Effectiveness in achieving mission is fundamental to evaluating charity performance, and is of central concern to stakeholders who fund, regulate and otherwise engage with such organisations. Exploring the meaning of transparency in the context of stakeholder engagement, and utilising previous research and authoritative sector discussion, this paper develops a novel framework of transparent, stakeholder-focused effectiveness reporting. It is contended that such reporting can assist the charity sector in discharging accountability, gaining legitimacy, and in sharpening mission-centred managerial decision making. Then applying this to UK charities’ publicly-available communications, it highlights significant challenges and weaknesses in current effectiveness reporting.

This paper presents the results of a study aimed at identifying and assessing positive parenting programmes and activities carried out in the Autonomous Region of the Basque Country (ARBC), Spain. The study is a development of the III Inter-institutional Family Support Plan (2011), drafted by the Basque Government's Department of Family Policy and Community Development, and its aim is to offer a series of sound criteria for improving existing programmes and ensuring the correct design and implementation of new ones in the future. It analyses 129 programmes and gathers data relative to institutional management and coordination, format, quality of the established aims, adaptation to the theoretical proposal for an Optimal Positive Parenting Curriculum, scientific base, use of the framework of reference for competences, working method, assessment techniques, budgets and publicity, among others. The results highlight the good quality of the programmes' aims and content, and the poor systematic assessment of these same aspects. The study concludes with a series of recommendations for improving the initiatives, integrated into a proposal for a system of indicators to assess and implement positive parenting programmes.

Long-term monitoring of data of ambient mercury (Hg) on a global scale to assess its emission, transport, atmospheric chemistry, and deposition processes is vital to understanding the impact of Hg pollution on the environment. The Global Mercury Observation System (GMOS) project was funded by the European Commission (http://www.gmos.eu) and started in November 2010 with the overall goal to develop a coordinated global observing system to monitor Hg on a global scale, including a large network of ground-based monitoring stations, ad hoc periodic oceanographic cruises and measurement flights in the lower and upper troposphere as well as in the lower stratosphere. To date, more than 40 ground-based monitoring sites constitute the global network covering many regions where little to no observational data were available before GMOS. This work presents atmospheric Hg concentrations recorded worldwide in the framework of the GMOS project (2010–2015), analyzing Hg measurement results in terms of temporal trends, seasonality and comparability within the network. Major findings highlighted in this paper include a clear gradient of Hg concentrations between the Northern and Southern hemispheres, confirming that the gradient observed is mostly driven by local and regional sources, which can be anthropogenic, natural or a combination of both.

Single-cell functional proteomics assays can connect genomic information to biological function through quantitative and multiplex protein measurements. Tools for single-cell proteomics have developed rapidly over the past 5 years and are providing unique opportunities. This thesis describes an emerging microfluidics-based toolkit for single cell functional proteomics, focusing on the development of the single cell barcode chips (SCBCs) with applications in fundamental and translational cancer research.
The microchip designed to simultaneously quantify a panel of secreted, cytoplasmic and membrane proteins from single cells will be discussed at the beginning, which is the prototype for subsequent proteomic microchips with more sophisticated design in preclinical cancer research or clinical applications. The SCBCs are a highly versatile and information rich tool for single-cell functional proteomics. They are based upon isolating individual cells, or defined number of cells, within microchambers, each of which is equipped with a large antibody microarray (the barcode), with between a few hundred to ten thousand microchambers included within a single microchip. Functional proteomics assays at single-cell resolution yield unique pieces of information that significantly shape the way of thinking on cancer research. An in-depth discussion about analysis and interpretation of the unique information such as functional protein fluctuations and protein-protein correlative interactions will follow.
The SCBC is a powerful tool to resolve the functional heterogeneity of cancer cells. It has the capacity to extract a comprehensive picture of the signal transduction network from single tumor cells and thus provides insight into the effect of targeted therapies on protein signaling networks. We will demonstrate this point through applying the SCBCs to investigate three isogenic cell lines of glioblastoma multiforme (GBM).
The cancer cell population is highly heterogeneous with high-amplitude fluctuation at the single cell level, which in turn grants the robustness of the entire population. The concept that a stable population existing in the presence of random fluctuations is reminiscent of many physical systems that are successfully understood using statistical physics. Thus, tools derived from that field can probably be applied to using fluctuations to determine the nature of signaling networks. In the second part of the thesis, we will focus on such a case to use thermodynamics-motivated principles to understand cancer cell hypoxia, where single cell proteomics assays coupled with a quantitative version of Le Chatelier's principle derived from statistical mechanics yield detailed and surprising predictions, which were found to be correct in both cell line and primary tumor model.
The third part of the thesis demonstrates the application of this technology in the preclinical cancer research to study the GBM cancer cell resistance to molecular targeted therapy. Physical approaches to anticipate therapy resistance and to identify effective therapy combinations will be discussed in detail. Our approach is based upon elucidating the signaling coordination within the phosphoprotein signaling pathways that are hyperactivated in human GBMs, and interrogating how that coordination responds to the perturbation of targeted inhibitor. Strongly coupled protein-protein interactions constitute most signaling cascades. A physical analogy of such a system is the strongly coupled atom-atom interactions in a crystal lattice. Similar to decomposing the atomic interactions into a series of independent normal vibrational modes, a simplified picture of signaling network coordination can also be achieved by diagonalizing protein-protein correlation or covariance matrices to decompose the pairwise correlative interactions into a set of distinct linear combinations of signaling proteins (i.e. independent signaling modes). By doing so, two independent signaling modes – one associated with mTOR signaling and a second associated with ERK/Src signaling have been resolved, which in turn allow us to anticipate resistance, and to design combination therapies that are effective, as well as identify those therapies and therapy combinations that will be ineffective. We validated our predictions in mouse tumor models and all predictions were borne out.
In the last part, some preliminary results about the clinical translation of single-cell proteomics chips will be presented. The successful demonstration of our work on human-derived xenografts provides the rationale to extend our current work into the clinic. It will enable us to interrogate GBM tumor samples in a way that could potentially yield a straightforward, rapid interpretation so that we can give therapeutic guidance to the attending physicians within a clinical relevant time scale. The technical challenges of the clinical translation will be presented and our solutions to address the challenges will be discussed as well. A clinical case study will then follow, where some preliminary data collected from a pediatric GBM patient bearing an EGFR amplified tumor will be presented to demonstrate the general protocol and the workflow of the proposed clinical studies.

