This article is a review of our work related to Raman studies of single layer and bilayer graphenes as a function Fermi level shift achieved by electrochemically top gating a field effect transistor. Combining the transport and in situ Raman studies of the field effect devices, a quantitative understanding is obtained of the phonon renormalization due to doping of graphene. Results are discussed in the light of time dependent perturbation theory, with electron phonon coupling parameter as an input from the density functional theory. It is seen that phonons near and Gamma and K points of the Brillouin zone are renormalized very differently by doping. Further, Gamma-phonon renormalization is different in bilayer graphene as compared to single layer, originating from their different electronic band structures near the zone boundary K-point. Thus Raman spectroscopy is not only a powerful probe to characterize the number of layers and their quality in a graphene sample, but also to quantitatively evaluate electron phonon coupling required to understand the performance of graphene devices.

Redox imbalance generates multiple cellular damages leading to oxidative stress-mediated pathological conditions such as neurodegenerative diseases and cancer progression. Therefore, maintenance of reactive oxygen species (ROS) homeostasis is most important that involves well-defined antioxidant machinery. In the present study, we have identified for the first time a component of mammalian protein translocation machinery Magmas to perform a critical ROS regulatory function. Magmas overexpression has been reported in highly metabolically active tissues and cancer cells that are prone to oxidative damage. We found that Magmas regulates cellular ROS levels by controlling its production as well as scavenging. Magmas promotes cellular tolerance toward oxidative stress by enhancing antioxidant enzyme activity, thus preventing induction of apoptosis and damage to cellular components. Magmas enhances the activity of electron transport chain (ETC) complexes, causing reduced ROS production. Our results suggest that J-like domain of Magmas is essential for maintenance of redox balance. The function of Magmas as a ROS sensor was found to be independent of its role in protein import. The unique ROS modulatory role of Magmas is highlighted by its ability to increase cell tolerance to oxidative stress even in yeast model organism. The cytoprotective capability of Magmas against oxidative damage makes it an important candidate for future investigation in therapeutics of oxidative stress-related diseases.

DNA damage is extremely detrimental to the cell and must be repaired to protect the genome. DNA is capable of conducting charge through the overlapping π-orbitals of stacked bases; this phenomenon is extremely sensitive to the integrity of the π-stack, as perturbations attenuate DNA charge transport (CT). Based on the E. coli base excision repair (BER) proteins EndoIII and MutY, it has recently been proposed that redox-active proteins containing metal clusters can utilize DNA CT to signal one another to locate sites of DNA damage.

To expand our repertoire of proteins that utilize DNA-mediated signaling, we measured the DNA-bound redox potential of the nucleotide excision repair (NER) helicase XPD from Sulfolobus acidocaldarius. A midpoint potential of 82 mV versus NHE was observed, resembling that of the previously reported BER proteins. The redox signal increases in intensity with ATP hydrolysis in only the WT protein and mutants that maintain ATPase activity and not for ATPase-deficient mutants. The signal increase correlates directly with ATP activity, suggesting that DNA-mediated signaling may play a general role in protein signaling. Several mutations in human XPD that lead to XP-related diseases have been identified; using SaXPD, we explored how these mutations, which are conserved in the thermophile, affect protein electrochemistry.

To further understand the electrochemical signaling of XPD, we studied the yeast S. cerevisiae Rad3 protein. ScRad3 mutants were incubated on a DNA-modified electrode and exhibited a similar redox potential to SaXPD. We developed a haploid strain of S. cerevisiae that allowed for easy manipulation of Rad3. In a survival assay, the ATPase- and helicase-deficient mutants show little survival, while the two disease-related mutants exhibit survival similar to WT. When both a WT and G47R (ATPase/helicase deficient) strain were challenged with different DNA damaging agents, both exhibited comparable survival in the presence of hydroxyurea, while with methyl methanesulfonate and camptothecin, the G47R strain exhibits a significant change in growth, suggesting that Rad3 is involved in repairing damage beyond traditional NER substrates. Together, these data expand our understanding of redox-active proteins at the interface of DNA repair.

All major geochemical cycles on the Earth’s surface are mediated by microorganisms. Our understanding of how these microbes have interacted with their environments (and vice versa) throughout Earth's history, and how they will respond to changes in the future, is primarily based on studying their activity in different environments today. The overarching questions that motivate the research presented in the two parts of this thesis -- how do microorganisms shape their environment (and vice versa)? and how can we best study microbial activity in situ? -- have arisen from the ultimate goal of being able to predict microbial activity in response to changes within their environments both past and future.

