New antibacterial compounds, preferentially exploiting novel cellular targets, are urgently needed to fight the increasing resistance of pathogens against conventional antibiotics. Here we demonstrate that Carolacton, a myxobacterial secondary metabolite previously shown to damage Streptococcus mutans biofilms, inhibits planktonic growth of Streptococcus pneumoniae TIGR4 and multidrug-resistant clinical isolates of serotype 19A at nanomolar concentrations. A Carolacton diastereomer is inactive in both streptococci, indicating a highly specific interaction with a conserved cellular target. S. mutans requires the eukaryotic-like serine/threonine protein kinase PknB and the cysteine metabolism regulator CysR for susceptibility to Carolacton, whereas their homologues are not needed in S. pneumoniae, suggesting a specific function for S. mutans biofilms only. A bactericidal effect of Carolacton was observed for S. pneumoniae TIGR4, with a reduction of cell numbers by 3 log units. The clinical pneumonia isolate Sp49 showed immediate growth arrest and cell lysis, suggesting a bacteriolytic effect of Carolacton. Carolacton treatment caused a reduction in membrane potential, but not membrane integrity, and transcriptome analysis revealed compensatory reactions of the cell. Our data show that Carolacton might have potential for treating pneumococcal infections.

La caries dental es la principal enfermedad oral que padece la población a nivel mundial. Su prevención se basa en educación sobre higiene oral, además del uso complementario de agentes antimicrobianos. Entre las alternativas de prevención una área prometedora incluye el empleo de extractos de plantas como agentes antimicrobianos incorporados en nanopartículas poliméricas (NP), las cuales, puedan funcionar como vehículos de liberación de extractos, mejorando el desempeño de estos agentes activos naturales. El objetivo de este trabajo fue obtener extractos de plantas de Ocimum basilicum (Albahaca) Calendula officinalis (Cálendula) y aceites esenciales de Syzigium aromaticum (Clavo) y Thymus vulgaris (Tomillo). Se realizó su caracterización fitoquímica y se evaluó su actividad antimicrobiana mediante dilución en tubo contra células plantónicas de Streptococcus gordonii, Streptococcus mutans y Candida albicans (ATCC), determinando la concentración mínima inhibitoria (CMI). El extracto con mayor actividad fue incorporado en NP mediante el método de nanoprecipitación (NP) y las nanopartículas se caracterizaron en base a su tamaño e índice de polidispersidad por espectroscopia de relación fotónica; el porcentaje y la eficiencia de encapsulación y mecanismo de liberación del eugenol contenido en el aceite encapsulado, se realizó mediante cromatografía de gases acoplado a espectrometría de masas (GSMC). En la presente investigación, se determinó que el aceite esencial de clavo mostró la mayor actividad antimicrobiana con respecto a los demás extractos evaluados. Por medio de GSMC se identificó al eugenol como el componente principal del aceite esencial obtenido por hidrodestilación. Se obtuvieron NP esféricas con tamaño alrededor de 157 nm, el porcentaje y la eficiencia de encapsulación de eugenol presente en el aceite encapsulado fue de 73.16 % y 47 % respectivamente; además de mostrar una liberación in vitro de eugenol del 50 % a las 24 horas. Finalmente, la CMI del aceite de clavo sin encapsular e incorporado en NP, correspondió a 125 y 75 μg/mL respectivamente. En base a los resultados obtenidos, es factible incorporar extractos vegetales en NP para su liberación sostenida, siendo una terapia antimicrobiana prometedora dentro del área odontológica.