Increasing in resolution of numerical weather prediction models has allowed more and more realistic forecasts of atmospheric parameters. Due to the growing variability into predicted fields the traditional verification methods are not always able to describe the model ability because they are based on a grid-point-by-grid-point matching between observation and prediction. Recently, new spatial verification methods have been developed with the aim of show the benefit associated to the high resolution forecast. Nested in among of the MesoVICT international project, the initially aim of this work is to compare the newly tecniques remarking advantages and disadvantages. First of all, the MesoVICT basic examples, represented by synthetic precipitation fields, have been examined. Giving an error evaluation in terms of structure, amplitude and localization of the precipitation fields, the SAL method has been studied more thoroughly respect to the others approaches with its implementation in the core cases of the project. The verification procedure has concerned precipitation fields over central Europe: comparisons between the forecasts performed by the 00z COSMO-2 model and the VERA (Vienna Enhanced Resolution Analysis) have been done. The study of these cases has shown some weaknesses of the methodology examined; in particular has been highlighted the presence of a correlation between the optimal domain size and the extention of the precipitation systems. In order to increase ability of SAL, a subdivision of the original domain in three subdomains has been done and the method has been applied again. Some limits have been found in cases in which at least one of the two domains does not show precipitation. The overall results for the subdomains have been summarized on scatter plots. With the aim to identify systematic errors of the model the variability of the three parameters has been studied for each subdomain.

Existing bridges built in the last 50 years face challenges due to states far different than those envisaged when they were designed, due to increased loads, ageing of materials, and poor maintenance. For post-tensioned bridges, the need emerged for reliable engineering tools for the evaluation of their capacity in case of steel corrosion due to lack of mortar injection. This can lead to sudden brittle collapses, highlighting the need for proper maintenance and monitoring. This thesis proposes a peak strength model for corroded strands, introducing a “group coefficient” that aims at considering corrosion variability in the wires constituting the strands. The application of the introduced model in a deterministic approach leads to the proposal of strength curves for corroded strands, which represent useful engineering tools for estimating their maximum strength considering both geometry of the corrosion and steel material parameters. Together with the proposed ultimate displacement curves, constitutive laws of the steel material reduced by the effects of corrosion can be obtained. The effects of corroded strands on post-tensioned beams can be evaluated through the reduced bending moment-curvature diagram accounting for these reduced stress-strain relationships. The application of the introduced model in a probabilistic approach allows to estimate peak strength probability functions and consecutive design-oriented safety factors to consider corrosion effects in safety assessment verifications. Both approaches consider two procedures that are based on the knowledge level of the corrosion in the strands. On the sidelines of this main research line, this thesis also presents a study of a seismic upgrading intervention of a case-study bridge through HDRB isolators providing a simplified procedure for the identification of the correct device. The study also investigates the effects due to the variability of the shear modulus of the rubber material of the HDRB isolators on the structural response of the isolated bridge.