Part one focuses on work related to microbial processes in iron-rich Lake Matano and, more broadly, microbial interactions with the biogeochemical cycling of iron. Primarily, we find that the chelation of ferrous iron by organic ligands can affect the role of iron in anoxic environmental systems, enabling photomixotrophic growth of anoxygenic microorganisms with ferrous iron, as well as catalyzing the oxidation of ferrous iron by denitrification intermediates. These results imply that the ability to grow photomixotrophically on ferrous iron might be more widespread than previously assumed, and that the co-occurrence of chemical and biological processes involved in the coupled biogeochemical cycling of iron and nitrogen likely dominate organic-rich environmental systems.

Part two switches focus to in situ measurements of growth activity and comprises work related to microbial processes in the Cystic Fibrosis lung, and more broadly, the physiology of slow growth. We introduce stable isotope labeling of microbial membrane fatty acids and whole cells with heavy water as a new technique to measure microbial activity in a wide range of environments, demonstrate its application in continuous culture in the laboratory at the population and single cell level, and apply the tool to measure the in situ activity of the opportunistic pathogen Staphylococcus aureus within the environment of expectorated mucus from cystic fibrosis patients. We find that the average in situ growth rates of S. aureus fall into a range of generation times between ~12 hours and ~4 days, with substantial heterogeneity at the single-cell level. These data illustrate the use of heavy water as a universal environmental tracer for microbial activity, and highlight the crucial importance of studying the physiology of slow growth in representative laboratory systems in order to understand the role of these microorganisms in their native environments.

Background: Excessive apoptosis induces unwanted cell death and promotes pathological conditions. Drug discovery efforts aimed at decreasing apoptotic damage initially targeted the inhibition of effector caspases. Although such inhibitors were effective, safety problems led to slow pharmacological development. Therefore, apoptosis inhibition is still considered an unmet medical need. Methodology and Principal Findings: The interaction between Apaf-1 and the inhibitors was confirmed by NMR. Target specificity was evaluated in cellular models by siRNa based approaches. Cell recovery was confirmed by MTT, clonogenicity and flow cytometry assays. The efficiency of the compounds as antiapoptotic agents was tested in cellular and in vivo models of protection upon cisplatin induced ototoxicity in a zebrafish model and from hypoxia and reperfusion kidney damage in a rat model of hot ischemia. Conclusions: Apaf-1 inhibitors decreased Cytc release and apoptosome-mediated activation of procaspase-9 preventing cell and tissue damage in ex vivo experiments and in vivo animal models of apoptotic damage. Our results provide evidence that Apaf-1 pharmacological inhibition has therapeutic potential for the treatment of apoptosis-related diseases.

A chuva de sementes é a dinâmica de dispersão de uma floresta, representando o que chega ao solo por meio dos agentes dispersores. Na comunidade de plantas, as síndromes de dispersão predominantes permitem entender os estádios sucessionais, estrutura e o nível de conservação da vegetação. No Brasil, a jaqueira, Artocarpus heterophyllus, é considerada uma espécie exótica invasora, que foi introduzida no período colonial. Essa invasão biológica pode comprometer a dispersão dos diásporos de espécies nativas pela chuva de sementes, já que a dispersão é influenciada pela distribuição espacial das plantas e pelas barreiras presentes no ambiente. O trabalho teve por objetivo verificar se a jaqueira, por ser uma espécie exótica invasora, influência na composição e abundância da chuva de sementes. O estudo foi realizado em floresta Atlântica, localizada no Parque Estadual da Ilha Grande, RJ. Foram utilizados para a coleta da chuva de semente 16 coletores de 1m de diâmetro, em áreas com (C) e sem (S) jaqueira, totalizando 32 coletores verificados mensalmente durante um ano de coleta. As análises estatísticas realizadas foram ANOVA, Regressão e NMDS, com o objetivo de verificar se a presença da jaqueira estaria influenciando na chuva de sementes quanto a riqueza, composição e abundância. Foram coletadas 320 amostras e contabilizadas 166.376 sementes, sendo encontrados 84 morfoespécies, 29 espécies em 25 famílias e 55 morfoespécies não identificadas. As áreas sem (S) jaqueira apresentaram maior riqueza, menor abundância e maior diversidade. Nas áreas com jaqueira (C) a abundância de sementes nativas apresentou uma relação negativa com a abundância dessa exótica nas parcelas. A composição e a abundância dos diásporos não tiveram uma separação nítida entre as áreas com e sem jaqueiras, como mostrado no NMDS. Porém, quando analisado o primeiro eixo do NMDS com a abundância de jaqueiras, a ordenação das grades mostrou ter uma relação com a abundância de jaqueiras. As principais síndromes de dispersão observadas no trabalho, referente às espécies que foram identificadas, foram à anemocoria e a zoocoria, sendo esta ultima síndrome a mais abundante entre as áreas. Os resultados mostraram que a presença das jaqueiras afeta, negativamente a chuva de sementes no Parque Estadual da Ilha Grande. Uma menor riqueza e abundância de sementes nativas alcançaram o solo nas áreas com jaqueira (C) podendo afetar a sucessão e a dinâmica da floresta.

