The respiratory rates of mycelia of the mesophilic fungus, Aspergillus niger, and the thermophilic fungus, Thermomyces lanuginosus, were comparable at their respective temperature optima for growth. The respiratory rate of A. niger was independent of changes in temperature between 15 and 40 C. The respiratory rate of T. lanuginosus increased with increase in temperature between 25 and 55 C.

Mycotoxins are secondary metabolites of filamentous fungi. They pose a health risk to humans and animals due to their harmful biological properties and common occurrence in food and feed. Liquid chromatography/mass spectrometry (LC/MS) has gained popularity in the trace analysis of food contaminants. In this study, the applicability of the technique was evaluated in multi-residue methods of mycotoxins aiming at simultaneous detection of chemically diverse compounds. Methods were developed for rapid determination of toxins produced by fungal genera of Aspergillus, Fusarium, Penicillium and Claviceps from cheese, cereal based agar matrices and grains. Analytes were extracted from these matrices with organic solvents. Minimal sample clean-up was carried out before the analysis of the mycotoxins with reversed phase LC coupled to tandem MS (MS/MS). The methods were validated and applied for investigating mycotoxins in cheese and ergot alkaloid occurrence in Finnish grains. Additionally, the toxin production of two Fusarium species predominant in northern Europe was studied. Nine mycotoxins could be determined from cheese with the method developed. The limits of quantification (LOQ) allowed the quantification at concentrations varying from 0.6 to 5.0 µg/kg. The recoveries ranged between 96 and 143 %, and the within-day repeatability (as relative standard deviation, RSDr) between 2.3 and 12.1 %. Roquefortine C and mycophenolic acid could be detected at levels of 300 up to 12000 µg/kg in the mould cheese samples analysed. A total of 29 or 31 toxins could be analysed with the method developed for agar matrices and grains, with the LOQs ranging overall from 0.1 to 1250 µg/kg. The recoveries ranged generally between 44 and 139 %, and the RSDr between 2.0 and 38 %. Type-A trichothecenes and beauvericin were determined from the cereal based agar and grain cultures of F. sporotrichioides and F. langsethiae. T-2 toxin was the main metabolite, the average levels reaching 22000 µg/kg in the grain cultures after 28 days of incubation. The method developed for ten ergot alkaloids from grains allowed their quantification at levels varying from 0.01 to 10 µg/kg. The recoveries ranged from 51 to 139 %, and the RSDr from 0.6 to 13.9 %. Ergot alkaloids were measured in barley and rye at average levels of 59 and 720 µg/kg, respectively. The two most prevalent alkaloids were ergocornine and ergocristine. The LC/MS methods developed enabled rapid detection of mycotoxins in such applications where several toxins co-occurred. Generally, the performance of the methods was good, allowing reliable analysis of the mycotoxins of interest with sufficiently low quantification limits. However, the variation in validation results highlighted the challenges related to optimising this type of multi-residue methods. New data was obtained about the occurrence of mycotoxins in mould cheeses and of ergot alkaloids in Finnish grains. In addition, the study revealed the high mycotoxin-producing potential of two common fungi in Finnish crops. The information can be useful when risks related to fungal and mycotoxin contamination will be assessed.

A microbial survey of Jamnagar bauxite mines in Gujarat, India, revealed the indigenous presence of a variety of autotrophic and heterotrophic bacteria and fungi associated with the ore body and water ponds in the vicinity. Among these, bacteria belonging to the genera Thiobacillus, Bacillus and Pseudomonas are implicated in the weathering of aluminosilicates; the precipitation of iron oxyhydroxides; the dissolution and conversion of alkaline metal species; and the formation of alumina, silica and calcite minerals. Fungi belonging to the genus Cladosporium can reduce ferric iron and dissolve alumina silicates. Biogenesis thus plays a significant role in bauxite mineralization. Various types of bacteria and fungi, such as Bacillus polymyxa, Bacillus coagulans and Aspergillus niger, were found to be efficient in significant calcium solubilization and partial iron removal from bauxite ore. Probable mechanisms in the biobeneficiation process are analyzed. Biobeneficiation is shown to be an effective technique for the removal of iron and calcium from bauxite ores for use in refractories and ceramics.

