Due to the large amount of pesticides applied in agriculture, mainly herbicides, there is a growing concern about a possible environmental contamination with these products, including water bodies. Given the above, the aim of the present work was to detect and quantify herbicides through multiresidue analysis in water samples collected in semi-artesian wells and springs in a rural area of the city of Jaboticabal (SP). Samples were collected from 32 wells and 13 water springs, in three different seasons: October 2010, February 2011 and May 2011. Additionally, samples at a residence in the urban area were also collected. Analysis using high performance liquid chromatography coupled to mass spectrometry was performed and herbicides ametryn, amicarbazone, clomazone, diclosulan, diuron, hexazinone, imazapic, imazapyr, isoxaflutole, S-metolachlor, sulfentrazone, sulfometuron-methyl, and tebuthiuron were evaluated. In semi-artesian wells, an incresed quantity of herbicides was found in comparison with the water springs. Among the tested herbicides, hexazinone, imazapyr and sulfentrazone were detected in measurable amounts in accordance with the analytical method applied, while clomazone was the most common herbicide being detected in more than 60% of the samples. Ametryn, diuron and amicarbazone herbicides were also detected. Diclosulan, imazapic, isoxaflutole, S-metolachlor, sulfometuron-methyl, and tebuthiuron were not detected in any sample. Inappropriate use of these products without prior knowledge of the behavior of the soil can lead to groundwaters and water springs contamination, thus an ongoing monitoring of this resource becomes very important.

The use of medicinal plants is common in many Brazil's ethnic groups, because the diversity of plant species in the country is large. But the utilization of these plants as standardized herbal doesn't occur with de same frequency, due the lack of studies related to these therapies, classified as alternatives. Due to the high cost of some synthetic medicines for the treatment of chronic diseases, phytotherapy is a low-cost alternative and efficient, since the phytomedication has proven scientific studies and its production meets strict criteria for manufacturing and quality control to ensure its safety and efficacy. Among the medicinal species used for the treatment of chronic diseases, such as pain and inflammation, is Eugenia speciosa Cambess, popularly known as the bush laranjinha. Against this context, this paper held preliminary studies and developed the chromatographic profile of the hydroalcoholic extract of the leaves of E. speciosa by High Performance Liquid Chromatography coupled to photodiode array detector (CLAE-DAD). Preliminary qualitative results and the chromatographic profile have identified some metabolites classes present and some constituent as: homogentisic acid, homorientina, isovitexin and quercetrin

In the present study, a fast, sensitive and robust method to quantify dextromethorphan, dextrorphan and doxylamine in human plasma using deuterated internal standards (IS) is described. The analytes and the IS were extracted from plasma by a liquid-liquid extraction (LLE) using diethyl-ether/hexane (80/20, v/v). Extracted samples were analyzed by high performance liquid chromatography coupled to electrospray ionization tandem mass spectrometry (HPLC-ESI-MS/MS). Chromatographic separation was performed by pumping the mobile phase (acetonitrile/water/formic acid (90/9/1, v/v/v) during 4.0 min at a flow-rate of 1.5 mL min(-1) into a Phenomenex Gemini (R) C18, 5 mu m analytical column (150 x 4.6 mm id.). The calibration curve was linear over the range from 0.2 to 200 ng mL(-1) for dextromethorphan and doxylamine and 0.05 to 10 ng mL(-1) for dextrorphan. The intra-batch precision and accuracy (%CV) of the method ranged from 2.5 to 9.5%, and 88.9 to 105.1%, respectively. Method inter-batch precision (%CV) and accuracy ranged from 6.7 to 10.3%, and 92.2 to 107.1%, respectively. The run-time was for 4 min. The analytical procedure herein described was used to assess the pharmacokinetics of dextromethorphan, dextrorphan and doxylamine in healthy volunteers after a single oral dose of a formulation containing 30 mg of dextromethorphan hydrobromide and 12.5 mg of doxylamine succinate. The method has high sensitivity, specificity and allows high throughput analysis required for a pharmacokinetic study. (C) 2012 Elsevier B.V. All rights reserved.