A presente dissertação se insere no campo de estudo sobre o trabalho informal, tendo como universo empírico de investigação a experiência de trabalho dos trabalhadores ambulantes integrantes do Programa Renda Alternativa, em Rio das Ostras/RJ. Portanto, a pesquisa foi desdobrada em três dimensões: 1) levantamento e estudo bibliográfico; 2) levantamento de dados sobre a formação sócio-histórica da cidade de Rio das Ostras; e, 3) trabalho de campo com ambulantes. Por meio desses dispositivos, a pesquisa buscou compreender os laços entre informalidade do trabalho e dinâmica capitalista, por meio da tradição de estudos das Ciências Sociais nessa área. Seguidamente, a pesquisa se ocupou do estudo de outras investigações sobre trabalho ambulante visando conhecer e ampliar o escopo de análise das peculiaridades desse tipo de trabalho no contexto de reprodução social da chamada superpopulação relativa. Esse universo de questões teóricas, analíticas e empíricas foi confrontado com as especificidades do contexto socioeconômico da cidade de Rio das Ostras, recentemente recomposta como um dos elos da cadeia produtiva do petróleo. Outro eixo da investigação de campo foi conhecer o trabalho, o que foi realizado por meio de entrevistas semi-estruturadas com os trabalhadores ambulantes do município, buscando perceber a relação do trabalhador com o trabalho ao longo de sua vida, as marcas das conjunturas socioeconômicas nas trajetórias e o papel da família e do associativismo na ocupação ambulante. Os achados da pesquisa mostram que o trabalho ambulante acomete gerações de trabalhadores como consequência do desenvolvimento desigual e dependente do capitalismo que limita a oferta de emprego. Somado a isso temos os processos de transformações socioeconômicas das últimas décadas que intensificam as formas de precarização do trabalho potencializando a chamada informalidade do trabalho. Esse processo se intensifica na região em estudo através dos impactos trazidos pelo intenso incremento das indústrias de petróleo e gás que não absorvem a força de trabalho disponível no local e atraída, engendrando uma densa superpopulação relativa para o capital, no município. A experiência de trabalho dos ambulantes que participaram da pesquisa mostra que a mesma é entrecortada por formas laborativas variadas, baixa qualificação profissional, além de dependência dos familiares, de ações públicas do Estado e do associativismo. A expropriação do direito ao emprego empurra os trabalhadores para experiências laborativas inseguras, incertas e sem proteção social. A repetição dessa história entre familiares amplia a percepção da perenidade da expropriação social entre gerações de trabalhadores.

This baseline assessment of Jobos Bay and surrounding marine ecosystems consists of a two part series. The first report (Zitello et al., 2008) described the characteristics of the Bay and its watershed, including modeling work related to nutrients and sediment fluxes, based on existing data. The second portion of this assessment, presented in this document, presents the results of new field studies conducted to fill data gaps identified in previous studies, to provide a more complete characterization of Jobos Bay and the surrounding coral reef ecosystems. Specifically, the objective was to establish baseline values for the distribution of habitats, nutrients, contaminants, fi sh, and benthic communities. This baseline assessment is the first step in evaluating the effectiveness in changes in best management practices in the watershed. This baseline assessment is part of the Conservation Effects Assessment Project (CEAP), which is a multi-agency effort to quantify the environmental benefits of conservation practices used by agricultural producers participating in selected U.S. Department of Agriculture (USDA) conservation programs. Partners in the CEAP Jobos Bay Special Emphasis Watershed (SEW) included USDA’s Agricultural Research Service (ARS) and the Natural Resources Conservation Service (NRCS), National Oceanic and Atmospheric Administration (NOAA) and the Government of Puerto Rico. The project originated from an on-going collaboration between USDA and NOAA on the U.S. Coral Reef Task Force. The Jobos Bay watershed was chosen because the predominant land use is agriculture, including agricultural lands adjacent to the Jobos Bay National Estuarine Research Reserve (JBNERR or Reserve), one of NOAA’s 26 National Estuarine Research Reserves (NERR). This report is organized into six chapters that represent a suite of interrelated studies. Chapter 1 provides a short introduction to Jobos Bay, including the land use and hydrology of the watershed. Chapter 2 is focused on benthic mapping and provides the methods and results of newly created benthic maps for Jobos Bay and the surrounding coral reef ecosystem. Chapter 3 presents the results of new surveys of fish, marine debris, and reef communities of the system. Chapter 4 is focused on the distribution of chemical contaminants in sediments within the Bay and corals outside of the Bay. Chapter 5 focuses on quantifying nutrient and pesticide concentrations in the surface waters at the Reserve’s System-Wide Monitoring Program (SWMP) sites. Chapter 6 is a brief summary discussion that highlights key findings of the entire suite of studies.