De acuerdo con un anuncio publicado por el NIH (National Institute of Health, EUA), los biofilms o biocapas son un problema de salud pública muy importante en el área médica, ya que representan más del 80% de las infecciones microbianas en el cuerpo humano. Los tratamientos antimicrobianos estándar, típicamente fallan para erradicarlos. Por tanto, existe una creciente necesidad de desarrollo de nuevos fármacos con nuevas y mejores capacidades que puedan controlar a los biofilms. Hoy en día, la nanoterapeútica ofrece opciones innovadoras para controlar enfermedades infecciosas incluyendo la cavidad oral, contrarrestando el crecimiento de microorganismos patógenos tales como, Streptococcus mutans, Candida albicans y herpesvirus. En el área médica, algunos derivados del bismuto, como el subsalicilato, han sido empleados en el área médica para contrarrestar el vómito, náuseas, diarrea y dolor de estómago. En esta investigación determinamos la actividad antimicrobiana de nanopartículas de bismuto (BiNPs) contra patógenos orales. La actividad antibacteriana y antifúngica lo evaluamos por el ensayo de MTT y microscopía de fluorescencia. Demostramos que las BiNPs inhibieron el crecimiento de Streptococcus mutans con un 69% de efectividad así mismo obtuvimos un 85% de inhibición con Candida albicans, además fueron capaces de inhibir la formación del biofilm de ambas cepas. Por otra parte, determinamos la actividad antiviral en un modelo de rotavirus mediante el ensayo de inmunoperoxidasa presentando 90% de control de infección con el tratamiento de BiNPs. Finalmente, se evaluó la posible citotoxicidad de BiNPs en células epiteliales de riñón de mono mediante microscopía de fluorescencia y no mostró evidencias de citotoxicidad a 24 horas de exposición.

A cárie, na atualidade, pode ser definida como a falta de equilíbrio nos processos de desmineralização e remineralização do dente, na presença de microorganismos, os Streptococcus mutans e de dieta. Esse processo dinâmico pode ser mediado pela ação do flúor por meio de diversos veículos seja na dentição decídua ou na dentição permanente. A fluoretação da água do sistema de abastecimento é considerada a medida mais efetiva de controle da prevalência da doença quando se consideram medidas de impacto coletivo. Entretanto, no Brasil, ainda existem municípios sem fluoretação da água e com alto índice de cárie em ambas as dentições. O presente estudo propôs uma revisão de literatura sobre as alternativas de controle de cárie na dentição decídua em municípios sem fluoretação de água e se deparou com medidas educacionais, materno-infantis, além da necessidade de adoção de programas preventivos que incluam uma abordagem multiprofissional com trabalho educativo e motivacional para os pais ou responsáveis. A importância ao acesso ao serviço de saúde no primeiro ano de vida e das consultas subseqüentes regulares, associadas a melhorias das condições socioeconômicas e culturais da população sugerem ser as medidas necessárias para o controle da doença.

FUNDAMENTO: Penicilina G benzatina a cada 3 semanas é o protocolo padrão para a profilaxia secundária para febre reumática recorrente. OBJETIVO: Avaliar o efeito da penicilina G benzatina em Streptococcus sanguinis e Streptococcus oralis em pacientes com doença valvular cardíaca, devido à febre reumática com recebimento de profilaxia secundária. MÉTODOS: Estreptococos orais foram avaliados antes (momento basal) e após 7 dias (7º dia) iniciando-se com penicilina G benzatina em 100 pacientes que receberam profilaxia secundária da febre reumática. Amostras de saliva foram avaliadas para verificar a contagem de colônias e presença de S. sanguinis e S. oralis. Amostras de saliva estimulada pela mastigação foram serialmente diluídas e semeadas em placas sobre agar-sangue de ovelhas seletivo e não seletivo a 5% contendo penicilina G. A identificação da espécie foi realizada com testes bioquímicos convencionais. Concentrações inibitórias mínimas foram determinadas com o Etest. RESULTADOS: Não foram encontradas diferenças estatísticas da presença de S. sanguinis comparando-se o momento basal e o 7º dia (p = 0,62). No entanto, o número existente de culturas positivas de S. oralis no 7º dia após a Penicilina G benzatina apresentou um aumento significativo em relação ao valor basal (p = 0,04). Não houve diferença estatística existente entre o momento basal e o 7º dia sobre o número de S. sanguinis ou S. oralis UFC/mL e concentrações inibitórias medianas. CONCLUSÃO: O presente estudo mostrou que a Penicilina G benzatina a cada 3 semanas não alterou a colonização por S. sanguinis, mas aumentou a colonização de S. oralis no 7º dia de administração. Portanto, a susceptibilidade do Streptococcus sanguinis e Streptococcus oralis à penicilina G não foi modificada durante a rotina de profilaxia secundária da febre reumática utilizando a penicilina G.

