Two experiments were conducted to measure urea recycling and rumen flow dynamics in young rusa deer fed low (LP) or high (HP) protein diets. Pool size and flux rate of labelled urea. into and out of the blood pool were measured using single intravenous (i.v.) injection solutions containing [C-14] - and [N-15]-urea. A curve peeling technique was used to fit the enrichment of N-15 or specific radioactivity (SRA) of C-14 to exponential equations. Body urea-N pool size was significantly greater (P < 0.05) when a HP, compared to a LP diet, was fed. Urea space, expressed as a percent of live weight, total flux rate of urea through the blood pool and the irreversible loss of urea was similar for both diets. The mean (+/- S.E.M.) concentration of plasma urea-N was greater when animals were fed the HP diet compared to the LP diet (2 1.1 +/- 0.3 versus 14.4 +/- 1.4 mg/100 ml, respectively). Voluntary feed intake and digestibility of dietary components were also measured. Daily dry matter intakes were not affected by the crude protein (CP) content of the diet, although apparent DM digestibility was significantly greater for HP diet fed in both experiments. An intraruminal infusion of CrEDTA was used to determine rumen flow dynamics. Ruminal mean retention time, relative net outflow rate of water and passage rate constant (k(w)) were significantly greater (P < 0.05) when the HP diet was fed compared to the LP diet. The extent of urea metabolism and flux rates of urea between the blood and secondary pools appear similar to those previously reported for other ruminants fed diets contrasting in CP content. The effects of three vehicles, phosphate-buffered saline (PBS), ethanol (50% in PBS w/w) and propylene glycol (50% in PBS w/w) on in vitro transdermal penetration of testosterone was investigated in the horse. Skin was harvested from the thorax of five Thoroughbred horses after euthanasia and stored at -20 degrees C until required. The skin was then defrosted and placed into Franz-type diffusion cells, which were maintained at approximately 32 degrees C by a water bath. Saturated solutions of testosterone, containing trace amounts of radiolabelled [C-14]testosterone, in each vehicle were applied to the outer (stratum corneum) surface of each skin sample and aliquots of receptor fluid were collected at 0, 2, 4, 8, 16, 20, 22 and 24 h and analysed for testosterone by scintillation counting. The maximum flux (J(max)) of testosterone was significantly higher for all sites when testosterone was dissolved in a vehicle containing 50% ethanol or 50% propylene glycol, compared to PBS. In contrast, higher residues of testosterone were found remaining within the skin when PBS was used as a vehicle. This study shows that variability in clinical response to testosterone could be expected with formulation design.