Die Erdatmosphäre besteht hauptsächlich aus Stickstoff (78%), Sauerstoff (21%) und Edelga¬sen. Obwohl Partikel weniger als 0,1% ausmachen, spielen sie eine entscheidende Rolle in der Chemie und Physik der Atmosphäre, da sie das Klima der Erde sowohl direkt als auch indirekt beeinflussen. Je nach Art der Bildung unterscheidet man zwischen primären und sekundären Partikeln, wobei primäre Partikel direkt in die Atmosphäre eingetragen werden. Sekundäre Partikel hingegen entstehen durch Kondensation von schwerflüchtigen Verbindungen aus der Gasphase, welche durch Reaktionen von gasförmigen Vorläufersubstanzen (volatile organic compounds, VOCs) mit atmosphärischen Oxidantien wie Ozon oder OH-Radikalen gebildet werden. Da die meisten Vorläufersubstanzen organischer Natur sind, wird das daraus gebil¬dete Aerosol als sekundäres organisches Aerosol (SOA) bezeichnet. Anders als die meisten primären Partikel stammen die VOCs überwiegend aus biogenen Quellen. Es handelt sich da¬bei um ungesättigte Kohlenwasserstoffe, die bei intensiver Sonneneinstrahlung und hohen Temperaturen von Pflanzen emittiert werden. Viele der leichtflüchtigen Vorläufersubstanzen sind chiral, sowohl die Vorläufer als auch die daraus gebildeten Partikel werden aber in den meisten Studien als eine Verbindung betrachtet und gemeinsam analysiert. Die mit Modellen berechneten SOA-Konzentrationen, welche auf dieser traditionellen Vorstellung der SOA-Bil¬dung beruhen, liegen deutlich unterhalb der in der Atmosphäre gefundenen, so dass neben diesem Bildungsweg auch noch andere SOA-Bildungsarten existieren müssen. Aus diesem Grund wird der Fokus der heutigen Forschung vermehrt auf die heterogene Chemie in der Partikelphase gerichtet. Glyoxal als Modellsubstanz kommt hierbei eine wichtige Rolle zu. Es handelt sich bei dieser Verbindung um ein Molekül mit einem hohen Dampfdruck, das auf Grund dieser Eigenschaft nur in der Gasphase zu finden sein sollte. Da es aber über zwei Alde¬hydgruppen verfügt, ist es sehr gut wasserlöslich und kann dadurch in die Partikelphase über¬gehen, wo es heterogenen chemischen Prozessen unterliegt. Unter anderem werden in An¬wesenheit von Ammoniumionen Imidazole gebildet, welche wegen der beiden Stickstoff-He¬teroatome lichtabsorbierende Eigenschaften besitzen. Die Verteilung von Glyoxal zwischen der Gas- und der Partikelphase wird durch das Henrysche Gesetz beschrieben, wobei die Gleichgewichtskonstante die sogenannte Henry-Konstante ist. Diese ist abhängig von der un¬tersuchten organischen Verbindung und den im Partikel vorhandenen anorganischen Salzen. Für die Untersuchung chiraler Verbindungen im SOA wurde zunächst eine Filterextraktions¬methode entwickelt und die erhaltenen Proben anschließend mittels chiraler Hochleistungs-Flüssigchromatographie, welche an ein Elektrospray-Massenspektrometer gekoppelt war, analysiert. Der Fokus lag hierbei auf dem am häufigsten emittierten Monoterpen α-Pinen und seinem Hauptprodukt, der Pinsäure. Da bei der Ozonolyse des α-Pinens das cyclische Grund¬gerüst erhalten bleibt, können trotz der beiden im Molekül vorhanden chiralen Zentren nur zwei Pinsäure Enantiomere gebildet werden. Als Extraktionsmittel wurde eine Mischung aus Methanol/Wasser 9/1 gewählt, mit welcher Extraktionseffizienzen von 65% für Pinsäure Enan¬tiomer 1 und 68% für Pinsäure Enantiomer 2 erreicht werden konnten. Des Weiteren wurden Experimente in einer Atmosphärensimulationskammer durchgeführt, um die Produkte der α-Pinen Ozonolyse eindeutig zu charakterisieren. Enantiomer 1 wurde demnach aus (+)-α-Pinen gebildet und Enantiomer 2 entstand aus (-)-α-Pinen. Auf Filterproben aus dem brasilianischen Regenwald konnte ausschließlich Pinsäure Enantiomer 2 gefunden werden. Enantiomer 1 lag dauerhaft unterhalb der Nachweisgrenze von 18,27 ng/mL. Im borealen Nadelwald war das Verhältnis umgekehrt und Pinsäure Enantiomer 1 überwog vor Pinsäure Enantiomer 2. Das Verhältnis betrug 56% Enantiomer 1 zu 44% Enantiomer 2. Saisonale Verläufe im tropischen Regenwald zeigten, dass die Konzentrationen zur Trockenzeit im August höher waren als wäh¬rend der Regenzeit im Februar. Auch im borealen Nadelwald wurden im Sommer höhere Kon¬zentrationen gemessen als im Winter. Die Verhältnisse der Enantiomere änderten sich nicht im jahreszeitlichen Verlauf. Die Bestimmung der Henry-Konstanten von Glyoxal bei verschiedenen Saataerosolen, nämlich Ammoniumsulfat, Natriumnitrat, Kaliumsulfat, Natriumchlorid und Ammoniumnitrat sowie die irreversible Produktbildung aus Glyoxal in Anwesenheit von Ammoniak waren Forschungs¬gegenstand einer Atmosphärensimulationskammer-Kampagne am Paul-Scherrer-Institut in Villigen, Schweiz. Hierzu wurde zunächst das zu untersuchende Saataerosol in der Kammer vorgelegt und dann aus photochemisch erzeugten OH-Radikalen und Acetylen Glyoxal er¬zeugt. Für die Bestimmung der Glyoxalkonzentration im Kammeraerosol wurde zunächst eine beste¬hende Filterextraktionsmethode modifiziert und die Analyse mittels hochauflösender Mas¬senspektrometrie realisiert. Als Extraktionsmittel kam 100% Acetonitril, ACN zum Einsatz wo¬bei die Extraktionseffizienz bei 85% lag. Für die anschließende Derivatisierung wurde 2,4-Di¬nitrophenylhydrazin, DNPH verwendet. Dieses musste zuvor drei Mal mittels Festphasenex¬traktion gereinigt werden um störende Blindwerte ausreichend zu minimieren. Die gefunde¬nen Henry-Konstanten für Ammoniumsulfat als Saataerosol stimmten gut mit in der Literatur gefundenen Werten überein. Die Werte für Natriumnitrat und Natriumchlorid als Saataerosol waren kleiner als die von Ammoniumsulfat aber größer als der Wert von reinem Wasser. Für Ammoniumnitrat und Kaliumsulfat konnten keine Konstanten berechnet werden. Alle drei Saataerosole führten zu einem „Salting-in“. Das bedeutet, dass bei Erhöhung der Salzmolalität auch die Glyoxalkonzentration im Partikel stieg. Diese Beobachtungen sind auch in der Litera¬tur beschrieben, wobei die Ergebnisse dort nicht auf der Durchführung von Kammerexperi¬menten beruhen, sondern mittels bulk-Experimenten generiert wurden. Für die Trennung der Imidazole wurde eine neue Filterextraktionsmethode entwickelt, wobei sich ein Gemisch aus mit HCl angesäuertem ACN/H2O im Verhältnis 9/1 als optimales Extrak¬tionsmittel herausstellte. Drei verschiedenen Imidazole konnten mit dieser Methode quanti¬fiziert werden, nämlich 1-H-Imidazol-4-carbaldehyd (IC), Imidazol (IM) und 2,2‘-Biimidazol (BI). Die Effizienzen lagen für BI bei 95%, für IC bei 58% und für IM bei 75%. Kammerexperimente unter Zugabe von Ammoniak zeigten höhere Imidazolkonzentrationen als solche ohne. Wurden die Experimente ohne Ammoniak in Anwesenheit von Ammoni¬umsulfat durchgeführt, wurden höhere Imidazol-Konzentrationen gefunden als ohne Ammo¬niumionen. Auch die relative Luftfeuchtigkeit spielte eine wichtige Rolle, da sowohl eine zu hohe als auch eine zu niedrige relative Luftfeuchtigkeit zu einer verminderten Imidazolbildung führte. Durch mit 13C-markiertem Kohlenstoff durchgeführte Experimente konnte eindeutig gezeigt werden, dass es sich bei den gebildeten Imidazolen und Glyoxalprodukte handelte. Außerdem konnte der in der Literatur beschriebene Bildungsmechanismus erfolgreich weiter¬entwickelt werden. Während der CYPHEX Kampagne in Zypern konnten erstmalig Imidazole in Feldproben nach¬gewiesen werden. Das Hauptprodukt IC zeigte einen tageszeitlichen Verlauf mit höheren Kon¬zentrationen während der Nacht und korrelierte signifikant aber schwach mit der Acidität und Ammoniumionenkonzentration des gefundenen Aerosols.