Streptococcus sanguinis is a commensal pioneer colonizer of teeth and an opportunistic pathogen of infectious endocarditis. The establishment of S. sanguinis in host sites likely requires dynamic fitting of the cell wall in response to local stimuli. In this study, we investigated the two-component system (TCS) VicRK in S. sanguinis (VicRKSs), which regulates genes of cell wall biogenesis, biofilm formation, and virulence in opportunistic pathogens. A vicK knockout mutant obtained from strain SK36 (SKvic) showed slight reductions in aerobic growth and resistance to oxidative stress but an impaired ability to form biofilms, a phenotype restored in the complemented mutant. The biofilm-defective phenotype was associated with reduced amounts of extracellular DNA during aerobic growth, with reduced production of H2O2, a metabolic product associated with DNA release, and with inhibitory capacity of S. sanguinis competitor species. No changes in autolysis or cell surface hydrophobicity were detected in SKvic. Reverse transcription-quantitative PCR (RT-qPCR), electrophoretic mobility shift assays (EMSA), and promoter sequence analyses revealed that VicR directly regulates genes encoding murein hydrolases (SSA_0094, cwdP, and gbpB) and spxB, which encodes pyruvate oxidase for H2O2 production. Genes previously associated with spxB expression (spxR, ccpA, ackA, and tpK) were not transcriptionally affected in SKvic. RT-qPCR analyses of S. sanguinis biofilm cells further showed upregulation of VicRK targets (spxB, gbpB, and SSA_0094) and other genes for biofilm formation (gtfP and comE) compared to expression in planktonic cells. This study provides evidence that VicRKSs regulates functions crucial for S. sanguinis establishment in biofilms and identifies novel VicRK targets potentially involved in hydrolytic activities of the cell wall required for these functions.

A wild strain of Streptococcus thermophilus isolated from pasteurized milk was evaluated using an experimental model with respect to its adhesion onto stainless steel surfaces and its behaviour when submitted to cleansing and sanification. In milk, the adhesion of the microorganism on to stainless steel surfaces was studied after 6 hours of contact at 45°C with agitation, and after a cleansing process involving cleaning stages with alkaline and acid detergents followed by sanification, in order to evaluate the resistance of the adhered cells. The microorganism adhered to stainless steel surfaces producing a cell load of 10(4) CFU/cm². After alkaline cleansing, no adhered cells were detected but 6 CFU/cm² were still detected on the surfaces after acid cleansing. Cleansing, followed by sanification with sodium hypochlorite, was sufficient to reduce the load of wild S. thermophilus on the stainless steel surfaces to non-detectable levels. The experimental model proved adequate for the study indicating that the wild microorganism S. thermophilus produces biofilms on stainless steel surfaces. Alkaline cleansing remove more that 99.9% of the adhered cells. The few cells adhered on the surface are removed by acid cleansing demonstrating the need to use different steps and types of detergent for efficient cleansing. The best results for the removal of these biofilms are obtained by using alkaline cleansing followed by acid cleaning, this procedure being more efficient when complemented by sanification with sodium hypochlorite.

Objective: The purpose of this in vitro study was to evaluate the dentine root surface roughness and the adherence of Streptococcus sanguinis (ATCC 10556) after treatment with an ultrasonic system, Er:YAG laser, or manual curette. Background Data: Bacterial adhesion and formation of dental biofilm after scaling and root planing may be a challenge to the long-term stability of periodontal therapy. Materials and Methods: Forty flattened bovine roots were randomly assigned to one of the following groups: ultrasonic system (n = 10); Er:YAG laser (n = 10); manual curette (n = 10); or control untreated roots (n = 10). The mean surface roughness (Ra, mu m) of the specimens before and after exposure to each treatment was determined using a surface profilometer. In addition, S. sanguinis was grown on the treated and untreated specimens and the amounts of retained bacteria on the surfaces were measured by culture method. Results: All treatments increased the Ra; however, the roughest surface was produced by the curettes. In addition, the specimens treated with curettes showed the highest S. sanguinis adhesion. There was a significant positive correlation between roughness values and bacterial cells counts. Conclusion: S. sanguinis adhesion was the highest on the curette-treated dentine root surfaces, which also presented the greatest surface roughness.

The incidence of Streptococcus agalactiae is widely recognized as increasing in nonpregnant adults. This report describes a retrospective case series of S. agalactiae endocarditis at the Heart Institute of Sao Paulo, Brazil, between 2000 and 2006. Early diagnosis and valve surgery seem to have contributed to improved patient outcomes. (Heart Lung (R) 2010;39:331-334.)

