Dexamethasone is a synthetic glucocorticoid widely used to treat allergic and inflammatory processes. This drug is used in three main situations, are used to contain acute or chronic inflammatory processes, or like immunosuppressive drug's. In these cases the patient will receive high doses for a chronic period and, therefore, has a much greater chance of adverse side effects, such as hypertension, diabetes and dyslipidemia. Dexamethasone promotes deleterious effects on the arachidonic acid pathway, when administered in high doses, because it is a potent anti-inflammatory drug. We recently demonstrated that dexamethasone significantly reduces the protein expression of vascular endothelial growth factor (VEGF) in both skeletal muscle and heart, but the mechanisms involved remain unclear. Meanwhile, exercise has been shown to be effective against high blood pressure, diabetes and dyslipidemia, promoting, among other factors, the increase in VEGF and angiogenesis. One possible explanation for these effects would be the creation of new vessels mediated by inflammation, or by the stimulation of the formation of products of the metabolism of arachidonic acid (AA), such as prostaglandin E2 (PGE2) and VEGF, by increasing the stimulation of the enzymes cyclooxygenase 1 and 2 (COX-1 and COX-2). Little is known about the preventive effects of training on the action of dexamethasone in the arachidonic acid pathway. Therefore, the aim of this study was to determine whether aerobic exercise training, performed before and concomitant treatment with dexamethasone, was able to prevent the effects of the dexamethasone in the protein expression of COX-2 and VEGF. Dexamethasone (DEXA) is a synthetic glucocorticoid widely used in the handling of several drugs, for its proven benefits in fighting inflammation and allergies. Despite their benefits, their chronic use leads to several side effects that include changes in the body in the metabolism of carbohydrates, lipids and proteins. Moreover, being an anti-inflammatory, acts on the arachidonic acid pathway, reducing the expression of the enzyme cyclooxygenase (COX-2) and growth factor derived from the endothelium of blood vessels (VEGF) in various tissues. However, its effects on the myocardium are still uncertain. The physical training (PT), in turn, promotes effects contrary to those caused by chronic use of DEXA, however, little is known about the preventive effects of TF in the side effects of Dexa in the myocardium. Therefore, the aim of this study was to determine if the TF has the ability to prevent and/or mitigate the effects of Dexa in protein expression of COX-2 and VEGF in the myocardium. Forty animals were divided into 4 groups: sedentary control (SC), sedentary treated with Dexa (SD), trained control (TC) and Trained treated with Dexa (TD) and submitted to a protocol of physical training on the treadmill for 70 days (1 h/day-5 days per week, 60% of physical capacity) or kept sedentary. Over the past 10 days, rats were treated with Dexa (Decadron, 0.5 mg/kg per day, ip) or saline. During training the animals were weighed weekly and during treatment daily. At the end of treatment was made to measure fasting glucose levels of animals. The rats were killed with excess anesthesia and cardiac muscle was removed, weighed, homogenized, centrifuged and stored at -20° C for analysis of protein expression of VEGF and COX-2 by Western blotting technique. The transgenic application of green fluorescent protein (GFP) as fetal cell marker on cattle cloned placenta could provide an exclusive model for studying the morphologic and immunologic maternal-fetal interactions, providing information about its mapping, distinguishing the fetal from maternal cells. This model will have direct application, mainly because these animals present problems during its development. With this model's support, we intend to verify the substances transport between mother and fetus during endocytosis, through the immunolocalization of protein named caveolae. For these, we used 06 cloned bovine and 30 cattle samples of artificial insemination (AI) with 90 days of pregnancy, which had been their development interrupted by humanitarian slaughter of the recipient and recovery of the pregnant uterus. We collected the placentome and the chorion. A part of the samples was cut and fixed, by immersion, on a solution containing 4% of parafomaldehyde or 10% of formaldehyde on a sodium phosphate buffer (PBS), at 0,1 M pH 7.4, Zamboni solution (4% of paraformaldehyde, 15% of picric acid, on sodium phosphate buffer 0,1 M pH 7.4), metacarn (60% of metanol, 30% of chloroform, and 10% glacial acetic acid), for morphologic and immunohistochemistry verification for caveolinas proteins -1 and -2 (CAV -1 and CAV-2). The caveolins -1 were found in fetal and maternal villi, but its strongest staining was observed in the endometrial stroma. The caveolins -2 had positive staining in trophoblast and chorioallantoic membrane, and specifically in giant trophoblastic binucleated cell. Therefore the results were compared between cloned cattle and from AI or natural mating, for assisting on detection of the reason of many placental alterations, embryonic losses, spontaneous abortion, post-natal mortality and large offspring syndrome on laboratory-manipulated animals. The result suggests that the proteins caveolins -1 and -2 (CAV-1 and CAV-2) are part of the caveolae composition and important structures related to the molecule transfer to the fetus, nourish it through endocytosis and pinocytosis.

Hemogram values may have variations caused by intrinsic or extrinsic factors, which can take healthy animals to present values outside of the normal reference ranges and it is important to the veterinarian to know these variation factors for a correct results interpretation. The influence of the period of the day is among them. The objective of this study was to test the hypothesis that there are hematological parameters variations, such as erythrogram and plasmatic proteins during the day. The study was carried out with 10 non-pregnant adult goats in Araçatuba/SP/Brazil. The blood sampling and physical exam occurred at three moments between 07:00 and 08:00 a.m. (M1), 12:30 and 01:30 p.m. (M2), 5:00 and 6:00 p.m. (M3). We noted differences in the values of packed cell volume (PCV), hemoglobin, and rectal temperature through the moments, with reduction of PCV and hemoglobin were related to increase in rectal and air temperature, leading to water intake increase and, consequently, decreasing blood concentration. These variations could lead to an error of interpretation of the results due to values below the reference ranges.

A perda de massa muscular observada no diabetes mellitus (DM) tipo 1 é consequência da combinação entre redução na velocidade de síntese proteica e aumento na velocidade de proteólise. A curcumina, pigmento amarelo extraído dos rizomas de Curcuma longa L., promove diversos benefícios no metabolismo de carboidratos e lipídeos no DM. Deste modo, buscamos avaliar o efeito do tratamento de ratos diabéticos com curcumina incorporada em iogurte sobre o metabolismo proteico muscular. Ratos Wistar machos (150±10 g) receberam estreptozotocina (40 mg/kg, i.v.) para indução do DM e foram divididos nos grupos (n=8): diabético tratado com iogurte (DIOG), 90 mg/kg de curcumina (DC90), 4U de insulina (DINS) e ratos normais, não diabéticos, tratados com iogurte (NIOG). Após 35 dias de tratamento, os animais foram eutanasiados e os músculos esqueléticos soleus e extensor digitorium longus (EDL) foram retirados e utilizados para a determinação das atividades proteolíticas de caspase-3, calpaína e proteassoma (atividade quimiotripsina-like). O tratamento de animais diabéticos com curcumina incorporada em iogurte reduziu a glicemia, os níveis de ureia urinária e promoveu um maior ganho de peso corporal em relação aos animais diabéticos tratados somente com iogurte (DIOG). Animais DIOG apresentaram um aumento nas atividades de calpaína e proteassoma em músculos soleus e EDL em relação aos valores encontrados em músculos de animais NIOG; já o tratamento com curcumina reduziu as atividades de calpaína e proteassoma em EDL de ratos diabéticos, o que explica, pelo menos em parte, a menor perda de massa deste músculo em ratos DC90. Houve uma redução na atividade de caspase-3 em músculos de animais DIOG em comparação aos grupos...