To enhance understanding of the metabolic indicators of type 2 diabetes mellitus (T2DM) disease pathogenesis and progression, the urinary metabolomes of well characterized rhesus macaques (normal or spontaneously and naturally diabetic) were examined. High-resolution ultra-performance liquid chromatography coupled with the accurate mass determination of time-of-flight mass spectrometry was used to analyze spot urine samples from normal (n = 10) and T2DM (n = 11) male monkeys. The machine-learning algorithm random forests classified urine samples as either from normal or T2DM monkeys. The metabolites important for developing the classifier were further examined for their biological significance. Random forests models had a misclassification error of less than 5%. Metabolites were identified based on accurate masses (<10 ppm) and confirmed by tandem mass spectrometry of authentic compounds. Urinary compounds significantly increased (p < 0.05) in the T2DM when compared with the normal group included glycine betaine (9-fold), citric acid (2.8-fold), kynurenic acid (1.8-fold), glucose (68-fold), and pipecolic acid (6.5-fold). When compared with the conventional definition of T2DM, the metabolites were also useful in defining the T2DM condition, and the urinary elevations in glycine betaine and pipecolic acid (as well as proline) indicated defective re-absorption in the kidney proximal tubules by SLC6A20, a Na(+)-dependent transporter. The mRNA levels of SLC6A20 were significantly reduced in the kidneys of monkeys with T2DM. These observations were validated in the db/db mouse model of T2DM. This study provides convincing evidence of the power of metabolomics for identifying functional changes at many levels in the omics pipeline.