Midbrain dopaminergic neurons in the substantia nigra, pars compacta and ventral tegmental area are critically important in many physiological functions. These neurons exhibit firing patterns that include tonic slow pacemaking, irregular firing and bursting, and the amount of dopamine that is present in the synaptic cleft is much increased during bursting. The mechanisms responsible for the switch between these spiking patterns remain unclear. Using both in-vivo recordings combined with microiontophoretic or intraperitoneal drug applications and in-vitro experiments, we have found that M-type channels, which are present in midbrain dopaminergic cells, modulate the firing during bursting without affecting the background low-frequency pacemaker firing. Thus, a selective blocker of these channels, 10,10-bis(4-pyridinylmethyl)-9(10H)- anthracenone dihydrochloride, specifically potentiated burst firing. Computer modeling of the dopamine neuron confirmed the possibility of a differential influence of M-type channels on excitability during various firing patterns. Therefore, these channels may provide a novel target for the treatment of dopamine-related diseases, including Parkinson's disease and drug addiction. Moreover, our results demonstrate that the influence of M-type channels on the excitability of these slow pacemaker neurons is conditional upon their firing pattern. © 2010 Federation of European Neuroscience Societies and Blackwell Publishing Ltd.

线粒体是细胞内提供能量的细胞器,并负责调节细胞的程序化死亡。因遗传 缺陷引起的线粒体功能障碍会导致ATP 合成障碍、能量产生不足而出现一系列 病症。线粒体DNA 相关疾病目前日益受到广泛的关注,然而在线粒体DNA 相 关疾病领域常用的病例对照法容易受到遗传背景、群体分层、数据质量等方面因 素的影响以致经常得到假阳性结果。系统发育分析方法有助于解决这些方面的问 题,因此我们以此分析方法开展了如下工作: 首先我们对线粒体DNA C1494T 突变及其所属单倍型类群和氨基糖苷类药 物性耳聋之间的关联进行了研究。之前有研究报道了两个中国氨基糖苷类药物性 耳聋的家系,经过对先征者的线粒体DNA 全序列测定在这两个家系中都发现了 C1494T 突变。我们采用系统发育方法分析这两个家系先征者的全序列后,发现 这两个个体都属于线粒体单倍型A。巧合的是,在我们之前的研究中,在一个来 自武汉的汉族样本WH6980 中也有C1494T 突变,而且该个体同样也属于单倍型 类群A。这不由得使人想到:C1494T 突变可能是单倍型类群A 中一个分支的界 定位点,或者,受母系遗传背景的影响,该突变偏好于在A 类群中发生。