Este trabalho tem por objetivo avaliar o comportamento dinâmico de pisos mistos (aço-concreto) sob a ação de cargas provenientes das atividades humanas rítmicas, especificamente a prática de ginástica aeróbica, sob o ponto de vista do conforto humano. Tal avaliação torna-se necessária por crescentes problemas estruturais associados às vibrações excessivas, decorrentes da concepção de sistemas estruturais com baixos níveis de amortecimento e com frequências naturais cada vez mais baixas e bastante próximas das faixas de frequência das excitações associadas às atividades humanas rítmicas. O modelo estrutural investigado baseiase em um piso misto (aço-concreto) submetido a aulas de ginástica aeróbica. A modelagem numérica do piso misto investigado foi realizada com base no emprego do programa ANSYS e foram utilizadas técnicas de discretização por meio do método dos elementos finitos (MEF). As cargas aplicadas sobre o piso, oriundas das atividades aeróbicas, são simuladas através de dois modelos de carregamentos dinâmicos distintos. Uma extensa análise paramétrica foi desenvolvida sobre o modelo estrutural investigado e a resposta dinâmica do sistema foi obtida, em termos dos deslocamentos e das acelerações, e comparada com os limites recomendados por normas e critérios de projeto. A resposta dinâmica do piso estudado viola os critérios de projeto relativos ao conforto humano e indica níveis de vibrações excessivas nos casos de carregamento dinâmicos analisados nesta dissertação.

Essa tese teve como objetivo avaliar a associação do consumo de padrões alimentares e produtos alimentícios ultraprocessados (UP) sobre indicadores antropométricos e sua variação entre adolescentes. Para tal, foram analisados os dados de 1.031 estudantes de quatro escolas privadas e duas escolas públicas da região metropolitana do Rio de Janeiro que foram acompanhados por três anos consecutivos (2010 a 2012) no Estudo Longitudinal de Avaliação Nutricional de adolescentes (ELANA). Os resultados estão apresentados em três manuscritos que compõem a tese. O primeiro manuscrito identificou três padrões alimentares: Ocidental, composto por massas, embutidos e bacon, carnes, doces, bebidas açucaradas, salgados e fast food, biscoito, pães e cereais, leite e derivados e ovos, o Saudável, caracterizado por peixes, hortaliças, verduras, raízes e feculentos, frutas e sucos e o Arroz/Feijão, composto pelo consumo de arroz, feijão e café. Na análise dos dados na linha de base o padrão Ocidental associou-se a um menor índice de massa corporal (IMC) e não foi observada associação estatisticamente significativa em relação ao percentual de gordura corpórea (%GC) e o padrão em questão. Adicionalmente, foi verificada uma associação negativa entre o padrão Arroz/Feijão e ambos os indicadores antropométricos sugerindo que esse padrão de consumo tradicional possa ser protetor para a obesidade. No segundo manuscrito avaliou-se a relação entre o consumo de UP e indicadores de qualidade da dieta. O consumo de UP foi avaliado em gramas por dia e em gramas/total de calorias. Observou-se um aumento das variáveis marcadoras de alimentação não saudável e saudável com o aumento do consumo de UP. No entanto, quando relacionados ao aumento de consumo de UP/energia observou-se um aumento de marcadores de alimentação não saudável e, com exceção da vitamina C, uma redução de marcadores de alimentação saudável. Assim, a densidade do consumo de UP mostrou-se um bom indicador de qualidade da dieta entre adolescentes. No terceiro manuscrito avaliou-se a associação entre o consumo de UP e a evolução temporal do IMC e %GC. A análise longitudinal por modelos lineares mistos, que levam em conta todas as observações ao longo do tempo, indicou que nas análises não ajustados havia uma associação negativa entre consumo de UP e o IMC e %GC, porém nas análises de UP/energia e nas análises ajustadas por atividade física e uma variável de controle para a qualidade do relato não mais foram observadas associações com o consumo de UP e as medidas antropométricas de adiposidade. Pode-se concluir que não evidenciamos associação do consumo de UP com medidas antropométricas de obesidade uma vez que o consumo elevado de UP associado ao crescimento do %GC nas análises transversais, não se manteve nas análises longitudinais. A discrepância entre os achados dos estudos seccionais e os longitudinais pode decorrer de confundimento residual nos estudos seccionais. Conclui-se, que para redução do ganho excessivo de peso, orientações relativas exclusivamente a um padrão de alimentação saudável que incorpora a ideia de UP, devem ser acompanhadas de redução da quantidade de alimentos consumidos.