This PhD thesis focused on the analysis and application of microbial membrane lipids as biomarkers in marine sediments. Existing protocols for lipid extraction from marine sediments and biomass were modified. In addition, recent protocols based on high-performance liquid chromatography coupled to mass spectrometry (HPLC-MS) as well as state-of-the-art mass spectrometric analysis by quadrupole time-of-flight (MSqTOF) and the triple quadrupole (MSQQQ) mass spectrometer were used to investigate matrix effects and evaluate the reliability of quantitative analysis in marine environmental samples. The improved lipid extraction and quantification were used to analyze Black Sea water column and sediments samples to a depth of 8 meters from site GeoB 15105 taken during cruise M84/1 (DARCSEAS) with R/V Meteor to apply lipid analysis in benthic bio systems. With this component specific differentiation between planktonic and benthic lipid signature we assessed possible lipid sources. Here, this high detail lipid fingerprinting allowed us to observe changes in the head group and lipid core structures of the intact polar lipids according to the geochemical zonation.

We report that silver ion HPLC provides remarkable separations of C27 sterols differing only in the number or location of olefinic double bonds. This technique has been extended to LC-MS, analysis of purified components by GC, GC-MS, and 1H NMR, and to its use on a semipreparative scale. The application of this methodology for the demonstration of the catalysis, by rat liver microsomes, of the conversion of 7-dehydrocholesterol to cholesta-5,8-dien-3 beta-ol is also presented.

During the analytical method development for BAY 11-7082 ((E)-3-[4-methylphenylsulfonyl]-2-propenenitrile), using HPLC-MS-MS and HPLC-UV, we observed that the protein removal process (both ultrafiltration and precipitation method using organic solvents) prior to HPLC brought about a significant reduction in the concentration of this compound. The use of a structurally similar internal standard, BAY 11-7085 ((E)-3-[4-t-butylphenylsulfonyl]-2-propenenitrile), was not effective in compensating for the loss of analyte as the extent of reduction was different to that of the analyte. A high performance liquid-level sensor based on microstructured polymer optical fiber Bragg grating (mPOFBG) array sensors is reported in detail. The sensor sensitivity is found to be 98pm/cm of liquid, enhanced by more than a factor of 9 compared to a reported silica fiber-based sensor.