A pesquisa foi realizada por meio de metodologia qualitativa, explorat??ria e adotando a estrat??gia de investiga????o do estudo de caso. A formação profissional, nomeadamente a formação profissionalizante (2ºCiclo de Estudos no âmbito de Bolonha) tem vindo a ser alvo de discussão entre as diferentes áreas do conhecimento, nomeadamente no que respeita às possíveis metodologias de avaliação a adotar (práticas profissionais supervisionadas, elaboração de portefólios individuais, realização de trabalhos de grupo com acompanhamento tutorial, redação de Relatórios de Estágio, entre outras). A presente comunicação tem como principal objetivo discutir e analisar as conceções dos educadores de infância recém-formados acerca da educação de infância e das aprendizagens realizadas no âmbito da Prática Profissional Supervisionada (PPS), através da análise dos Relatórios de Estágio elaborados no âmbito do Mestrado em Educação Pré-Escolar de uma escola superior de educação, especificamente os que dizem respeito a um grupo de mestrandos dos anos letivos 2010-2011, 2011-2012 e 2012-2013. Nesta análise, serão privilegiados os seguintes eixos: i) a caracterização reflexiva do contexto socioeducativo; ii) a análise reflexiva da intervenção; iii) as considerações finais realizadas. Pretende-se, a partir desta análise, identificar e compreender os potenciais desafios (que elementos e critérios de avaliação? Como caracterizar, de forma reflexiva, um determinado contexto socioeducativo?) trazidos ao processo de avaliação das aprendizagens dos mestrandos do curso em questão. O quadro teórico de referência centra-se numa revisão de literatura sobre a formação profissional dos educadores de infância e sobre a avaliação das aprendizagens dos adultos. A metodologia utilizada, de natureza qualitativa, inclui análise documental (objetivos do curso, perfil profissional de saída, guião para a elaboração dos Relatórios de Estágio), e a subsequente análise de conteúdo. Os resultados obtidos apontam para asseguintes conclusões: se por um lado, todos (12) os mestrandos do estudo seguiram, escrupulosamente, o guião fornecido pela equipa de docentes da Prática Profissional Supervisionada para a elaboração dos Relatórios de Estágio, por outro lado e no que se relaciona com a caracterização reflexiva do contexto socioeducativo, a análise reflexiva da intervenção e as considerações finais realizadas, constata-se que existem diferenças significativas na estrutura organizativa do discurso escrito, na escolha do quadro teórico de referência bem como nas reflexões pessoais realizadas. Enquanto que os mestrandos (5) do ano letivo 2010-2011 demonstram ter optado por construir um quadro teórico de referência ancorado, preferencialmente, por referências bibliográficas relacionadas com a Educação de Infância e facultadas pela equipa de docentes do mestrado em estudo, os mestrandos do ano letivo 2011-2012 (3) optaram por recorrer a um quadro teórico de referência mais alargado e abrangente, no qual se pode verificar, também, a existência de uma consulta e análise documental frequente sobre a legislação em vigor para o âmbito da Educação de Infância. Por sua vez, os mestrandos que terminaram a sua formação no ano letivo 2012-2013 (4) demonstraram, na elaboração dos seus relatórios de estágio, uma preocupação e intenção claras em dar resposta a todas as questões colocadas, fazendo emergir algumas questões de fundo: 1) por que razão os relatórios de estágio analisados apresentam diferenças significativas nas aprendizagens realizadas pelos mestrandos em estudo? 2) Quais as possíveis razões para estas diferenças? 3) Que aprendizagens realizaram estes mestrandos e quais foram as mais significativas? Estes são alguns dos possíveis desafios colocados à avaliação das aprendizagens.

No âmbito do Mestrado em Educação Pré-Escolar e Ensino do 1º Ciclo do Ensino Básico e da Prática Pedagógica Supervisionada, realizada em contexto de Creche, foi desenvolvido o presente relatório de estágio de qualificação profissional na Educação Pré-Escolar. A prática educativa sustentou-se num quadro teórico-concetual de referência, com vista à construção de saberes para a Educação de Infância, pelo compromisso e responsabilização progressiva da ação docente, tendo sido encarado como um momento de singular importância. Desta forma, este documento procura ilustrar o processo de desenvolvimento de competências profissionais pela estudante para este nível de educação. Nesta linha de pensamento, importa referenciar a metodologia de investigação-ação, que fundamentou a prática pedagógica da estudante ao longo das várias etapas do processo educativo: Observação, Planificação, Ação, Avaliação, Comunicação e Articulação. Assim, apoiada em estratégias e atitudes investigativas e reflexivas, esta metodologia potenciou a transformação, melhoria e adequação das suas práticas. O contexto de formação assumiu-se, portanto, como um lugar privilegiado de articulação entre teoria e prática, onde o processo de ensino e aprendizagem ficou pautado por intencionalidades educativas com vista ao desenvolvimento integral de cada criança, bem como pela construção de saberes e competências profissionais pela estagiária, elencadas nos Decretos-Lei n.º 240 e 241 de 2001. Tomando em consideração o processo desenvolvido, importa realçar a importância de uma formação profissional ao longo da vida, de modo a potenciar o desenvolvimento de uma atitude perante a Educação cada vez mais crítica, problematizadora, indagadora, reflexiva e investigativa, em prol do desenvolvimento holístico de cada criança.