[Objective] The research aimed to reveal the effects of excessive Mg2+ on the germination characteristics of maize and soybean. [Method] Eleven concentration gradients of Mg2+ including 0,40,80,120,160,200,240,280,320,360 and 400 mmol/L were set up to study the effects of different treatments on such indices as the germination potential,germination rate,fresh weight,dry weight,root length,plant height and stem diameter of maize and soybean. [Result] With the increasing of Mg2+ concentration,the germination of maize delayed and the germination rate of soybean obviously decreased,reaching the significant difference at 0.05 level. When Mg2+ concentration increased,the fresh weight and dry weight of maize and soybean decreased,but maize and soybean showed some adaptability. The growth of radicles and seedlings in maize and soybean were obviously inhibited by Mg2+,and atrophy and dysplasia phenomena appeared. [Conclusion] Excessive Mg2+ stress has different effects on the germination potential and germination rate of different crops and has obvious poisoning effects on the root and stem growth of crops.

基于车轮滑转率和车轮地面力学,研究了月球车在松软月面行驶时的车轮过度下陷问题.将月球车车轮下陷和车轮—土壤作用力表达为车轮滑转率的函数,结合车辆地面力学理论并考虑纵列式车轮多通过性土壤参数的修正,建立了月球车的动力学模型.判断车轮是否发生过度下陷的标准为土壤所提供给驱动轮的土壤推力能否克服土壤对车轮的阻力.利用建立的动力学模型,计算出能够保证车轮不会过度下陷的期望滑转率.考虑到月球车动力学系统的非线性和不确定性,设计了以车轮滑转率为状态变量的滑模驱动控制器.仿真结果表明,采用该控制器可以较快地跟踪期望滑转率,避免车轮的过度滑转下陷,保证月球车能够在软质路面上正常行驶.

结合车轮沙土相互作用的力学分析,研究解决轮式移动机器人沙地行驶车轮过度滑转下陷问题。考虑纵列式重复通过车轮沙土力学参数的修正,建立六轮式沙地移动机器人的动力学模型,以车轮滑转率为状态变量,设计了移动机器人沙地行驶的滑模驱动控制器跟踪车轮期望滑转率。MATLAB/Simulink仿真结果表明,采用该控制器可以较快地跟踪期望滑转率,有效地限制机器人车轮的滑转,避免车轮的过度下陷。

74 Front Street is intended as a musical response to the work of American sculptor Fred Sandback (1943-2003), in particular his string sculptures which are intentionally uncomplicated, minimal and strikingly beautiful. The relationship between these string works and music is rather distinct; indeed, before formally studying sculpture Sandback made a number of string instruments. He has said that his string works evolved from a desire to rid art of excessive decoration and, while there are instances of activity in 74 Front Street, much of the piece aims to reflect the simplicity of Sandback’s work.

Systemic lupus erythematosus (SLE) is a prototypical autoimmune disease characterized by polyclonal B cell activation and by the production of anti-double-stranded (ds) DNA antibodies. Given the inhibitory effects of IL-12 on humoral immune responses, we investigated whether IL-12 displayed such an activity on in vitro immunoglobulin production by SLE PBMC. Spontaneous IgG, IgG1, IgG2, IgG3 and IgM antibody production was dramatically reduced by addition of IL-12. These results were confirmed by Elispot assays detecting IgG- and anti-dsDNA-secreting cells. While IL-6 and TNF titres measured in PBMC supernatants were not modified by addition of IL-12, interferon-gamma (IFN-gamma) titres were up-regulated and IL-10 production down-regulated. Since addition of IFN-gamma did not down-regulate immunoglobulin production and since the inhibitory activity of IL-12 on immunoglobulin synthesis was not suppressed by anti-IFN-gamma antibody, we concluded that the effect of IL-12 on immunoglobulin production was not mediated through IFN-gamma. Our data also argue against the possibility that down-regulation of endogenous IL-10 production was responsible for the effect of IL-12. Thus, inhibition of IL-10 production by IFN-gamma was not accompanied by inhibition of immunoglobulin production, and conversely, restoration of IL-10 production by anti-IFN-gamma antibody did not suppress the inhibitory activity exerted by IL-12 on immunoglobulin production. Taken together, our data indicate that reduction of excessive immunoglobulin and anti-dsDNA antibody production by lupus PBMC can be achieved in vitro by IL-12, independently of IFN-gamma and IL-10 modulation.