那么 很有可能单倍型类群A对能够引发氨基糖苷类药物性耳聋的C1494T 突变的发生 有促进作用。 为了验证这个假设,我们从三个省份随机选取了553 个正常个体来检测 C1494T 突变,以调查该突变在普通人群中的发生频率。另外,我们从1823 个中 国人样本中筛选出属于单倍型类群A 的111 个体,在这111 个个体中检测C1494T 突变,以调查该突变是否为单倍型类群A 特有的变异位点。我们的结果表明: 在553 个随机样本群中没有检测到C1494T 突变,这说明该突变是一个稀有的突 变,在正常人群中发生的频率极低。另外,在111 个属于单倍型类群A 的样本 群中,我们也没有检测到C1494T 突变的存在,这说明该突变并非单倍型类群A 特有的变异位点。经过对带有C1494T 突变的全序列进行综合的系统发育学分析 表明,这三条序列的C1494T 突变应是来源于同一次突变事件。同时,根据序列 之间共享变异位点的状况推断,两个耳聋家系具有很近的母系亲缘关系,我们推 测这两个家系的C1494T 突变是来源于一位共同的近期母系祖先。综合这些结果 表明,C1494T 突变的发生与单倍型类群即母系遗传背景没有相关性,是属于在人群中频率极低的散发性突变。 我们又将系统发育分析的方法应用于肿瘤相关的线粒体DNA 突变的研究 中。线粒体在细胞的自由基产生以及细胞凋亡中扮演重要角色;有研究报道线粒 体功能的缺陷能导致癌症的发生,同时又有很多报道指出癌变组织的线粒体基因 组存在异常。为探讨癌组织中线粒体基因组的变异情况及其在癌症的发生和发展 中所扮演的角色,我们选取乳腺癌早期患者为研究对象。 近年来,有大量的研究都报导了在癌组织中存在高频率高密度的线粒体 DNA 体细胞突变,并认为这些突变在肿瘤发生过程中可能具有功能相关性。但 这些研究存在着不可回避的问题,总结来讲归为三个方面:①数据质量差:很多 前面报道的突变数据,经系统发育分析,发现存在不少因样本交叉污染所造成的 假阳性突变。②所测片断太短。大多数研究只是检测了D-loop 区,只占了整个 基因组的约十六分之一,很难全面反映问题。③对照设置有问题,许多研究进行 简单的case-control 分析,忽略了不同个体间母系遗传背景的差异,导致大量假 阳性突变的产生。 针对以上问题的存在,我们基于系统发育分析的方法在中国的乳腺癌病人中 开展了一项研究。分别取得10 例乳腺癌早期患者的癌组织、癌旁组织、以及远 端正常组织;提取了总DNA,对每一份组织样品进行线粒体全基因组测序。这 样做有两个研究目的:第一是调查在严格的质量控制手段下,排除交叉污染所造 成的假阳性突变后,在癌症病人中是否仍然能观察到高频率高密度的线粒体 DNA 体细胞突变。第二个目的是调查中国乳腺癌患者线粒体DNA 体细胞突变情 况,为乳腺癌早期诊断提供有效的信息。 为了避免过去部分研究中出现的问题,我们采取了相应的措施。首先是在严 格的质量控制下,我们对同一个病人的三种不同组织,即癌组织,癌旁组织,正 常组织中的线粒体基因组进行全序列测定的方式来检测体细胞突变。同时将所测 得线粒体全基因组序列进行系统发育的分析;结果表明属于同一个病人的不同组 织都能忠实的聚到一支,而且每一条全序列都完整的带有所属单倍型特有的界定 位点,不存在任何样本交叉或者污染的情况;这样进一步确证了我们数据的可靠 性。同时经过这样的比较,可以非常清晰的筛选出在某一种组织类型中所发生的 体细胞突变。来自10 例病人的癌组织,癌旁组织和正常组织的29 条(7#患者远端正常组 织没有得到)线粒体基因组全序列中,我们只检测到了两个体细胞突变(T2275C 和A8601G)。这两个突变经PCR 克隆试验的验证,在远端正常组织中均不存在, 是真实的体细胞突变。突变A8601G 同时出现在3#患者的癌组织和癌旁组织中; 提示线粒体DNA 的体细胞突变可能先于细胞的癌变,这可能对乳腺癌的早期临 床诊断具有一定的意义。突变T2275C 只出现在6#患者的癌组织中,该突变位于 16S rRNA 基因非常保守的位点上,我们对该突变位点潜在的生物学作用作了进 一步的讨论。 在我们研究的病例中体细胞突变的比率要远远低于先前的报道;造成差异的 主要原因可能是因为我们采取了严格的质量控制手段。综合我们的研究结果表 明,先前所报道的乳腺癌组织中的线粒体基因体细胞突变的频率存在被高估的现 象。目前线粒体突变与肿瘤发生的关系还有待于更深入的研究,同时我们呼吁本 领域的研究要加强数据质量的可靠性。我们的研究表明,系统发育的分析方法在 线粒体DNA 相关疾病的研究中是有效的,因此我们推荐在线粒体DNA 相关疾 病的研究中进行推广。

