A área septal medial (ASM), situada no prosencéfalo, está envolvida na regulação cardiovascular e no controle do balanço hidroeletrolítico. Esta área é rica em receptores colinérgicos e a ativação dos mesmos induz ingestão de água, natriurese e antidiurese. A ASM também envia projeções aos núcleos paraventricular (NPV) supra-óptico (NSO), os quais contêm os neurônios que secretam vasopressina e ocitocina. Existem evidências experimentais demonstrando que as espécies reativas de oxigênio podem participar do controle de respostas fisiológicas. Resultados recentes de nosso laboratório demonstraram que uma espécie reativa de oxigênio, o peróxido de hidrogênio (H2O2), injetado no ventrículo lateral (VL) reduz a ingestão de água e a resposta pressora induzida por ANG II e carbacol (agonista colinérgico) também injetados no VL. Por isso, o presente estudo teve como objetivo estudar os efeitos da injeção de H2O2 na ASM sobre a ingestão de água, sobre a excreção renal de água e eletrólitos e sobre a expressão da proteína c-Fos no NSO produzidos pela injeção de carbacol também na ASM. Para realizar este trabalho, foram utilizados ratos com cânulas de aço inoxidável implantadas na ASM. A ingestão de água e a excreção renal de água e eletrólitos foram estudadas em ratos que receberam injeções de H2O2 (5 mol/0,5 μl) ou PBS (veículo, 0,5 l) na ASM e, após um minuto, injeção de carbacol (4 nmol/0,5 l) ou salina (NaCl 0,15 M / 0,5 l) também na ASM. A ingestão de água induzida pelo carbacol, através da estimulação colinérgica, foi menor nos ratos que receberam a injeção prévia de peróxido de hidrogênio (8 ± 2,0 ml / 1 h, p<0,05) comparado àqueles que receberam veículo, também na ASM (16,6 ± 1,9 ml / 1 h, p<0,05). Apocynin has been used as an efficient inhibitor of the multi-enzymatic complex NADPH oxidase in many experimental models involving phagocytic and nonphagocytic cells. The mechanism of inhibition has been linked with the previous activation of apocynin through the action of cellular peroxidases leading to the formation of a dimeric oxidation product, diapocynin. In this study we compared apocynin with pure diapocynin regarding their effects as scavenger of hydrogen peroxide and hypochlorous generated by glucose/glucose oxidase and myeloperoxidase respectively, and as inhibitors of the production of hydrogen peroxide and hypochlorous acid by activated neutrophils. The production of hydrogen peroxide was measured by the oxidation of the fluorescent substance Amplex Red and the production of hypochlorous acid by was measured as taurine-chloramine derivative using the chromogenic substrate 3,3’,5,5’- tetramethylbenzidine (TMB). Neutrophils (1 x106 cells/mL) were pre-incubated in PBS buffer supplemented with 1 mM calcium chloride, 0.5 mM magnesium chloride, 1 mg/mL glucose and 5 mM taurine in the presence or absence of inhibitors. The reactions were triggered by adding the soluble stimulus Forbol Miristate Acetate PMA or zymosan and incubated by additional 30 minutes. We found that pure diapocynin was not better than apocynin regarding its scavenger and inhibitory properties. These results suggest that the formation of diapocynin is not essential for the action of apocynin as inhibitor of NADPH oxidase activation

Este trabalho apresenta o estudo de dois sistemas de liberação prolongada, microemulsão e lipossomas, contendo peptídeo regulatório de fatores de crescimento, “osteogenic growth peptide” OGP, para aplicação em regeneração óssea. A base de adsorção para estes sistemas de liberação foi a celulose bacteriana (CB) produzida pela bactéria Gluconacetobacter xylinus. Foi escolhida devido às suas propriedades físicas e químicas, tais como como alta resistência à corrosão química, bio absorção, biocompatibilidade, porosidade e ainda boa resistência mecânica, o que a torna um biopolímero com grande potencial a ser explorado pela ciência biomédica. Estudos in vitro foram realizados para avaliar o perfil de liberação do peptídeo dos diferentes sistemas de liberação prolongada. O peptídeo OGP foi sintetizado pelo método da fase sólida (estratégia SPFS Fmoc); foi purificado e caracterizado por HPLC, espectrometria de massas e análise de aminoácidos e, em seguida, marcados com 5,6 carboxifluoresceína (CF) para análise por espectroscopia de fluorescência. O peptídeo marcado foi incorporado aos sistemas de liberação no momento do respectivo preparo, foram adsorvidos na CB por um período de 72h, seguido de sua liberação prolongada em sistema fechado de fluxo constante contendo tampão PBS pH 7,4, por um período de 24h. Após a análise da liberação, observou se que o sistema que obteve melhor resultado foi a microemulsão, sendo sua liberação prolongada nas primeiras 6,5 h, liberando 21,5% do valor teórico de peptídeo incorporado, seguido de uma liberação constante a partir desse período. Dessa forma, tem se que a microemulsão pode ser um sistema promissor para liberação prolongada do OGP em processos de regeneração óssea