The initiation of the Benguela upwelling has been dated to the late Miocene, but estimates of its sea surface temperature evolution are not available. This study presents data from Ocean Drilling Program (ODP) Site 1085 recovered from the southern Cape Basin. Samples of the middle Miocene to Pliocene were analyzed for alkenone-based (UK'37, SSTUK) and glycerol dialkyl glycerol tetraether (GDGT) based (TEX86, TempTEX) water temperature proxies. In concordance with global cooling during the Miocene, SSTUK and TempTEX exhibit a decline of about 8°C and 16°C, respectively. The temperature trends suggest an inflow of cold Antarctic waters triggered by Antarctic ice sheet expansion and intensification of Southern Hemisphere southeasterly winds. A temperature offset between both proxies developed with the onset of upwelling, which can be explained by differences in habitat: alkenone-producing phytoplankton live in the euphotic zone and record sea surface temperatures, while GDGT-producing Thaumarchaeota are displaced to colder subsurface waters in upwelling-influenced areas and record subsurface water temperatures. We suggest that variations in subsurface water temperatures were driven by advection of cold Antarctic waters and thermocline adjustments that were due to changes in North Atlantic deep water formation. A decline in surface temperatures, an increased offset between temperature proxies, and an increase in primary productivity suggest the establishment of the Benguela upwelling at 10 Ma. During the Messinian Salinity Crisis, between 7 and 5 Ma, surface and subsurface temperature estimates became similar, likely because of a strong reduction in Atlantic overturning circulation, while high total organic carbon contents suggest a "biogenic bloom." In the Pliocene the offset between the temperature estimates and the cooling trend was reestablished.

This dissertation presents the design of three high-performance successive-approximation-register (SAR) analog-to-digital converters (ADCs) using distinct digital background calibration techniques under the framework of a generalized code-domain linear equalizer. These digital calibration techniques effectively and efficiently remove the static mismatch errors in the analog-to-digital (A/D) conversion. They enable aggressive scaling of the capacitive digital-to-analog converter (DAC), which also serves as sampling capacitor, to the kT/C limit. As a result, outstanding conversion linearity, high signal-to-noise ratio (SNR), high conversion speed, robustness, superb energy efficiency, and minimal chip-area are accomplished simultaneously. The first design is a 12-bit 22.5/45-MS/s SAR ADC in 0.13-μm CMOS process. It employs a perturbation-based calibration based on the superposition property of linear systems to digitally correct the capacitor mismatch error in the weighted DAC. With 3.0-mW power dissipation at a 1.2-V power supply and a 22.5-MS/s sample rate, it achieves a 71.1-dB signal-to-noise-plus-distortion ratio (SNDR), and a 94.6-dB spurious free dynamic range (SFDR). At Nyquist frequency, the conversion figure of merit (FoM) is 50.8 fJ/conversion step, the best FoM up to date (2010) for 12-bit ADCs. The SAR ADC core occupies 0.06 mm2, while the estimated area the calibration circuits is 0.03 mm2. The second proposed digital calibration technique is a bit-wise-correlation-based digital calibration. It utilizes the statistical independence of an injected pseudo-random signal and the input signal to correct the DAC mismatch in SAR ADCs. This idea is experimentally verified in a 12-bit 37-MS/s SAR ADC fabricated in 65-nm CMOS implemented by Pingli Huang. This prototype chip achieves a 70.23-dB peak SNDR and an 81.02-dB peak SFDR, while occupying 0.12-mm2 silicon area and dissipating 9.14 mW from a 1.2-V supply with the synthesized digital calibration circuits included. The third work is an 8-bit, 600-MS/s, 10-way time-interleaved SAR ADC array fabricated in 0.13-μm CMOS process. This work employs an adaptive digital equalization approach to calibrate both intra-channel nonlinearities and inter-channel mismatch errors. The prototype chip achieves 47.4-dB SNDR, 63.6-dB SFDR, less than 0.30-LSB differential nonlinearity (DNL), and less than 0.23-LSB integral nonlinearity (INL). The ADC array occupies an active area of 1.35 mm2 and dissipates 30.3 mW, including synthesized digital calibration circuits and an on-chip dual-loop delay-locked loop (DLL) for clock generation and synchronization.