本论文由四部分组成,前三部分为实验论文,第四部分为文献综述。第一、二部分分别报道了中药西藏胡黄连和鸡矢藤的化学成分研究结果。从两种药用植物中共分离和鉴定了32个化学成分,其中3个为新化合物。第三部分为黄芪多糖的提取工艺研究。第四部分概述了近年来植物多糖的研究进展。 第一章为西藏胡黄连化学成分研究。通过正、反相硅胶柱层析等分离方法从药用植物西藏胡黄连(Picrorhiza scrophulariiflora Pennell)的根茎中共分离纯化出7个化合物。运用MS、1H-NMR、13C-NMR、DEPT、HSQC和HMBC等现代谱学方法,结合理化分析对这些化合物的结构进行了分析鉴定。7个化合物中有两个是酚性的葡萄糖苷类成分:西藏胡黄连酚苷D (1)、4-O-β-D-(6-O-vanilloyl glucopyranosyl) vanillic acid (6);四个苯乙基苷类化合物:plantamajoside (2)、plantainoside D (3)、西藏胡黄连苷A (4) 和西藏胡黄连苷F (5);一个苯基小分子化合物:香豆酸甲酯 (7)。其中化合物1和5未见文献报道,确定为新化合物;化合物3为首次从该种植物中分到。 第二章为鸡矢藤化学成分研究。从鸡矢藤(Paederia scandense (Lour) Merrill)全草中分离出25个化合物,通过理化常数和波谱数据鉴定了它们的结构。25个化合物中包括一个蒽醌类成分:茜根定-1-甲醚 (1);两个香豆素:异东莨菪香豆素 (2)和5-羟基-8-甲氧基吡喃香豆素 (3);两个香豆素-木脂素化合物:臭矢菜素 B (4)和臭矢菜素 D (5);一个木脂素:异落叶松树脂醇 (6);两个黄酮:diadzein (7)和蒙花苷 (8);三个三萜类化合物:齐墩果酸 (9)、乌苏酸 (10)和 3-O-β-D-吡喃葡萄糖基乌苏烷 (11);三个甾体及其糖苷:b-谷甾醇 (12)、胡萝卜苷 (13)和(24R)-豆甾-4-烯-3-酮 (14);六个小分子化合物:对羟基苯甲酸 (15),咖啡酸 (16),香豆酸 (17),丁烯二酸 (18),3,5-二甲氧基-4-羟基苯甲酸(19),咖啡酸-4-O-β-D-吡喃葡萄糖苷(20);五个环烯醚萜类化合物:鸡矢藤苷 (21),鸡矢藤酸 (22),鸡矢藤酸甲酯 (23),saprosmoside E (24)和paederoside B (25)。其中化合物25未见文献报道,为新化合物。化合物1~8、11、14、15~20为首次从该化合物中分离得到。同时对鸡矢藤中环烯醚萜类化合物做了高效液相-串联质谱(HPLC-MSn)分析,探讨了这类化合物的质谱裂解规律。 第三章为黄芪多糖的提取工艺研究。首先确定了黄芪多糖含量的测定方法,并进行了方法学验证;其次探讨了黄芪中黄芪多糖的提取工艺,确定以酶法-Sevag法联用来去除黄芪多糖中的蛋白质,可使其提取物中黄芪多糖总含量达到70%以上。 第四章为近年来植物多糖的研究进展。主要包括植物多糖的提取纯化、多糖的定性定量检测方法、多糖的结构分析和多糖的药理活性。 This dissertation consists of four parts. The first and second parts reports the studies on the chemical constituents of medicinal plants of Picrorhiza Scrophulariiflora and Paederia scandens. The third part is about the extract technique of Astragalan Polysaccharide (APS). The last part reviews the progress of the studies on plant polysaccharides.   The first chapter is about the chemical constituents of P. Scrophulariiflora which is widely used as an important medicine to treat various immune-related diseases. A new phenyl glycoside, scrophenoside D (1) and a new phenylethyl glycoside, scroside F (5), together with five known compounds, plantamajoside (2), plantainoside D (3), scroside A (4), 4-O-β-D-(6-O-vanilloylglucopyranosyl) vanillic acid (6); and methyl-p-coumarate (7) were isolated from the stems of P. scrophulariiflora. Their structures were elucidated by spectroscopic and chemical methods. The second chapter is about the chemical constituents of medicinal herb of P. scandens. Twenty-five compounds were isolated and purified by normal and reversed phase silica gel column chromatography. By physicochemical properties and spectral analysis, their structures were identified as rubiadin-1-methylether (1), isoscopoletin (2), 5-hydroxyl-8-methoxyl-coumarin (3), cleomiscosin B (4), cleomiscosin D (5), isolariciresinol (6), diadzein (7), linarin (8), oleanolic acid (9), ursolic acid (10), 3-O-β-D-glucopyranosyloxyl-ursane (11), b-sitosterol (12), b-daucosterol (13), (24R)-stigmast-4-ene-3-one (14), p-hydroxyl-benzoic acid (15), caffic acid (16), coumaric acid (17), trans-butenedioic acid (18), 3,5-dimethoxyl-4-hydroxylbenzoic acid (19), caffeic acid 4-O-β-D-glucopyranoside (20), paederoside (21), paederosidic acid (22), paederosidic acid methyl ester (23), saprosmoside E (24), paederoside B (25). Among them, compound 25 is a new compound. Compounds 1~8、11、14、15~20 were isolated from this plant for the first time. Futhermore, we studied the HPLC-MSn analysis and investigation of fragmentation behavior of the sulfur-containing iridoid glucosides. The third chapter is about the extracting process of Astragalan Polysaccharide (APS). The method of the content determination is built. The optimum condition of extraction of polysaccharides from Radix Astragali is defined and the more effective way to remove protein is combined enzyme method with Sevag method, by which the content of polysaccharides extract can be up to 70%. The last part is a review of the research progress of the plant polysaccharides, which includes its extraction, isolation, purification, determination, structure analysis, and pharmacology.

