Existing data supports Portugal as the Western Europe country with highest HIV-1 subtype diversity. However, detailed phylogenetic studies of Portuguese HIV-1 epidemics are still scarce. Thus, our main goal was to analyze the phylodynamics of a local HIV-1 infection in the Portuguese region of Minho. Molecular epidemiological analysis was applied to data from 289 HIV-1 infected individuals followed in the reference Hospital of the province of Minho, Portugal, in which isolated viruses had been sequenced between 2000 and 2012. Viruses of the G (29.1%) and B (27.0%) subtypes were the most frequent, followed by recombinant forms (17.6%), C (14.5%), F1 (7.3%) and A1 (4.2%) subtypes. Multinomial logistic regression revealed that the odds of being infected with A1 and F1 subtype increased over the years when compared with B, G, C or recombinant viruses. As expected, polyphyletic patterns suggesting multiple and old introductions of subtypes B and G were found. However, transmission clusters of non-B and -G viruses among native individuals were also found with the dates of the most recent common ancestor estimated to the early 2000s. Our study supports that the HIV-1 subtype diversity in the Portuguese region of Minho is high and has been increasing in a manner that is apparently driven by factors other than immigration and international travel. Infections with A1 and F1 viruses in the region of Minho are becoming established and were mainly found in sexually transmitted clusters, reinforcing the need for more efficacious control measures targeting this infection route.

Sirtuins (Sirts) regulate several cellular mechanisms through deacetylation of several transcription factors and enzymes. Recently, Sirt2 was shown to prevent the development of inflammatory processes and its expression favors acute Listeria monocytogenes infection. The impact of this molecule in the context of chronic infections remains unknown. We found that specific Sirt2 deletion in the myeloid lineage transiently increased Mycobacterium tuberculosis load in the lungs and liver of conditional mice. Sirt2 did not affect long-term infection since no significant differences were observed in the bacterial burden at days 60 and 120 post-infection. The initial increase in M. tuberculosis growth was not due to differences in inflammatory cell infiltrates in the lung, myeloid or CD4+ T cells. The transcription levels of IFN-?, IL-17, TNF, IL-6 and NOS2 were also not affected in the lungs by Sirt2-myeloid specific deletion. Overall, our results demonstrate that Sirt2 expression has a transitory effect in M. tuberculosis infection. Thus, modulation of Sirt2 activity in vivo is not expected to affect chronic infection with M. tuberculosis.

The fate of infected macrophages is a critical aspect of immunity to mycobacteria. By depriving the pathogen of its intracellular niche, apoptotic death of the infected macrophage has been shown to be an important mechanism to control bacterial growth. Here, we show that IL-17 inhibits apoptosis of Mycobacterium bovis BCG- or Mycobacterium tuberculosis-infected macrophages thus hampering their ability to control bacterial growth. Mechanistically, we show that IL-17 inhibits p53, and impacts on the intrinsic apoptotic pathway, by increasing the Bcl2 and decreasing Bax expression, decreasing cytochrome c release from the mitochondria, and inhibiting caspase-3 activation. The same effect of IL-17 was observed in infected macrophages upon blockade of p53 nuclear translocation. These results reveal a previously unappreciated role for the IL-17/p53 axis in the regulation of mycobacteria-induced apoptosis and can have important implications in a broad spectrum of diseases where apoptosis of the infected cell is an important host defense mechanism.

Na região amazônica, algumas indústrias despejam cobre nos corpos receptores que, em elevadas concentrações, é tóxico para os seres vivos. A remoção de cobre de efluentes industriais é realizada por diversos processos como a adsorção. Neste trabalho mostrou-se o resultado da adsorção de cobre (II) em carvão ativado de caroço de buriti carbonizado a 400 °C e ativado a 900 °C. O carvão ativado foi caracterizado em termos de área específica, tamanho dos poros, densidades aparente e real, porosidade, microscopia eletrônica de varredura, conteúdo de cinzas, pH, umidade, carbono fixo e grupos funcionais de superfície. O estudo de equilíbrio de adsorção avaliou a influência do diâmetro das partículas do carvão, do tempo de contato adsorvente/adsorbato, do pH e da concentração inicial da solução de cobre (II) sobre a remoção de cobre (II). Com base nos resultados, concluiu-se que há uma maior eficiência de remoção de cobre (II) para diâmetro < 0,595 mm, tempo de contato de 300 minutos, pH 4,0 e concentrações iniciais de cobre (II) de 50 e 80 mg L-1. O modelo matemático de Langmuir foi o que melhor se ajustou aos dados de equilíbrio de adsorção. A partir do tempo de contato de 15 minutos todas as concentrações de equilíbrio ficaram abaixo do máximo permitido de 1,0 mg L-1 previsto pela legislação vigente para lançamento de efluentes em corpos receptores.

