseases, use of broad-spectrum antibiotics, renal and urinary tract malformations, mechanical ventilation, parenteral nutrition and intravascular catheter) were more frequent in Group II (p<0.05). Through relative risk analysis, risk factors were, in decreasing importance: parenteral nutrition, intravascular catheter, associated infectious diseases, use of broad-spectrum antibiotics, mechanical ventilation, and renal and urinary tract malformations. CONCLUSION: The results showed that parenteral nutrition, intravascular catheter, and associated infectious diseases contributed to increase the frequency of neonatal urinary tract infection, and in the presence of more than one risk factor, the occurrence of urinary tract infection rose up to 11 times.

OBJECTIVES: To determine the presence of immunoglobulin E-rheumatoid factor in patients with juvenile rheumatoid arthritis and to correlate it with clinical and laboratory parameters. METHODS: A multicenter prospective study was carried out from January 1993 to January 1999 with the enrollment of 3 centers of pediatric rheumatology. Ninety-one children with juvenile rheumatoid arthritis diagnosed according to the American College of Rheumatology criteria were studied: 38 (42%) with systemic, 28 (31%) with pauciarticular, and 25 (27%) with polyarticular onset. Ages ranged from 2.1 years to 22.6 years (mean 10.5 ± 4.7), with 59 (65%) girls. The control group consisted of 45 healthy children. The detection of immunoglobulin E-rheumatoid factor was carried out utilizing an enzyme-linked immunosorbent assay. Associations of immunoglobulin E-rheumatoid factor with immunoglobulin M-rheumatoid factor (latex agglutination test), total serum immunoglobulin E, erythrocyte sedimentation rate, antinuclear antibody, and functional and radiological classes III or IV were analyzed. RESULTS: Positive immunoglobulin E-rheumatoid factor was found in 15 (16.5%) of the 91 children with juvenile rheumatoid arthritis: 7 (18.5%) with systemic, 5 (18%) with pauciarticular, and 3 (12%) with polyarticular onset. A significant correlation was observed between immunoglobulin E-rheumatoid factor and total serum immunoglobulin E in the juvenile rheumatoid arthritis patients. No correlation was found between immunoglobulin E-rheumatoid factor and positive latex agglutination slide test, erythrocyte sedimentation rate, antinuclear antibody, or the functional and radiological classes III or IV in any disease onset group. In 4 out of 45 control children (8.9%), immunoglobulin E-rheumatoid factor was positive but with no correlation with total serum immunoglobulin E levels. CONCLUSIONS: Immunoglobulin E-rheumatoid factor could be detected in 16.5% of juvenile rheumatoid arthritis patients, particularly in those with high levels of total serum immunoglobulin E, and immunoglobulin E-rheumatoid factor appears not to be associated with disease activity or severity.

PURPOSE: The differential diagnosis between benign and malignant adrenal cortical tumors circumscribed to the gland is controversial. One hundred and seven patients with adrenal cortex tumors (excluding those with primary hyperaldosteronism) were studied to assess the 5-year survival rate of adults, children, patients stratified by pathological stage, and patients stratified according to Weiss's score of <3 or >3. METHODS: The patients were evaluated both clinically and biochemically. One hundred and five patients underwent surgery and were classified pathologically as stages I, II, III, or IV. The tumors were weighed, measured, and classified according to Weiss's criteria and divided into 2 groups: <3 and >3. RESULTS: After 5 years, the survival rate was 77.5% for the whole group, 74.61% for the adults, 84.3% for the children, 100% for stage I, 83.9% for stage II, 33% for stage III, and 11.7% for stage IV groups. Additionally, after 5 years, 100% of the patients with tumors with Weiss's score <3 were alive compared to 61.65% of those with Weiss's score >3. The average weights of the tumors of score <3 and >3 were 23.38 g ± 41.36 g and 376.3 ± 538.76 g, respectively, which is a statistically significant difference. The average sizes of tumors of Weiss's score <3 and >3 were 3.67 ± 2.2 cm and 9.64 ± 5.8 cm, respectively, which is also a statistically significant difference. CONCLUSIONS: Weiss's score may be a good prognostic factor for tumors of the adrenal cortex. Additionally, there was a statistically significant difference between the average weight and size of tumors with benign behavior (Weiss's score <3) and those with malignant behavior (Weiss's score >3).

Measurements of inclusive jet production are performed in pp and Pb+Pb collisions at sNN−−−√=2.76 TeV with the ATLAS detector at the LHC, corresponding to integrated luminosities of 4.0 pb−1 and 0.14 nb−1 , respectively. The jets are identified with the anti-kt algorithm with R=0.4, and the spectra are measured over the kinematic range of jet transverse momentum 32factor, RAA, is evaluated and jets are found to be suppressed by approximately a factor of two in central collisions compared to pp collisions. The RAA shows a slight increase with pT and no significant variation with rapidity.

Tau-mediated neurodegeneration is a central event in Alzheimer's disease (AD) and other tauopathies. Consistent with suggestions that lifetime stress may be a clinically-relevant precipitant of AD pathology, we previously showed that stress triggers tau hyperphosphorylation and accumulation; however, little is known about the etiopathogenic interaction of chronic stress with other AD risk factors, such as sex and aging. This study focused on how these various factors converge on the cellular mechanisms underlying tau aggregation in the hippocampus of chronically stressed male and female (middle-aged and old) mice expressing the most commonly found disease-associated Tau mutation in humans, P301L-Tau. We report that environmental stress triggers memory impairments in female, but not male, P301L-Tau transgenic mice. Furthermore, stress elevates levels of caspase-3-truncated tau and insoluble tau aggregates exclusively in the female hippocampus while it also alters the expression of the molecular chaperones Hsp90, Hsp70, and Hsp105, thus favoring accumulation of tau aggregates. Our findings provide new insights into the molecular mechanisms through which clinically-relevant precipitating factors contribute to the pathophysiology of AD. Our data point to the exquisite sensitivity of the female hippocampus to stress-triggered tau pathology.

