Résumé : Une dysrégulation de la lipolyse des tissus adipeux peut conduire à une surexposition des tissus non-adipeux aux acides gras non-estérifiés (AGNE), qui peut mener à un certain degré de lipotoxicité dans ces tissus. La lipotoxicité constitue, par ailleurs, l’une des causes majeures du développement de la résistance à l’insuline et du diabète de type 2. En plus de ses fonctions glucorégulatrices, l’insuline a pour fonction d’inhiber la lipolyse et donc de diminuer les niveaux d’AGNE en circulation, prévenant ainsi la lipotoxicité. Il n’y a pas d’étalon d’or pour mesurer la sensibilité de la lipolyse à l’insuline. Le clamp euglycémique hyperinsulinémique constitue la méthode étalon d’or pour évaluer la sensibilité du glucose à l’insuline mais il est aussi utilisé pour mesurer la suppression de la lipolyse par l’insuline. Par contre, cette méthode est couteuse et laborieuse, et ne peut pas s’appliquer à de grandes populations. Il existe aussi des indices pour estimer la fonction antilipolytique de l’insuline dérivés de l’hyperglycémie provoquée par voie orale (HGPO), un test moins dispendieux et plus simple à effectuer à grande échelle. Cette étude vise donc à : 1) Étudier la relation entre les indices de suppressibilité des AGNE par l’insuline dérivés du clamp et ceux dérivés de l’HGPO; et 2) Déterminer laquelle de ces mesures corrèle le mieux avec les facteurs connus comme étant reliés à la dysfonction adipeuse : paramètres anthropométriques et indices de dysfonction métabolique. Les résultats montrent que dans le groupe de sujets étudiés (n=29 femmes, 15 témoins saines et 14 femmes avec résistance à l’insuline car atteintes du syndrome des ovaires polykystiques), certains indices de sensibilité à l’insuline pour la lipolyse dérivés de l’HGPO corrèlent bien avec ceux dérivés du clamp euglycémique hyperinsulinémique. Parmi ces indices, celui qui corrèle le mieux avec les indices du clamp et les paramètres anthropométriques et de dysfonction adipeuse est le T50[indice inférieur AGNE] (temps nécessaire pour diminuer de 50% le taux de base – à jeun – des AGNE). Nos résultats suggèrent donc que l’HGPO, facile à réaliser, peut être utilisée pour évaluer la sensibilité de la lipolyse à l’insuline. Nous pensons que la lipo-résistance à l’insuline peut être facilement quantifiée en clinique humaine.