Os Serviços de Documentação da Universidade do Minho (SDUM) estabeleceram para os anos de 2014 e 2015 um plano de intervenção para os serviços de suporte à Gestão dos Dados Científicos na UMinho. Esta apresentação pretende resumir o plano traçado e a metodologia para a elaboração desta estratégia de atuação, identificando as áreas e os eixos de intervenção definidos pelos SDUM. Descreve algumas das ações já realizadas ou em desenvolvimento e destaca os principais resultados obtidos através de um estudo diagnóstico, realizado por questionário junto dos investigadores e centros de investigação da universidade, com o intuito de se obter um melhor conhecimento sobre o tipo de dados produzidos, as práticas de gestão associadas e averiguar a pertinência de serviços de suporte neste domínio.
Constituintes químicos isolados dos galhos e cascas de amapazeiro (Parahancornia amapa, Apocynaceae)

Parahancornia amapa (Apocynaceae) é típica da região amazônica e conhecida popularmente como amapazeiro e muito utilizada na medicina popular da região Amazônica. O objetivo desse trabalho foi o estudo fitoquímico dos galhos e cascas dessa espécie. Do extrato diclorometânico dos galhos foram isolados o β-sitosterol, estigmasterol, além de triterpenóides pentacíclicos, α-amirina, β-amirina, lupeol e friedelina. Do extrato metanólico das cascas foi isolado o alcaloide indólico isositsiriquina. As estruturas desses compostos foram identificadas por análise dos espectros de massa de alta resolução, Ressonância Magnética Nuclear de ¹H e 13C e comparações com dados da literatura. Esse é o primeiro relato de isolamento de alcaloide neste gênero.

Este artigo aborda a presença e a apresentação clínica da disfunção executiva em pacientes com TDAH. Na medida em que sintomas de disfunção executiva determinam as alterações de funcionamento desses pacientes nas atividades do dia-a-dia, torna-se relevante o estudo das relações entre alterações em testes neuropsicológicos formais de funções executivas e a ocorrência desses sintomas. A constatação relativamente recente do papel da disfunção executiva no TDAH tem impulsionado a pesquisa neuropsicológica, mas os dados até agora disponíveis são ainda pouco consistentes. Uma visão atualizada dessas questões sugere que a semiologia neuropsicológica disponível ainda não alcança plenamente a constelação de sintomas práticos do TDAH no dia-a-dia, mostrando a necessidade de avanços na pesquisa dessas relações.

A spreadsheet usually starts as a simple and singleuser software artifact, but, as frequent as in other software systems, quickly evolves into a complex system developed by many actors. Often, different users work on different aspects of the same spreadsheet: while a secretary may be only involved in adding plain data to the spreadsheet, an accountant may define new business rules, while an engineer may need to adapt the spreadsheet content so it can be used by other software systems.Unfortunately,spreadsheetsystemsdonotoffermodular mechanisms, and as a consequence, some of the previous tasks may be defined by adding intrusive “code” to the spreadsheet. In this paper we go through the design and implementation of an aspect-oriented language for spreadsheets so that users can work on different aspects of a spreadsheet in a modular way. For example, aspects can be defined in order to introduce new business rules to an existing spreadsheet, or to manipulate the spreadsheet data to be ported to another system. Aspects are defined as aspect-oriented program specifications that are dynamically woven into the underlying spreadsheet by an aspect weaver. In this aspect-oriented style of spreadsheet development, differentusers develop,orreuse,aspects withoutaddingintrusive code to the original spreadsheet. Such code is added/executed by the spreadsheet weaving mechanism proposed in this paper.

This paper proposes and validates a model-driven software engineering technique for spreadsheets. The technique that we envision builds on the embedding of spreadsheet models under a widely used spreadsheet system. This means that we enable the creation and evolution of spreadsheet models under a spreadsheet system. More precisely, we embed ClassSheets, a visual language with a syntax similar to the one offered by common spreadsheets, that was created with the aim of specifying spreadsheets. Our embedding allows models and their conforming instances to be developed under the same environment. In practice, this convenient environment enhances evolution steps at the model level while the corresponding instance is automatically co-evolved.Finally,wehave designed and conducted an empirical study with human users in order to assess our technique in production environments. The results of this study are promising and suggest that productivity gains are realizable under our model-driven spreadsheet development setting.

Renal cell tumors (RCTs) are the most lethal of the common urological cancers. The widespread use of imaging entailed an increased detection of small renal masses, emphasizing the need for accurate distinction between benign and malignant RCTs, which is critical for adequate therapeutic management. Histone methylation has been implicated in renal tumorigenesis, but its potential clinical value as RCT biomarker remains mostly unexplored. Hence, the main goal of this study was to identify differentially expressed histone methyltransferases (HMTs) and histone demethylases (HDMs) that might prove useful for RCT diagnosis and prognostication, emphasizing the discrimination between oncocytoma (a benign tumor) and renal cell carcinoma (RCC), especially the chromophobe subtype (chRCC). We found that the expression levels of three genes-SMYD2, SETD3, and NO66-was significantly altered in a set of RCTs, which was further validated in a large independent cohort. Higher expression levels were found in RCTs compared to normal renal tissues (RNTs) and in chRCCs comparatively to oncocytomas. SMYD2 and SETD3 mRNA levels correlated with protein expression assessed by immunohistochemistry. SMYD2 transcript levels discriminated RCTs from RNT, with 82.1% sensitivity and 100% specificity (AUC=0.959), and distinguished chRCCs from oncocytomas, with 71.0% sensitivity and 73.3% specificity (AUC: 0.784). Low expression levels of SMYD2, SETD3, and NO66 were significantly associated with shorter disease-specific and disease-free survival, especially in patients with non-organ confined tumors. We conclude that expression of selected HMTs and HDMs might constitute novel biomarkers to assist in RCT diagnosis and assessment of tumor aggressiveness.