Here we focus on factor analysis from a best practices point of view, by investigating the factor structure of neuropsychological tests and using the results obtained to illustrate on choosing a reasonable solution. The sample (n=1051 individuals) was randomly divided into two groups: one for exploratory factor analysis (EFA) and principal component analysis (PCA), to investigate the number of factors underlying the neurocognitive variables; the second to test the "best fit" model via confirmatory factor analysis (CFA). For the exploratory step, three extraction (maximum likelihood, principal axis factoring and principal components) and two rotation (orthogonal and oblique) methods were used. The analysis methodology allowed exploring how different cognitive/psychological tests correlated/discriminated between dimensions, indicating that to capture latent structures in similar sample sizes and measures, with approximately normal data distribution, reflective models with oblimin rotation might prove the most adequate.

Una de las principales causas de pérdida reproductiva en condiciones normales, es la mortalidad embrionaria basal. Por lo tanto, estaría ampliamente justificado el diseño de experiencias que nos permitan encontrar las causas de las pérdidas y detectar la preñez tan precozmente, como fuere posible. Las causas de las pérdidas embrionarias en muchos casos se relacionan con fallas en las señales del embrión dirigidas a la madre con el fin de establecer inmunotolerancia, fenómeno imprescindible para la sobrevida del embrión porque modifica la respuesta inmune de modo favorable para éste. La inmunomodulación es ejercida por sustancias de naturaleza proteica detectadas durante la gestación. Entre estas se encuentra el Factor Precoz de Preñez (EPF). Durante la preñez, el EPF tiene dos fuentes de producción: una materna y otra embrionaria. El EPF materno, también llamado temprano, se produce durante los períodos pre y periimplantacional y persiste hasta el último tercio de la preñez. Hasta el presente, la única técnica desarrollada para la detección del EPF, es un ensayo in vitro propuesto por Morton que mide de manera indirecta la actividad del factor. Dicha técnica se basa en la capacidad que tiene el EPF de potenciar la acción de un suero antilinfocitario (SAL), para inhibir la formación de rosetas entre linfocitos T y eritrocitos heterólogos en presencia de complemento. Esta técnica es efectiva pero muy compleja para realizarse rutinariamente en el laboratorio. De todo lo expuesto surge claramente la necesidad del aislamiento y purificación del EPF, lo que permitiría desarrollar metodologías inmunológicas aplicables para el diagnóstico de rutina de detección de preñez en cerdos y un notable adelanto en el estudio de alguno de los factores que afectan los sucesos de concepción y el curso de preñez, momentos claves en la fisiología reproductiva. Objetivos Generales - Desarrollar una técnica de detección rápida y precisa del Factor Precoz de Preñez (EPF) en cerdos. Objetivos Específicos - Aislar, purificar y caracterizar al Factor Precoz de Preñez (EPF) a partir de suero y placenta de cerdas gestantes. - Obtener y valorar un antisuero policlonal anti-EPF en conejos a partir de la fracción purificada con mayor índice de actividad EPF.

Numerosos estudios sugieren que los efectos conductuales y neuroquímicos del etanol y las benzodiazepinas (BDZs) están estrechamente relacionados. La administración aguda de BDZs y etanol potencia el efecto del GABA, sugiriendo que ambas drogas producirían efectos depresores del sistema nervioso central por incremento de la neurotransmisión GABAérgica. La administración prolongada de etanol y BDZS produce tolerancia a alguno de los efectos agudos tanto de la droga primaria, como de un agente relacionado (tolerancia cruzada). Este hecho se ha observado tanto en humanos como en animales. La interrupción del tratamiento crónico con dichas drogas provoca la aparición del síndrome de abstinencia. (...) Considerando que la ansiedad es un factor crítico tanto en la abstinencia a BDZs como en la génesis y mantenimiento del alcoholismo y que la abstinencia a diazepam podría provocar alteraciones a largo plazo en la capacidad de adaptación al estrés, se podría hipotetizar que esos efectos podrían estar implicados en la recurrencia al consumo de BDZs, etanol u otras drogas de abuso, luego de la interrupción de un tratamiento crónico. De lo anteriormente expuesto se deduce que fármacos que normalicen potenciales alteraciones en los mecanismos adaptativos frente al estrés podrían ser de utilidad para revertir las alteraciones observadas durante la expresión del sindrome de abstinencia inducido por etanol y BDZs. En relación a este tema, hemos demostrado recientemente que carbamazepina (CBZ), droga efectiva para el tratamiento del sindrome de abstinencia inducido por BDZs y etanol, revierte las alteraciones conductuales y neuroquímicas descriptas en animales con abstinencia a BDZs. Estos resultados sugieren que el tratamiento con dicho fármaco podría ser de utilidad terapéutica para la normalización de las alteraciones inducidas por la interrupción del consumo. Objetivos El objetivo general del presente proyecto es investigar una posible facilitación del desarrollo de dependencia al etanol en animales previamente sometidos a la abstinencia de benzodiazepinas. Este objetivo se extiende al estudio de nuevas alternativas terapéuticas que tengan capacidad potencial para revertir las alteraciones producidas por la abstinencia a etanol.