La littérature suggère que le sommeil paradoxal joue un rôle dans l'intégration associative de la mémoire émotionnelle. De plus, les rêves en sommeil paradoxal, en particulier leur nature bizarre et émotionnelle, semblent refléter cette fonction associative et émotionnelle du sommeil paradoxal. La conséquence des cauchemars fréquents sur ce processus est inconnue, bien que le réveil provoqué par un cauchemar semble interférer avec les fonctions du sommeil paradoxal. Le premier objectif de cette thèse était de reproduire conceptuellement des recherches antérieures démontrant que le sommeil paradoxal permet un accès hyper-associatif à la mémoire. L'utilisation d'une sieste diurne nous a permis d'évaluer les effets du sommeil paradoxal, comparativement au sommeil lent et à l’éveil, sur la performance des participants à une tâche sémantique mesurant « associational breadth » (AB). Les résultats ont montré que seuls les sujets réveillés en sommeil paradoxal ont répondu avec des associations atypiques, ce qui suggère que le sommeil paradoxal est spécifique dans sa capacité à intégrer les traces de la mémoire émotionnelle (article 1). En outre, les rapports de rêve en sommeil paradoxal étaient plus bizarres que ceux en sommeil lent, et plus intenses émotionnellement ; ces attributs semblent refléter la nature associative et émotionnelle du sommeil paradoxal (article 2). Le deuxième objectif de la thèse était de préciser si et comment le traitement de la mémoire émotionnelle en sommeil paradoxal est altéré dans le Trouble de cauchemars fréquents (NM). En utilisant le même protocole, nos résultats ont montré que les participants NM avaient des résultats plus élevés avant une sieste, ce qui correspond aux observations antérieures voulant que les personnes souffrant de cauchemars soient plus créatives. Après le sommeil paradoxal, les deux groupes, NM et CTL, ont montré des changements similaires dans leur accès associatif, avec des résultats AB-négatif plus bas et AB-positif plus grands. Une semaine plus tard, seul les participants NM a maintenu ce changement dans leur réseau sémantique (article 3). Ces résultats suggèrent qu’au fil du temps, les cauchemars peuvent interférer avec l'intégration de la mémoire émotionnelle pendant le sommeil paradoxal. En ce qui concerne l'imagerie, les participants NM avaient plus de bizarrerie et plus d’émotion positive, mais pas négative, dans leurs rêveries (article 4). Ces attributs intensifiés suggèrent à nouveau que les participants NM sont plus imaginatifs et créatifs à l’éveil. Dans l'ensemble, les résultats confirment le rôle du sommeil paradoxal dans l'intégration associative de la mémoire émotionnelle. Cependant, nos résultats concernant le Trouble de cauchemars ne sont pas entièrement en accord avec les théories suggérant que les cauchemars sont dysfonctionnels. Le groupe NM a montré plus d’associativité émotionnelle, de même que plus d'imagerie positive et bizarre à l’éveil. Nous proposons donc une nouvelle théorie de sensibilité environnementale associée au Trouble de cauchemar, suggérant qu'une sensibilité accrue à une gamme de contextes environnementaux sous-tendrait les symptômes uniques et la richesse imaginative observés chez les personnes souffrant de cauchemars fréquents. Bien que davantage de recherches doivent être faites, il est possible que ces personnes puissent bénéficier e milieux favorables, et qu’elles puissent avoir un avantage adaptatif à l'égard de l'expression créative, ce qui est particulièrement pertinent lorsque l'on considère leur pronostic et les différents types de traitements.

Background: Cervicocephalic kinesthetic deficiencies have been demonstrated in patients with chronic neck pain (NP). On the other hand, authors emphasized the use of different motion speeds for assessing functional impairment of the cervical spine. Purpose: The objectives of this study were (1) to investigate the head repositioning accuracy in NP patients and control subjects and (2) to assess the influence of target distance, motion speed, motion direction and pain. Materials and methods: Seventy-one subjects (36 healthy subjects and 35 NP patients; age 30–55 years) performed the head repositioning test (HRT) at two different speeds for horizontal and vertical movements and at two different distances. For each condition, six consecutive trials were sampled. Results: The study showed the validity and reproducibility of the HRT, confirming a dysfunctional threshold of 4.5°. Normative values of head repositioning error up to 3.6° and 7.1° were identified for healthy and NP subjects, respectively. A distance of 180 cm from the target and a natural motion speed increased HRT accuracy. Repositioning after extension movement showed a significantly larger error in both groups. Intensity, duration of pain as well as pain level did not significantly alter head repositioning error. Conclusions: The assessment of proprioceptive performance in healthy and NP subjects allowed the validation of the HRT. The HRT is a simple, not expensive and fast test, easily implementable in daily practice to assess and monitor treatment and evolution of proprioceptive cervical deficits.

Background: Friedreich ataxia (FRDA) is a progressive inherited neurodegenerative disorder caused by mutation of the FXN gene, resulting in decreased frataxin expression, mitochondrial dysfunction and oxidative stress. A recent study has identified shorter telomeres in FRDA patient leukocytes as a possible disease biomarker. Results: Here we aimed to investigate both telomere structure and function in FRDA cells. Our results confirmed telomere shortening in FRDA patient leukocytes and identified similar telomere shortening in FRDA patient autopsy cerebellar tissues. However, FRDA fibroblasts showed significantly longer telomeres at early passage, occurring in the absence of telomerase activity, but with activation of an alternative lengthening of telomeres (ALT)-like mechanism. These cells also showed accelerated telomere shortening as population doubling increases. Furthermore, telomere dysfunction-induced foci (TIF) analysis revealed that FRDA fibroblasts have dysfunctional telomeres. Conclusions: Our finding of dysfunctional telomeres in FRDA cells provides further insight into FRDA molecular disease mechanisms, which may have implications for future FRDA therapy.