This article is to discuss the evaluative perspectives from which it is possible and convenient evaluate the Participatory Budgets (PBs) and the participatory institutions (PIs) that are being experienced in Brazil since the last two decades of the last century. It is proposed an integrated perspective of evaluation of PBs and PIs, in order to avoid partial and biased conclusions, especially those which are based solely or primarily on quantitative methodologies. This discussion is necessary since, in the past five years, the criticism of these experiments has been increasing. Through critical and creative reflection, in essay form, and based on the recent literature on the topic, it is concluded by the need to adopt, in the empirical evaluation, an integrated approach of evaluation of the PBs and the PIs, such as it is proposed in this article. It is conclude also which is fundamental for the theoretical advancement on the subject dedicate special attention to the desirability or not of adopting the criterion of popular participation in government decisions under representative democracy, since it is refuted by public choice and defended by neo-republicanism.

This study analyzes the clot stabilization on root surfaces of teeth impregnated with cotinine and nicotine and the influence of the scaling in the adhesion of blood components, observing the influence of new exposition to nicotine and/or cotinine after scaling. Fifteen human teeth extracted due to periodontal disease of non-smokers patients were selected and manually scaled. Four dentin blocks were obtained from each tooth (n = 60). Samples received blood application or reimpregnation with nicotine and/or cotinine, depending on the groups. Group 1: PBS immersion + root scaling + blood; group 2: nicotine + root scaling + blood; group 3: nicotine + root scaling + nicotine reapplication + blood; group 4: cotinine + root scaling + blood; group 5: cotinine + root scaling + cotinine reapplication+ blood; group 6: nicotine and cotinine + root scaling + nicotine and cotinine + blood. Samples were kept in 2 ml of each substance for 24 hours. Each group received a blood drop and was analyzed by SEM. The higher amount of blood components was present in teeth exposed to cotinine and the groups submitted to scaling and blood application in comparison with groups that received reapplication of toxic substances after scaling. The greater toxic effect on root dentin surface was after the exposure to nicotine and cotinine. Results suggest that periodontal healing may be delayed in smokers due to the direct inhibition of clot stabilization on the root surface when nicotine and cotinine are present concomitantly.

The transgenic application of green fluorescent protein (GFP) as fetal cell marker on cattle cloned placenta could provide an exclusive model for studying the morphologic and immunologic maternal-fetal interactions, providing information about its mapping, distinguishing the fetal from maternal cells. This model will have direct application, mainly because these animals present problems during its development. With this model's support, we intend to verify the substances transport between mother and fetus during endocytosis, through the immunolocalization of protein named caveolae. For these, we used 06 cloned bovine and 30 cattle samples of artificial insemination (AI) with 90 days of pregnancy, which had been their development interrupted by humanitarian slaughter of the recipient and recovery of the pregnant uterus. We collected the placentome and the chorion. A part of the samples was cut and fixed, by immersion, on a solution containing 4% of parafomaldehyde or 10% of formaldehyde on a sodium phosphate buffer (PBS), at 0,1 M pH 7.4, Zamboni solution (4% of paraformaldehyde, 15% of picric acid, on sodium phosphate buffer 0,1 M pH 7.4), metacarn (60% of metanol, 30% of chloroform, and 10% glacial acetic acid), for morphologic and immunohistochemistry verification for caveolinas proteins -1 and -2 (CAV -1 and CAV-2). The caveolins -1 were found in fetal and maternal villi, but its strongest staining was observed in the endometrial stroma. The caveolins -2 had positive staining in trophoblast and chorioallantoic membrane, and specifically in giant trophoblastic binucleated cell. Therefore the results were compared between cloned cattle and from AI or natural mating, for assisting on detection of the reason of many placental alterations, embryonic losses, spontaneous abortion, post-natal mortality and large offspring syndrome on laboratory-manipulated animals. The result suggests that the proteins caveolins -1 and -2 (CAV-1 and CAV-2) are part of the caveolae composition and important structures related to the molecule transfer to the fetus, nourish it through endocytosis and pinocytosis.