The effects of the vehicles phosphate-buffered saline (PBS), ethanol (EtOH; 50% in PBS w/w) and propylene glycol (PG; 50% in PBS w/w) and the region of administration on in vitro transdermal penetration of testosterone was investigated in the dog. Skin was harvested from the thorax, neck (dorsal part) and groin regions of greyhounds after euthanasia and stored at -20 degrees C until required. The skin was then de-frosted and placed into Franz-type diffusion cells which were maintained at approximately 32 degrees C by a water-bath. Saturated solutions of testosterone, containing trace amounts of radiolabelled (C-14) testosterone, in each vehicle were applied to the outer (stratum corneum) surface of each skin sample and aliquots of receptor fluid were collected at 0, 2, 4, 8, 16, 20, 22 and 24 h and analysed for testosterone by scintillation counting. The maximum flux (J(max)) of testosterone was significantly higher for all sites when dissolved in a vehicle containing 50% EtOH or 50% PG, compared to PBS. In contrast, higher residues of testosterone were found remaining within the skin when PBS was used as a vehicle. The degree to which palaeoclimatic changes in the Southern Hemisphere co-varied with events in the high latitude Northern Hemisphere during the Last Termination is a contentious issue, with conflicting evidence for the degree of 'teleconnection' between different regions of the Southern Hemisphere. The available hypotheses are difficult to test robustly, however, because there are few detailed palaeoclimatic records in the Southern Hemisphere. Here we present climatic reconstructions from the southwestern Pacific, a key region in the Southern Hemisphere because of the potentially important role it plays in global climate change. The reconstructions for the period 20-10 kyr BP were obtained from five sites along a transect from southern New Zealand, through Australia to Indonesia, supported by 125 calibrated C-14 ages. Two periods of significant climatic change can be identified across the region at around 17 and 14.2 cal kyr BP, most probably associated with the onset of warming in the West Pacific Warm Pool and the collapse of Antarctic ice during Meltwater Pulse-1A, respectively. The severe geochronological constraints that inherently afflict age models based on radiocarbon dating and the lack of quantified climatic parameters make more detailed interpretations problematic, however. There is an urgent need to address the geochronological limitations, and to develop more precise and quantified estimates of the pronounced climate variations that clearly affected this region during the Last Termination. The bioavailability of iron, in combination with essential macronutrients such as phosphorus, has been hypothesised to be linked to nuisance blooms of the toxic cyanobacterium Lyngbya majuscula. The present laboratory study used two biological assay techniques to test whether various concentrations of added iron (inorganic and organically chelated) enhanced L. majuscula filament growth and productivity (C-14-bicarbonate uptake rate). Organically chelated iron (FeEDTA) with adequate background concentrations of phosphorus and molybdenum caused the largest increases (up to 4.5 times the control) in L. majuscula productivity and filament growth. The addition of inorganic iron (without added phosphorus or molybdenum) also stimulated L. majuscula filament growth. However, overall the FeEDTA was substantially and significantly more effective in promoting L. majuscula growth than inorganic iron (FeCl3). The organic chelator (EDTA) alone and molybdenum alone also enhanced L. majuscula growth but to a lesser extent than the chelated iron. The results of the present laboratory study support the hypothesis that iron and chelating organic compounds may be important in promoting blooms of L. majuscula in coastal waters of Queensland, Australia.

Research surrounding the transition from II to I millennium cal BC in Eastern Iberian Peninsula has a large and extensive tradition of investigation. However, the chances to do research on this historical process have been limited by the lack of a well contextualized and dated stratigraphic sequence. For this reason, recent studies in this topic have followed the periodic proposals which were developed in closer regions and areas, especially in the South East and North East of the Peninsula. The investigation perspective about the Late Bronze in Eastern Iberian has however now improved, with the development of several archaeological investigations, the increase in the number of sites being dated and more recent studies into the region helping to bring about this change. As such, it is now in the correct state to be able to propose a new periodization and delve into the changes which occurred in the transition between 1500 and 725 cal BC.

Durante el siglo XIII se produjo una sucesión de revueltas que supuso la desaparición del Imperio almohade y su sustitución por poderes regionales en al-Andalus, el Magreb y el Magreb al-Aqsà. La historiografía ha presentado el surgimiento y pugna entre estos poderes como un fenómeno social, político e, incluso, cultural y religioso, con el que se ha podido explicar su aniquilación o marginalización. Este trabajo pretende contextualizar los hechos desde una perspectiva medioambiental, de forma que la desintegración del califato almohade, el surgimiento de aquellos poderes y la progresión de los reinos cristianos en la península ibérica puedan entenderse desde una visión global de cambio climático y una posible crisis agrícola.

The not criminally responsible on account of mental disorder (NCRMD) defence is used when claims can be made that offenders are not responsible for their actions due to symptoms of a mental disorder. Bill C-14, now enacted in Canada, has implemented changes making it more difficult for NCRMD defendants to be released back into the public. This enactment appears to have been primarily due to public perceptions rather than actual knowledge of the defence. Thus it seems important to assess what members of the public actually know about the defence. To assess this, 127 participants completed a survey assessing their knowledge of the illnesses generally involved in the NCRMD defence, crimes committed, and punishments received. On average, only 31.6% of responses were answered within 20% of the factual statistics. Results suggest a general lack of knowledge about the defence and demonstrate why important changes should be based on factual information rather than public opinion.