O objetivo do presente trabalho foi estudar a relação entre representações parentais e traços disfuncionais de personalidade em sujeitos adultos da população não-clínica. Participaram 177 estudantes universitários, dos quais 104 eram mulheres e 73 eram homens, com idades entre os 18 e os 43 anos (M=22,66; DP=4,054). Foi aplicado o Millon Clínical Multiaxial Inventory-II, o Parental Bonding Instrument e o Object Relations lnventory. De acordo com os resultados, existe uma relação entre alguns tipos de traços disfuncionais da personalidade e experiências disruptivas na relação precoce com ambas as figuras parentais, como a falta de autonomia psicológica e instrumental sentida, e com determinados elementos das representações das figuras parentais, como a perceção de uma menor benevolência ou maior punição. Encontram-se também diferenças entre sujeitos do sexo masculino e do sexo feminino no que respeita ao padrão específico de correlações entre traços de personalidade e características da relação e das representações parentais. ABSTRACT: The purpose of this work was to study the relation between parental representations and dysfunctional personality traits in adult subjects from a non-clinical population. Participants were 177 university students, 104 of which were female and 73 were men. Ages ranged from 18 to 43 years old (M=22,66; DP=4,054). The Millon Clinical Multiaxial Inventory-II, the Parental Bonding lnstrument and the Object Relations Inventory were administered. According to the results, there is a relation between certain types of dysfunctional personality traits and disruptive experiences in the early relations with both caregivers, like a perceived lack of psychological and instrumental autonomy, and with certain elements of parental representations, like the perception of less benevolence or a more punitive attitude. Differences between male and female subjects, concerning the specific pattern of correlations between personality traits and parental relations' and representations' characteristics, were also obtained.

The hopelessness depression (HD) theory assumes a number of contributing factors to the development of hopelessness expectations and HD symptoms. Studies carried out to test HD theory have generally supported the prediction that a negative attributional style contributes to the development of hopelessness expectations and HD symptoms, although the evidence is less consistent for adolescents. Due to these inconsistent results, as well as the assumption advanced by HD theory that other factors aside from attributional style may contribute to the development of HD, in this study we examined the potential role of adolescents’ daily stress and coping styles as contributing factors to HD at these ages, and the effects of gender on these relationships. Seiffge-Krenke (1995) showed that stress in daily life plays a particularly relevant role during adolescence and described three coping styles used by adolescents to face daily stress: active and internal approach-oriented styles, which are considered functional and complementary, and avoidant style, considered dysfunctional. In this study, secondary students (N = 480; aged 13–17) completed the Hopelessness Scale (Beck, Weissman, Lester, & Trexler, 1974), the Hopelessness Depression Symptoms Questionnaire (Metalsky & Joiner, 1997), the Problem Questionnaire ((Seiffge-Krenke, 1995) and the Coping Across Situations Questionnaire (Seiffge-Krenke, 1995). To test the role of daily stress and coping styles in the prediction of hopelessness expectations and HD symptoms, two four-step hierarchical multiple regression analyses were conducted. In these analyses, gender was introduced first, daily stress was introduced second, the three coping styles were entered in a third step, and the moderating effects of gender on the associations of the predictor variables (daily stress and each coping style) with the criterion variables (hopelessness expectations or HD symptoms) were introduced in a fourth step. Results showed a moderating effect of gender on the relationship between daily stress and hopelessness expectations, which revealed a significant effect for boys despite the fact that girls experienced more daily stress than boys, thus suggesting an inoculation effect in girls. It was also found a gender-dependent role of coping styles in the prediction of HD symptoms, revealing a protective effect for the active style in girls and for the internal style in boys. This suggests that girls would benefit from being more action-prone and boys more reflection-prone in order to prevent HD. These findings indicate that programmes aimed to promote coping skills in adolescents would benefit from being gender-adapted.