人类的载脂蛋白A5(apolipoprotein A5,APOA5)是一个新近发现的载脂蛋白家族成员。它在血浆中的含量比其他载脂蛋白低1-2个数量级,但能显著影响血浆三酰甘油水平,对血脂代谢具有重要意义,可以作为降血脂药物治疗中一个强有力的潜在靶标。 由于APOA5在血浆中含量低,直接从血浆中分离纯化很困难,国内一直没有报道简易可靠的纯化方法。为进一步研究APOA5的生物学特性,探讨其与TG代谢中的其它关键成分之间的相互关系,揭示其在脂类代谢相关疾病中的重要地位,必须有大量的蛋白和抗体用于基础研究。因此本研究首先利用基因工程技术,诱导表达纯化APOA5蛋白,免疫动物制备多克隆抗体,为进一步研究人肝脏细胞中APOA5的相互作用蛋白,研究APOA5蛋白在肝脏细胞中的功能奠定基础。 为了深入研究APOA5在肝脏中如何行使功能,我们采用细菌双杂交技术寻找与APOA5相互作用的蛋白因子。并采用Pull-down技术,免疫荧光及免疫共沉淀技术进一步确证其在体外和体内的相互作用关系,为进一步阐明APOA5在体内的生理功能提供了新的线索。 第一部分 APOA5基因的克隆、原核表达、纯化及其多克隆抗体的制备 本研究首先应用基因克隆技术,从人肝癌细胞系SMMC-7721的cDNA中扩增出1.1 kb的ApoA5基因全长序列。然后将其克隆至表达载体pThioHisD,构建原核表达载体pTH-APOA5。该重组质粒转化至大肠杆菌 BL21(DE3),成功实现人APOA5融合蛋白在大肠杆菌中的表达。经发酵得到高效表达的融合蛋白。 融合蛋白在 IPGT 诱导下以包涵体的形式大量表达。利用融合蛋白上的一段组氨酸序列,用镍离子亲和柱进行纯化和复性后,获得较高纯度的人APOA5融合蛋白。利用该融合蛋白免疫新西兰大耳白兔,获得了高效价的兔抗人APOA5多克隆抗体,Western Blot结果显示此多克隆抗体与APOA5特异性结合。 第二部分 细菌双杂交筛选与APOA5相互作用的蛋白 本实验首先构建了pBT-APOA5重组质粒,经双酶切、PCR和测序鉴定证明重组诱饵质粒构建成功,并进行了表达、自激活鉴定。Western Blot鉴定证实报告菌株中表达了分子量为 68 kD左右的重组融合蛋白,与预测的分子量APOA5(41 kD)/lamda cI (27 kD)一致。自激活实验证明诱饵蛋白不能单独激活报告基因,可用于筛选人肝脏cDNA文库。经过双重抗性筛选和回复筛选,分离出10个阳性克隆。对结果进行生物信息学分析,得到7个与APOA5相互作用的蛋白,其中BI1为细胞凋亡调节因子;ATP6、CYTB、ND2、COX-1为线粒体表达蛋白; ALB、TTR为血清蛋白。 第三部分 APOA5与BI1相互作用的确证 首先构建了BI1的原核表达载体pGEX-5X-3-BI1,利用Pull-down实验检测了APOA5与BI1在体外具有相互作用。然后构建了BI1的真核表达载体pCDNA3.1-HA-BI1和APOA5的真核表达载体pCDNA3.1-APOA5,并验证其表达。通过免疫荧光细胞内共定位研究发现,靶蛋白APOA5主要分布于胞浆,与BI1在HEK293细胞有共定位,即APOA5与BI1存在相互作用的可能。最后利用免疫共沉淀手段,在HEK293细胞中确证了靶蛋白APOA5与BI1在体内的相互作用。 上述研究结果,为深入研究APOA5在体内的生物学功能提供了新的思路。 Apolipoprotein A5 (APOA5) is a newly discovered protein belongs to apolipoprotein family. APOA5’s concentration is 1-2 orders of magnitude lower than other apolipoproteins in the circulation. APOA5 significantly affected plasma triglyceride levels, which is important on lipid metabolism. APOA5 has strong potential to be used as a hypolipidemic drug target. Large amount of APOA5 protein and antibodies are needed in basic research, such as biological characteristics study of the APOA5, its relationship with other key components in TG metabolism, its role played in Lipid metabolism-related diseases. Due to its low concentration in plasma, separation and purification of APOA5 from the plasma is very difficult. Until now no report on simple and reliable method for purification has been published in China. In this study, we firstly got APOA5 recombinant protein using genetic engineering technology. The purified recombinant protein was used to immunize rabbits to get antiserum. It is important for further study of the APOA5 protein-interacting protein. And it lays the foundation for studing APOA5 function in liver. In order to study APOA5 function in liver, we used bacterial two-hybrid technology to find the APOA5 protein interactor. Pull-down, immunofluorescence and immunoprecipitation techniques were used to further confirm the interaction between APOA5 with its interactor in vitro and in vivo. All of these stdudies provided new clues on its physiological functions in vivo. Part I: Cloning, prokaryotic expression, purification and polyclonal antibody preparation of APOA5 First of all, we amplified APOA5 CDS sequence from the human hepatoma cell line SMMC-7721, and subcloned into Expression vector pThioHisD, and got the recombinants named pTH-APOA5. The plasmid was transformed to BL21 (DE3). E. coli BL21(DE3) cells bearing the pTH-APOA5 plasmid were cultured and APOA5 protein synthesis was induced by the addition of IPTG. Recombinant protein was expression in the form of inclusion. Inclusion bodies were dissolved in phosphate-buffered saline containing 8 M urea and 40 mM imidazole, then applied to a Ni2+ affinity column, and were eluted in a buffer containing 4 M urea and 200 mM imidazole. Fractions containing the APOA5 protein were pooled and dialyzed against buffer containing phosphate-buffered saline. Antiserum to recombinant human APOA5 was generated by immuning rabbit. Western Blot showed that this antiserum specific binding with APOA5. Part II Two-hybrid system screening protein interactions with the APOA5 The coding sequence of human APOA5 was amplified using synthetic oligonucleotide primers from pTH-APOA5 vector and was subcloned into the pBT plasmidc to yield pBT-APOA5 vector. DNA sequencing was performed to verify that no unwanted mutations occurred during the process of plasmid vector construction. We verified recombinant protein expression and tested self-activation by pBT-APOA5 prior to screening. Western Blot verified inducing a 68 kD band, consistent with the predicted molecular weight (APOA5 41 kD, lamda cI 27 kD). pBT-APOA5 can be used for screening human liver cDNA library because it can not self-activation. Totally 10 positive clones were isolated. The nucleotide sequence of the positive clones were determined and compared to NCBI nucleotide sequence databases. We got 7 protein which interact with APOA5, included BI1(Apoptosis regulator); ATP6, CYTB, ND2, COX-1(Mitochondrial protein) and ALB, TTR(Serum protein). Part III Confirming of interaction between APOA5 with BI1 pGEX-5X-3-BI1 vector was subcloned at first. Pull-down experiments were used to detect the interaction between APOA5 with BI1 in vitro. Later, pCDNA3.1-HA-BI1 and pCDNA3.1-APOA5 were subcloned. Through immunofluorescence co-localization study, we found APOA5 mainly distributed in the cytoplasm. APOA5 is co-localization with BI1 in HEK293 cells. Finally, we verified interaction between APOA5 with BI1 in vivo through immunoprecipitation.