Inulin was used as a prebiotic to improve the quality and consistency of skim milk fermented by co-cultures and pure Cultures of Lactobacillus acidophilus, Lactobacillus rhamnosus, Lactobacillus bulgaricus and Bifidobacterium lactis with Streptococcus thermophilus. We compared, either in the presence or absence of 4 g inulin/100 g, the results of the main kinetic parameters, specifically the generation time (t(g)), the maximum acidification rate (V(max)). and the times to reach V(max) (t(max)), to attain pH 5.0 (t(pH5.0)) and to complete the fermentation (t(pH4.5)). Post-acidification, lactic acid formation and cell counts were also determined and compared, either 1 day after the fermentation was complete or after 7 day storage at 4 degrees C. In general, inulin addition to the milk increased in co-cultures V(max), decreased t(max), t(g) and t(pH4.5), favored post-acidification, exerted a bifidogenic effect, and preserved almost intact cell viability during storage. In addition, S. thermophilus was shown to stimulate the metabolism of the other lactic bacteria. Contrary to co-cultures, most of the effects in pure Cultures were not statistically significant. The most important aspect of this paper is the use of the generation time as a toot to investigate the microbial response to inulin addition. (c) 2009 Elsevier Ltd. All rights reserved.

Inulin was used as a prebiotic to improve the quality and consistency of skim milk fermented by Lactobacillus acidophilus (La), Lactobacillus rhamnosus (Lr), Lactobacillus bulgaricus (Lb) and Bifidobacterium lactis (BI) with Streptococcus thermophilus (St), either in binary co-cultures or in cocktail containing all microorganisms. We compared, either in the presence of 40 mg inulin g(-1) or not, the results of the maximum acidification rate (V(max)) and the times to reach it (t(max)), to reach pH 5.0 (t(PH5.0)) and to complete the fermentation (t(f)). Post-acidification, lactic acid formation and cell counts were also compared after either 1 day (D1) or 7 days of storage at 4 degrees C (N). In co-culture, inulin addition to the milk increased V(max), decreased t(max) and t(f), favored post-acidification and exerted a bifidogenic effect. S. thermophilus proved to stimulate the metabolism of the other lactic bacteria and enhanced the product features. After D7, a significant prebiotic effect of inulin was observed in all co-cultures. Either after D1 or D7, the enumerations of Lr and BI in mixed culture markedly decreased compared to their respective co-cultures because of greater competition for the same substrates. The study compared the growth capability of probiotic (Lactobacillus acidophilus La05, Lactobacillus casei Lc01 and Bifidobacterium animalis Bb12) and non-probiotic (Lactobacillus delbrueckii subsp bulgaricus and Streptococcus thermophilus) cultures on twenty-one culture media grouped according to selectivity: nonselective agars, selective agars without antibiotics and MRS agars containing different combinations of lithium chloride, cystein, bile salts and antibiotics. Four of these media were selected for quantitative enumeration of L acidophilus La05, L casei Lc01, and B. animalis Bb12. The best culture media and incubation conditions for enumeration of the probiotic cultures were: B. animalis: MRS agar with dicloxacillin, 37 degrees C or 42 degrees C, anaerobiosis; L acidophilus: MRS agar with bile salts, 37 degrees C or 42 degrees C, aerobiosis; L casei: MRS agar with lithium chloride and sodium propionate, 37 degrees C or 42 degrees C, aerobiosis or anaerobiosis. Plating on MRS with glucose replaced by maltose, 37 degrees C or 42 degrees C, anaerobiosis, will distinguish probiotic from non-probiotic cultures. For enumeration of each probiotic in a mixed culture, the following media and incubation conditions were recommended: B. animalis: 4ABC-MRS, 42 degrees C, anaerobiosis, L acidophilus: LC medium, 42 degrees C, aerobiosis or anaerobiosis and L casei: LP-MRS, 42 degrees C, aerobiosis or anaerobiosis. The tannin-degrading species Streptococcus gallolyticus and Streptococcus caprinus have been shown to be subjective synonyms on the basis of their levels of 16S rRNA sequence similarity (98.3%) and DNA-DNA homology (>70%) and the phenotypes of their type strains. S. gallolyticus has nomenclatural priority according to Rule 24b(2) of the International Code of Nomenclature of Bacteria.