SOUTH ROAD, a novel told in third-person limited, follows Adrienne Harris as she navigates the trials of her coming-of-age summer and then must deal with the aftermath. 1997: seventeen-year-old Adrienne Harris wants nothing more than to flee her eccentric grandmother’s rule and leave Harbor Point and never look back. When she meets her new neighbors, Adrienne knows her life will never be the same. Adrienne quickly falls in love with the charismatic Quinn Merritt. They decide to keep their relationship a secret since both families disapprove. This secret starts a chain reaction that seemingly leads to the suicide of the troubled and poetic Lucas Merritt. The summer culminates with Adrienne running away, pregnant and heartbroken. 2011: thirty-one-year-old Adrienne is an out of work line cook and single mother. The story opens as Adrienne reluctantly returns home to Harbor Point to care for her ailing grandmother. Once home, Adrienne has to confront the things that haunt her—the summer she met and lost both Merritt brothers, and also her dysfunctional relationship with her grandmother—in order to heal and repair her own life and her relationship with her daughter. In the end, Adrienne discovers many truths that alter her perception of her past in Harbor Point. Adrienne is finally able to move forward and start to build a life for her and her daughter. Harbor Point, the last place in the world Adrienne Harris wanted to be, turns out to be the only place she wants to call home.

Social capital, or social cohesion or group connectedness, can influence both HIV risk behavior and substance use. Because recent immigrants undergo a change in environment, one of the consequences can be a change in social capital. There may be an association among changes in social capital, and HIV risk behavior and substance use post immigration. The dissertation focused on the interface of these three variables among recent Latino immigrants (RLIs) in South Florida. The first manuscript is a systematic review of social capital and HIV risk behavior, and served as a partial background for the second and third manuscripts. Twelve papers with a measure of social capital as an independent variable and HIV risk as the dependent variable were included in the analysis. Eleven studies measured social capital at the individual level, and one study measured social capital at the group level. HIV risk was influenced by social capital, but the type of influence was dependent on the type of social capital and on the study population. Cognitive social capital, or levels of collective action, was protective against HIV in both men and women. The role of structural social capital, or levels of civic engagement/group participation, on HIV risk was dependent on the type of structural social capital and varied by gender. Microfinance programs and functional group participation were protective for women, while dysfunctional group participation and peer-level support may have increased HIV risk among men. The second manuscript was an original study assessing changes in social capital and HIV risk behavior pre to post immigration among RLIs in South Florida (n=527). HIV risk behavior was assessed through the frequency of vaginal-penile condom use, and the number of sexual partners. It was a longitudinal study using secondary data analysis to assess changes in social capital and HIV risk behavior pre immigration to two years post immigration, and to determine if there was a relationship between the two variables. There was an 8% decrease in total social capital (p ˂ .05). Reporting of ‘Never use’ of condoms in the past 90 days increased in all subcategories (p ˂ .05). Single men had a decrease in number of sexual partners (p ˂ .05). Lower social capital measured on the dimension of ‘friend and other’ was marginally associated with fewer sexual partners. The third manuscript was another original study looking at the association between social capital and substance use among RLIs in South Florida (n=527). Substance use with measured by frequency of hazardous alcoholic drinking, and illicit drug use. It was a longitudinal study of social capital and substance-use from pre to two years post immigration. Post-immigration, social capital, hazardous drinking and illicit drug use decreased (p˂.001). After adjusting for time, compared to males, females were less likely to engage in hazardous drinking (OR=.31, p˂.001), and less likely to engage in illicit drug use (OR=.67, p=.01). Documentation status was a moderator between social capital and illicit drug use. ‘Business’ and ‘Agency’ social capital were associated with changes in illicit drug use for documented immigrants. After adjusting for gender and marital status, on average, documented immigrants with a one-unit increase in ‘business’ social capital were 1.2 times more likely to engage in illicit drug use (p˂.01), and documented immigrants with one-unit increase in ‘agency’ social capital were 38% less likely to engage in illicit drug use (p˂.01). ‘Friend and other’ social capital was associated with a decrease in illicit drug use among undocumented immigrants. After adjusting for gender and marital status, on average, undocumented immigrants with a one-unit increase in ‘friend and other’ social capital were 45% less likely to engage in hazardous drinking and 44% less likely to use illicit drugs (p˂.01, p˂.05). Studying these three domains is relevant because HIV continues to be a public health issue, particularly in Miami-Dade County, which is ranked among other U.S. regions with high rates of HIV/AIDS prevalence. Substance use is associated with HIV risk behavior; in most studies, increased substance use is associated with increased chances of HIV risk behavior. Immigration, which is the hypothesized catalyst for the change in social capital, has an impact on the dynamic of a society. Greater immigration can be burdensome on the host country’s societal resources; however immigrants are also potentially a source of additional skilled labor for the workforce. Therefore, successful adaption of immigrants can have a positive influence on receiving communities. With Florida being a major receiver of immigrants to the U.S, this dissertation attempts to address an important public health issue for South Florida and the U.S. at large.