Mobile devices offer a common platform for both leisure and work-related tasks but this has resulted in a blurred boundary between home and work. In this paper we explore the security implications of this blurred boundary, both for the worker and the employer. Mobile workers may not always make optimum security-related choices when ‘on the go’ and more impulsive individuals may be particularly affected as they are considered more vulnerable to distraction. In this study we used a task scenario, in which 104 users were asked to choose a wireless network when responding to work demands while out of the office. Eye-tracking data was obtained from a subsample of 40 of these participants in order to explore the effects of impulsivity on attention. Our results suggest that impulsive people are more frequent users of public devices and networks in their day-to-day interactions and are more likely to access their social networks on a regular basis. However they are also likely to make risky decisions when working on-the-go, processing fewer features before making those decisions. These results suggest that those with high impulsivity may make more use of the mobile Internet options for both work and private purposes but they also show attentional behavior patterns that suggest they make less considered security-sensitive decisions. The findings are discussed in terms of designs that might support enhanced deliberation, both in the moment and also in relation to longer term behaviors that would contribute to a better work-life balance.

The prevalence of obesity worldwide has increased dramatically over the last few decades. Poor dietary habits and low levels of exercise in adolescence are often maintained into adulthood where they can impact on the incidence of obesity and chronic diseases. A 3-year longitudinal study of anthropometric, dietary and exercise parameters was carried out annually (2005 - 2007) in 3 Irish secondary schools. Anthropometric measurements were taken in each year and analysed longitudinally. Overweight and obesity were at relatively low levels in these adolescents. Height, weight, BMI, waist and hip circumferences and TST increased significantly over the 3 years. Waist-to-hip ratio (WHR) decreased significantly over time. Boys were significantly taller than girls across the 3 years. A 3-day weighed food diary was used to assess food intake by the adolescents. Analysis of dietary intake data was determined using WISP©. Mean daily energy and nutrient intakes were reported. Mean daily energy and macronutrient intakes were analysed longitudinally. The adolescents’ diet was characterised by relatively high saturated fat intakes and insufficient fruit and vegetable consumption. The dietary pattern did not change significantly over the 3 years. Boys consumed more energy than girls over the study period. A validated questionnaire was used to assess physical activity and sedentary activity levels. Boys were substantially more active and had higher energy expenditure estimates than girls throughout the study. A significant longitudinal decrease in physical activity levels among the adolescents was observed. Both genders spent more than the recommended amount of time (hrs/day) pursing sedentary activities. The dietary pattern in these Irish adolescents is relatively poor. Of additional concern is the overall longitudinal decrease in physical activity levels. Promoting consumption of a balanced diet and increased exercise levels among adolescents will help to reduce future public health care costs due to weight-related diseases.