In podiatric clinics is very common to find inflamatory process in metatarsophalangeal joint capsule , plantar plate and collateral ligaments damage, but it is not clearly recognized. Many authors hipotetized with joint instability of multiple aetiology and his concomitant evolution in different stages with own joint disease. This pathology has more incidence in second metatarsophalangeal joint than third and others and it is a common etiology of metatarsal pain. Bad biomechanics alters forefoot function and can produce overload in capsular joint, decreasing mobility and getting worse the pathology.
LEARNING IMPULSE CONTROL IN A NOVEL ANIMAL MODEL: SYNAPTIC, CELLULAR, AND PHARMACOLOGICAL SUBSTRATES

Impulse control, an executive process that restrains inappropriate actions, is impaired in numerous psychiatric conditions. This thesis reports three experiments that utilized a novel animal model of impulse control, the response inhibition (RI) task, to examine the substrates that underlie learning this task. In the first experiment, rats were trained to withhold responding on the RI task, and then euthanized for electrophysiological testing. Training in the RI task increased the AMPA/NMDA ratio at the synapses of pyramidal neurons in the prelimbic, but not infralimbic, region of the medial prefrontal cortex. This enhancement paralleled performance as subjects underwent acquisition and extinction of the inhibitory response. AMPA/NMDA was elevated only in neurons that project to the ventral striatum. Thus, this experiment identified a synaptic correlate of impulse control. In the second experiment, a separate group of rats were trained in the RI task prior to electrophysiological testing. Training in the RI task produced a decrease in membrane excitability in prelimbic, but not infralimbic, neurons as measured by maximal spiking evoked in response to increasing current injection. Importantly, this decrease was strongly correlated with successful inhibition in the task. Fortuitously, subjects trained in an operant control condition showed elevated infralimbic, but not prelimbic, excitability, which was produced by learning an anticipatory signal that predicted imminent reward availability. These experiments revealed two cellular correlates of performance, corresponding to learning two different associations under distinct task conditions. In the final experiment, rats were trained on the RI task under three conditions: Short (4-s), long (60-s), or unpredictable (1-s to 60-s) premature phases. These conditions produced distinct errors on the RI task. Interestingly, amphetamine increased premature responding in the short and long conditions, but decreased premature responding in the unpredictable condition. This dissociation may arise from interactions between amphetamine and underlying cognitive processes, such as attention, timing, and conditioned avoidance. In summary, this thesis showed that learning to inhibit a response produces distinct synaptic, cellular, and pharmacological changes. It is hoped that these advances will provide a starting point for future therapeutic interventions of disorders of impulse control.

BACKGROUND: Although severe encephalopathy has been proposed as a possible contraindication to the use of noninvasive positive-pressure ventilation (NPPV), increasing clinical reports showed it was effective in patients with impaired consciousness and even coma secondary to acute respiratory failure, especially hypercapnic acute respiratory failure (HARF). To further evaluate the effectiveness and safety of NPPV for severe hypercapnic encephalopathy, a prospective case-control study was conducted at a university respiratory intensive care unit (RICU) in patients with acute exacerbation of chronic obstructive pulmonary disease (AECOPD) during the past 3 years. METHODS: Forty-three of 68 consecutive AECOPD patients requiring ventilatory support for HARF were divided into 2 groups, which were carefully matched for age, sex, COPD course, tobacco use and previous hospitalization history, according to the severity of encephalopathy, 22 patients with Glasgow coma scale (GCS) <10 served as group A and 21 with GCS = 10 as group B. RESULTS: Compared with group B, group A had a higher level of baseline arterial partial CO2 pressure ((102 +/- 27) mmHg vs (74 +/- 17) mmHg, P <0.01), lower levels of GCS (7.5 +/- 1.9 vs 12.2 +/- 1.8, P <0.01), arterial pH value (7.18 +/- 0.06 vs 7.28 +/- 0.07, P <0.01) and partial O(2) pressure/fraction of inspired O(2) ratio (168 +/- 39 vs 189 +/- 33, P <0.05). The NPPV success rate and hospital mortality were 73% (16/22) and 14% (3/22) respectively in group A, which were comparable to those in group B (68% (15/21) and 14% (3/21) respectively, all P > 0.05), but group A needed an average of 7 cm H2O higher of maximal pressure support during NPPV, and 4, 4 and 7 days longer of NPPV time, RICU stay and hospital stay respectively than group B (P <0.05 or P <0.01). NPPV therapy failed in 12 patients (6 in each group) because of excessive airway secretions (7 patients), hemodynamic instability (2), worsening of dyspnea and deterioration of gas exchange (2), and gastric content aspiration (1). CONCLUSIONS: Selected patients with severe hypercapnic encephalopathy secondary to HARF can be treated as effectively and safely with NPPV as awake patients with HARF due to AECOPD; a trial of NPPV should be instituted to reduce the need of endotracheal intubation in patients with severe hypercapnic encephalopathy who are otherwise good candidates for NPPV due to AECOPD.