A waste fungal biomass containing killed cells of Aspergillus niger was efficiently used in the removal of toxic metal ions such as nickel, calcium, iron and chromium from aqueous solutions. The role of different parameters such as initial metal ion concentration, solution pH and biomass concentration on biosorption capacity was established. The maximum metal uptake was found to be dependent on solution pH and increased with biomass loading upto 10g/L. The adsorption densities for various metal ions could be arranged as Ca>Cr (III)>Ni>Fe>Cr (VI). The effect of the presence of various metal ions in binary, ternary and quaternary combinations on biosorption was also assessed. Ni uptake was significantly affected, while that of Cr (VI) the least, in the presence of other metal ions. Uptake of base metals from an industrial cyanide effluent was studied using different species of fungi such as Aspergillus niger, Aspergillus terreus and Penicillium funiculosum and yeast such as Saccharomyces cerevisiae which were isolated from a gold mine. Traces of gold present in the cyanide effluent could be efficiently recovered. Among the four base metal contaminants present in the cyanide effluent, zinc was found to be most efficiently biosorbed, followed by iron, copper and lead. The role of both living and dead biomass on biosorption was distinguished and probable mechanisms illustrated.

In this Letter, we report the structure activity relationship (SAR) studies on series of positional isomers of 5(6)-bromo-1-(phenyl)sulfonyl]-2-(4-nitrophenoxy)methyl]-1H-benzim idazoles derivatives 7(a-j) and 8(a j) synthesized in good yields and characterized by H-1 NMR, C-13 NMR and mass spectral analyses. The crystal structure of 7a was evidenced by X-ray diffraction study. The newly synthesized compounds were evaluated for their in vitro antibacterial activity against Staphylococcus aureus, (Gram-positive), Escherichia coil and Klebsiella pneumoniae (Gram-negative), antifungal activity against Candida albicans, Aspergillus flavus and Rhizopus sp. and antitubercular activity against Mycobacterium tuberculosis H37Rv, Mycobacterium smegmatis, Mycobacterium fortuitum and MDR-TB strains. The synthesized compounds displayed interesting antimicrobial activity. The compounds 7b, 7e and 7h displayed significant activity against Mycobacterium tuberculosis H37Rv strain.

Immune responses during fungal infections are predominately mediated by 5/15-lipoxygenases (LO)-or cyclooxygenase (COX)-2-catalysed bioactive eicosanoid metabolites like leukotrienes, lipoxins and prostaglandins. Although few host mediators of fungi-triggered eicosanoid production have been established, the molecular mechanism of expression and regulation of 5-LO, 15-LO and COX-2 are not well-defined. Here, we demonstrate that, macrophages infected with representative fungi Candida albicans, Aspergillus flavus or Aspergillus fumigatus or those treated with Curdlan, a selective agonist of pattern recognition receptor for fungi Dectin-1, displays increased expression of 5-LO, 15-LO and COX-2. Interestingly, Dectin-1-responsive Syk pathway activates mTOR-sonic hedgehog (SHH) signaling cascade to stimulate the expression of these lipid metabolizing enzymes. Loss-of-function analysis of the identified intermediaries indicates that while Syk-mTOR-SHH pathway-induced 5-LO and 15-LO suppressed the Dectin-l-responsive pro-inflammatory signature cytokines like TNE-alpha, IL-1 beta and IL-12, Syk-mTOR-SHH-induced COX-2 positively regulated these cytokines. Dectin-1-stimulated IL-6, however, is dependent on 5-LO, 15-LO and COX-2 activity. Together, the current study establishes Dectin-1-arbitrated host mediators that direct the differential regulation of immune responses during fungal infections and thus are potential candidates of therapeutic intervention. Endophytic fungi isolated from Catharanthus roseus were screened for the production of vincristine and vinblastine. Twenty-two endophytic fungi isolated from various tissues of C. roseus were characterized taxonomically by sequence analysis of the internal transcribed spacer (ITS) region of rDNA and grouped into 10 genera: Alternaria, Aspergillus, Chaetomium, Colletotrichum, Dothideomycetes, Eutypella, Eutypa, Flavodon, Fusarium and Talaromyces. The antiproliferative activity of these fungi was assayed in HeLa cells using the MTT assay. The fungal isolates Eutypella sp-CrP14, obtained from stem tissues, and Talaromyces radicus-CrP20, obtained from leaf tissues, showed the strongest antiproliferative activity, with IC50 values of 13.5 mu g/ml and 20 mu g/ml, respectively. All 22 endophytic fungi were screened for the presence of the gene encoding tryptophan decarboxylase (TDC), the key enzyme in the terpenoid indole alkaloid biosynthetic pathway, though this gene could only be amplified from T. radicus-CrP20 (NCBI GenBank accession number KC920846). The production of vincristine and vinblastine by T. radicus-CrP20 was confirmed and optimized in nine different liquid media. Good yields of vincristine (670 mu g/l) in modified M2 medium and of vinblastine (70 mu g/l) in potato dextrose broth medium were obtained. The cytotoxic activity of partially purified fungal vincristine was evaluated in different human cancer cell lines, with HeLa cells showing maximum susceptibility. The apoptosis-inducing activity of vincristine derived from this fungus was established through cell cycle analysis, loss of mitochondrial membrane potential and DNA fragmentation patterns.