Este trabalho apresenta a síntese e caracterização de cinco ligantes e quatro complexos derivados de oximas e tiossemicarbazonas. Entre essas, discutem-se as estruturas cristalinas/moleculares determinadas por difração de raios-X em monocristais: do ligante 4-feniltiossemicarbazida-isatina (Ligante 5), do complexo piridina-salicilaldeído-4- feniltiossemicarbazona de niquel (II) (Complexo 1), e do bis-4-feniltiossemicarbazonaisatina de chumbo(II) (Complexo 2). A estrutura do Ligante 5 cristaliza no sistema monoclínico, grupo espacial P21/c, com parâmetros de cela a = 6,3227(2) Å, b = 15,7973(7) Å, c = 14,4572(6) Å, β = 93,9330(10)°, V = 1440,61(10) Å3 , Z = 4. O refinamento da estrutura convergiu aos índices de discordância finais R1 = 0,0520, wR2 = 0,1471. Observa-se ainda a ocorrência de interações intermoleculares do tipo ligações de hidrogênio clássicas [N18−H3---O1′ 2,907(2)Å], com a formação de estruturas dímeras inter-relacionadas por simetria dentro da cela cristalina. Para a estrutura cristalina do Complexo 1, observa-se NC=4, e geometria de coordenação quadrada plana, onde o ligante saliciladeído-4-feniltiossemicarbazida comporta-se como quelante tridentado, e completando a esfera de coordenação do centro metálico temos uma molécula de piridina. A estrutura cristaliza no sistema monoclínico, grupo espacial P21/m, parâmetros de cela a = 12,8211(2) Å, b = 5,73370(10) Å, c = 23,9950(4) Å, β = 101,0910(10)°, V = 1730,98(5) Å3 , índices de discordância finais R1= 0,0320, wR2 = 0,0888, Z=3. O Complexo 1 apresenta ainda interações intermoleculares do tipo [N(3)-H(3)---S(1) = 3,5838(17)º, N(3)–H(3A)---S(1) = 160,91(19)º], formando estruturas dímeras e ligação de hidrogênio intramolecular não-clássica do tipo [C(10)-H(10)---N(2) = 2,838(2)º e C(10) – H(10)---N(2) = 122º]. A estrutura cristalina do complexo 2, apresenta duas formas independentes (uma com centro representado por Pb1 e outra por Pb2). Para a unidade com Pb1 temos o complexo composto por duas unidades do Ligante 5, que comportam-se como quelantes tridentados, e a esfera de coordenação é completada por interações intermoleculares do tipo η 2 areno π e através da ligação polarizada com o O1 da moléculas vizinha, o que confere ao íon Pb1 NC=9. A unidade Pb2 apresenta apenas as duas unidades do Ligante 5 coordenadas conferindo-lhe NC=6. A estrutura cristaliza no sistema monoclínico, grupo espacial C2/c, parâmetros de cela a = 37,9747(6) Å, b= 9,51280(10) Å, c = 31,4378(5) Å, β = 125,951(2)°, V= 9193,5(2) Å3 , Z = 4, índices de discordância finais= R1 = 0,0643, wR2 = 0,1227.