HOX genes are evolutionarily highly conserved. The HOX proteins which they encode are master regulators of embryonic development and continue to be expressed throughout postnatal life. The 39 human HOX genes are located in four clusters (A-D) on different chromosomes at 7p15, 17q21 [corrected] 12q13, and 2q31 respectively and are assumed to have arisen by duplication and divergence from a primordial homeobox gene. Disorders of limb formation, such as hand-foot-genital syndrome, have been traced to mutations in HOXA13 and HOXD13. Evolutionary conservation provides unlimited scope for experimental investigation of the functional control of the Hox gene network which is providing important insights into human disease. Chromosomal translocations involving the MLL gene, the human homologue of the Drosophila gene trithorax, create fusion genes which exhibit gain of function and are associated with aggressive leukaemias in both adults and children. To date 39 partner genes for MLL have been cloned from patients with leukaemia. Models based on specific translocations of MLL and individual HOX genes are now the subject of intense research aimed at understanding the molecular programs involved, and ultimately the design of chemotherapeutic agents for leukaemia. Investigation of the role of HOX genes in cancer has led to the concept that oncology may recapitulate ontology, a challenging postulate for experimentalists in view of the functional redundancy implicit in the HOX gene network.

Purpose. The authors conducted an in vitro investigation of the role of Ca2+-dependent signaling in vascular endothelial growth factor (VEGF)-induced angiogenesis in the retina.
Methods. Bovine retinal endothelial cells (BRECs) were stimulated with VEGF in the presence or absence of 1,2-bis(2-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid-acetoxymethyl ester (BAPTA-AM; intracellular Ca2+ chelator), U73122 (phospholipase C (PLC) inhibitor), xestospongin C (Xe-C), and 2-aminoethoxydiphenyl borate (2APB) (inhibitors of inositol-1,4,5 triphosphate (IP3) signaling). Intracellular Ca2+ concentration ([Ca2+]i) was estimated using fura-2 Ca2+ microfluorometry, Akt phosphorylation quantified by Western blot analysis, and angiogenic responses assessed using cell migration, proliferation, tubulogenesis, and sprout formation assays. The effects of the Ca2+/calmodulin-dependent protein kinase II (CaMKII) inhibitor KN93 were also evaluated on VEGF-induced Akt signaling and angiogenic activity.
Results. Stimulation of BRECs with 25 ng/mL VEGF induced a biphasic increase in [Ca2+]i, with an initial transient peak followed by a sustained plateau phase. VEGF-induced [Ca2+]i increases were almost completely abolished by pretreating the cells with BAPTA-AM, U73122, Xe-C, or 2APB. These agents also inhibited VEGF-induced phosphorylation of Akt, cell migration, proliferation, tubulogenesis, and sprouting angiogenesis. KN93 was similarly effective at blocking the VEGF-induced activation of Akt and angiogenic responses.
Conclusions. VEGF increases [Ca2+]i in BRECs through activation of the PLC-IP3 signal transduction pathway. VEGF-induced phosphorylation of the proangiogenic protein Akt is critically dependent on this increase in [Ca2+]i and the subsequent activation of CaMKII. Pharmacologic inhibition of Ca2+-mediated signaling in retinal endothelial cells blocks VEGF-induced angiogenic responses. These results suggest that the PLC/IP3/Ca2+/CaMKII signaling pathway may be a rational target for the treatment of angiogenesis-related disorders of the eye.

This open learning zone article examines acid-base balance and the interpretation of arterial blood gases (ABG). The article begins with a brief revision of related physiology which leads on to the description of the primary disorders of acid-base balance. The normal ranges and the significance of abnormal ABG results are explored. The article concludes by providing an easy to follow 4 step guide to ABG interpretation with practice examples presented in the CPD task section.