Con el objetivo de determinar los principales agentes etiológicos de las dermatopatías en conejos y determinar el efecto terapéutico de tres tratamientos fitoterapéuticos 1) Jícaro Sabanero, 2) madero negro y 3) Neem, sobre las mismas, se realizó un estudio en el Rancho Agropecológico en especies menores, Ebenezer (RAEME), Niquinohomo, Nicaragua. El trabajo se realizó en 59 conejos divididos en cuatro grupos y sometidos a los diferentes tratamientos. Para evaluar la efectividad de los tratamientos se realizaron 3 aplicaciones de cada uno de ellos a sus respectivos grupos, con intervalos de siete días cada uno, durante 21 días; a excepción del jícaro que se aplicó tres días continuos, cada siete días, durante 21 días. Se determinó que los principales agentes etiológicos presentes en el estudio, fueron el ácaro Listrophorus sp, los hongos Mucor sp, Aspergillus sp, zygomices sp, Penicillium sp. De los tres tratamientos aplicados, el tratamiento a base de Neem (Azadirachta indica) fue el que presentó un efecto altamente significativo (P< 0.0001)sobre el control de las dermopatías y permitió un mejor grado de recuperación de los conejos tratados. El tratamiento a base de jícaro (Crescentia alata) presenta algún grado de efectividad para el control de las dermopatías encontradas. El tratamiento a base de madero negro (Gliricidia sepium ), presentó muy poca efectividad en el control de las dermopatías en estudio.

Entre las problemáticas de la producción de sorgo en Nicaragua está el deficiente manejo postcosecha del grano, como consecuencia es afectado por diferentes plagas que dañan su calidad (insectos, hongos y bacterias). Algunos hongos que afectan el grano en campo y almacén son productores de diferentes micotoxinas, causando micotoxicosis (intoxicaciones), mortales para la salud humana y animal. El estudio se realizó con el objetivo de conocer la calidad fitosanitaria y presencia de niveles de aflatoxinas en granos almacenados. Se recolectaron muestras a nivel de empresas e industrias almacenadoras y a nivel de campo. A cada una de las muestras se le realizó análisis organoléptico (olor, temperatura, apariencia); físico (impurezas), entomológico, patológico (hongos y bacterias) y análisis de aflatoxinas por medio del método de minicolumna. Las plagas primarias encontradas en los granos de almacén fueron los géneros: Rhizopertha dominica ( F. ), Sitophillus oryzae ( L. ), las plagas secundarias son: Tribolium castaneum (Herbst), Orizaephillus surinamensis ( L. ), Cryptolestes sp , además se encontraron insectos depredadores como Orius sp. y parasitoides de la familia Bethylidae y Pteromalidae. A nivel de campo el insecto que se encontró con mayor número fue el telarañero del sorgo ( Celama sp). Los géneros de hongos Fusarium spp, Helminthosporium sp. y la bacteria Pseudomonas syringae ocasionaron los más altos porcentajes de infección en granos de almacén y campo. Se identificaron siete especies de Aspergillus, de las cuales las especies A. flavus Link y A. parasiticus Speare se asocian a la presencia de aflatoxinas; sin embargo, este análisis de aflatoxinas resultó por debajo de 20 partes por billón (ppb).