A novel, anaerobic, chemo-organotrophic bacterium, designated strain Ra1766HT, was isolated from sediments of the Guaymas basin (Gulf of California, Mexico) taken from a depth of 2002 m. Cells were thin, motile, Gram-stain-positive, flexible rods forming terminal endospores. Strain Ra1766H(T) grew at temperatures of 25-45 degrees C (optimum 30 degrees C), pH 6.7-8.1 (optimum 7.5) and in a salinity of 5-60 g l(-1) NaCl (optimum 30 g l(-1)). It was an obligate heterotrophic bacterium fermenting carbohydrates (glucose and mannose) and organic acids (pyruvate and succinate). Casamino acids and amino acids (glutamate, aspartate and glycine) were also fermented. The main end products from glucose fermentation were acetate, butyrate, ethanol, H-2 and CO2. Sulfate, sulfite, thiosulfate, elemental sulfur, fumarate, nitrate, nitrite and Fe(III) were not used as terminal electron acceptors. The predominant cellular fatty acids were C-14 : 0, C-16:1 omega 7, C-16:1 omega 7 DMA and C-16:0. The main polar lipids consisted of phosphatidylglycerol, diphosphatidylglycerol, phosphatidylethanolamine and phospholipids. The G +C content of the genomic DNA was 33.7 molo/o. Phylogenetic analysis of the 16S rRNA gene sequence indicated that strain Ra1766H(T) was affiliated to cluster XI of the order Clostridia les, phylum Firmicutes. The closest phylogenetic relative of Ra1766H(T) was Geosporobacter subterraneus (94.2% 16S rRNA gene sequence similarity). On the basis of phylogenetic inference and phenotypic properties, strain Ra1766H(T) (=DSM 27501(T)=JCM 19377(T)) is proposed to be the type strain of a novel species of a novel genus, named Crassaminicella pro funda.

The marine dinoflagellate genus Dinophysis includes species that are the causative agents of diarrhetic shellfish poisoning (DSP). Recent findings indicate that some Dinophysis species are mixotrophic, i.e. capable of both autotrophic and heterotrophic nutrition. We investigated inorganic (and organic) carbon uptake by several species of Dinophysis in the Light and dark using the 'single-cell C-14 method', and compared uptake rates with those of photosynthetic Ceratium species and heterotrophic dinoflagellates in the genus Protoperidinium. Experiments were conducted with water from the Gullmar Fjord and from the Koster Strait (Swedish west coast). Nutrient-enriched phytoplankton from surface water samples were concentrated (20 to 70 mu m) and incubated at in situ temperature under artificial light conditions with high concentrations of inorganic C-14 (1 mu Ci ml(-1)). Individual cells of each desired species were manually isolated under a microscope and transferred to scintillation vials. C. tripes showed net C-14 uptake only during light periods, whereas both C. lineatum and C. furca showed C-14 uptake in the Light as well as uptake (and sometimes losses) in the dark. Dinophysis species had similar carbon fixation rates in Light compared to Ceratium species. For D. acuminata and D. norvegica, net carbon uptake occurred in both Light and dark periods. D. acuta showed a loss of carbon in the dark in one experiment, but in another, dark C uptake was significantly higher than uptake in Light. When exposed to Light, C. furca, D. norvegica and D. acuta had high specific carbon uptake rates. Growth rates for the different species were calculated from C-14 uptake by the cells during the first hours of incubation in light. D. acuminata and D. norvegica had similar maximum growth rates, 0.59 and 0.63 d(-1) (mu); the maximum growth rate of D. acuta was lower (0.41 d(-1)). The positive dark carbon uptake by Dinophysis may suggest a mixotrophic mode of nutrition. In one experiment, both D. norvegica and D. acuta showed a significantly higher carbon uptake in a dark bottle than in a Light bottle, which would be consistent with uptake of C-14-labeled organic matter by D. norvegica and D. acuta. Demonstration of direct uptake of dissolved and particulate organic matter would provide conclusive evidence of mixotrophy and this will require the development of new protocols for measuring organic matter uptake applicable to Dinophysis in the natural assemblages.