Syftet med studien var att jämföra grad av coping samt grad av livstillfredsställelse mellan religiösa personer och de som anser sig vara icke troende. För att testa skillnaderna mellan religiösa och ateister gjordes stickprov med data från 30 religiösa och 103 ateister. Mätinstrumentet REI skapades för att mäta graden av tro eller icke tro i avseende att jämföra resultatet mot den trosuppfattning som deltagarna själva hade angivit. Ett Mann–Whitney U test visade att det inte fanns en signifikant skillnad mellan deltagarnas självskattning av tro och resultatet från REI. Resultat från oberoende t-test avseende livstillfredsställelse visar att religiösa och ateister inte signifikant skilde sig åt. Signfikanta skillnader återfanns i oberoende t-test avseende emotionsbaserad, problemfokuserad och dysfunktionell coping. De religiösa och ateistiska grupperna skiljde sig mest i emotionsbaserad coping som hade högst effektstyrka enligt Cohens d (d = 1,39) i jämförelse med effektstyrkan för problemfokuserad och dysfunktionell coping.

This thesis explores perceptions and preferences on regional action in EU-related frameworks among regional actors in Western Sweden. Building upon the literature on Europeanisation and the Fusion approach, three dimensions of Europeanisation are clarified and explored– download, upload and crossload – and together with a set of five variables that constitute the Micro Fusion Framework; a comprehensive analytical tool is developed. The thesis analyses the intense debate among the members of West Sweden that took place from 2011 to 2013 that focused on how to functionally organise the regional office in Brussels in order to meet future challenges. Surprisingly, the members eventually decided to terminate their cooperation and close the jointly owned office in Brussels in spite of the fact that it has been widely regarded as successful and effective. Diverging perceptions and preferences is understood in terms of three positions on regional action; a download-, upload- and a coherent oriented position. Finally, the thesis presents the empirical findings and discusses in relation to three fusion scenarios, infusion, defusion and clustered fusion. In terms of Micro Fusion Framework, the dynamics shaping why West Sweden was finally regarded as a dysfunctional arena for regional action are explained by a shift of attention and action among regional actors in Western Sweden that led to pressure for further institutional adaptation in order to meet the demand of how ‘to get the best out of the EU’. Further, this redefinition of how to handle EU-affairs within the upload-oriented position was accompanied by positive attitudes towards the potential to bypass the state and thereby pursue regional priorities directly in Brussels given the compound nature of the EU. In contrast, those regional actors that are found to be more download-oriented often question the benefits of uploading activities in practice and advocate close relations to the state. A coherent oriented position recognises the importance of activities related to both of the vertical dimensions of Europeanisation.