Las plagas primarias insectiles encontradas en los granos en almacén son: el pequeño barrenador del grano ( Rhizo pertha dominica (F.) y el gorgojo del arroz ( Sitophillus oryzae (L.)). El gorgojo plano de los granos ( Cryptolestes sp.), predominó en todas las muestras de granos de almacén, considerado plaga secundaria. En granos de campo el insecto que se encontró en mayor número fue el telarañero del sorgo ( Celama sorghiella (Riley)). Los géneros de hongos Fusarium spp, Helminthosporium sp y las bacterias Pseudo monas syringae van Hall y Bacillus megaterium (De Bary) ocasio naron los mas altos porcentajes de infección en granos de almacén y campo. Se identificaron siete especies de Aspergillus de las cuales A. flavus Link y A. parasiticus Speare se asocian a la producción de aflatoxinas, sin embargo este análisis resultó por debajo de 20 partes por billón (ppb).

En la Universidad Nacional Agraria (UNA) ubicada en el kilómetro 12 ½ carretera Norte Managua, en el año 2013 , se realizó el estudio en una plantación de guayaba establecida desde el 2009; ubicada en el área de la parcela agroecológica. La finalidad de la investigación consistió en realizar análisis microbiológico del suelo, evaluar el rendimiento y rentabilidad del cultivo de guayaba variedad taiwanesa, usando tres dosis de humus de lombriz 10, 15 y 20 t (ha año) -1, aplicadas cada dos meses , en el período lluvioso y época seca 2013 – 2014.El diseño de campo utilizado fue el de franjas emparejadas en un área de 240m2, donde se encuentran establecidas 27 plantas distribuidas en 3 surcos con 9 plantas cada surco. Las variables evaluadas para el análisis microbiológico fueron el número de unidades formadoras de colonias (UFC) para hongos, bacterias y actinomicetos, organismos identificados, entre ellos los hongos. Aspergillus sp y Penicillium sp ; en el género de bacterias: Bacillums sp, Sarcina flava y Actinomycetes. Con las dosis aplicadas de humus de lombriz, no hubo incremento de la diversidad de microorganismos en el suelo. Las variables del rendimiento evaluadas fueron número de frutos cosechados, peso de frutos, diámetro ecuatorial y diámetro polar. Para el análisis económico se utilizó la metodología de CIMMYT (1988) modificada. Se realizó un análisis paramétrico (ANDEVA con separación de medias según Tukey al 95 % de confianza): mostrando que no hubo diferencias estadísticas significativas para las variables evaluadas de rendimiento. Resultando la mayor rentabilidad en el tratamiento de 10 t (ha)-1: C$ 1,250,868.68 afirmando que el tratamiento 10 t (ha)-1 es más rentable por generar C$ 2.13 por cada córdoba invertido en comparación con los demás tratamientos de 15-20 t (ha año) -1 los que obtuvieron valores inferiores de C$ 1.23 y C$ 1.19 por cada córdoba invertido

Survey of Fungal infestation of some species of fish in Tagwai Dam Minna was carried out from March to June 2002. Fungi were isolated from the scale/skin, gills and fins. Twenty-one fungi species were identified from 18 species of fish microbial growth was measured by direct cell count using Stuart colony counter. Most of the fungi encountered were of the mould group and infestation occurred among all the species sampled. The infestation was predominantly by Aspergillus species and the scale/skin was most widely affected. The study showed the identified fungi in order of frequency to be as follows. Aspergillius niger. Rrhizopus spp. Mmucor spp Aspergilus flavum. Aspergillus parasitous. Aspergillus fumigatus, Microsporum canis. Penicillum virridicalumand Fusarium spp respectively. Aspergillus niger occurred on all the species of fish sampled. Barrilius spa and Chrysichrhy auratus longifilis had significantly higher (P<0.05) mean fungal load on their fins and gills. There was no significant difference (P>0.05) in the mean fungal load in different parts of the body of other fish species sampled