Mushrooms are very perishable foods due to their high susceptibility to moisture loss, changes in color and texture, or microbiological spoilage. Drying is considered as the most appropriate method to prevent these alterations, but it has some limitations, such as shrinkage, enzymatic and non-enzymatic browning reactions, and oxidation of lipids and vitamins. Irradiation might effectively attenuate the undesirable changes caused by drying process, ensuring also higher shelf-life of mushrooms and their decontamination [I]. In the present work, the combined effects of electron-beam irradiation (at 0, 0.5, 1 and 6 kGy doses) and storage time (at 0, 6 and 12 months) were evaluated and compared. Macrolepiota procera (Scop.) Singer wild samples were obtained in Tnis-os-Montes, in the Northeast of Portugal, and dried at 30 •c in an oven. Subsequently, the samples were divided in four groups: control (non-irradiated, 0 kGy); sample 1 (0.5 kGy); sample 2 (1 kGy) and sample 3 (6 kGy). The irradiation was performed at the lNCTInstitute of Nuclear Chemistry and Technology (lNCT), in Warsaw, Poland. Moisture, protein, fat, carbohydrates and ash were determined following standard procedures. Free sugars and tocopherols were determined by high performance liquid chromatography coupled to a refraction index detector (HPLC-RI) and a fluorescence detector, respectively; fatty acids were determined by gas-liquid chromatography with flame ionization detection (GC-FID). Antioxidant activity was evaluated in the methanolic extracts by in vitro assays measuring DPPH (1,1-diphenyl-2- picrylhydrazyl) radical scavenging activity, reducing power, inhibition of ~-carotene bleaching and inhibition oflipid peroxidation using thiobarbituric acid reactive substances (TBARS) assay. Total phenolics were also determined by the Folin-Ciocalteu assay. All the parameters showed a decrease tendency with storage time. Trehalose and y-tocopherol were preserved with 1 kGy dose. Electron-beam irradiation did not impart additional changes to most of the chemical and antioxidant parameters of M. procera dried samples. This is a very promising result, since electron-beam irradiation might attenuate most unwanted changes caused by drying, maintaining its long-term effectiveness.

In recent years the interest in naturally occurring compounds has been increasing worldwide. Indeed, many of the bioactive compounds currently used as medicines have been synthesized based on the structure of natural compounds [1]. In order to obtain bioactive fractions and subsequently isolated compounds derived from natural matrices, several procedures have been carried out. One of these is to separate and assess the concentration of the active compound(s) present in the samples, a step in which the chromatographic techniques stand out [2]. In the present work the mushroom Sui/Ius granulatus (L.) Roussel was chemically characterized by chromatographic techniques coupled to different detectors, in order to evaluate the presence of nutritional and/or bioactive molecules. Some hydrophilic compounds, namely free sugars, were identified by high performance liquid chromatography coupled to a refraction index detector (HPLC-RI), and organic and phenolic acids were assessed by HPLC coupled to a photodiode array detector (HPLC-PDA). Regarding lipophilic compounds, fatty acids weredetermined by gas chromatography with a flame ionization detector (GC-FID) and tocopherols by HPLC-fluorescence detection. Mannitol and trehalose were the main free sugars detected. Different organic acids were also identified (i.e. oxalic, quinic and fumaric acids), as well as phenolic acids (i.e. gallic and p-hydroxybenzoic acids) and the related compound cinnamic acid. Mono- and polyunsaturated fatty acids were the prevailing fatty acids and a-, ~- and ~-tocopherol were the isoforms of vitamin E detected in the samples. Since this species proved to be a source of biologically active compounds, the antioxidant and antimicrobial properties were evaluated. The antioxidant activity was measured through the reducing power, free radical's scavenging activity and lipid peroxidation inhibition of its methanolic extract, and the antimicrobial activity was also tested in Gram positive and Gram negative bacteria and iri different fungi. S. granulatus presented antioxidant properties in all the performed assays, and proved to inhibit the growth of different bacterial and fungal strains.