The m-AAA protease is a hexameric complex involved in processing of specific substrates and turnover of misfolded polypeptides in the mitochondrial inner membrane. In humans, the m-AAA protease is composed of AFG3L2 and paraplegin. Mutations in AFG3L2 have been implicated in dominant spinocerebellar ataxia (SCA28) and recessive spastic ataxia-neuropathy syndrome (SPAX5). Mutations of SPG7, encoding paraplegin, are linked to hereditary spastic paraplegia. In the mouse, a third subunit AFG3L1 is expressed. Various mouse models recapitulate the phenotype of these neurodegenerative disorders, however, the pathogenic mechanism of neurodegeneration is not completely understood. Here, we studied several mouse models and focused on cell-autonomous role of the m-AAA protease in neurons and myelinating cells. We show that lack of Afg3l2 triggers mitochondrial fragmentation and swelling, tau hyperphosphorylation and pathology in Afg3l2 full-body and forebrain neuron-specific knockout mice. Moreover, deletion of Afg3l2 in adult myelinating cells causes early-onset mitochondrial abnormalities as in the neurons, but the survival of these cells is not affected, which is a contrast to early neuronal death. Despite the fact that myelinating cells have been previously shown to survive respiratory deficiency by glycolysis, total ablation of the m-AAA protease by deleting Afg3l2 in an Afg3l1 null background (DKO), leads to myelinating cell demise and subsequently progressive axonal demyelination. Interestingly, DKO mice show premature hair greying due to loss of melanoblasts. Together, our data demonstrate cell-autonomous survival thresholds to m-AAA protease deficiency, and an essential role of the m-AAA protease to prevent cell death independent from mitochondrial dynamics and the oxidative capacity of the cell. Thus, our findings provide novel insights to the pathogenesis of diseases linked to m-AAA protease deficiency, and also establish valuable mitochondrial dysfunctional mouse models to study other neurodegenerative diseases, such as tauopathies and demyelinating diseases.

Pretendemos estudar quais os traços desadaptativos de personalidade que se relacionam com os sintomas depressivos, a afectividade positiva e negativa, numa amostra de 187 estudantes universitários, entre os 18 e os 43 anos. Calculámos as correlações entre as escalas de padrões de personalidade e as escalas de padrões graves de personalidade do Inventário Multiaxial clínico de Millon II (MCMI-II) com os sintomas depressivos avaliados pela Escala de Depressão do Centro de Estudos Epidemiológicos e com a afectividade positiva e negativa avaliadas pela Escala de Afectividade Positiva e Afectividade Negativa. Realizámos uma análise da regressão múltipla utilizando as escalas de padrões de personalidade do MCMI-II como predictores e o resultado na CES-O, como variável dependente. Obtivemos uma correlação significativa entre os padrões de personalidade borderline, masoquista, esquizotípico, negativista e evitante e paranóide e os sintomas depressivos. A afectividade negativa apresenta-se também positivamente correlacionada com os padrões dependente e esquizóide. A afectividade positiva correlacionou-se significativa e positivamente com os padrões histriónico e narcisista e negativamente com os padrões esquizóide, esquizotípico, negativista, borderline, evitante e masoquista. Na análise de regressão, o padrão borderline prevê a presença de sintomas depressivos e os padrões sádico e dependente também, mas num sentido negativo. /ABSTRACT: We intendend to study wich personality traits relate to depressiva symptoms, and positive and negative affectivity, in a sample of 187 university students, between 18 and 43 years. We did the correlation between the personality patterns scales and the personality patterns of severe personality with the depressive symptoms measured by the Center for Epidemiologic Studies Depression Scale and the positive and negative affectivity measured by the Positive Affect and Negative Affect Shedule. We did a multiple regression analysis using the personality patterns scale of Millon Clinical Multiaxial lnventory (MCMI-II) as predictors and the result of CES-D, as dependent variable. We obtained a significant correlation between the borderline, masochist, schizotypal, negativistic, avoidant and paranoid personality patterns and the depressive symptoms. Negative affectivity relates also positively with dependent and schizoid patterns. Positive affectivity correlated significant and positively with histrionic and narcissistic personality patterns and negatively with schizoid, schizotypal, negativistic, borderline, avoidant and masochist personality patterns. ln the regression analysis, the borderline pattern predicts the presence of depressive symptoms as well as the sadistic and dependent, but in the negative sense.