A qualidade do ar é um importante indicador de saúde ambiental, sendo o seu monitoramento contínuo necessário. Apesar da relevância do tema, há muitos países em que os limites de exposição para agentes biológicos ainda não foram estabelecidos ou foram definidos de forma inadequada, podendo comprometer a qualidade ambiental. Os ambientes hospitalares, assim como as salas de necropsia podem apresentar problemas de contaminação do ar por agentes microbiológicos, necessitando de monitoramento contínuo a fim de evitar a ocorrência de doenças nos trabalhadores e na população em geral. Este estudo realizou a avaliação microbiológica do ar em hospitais públicos e IMLs da região metropolitana do Rio de Janeiro em salas cirúrgicas e de necropsia. A pesquisa exploratória e descritiva baseou-se em levantamento bibliográfico e investigação de campo, através de estudos de casos. Os dados foram obtidos por meio de entrevistas e observação direta nos locais de trabalho, onde foram realizadas as avaliações microbiológicas do ar. As variações em salas cirúrgicas para bactérias e fungos foram respectivamente de 14,99 ufc/m3 88,29 ufc/m3 e de 45,93 ufc/m3 - 742,09 ufc/m3. Já nas salas de necropsia os valores para bactérias e fungos variaram respectivamente de 18,96 ufc/m3 54,9 ufc/m3 e de 144,87 ufc/m3 - 1152,01 ufc/m3. Foram identificados tanto no ambiente cirúrgico como nas salas de necropsia a presença dos seguintes fungos: Aspergillus sp., Neurospora sp., Penicillium sp., Fusarium sp., Cladosporium sp., Curvularia sp., e Trichoderma sp. Já em relação às bactérias foram identificadas as presenças de Staphilococcus sp., Streptococcus sp. e Micrococcus sp. Foram traçadas recomendações para melhoria da qualidade ambiental e do ar. Os resultados indicaram que os valores são elevados quando comparados com as recomendações das normas internacionais. Foram encontrados valores inferiores aos sugeridos pela CP n. 109 da ANVISA. A presença de microrganismos patogênicos sugere adoção de medidas de controle ambiental. O estudo apontou a necessidade urgente do estabelecimento de valores de referência para ambientes hospitalares no Brasil a fim de garantir condições seguras que não venham a comprometer a saúde dos pacientes e profissionais de saúde envolvidos.

The present communication is a survey report carried out to assess the incidence of toxic mycoflora on seven types of agriculture products/by products incorporated during fish culture as supplementary dietary items. Samples were obtained from various sources at Darbhanga, Madhubani and Samashtipur districts during summer, winter and monsoon months. Out of the total 1774 samples, only 894 appeared to be fresh visually reflecting average incidence of contamination around 46.6%. However, the apparently fresh samples, when subjected to culture, 26.9% of them were found to be contaminated. Thus, degree of fungal spoilage in feed ingredients in parts of north Bihar appears to be significantly high (73.5%). The present study illustrates the facts with special reference to Aspergillus flavus, A. parasiticus (elaborating aflatoxins) A. ochraceous, Penicilium viradicatuin (elaborating ochratoxins) and A. versicolor (elaborating sterigmatocystin). The other strains already known for their toxigenic potentials that appeared on the present substrates included A. niger, A. fumigatus, A. candidus, P. islandicum, Rhizopus spp. and Mucur spp. Studies indicate that the prevalent climatic factors like temperature and relative humidity facilitate a congenial condition almost all through the year and in particular during summer and monsoon months. But water content of the substrates is a vital factor that further accelerates the pace of mycobial spoilage. A thorough sun-drying of the agricultural commodities before prolonged storage to bring water content below the "low risk limit" may significantly reduce the incidence of molds.