Los óxidos transparentes conductores (TCO′ s) son materiales compuestos conformados por oxígeno y un metal, que presentan una combinación única de alta estabilidad química, alta concentración electrónica y alta transparencia óptica. Por esta razón, el procesamiento de TCO′ s en película delgada va orientado hacia aplicaciones específicas tales como ventanas ópticas en celdas solares, sensores de gases, electrodos en dispositivos de pantallas planas, ventanas inteligentes. En este proyecto se trabajó en la síntesis experimental de dos TCO′ s relevantes tanto en investigación fundamental como en aplicaciones tecnológicas: el óxido de indio (In2O3) y el óxido de estaño (SnO2). Ambos TCO′ s se depositaron por la técnica de erosión iónica reactiva por corriente directa (DC). Para el análisis de las películas se utilizaron varias técnicas de caracterización: difracción de rayos X, espectroscopia UV-Visible, resistividad eléctrica, efecto Hall, así como microscopías electrónica de barrido y de fuerza atómica. Se fabricó también una bicapa de In2O3/SnO2, la cual se caracterizó además con espectroscopia de fotoemisión de rayos X (XPS).En esta tesis se reporta por primera vez la síntesis y caracterización de esta bicapa, la cual abre una línea de investigación en el área de interfaces. Asimismo, se desarrolló e implementó un procedimiento, basado en los modelos ópticos, tal que permite obtener parámetros que se utilizan para evaluar a cualquier película delgada TCO como potencial metamaterial. Las propiedades de las muestras se analizaron en función de la temperatura aplicada post-depósito: temperatura ambiente (TA), 100oC, 200oC, 300oC, bajo una atmósfera de argón o argón-oxígeno. Los resultados confirman que las películas presentan un crecimiento de tipo poli cristalino. Además, la calidad cristalina tiende a incrementarse como función del incremento de la temperatura. El In2O3 creció con estructura cúbica bcc (a=10.11 ˚A, ICDD #71-2195). A partir de 200C, se detectaron trazas de la fase romboédrica (a=5.490 ˚A, c=14.520 ˚A, ICDD #73-1809). Asimismo, el SnO2 creció con estructura tetragonal (a = 4.737 ˚A, c = 3.186 ˚A, ICDD #88-0287). Las películas de In2O3 poseen una transparencia promedio del 90 % en una ventana de 500 nm a 1100 nm. El borde de absorción se recorre al azul como función de la temperatura, de Eg=3.3 eV a Eg=3.7 eV por el efecto Burstein-Moss. Por otra parte, la bicapa presentó una interfaz claramente definida, sin difusión de especies metálicas. Al incrementarse la temperatura, de TA a 400oC, se detectaron dos fases de óxido de estaño: SnO2 y SnO, en un porcentaje atómico de ≈70 %:30 %, respectivamente. Se concluye que los parámetros y valores obtenidos de las películas como son el texturizado y espesor homogéneo, alta transparencia, crecimiento preferencial, ancho prohibido y resistividad eléctrica, son comparables a los que se requieren del In2O3 y SnO2 en película delgada para aplicaciones optoelectrónicas.

Eighteen Corynebacterium xerosis strains isolated from different animal clinical specimens were subjected to phenotypic and molecular genetic studies. On the basis of the results of the biochemical characterization, the strains were tentatively identified as C. xerosis. Phylogenetic analysis based on comparative analysis of the sequences of 16S rRNA and rpoB genes revealed that the 18 strains were highly related to C. xerosis, C. amycolatum, C. freneyi, and C. hansenii. There was a good concordance between 16S rRNA and partial rpoB gene sequencing results, although partial rpoB gene sequencing allowed better differentiation of C. xerosis. Alternatively, C. xerosis was also differentiated from C. freneyi and C. amycolatum by restriction fragment length polymorphism analysis of the 16S-23S rRNA gene intergenic spacer region. Phenotypic characterization indicated that besides acid production from D-turanose and 5-ketogluconate, 90% of the strains were able to reduce nitrate. The absence of the fatty acids C(14:0), C(15:0), C(16:1)omega 7c, and C(17:1)omega 8c can also facilitate the differentiation of C. xerosis from closely related species. The results of the present investigation demonstrated that for reliable identification of C. xerosis strains from clinical samples, a combination of phenotypic and molecular-biology-based identification techniques is necessary.

Tras conocer cuatro años de debate (dentro y fuera de La Habana), los borradores de dos puntos de la agenda y presenciar el crecimiento de bandas criminales y/u organizaciones sucesoras del paramilitarismo, de la mano de la expansión de economías ilegales y una polarización creciente (y preocupante por sus características) en torno a la paz y el conflicto social/armado en el país, el presente trabajo pretende analizar a profundidad el carácter y alcance de los discursos emitidos por los negociadores en La Habana.
