HLA-G is a non-classic Human Leukocyte Antigen (HLA-G) Class I of low polymorphism and restricted tissue distribution that displays tolerogenic functions. In heart transplantation and in combined liver/renal allograft transplantation, the expression of HLA-G has been associated with a lower incidence of acute graft rejection episodes and absence of chronic dysfunction. Since the expression of HLA-G in renal biopsies has been investigated only in few patients who received a combined kidney and liver transplant, in this study we performed a cross-sectional study, systematically comparing the expression of HLA-G in post-transplanted renal grafts, stratifying patients according to the presence or absence of rejection.Patients and Methods: Seventy-three renal specimens (10 with acute rejection and 13 with chronic allograft nephropathy, and 50 with no signs of rejection) were immunohistochemically evaluated for HLA-G expression.Results: In the group as a whole, HLA-G molecules were detected in 40 cases (54.8%). Among specimens that presented HLA-G expression, 2 out of 40 (5%) exhibited acute rejection, 2 (5%) exhibited chronic allograft nephropathy, and the remaining 36 (90%) exhibited no signs of rejection. The comparison between patients with rejection and those without rejection showed that the expression of HLA-G was significantly increased in specimens exhibiting no signs of rejection (p<0.0001). Considering only patients with acute rejection, 8 out of 10 patients showed no HLA-G expression in their kidney biopsies when compared to patients exhibiting no signs of rejection and absence of HLA-G was observed in 14 out of 50 (p=0.0032). Similarly, considering only patients with chronic allograft nephropathy, absence of HLA-G expression was observed in I I out of 13 specimens, whereas in patients without rejection absence of HLA-G was observed in 14 out of 50 (p=0.003). Therapy with tacrolimus was significantly associated with the expression of HLA-G and a better graft prognosis. Conclusions: Our results suggest that HLA-G expression in the kidney allograft and the use of tacrolimus are associated with a lower frequency of acute renal rejection and chronic allograft nephropathy. The significance of recruitment systems for community structure of epigaeic ants in a tropical upland forest in southern Brazil was evaluated by examining patterns of spatial occurrence at fixed points. Normal exploratory activity was evaluated with pitfall traps, while the effect of recruitment and diet was evaluated by using honey and sardine baits at the same points. Through techniques developed for environmental impact assessment, the significance of recruitment was evaluated following perturbation, or the placement of bait. Of the 46 species encountered, 15 were sufficiently frequent to study. Of these, only 6 showed significant spatial frequency changes at baits when compared with pitfall trap collections. In one analysis, monthly differences were important for a few smaller species, suggesting thermic limitations, while bait types either increased or decreased spatial point usage. The magnitude of spatial point variation is an index for the strength of recruitment in community organization. Bait types suggest nutritional possibilities of each species. Both recruitment and diet are probably functions of the species composition of the ant community.

The effects of chronic mild prenatal stress on leukocyte infiltration into the airways was investigated in rat offspring. The chronic prenatal stress consisted of transitory and variable changes in the rat's living conditions. Offspring at adult age were actively sensitized (day 0) and intratracheally challenged (day 14) with ovalbumin. Bronchoalveolar lavage was performed in the offspring at 48 h after intratracheal challenge with ovalbumin. A significant increase in total leukocyte infiltration was observed in the non-stressed offspring group and this was associated with a marked recruitment of eosinophils without a significant effect on the influx of neutrophils and mononuclear cells. In the prenatal stressed offspring, the counts of both total leukocyte and eosinophils, as well as mononuclear cells, was increased by 50% compared to the non-stressed offspring. We provide here the first experimental evidence that chronic mild unpredictable prenatal stress produces a marked increase in the allergen-induced airway inflammation in the rat offspring.

The effect of dietary supplementation with 0, 100 and 450 mg of vitamin E (DL-α tocopheryl acetate)/kg of a dry diet on the kinetics of macrophage recruitment and giant cell formation in the pacu, maintained at different stocking densities (5 kg/m3 and 20 kg/m3), was investigated by insertion of round glass coverslips into the subcutaneous connective tissue. After a feeding period of 18 weeks, the coverslips were implanted and later removed for examination at 2, 7 and 15 days post-implantation. Fish fed diets supplemented with 450 mg of vitamin E showed an increase (P<0.05) in the accumulation of macrophages, foreign body giant cells and Langhans type cells. The kinetics of macrophage recruitment and giant cell formation on the glass coverslips appeared to be strongly influenced by vitamin E supplementation, since fish fed a basal diet and held at high stocking densities showed low numbers of adhering cells on the coverslips, and high concentrations of plasma corticosteroids. On the other hand, fish given a diet supplemented with 450 mg of vitamin E did not show a similar difference in plasma cortisol concentrations related to stocking density. The effect of cortisol concentrations on carbohydrate metabolism, analysed by assessment of plasma glycaemia, was not clear. Blood glucose concentrations did not vary substantially with the different treatments examined. These results suggest that vitamin E may contribute to the efficiency of the fish's inflammatory response by increasing macrophage recruitment and giant cell formation in the foreign body granulomatous reaction. Vitamin E appeared to act on the stress response of pacus by preventing a stress-related immunosuppression. © 2005 Elsevier Ltd. All rights reserved.

The objective of this study was to investigate the spatial and temporal distribution of Libinia spinosa H. MILNE EDWARDS 1834 on unconsolidated sublittoral bottoms in two regions off the northern coast of the state of São Paulo, and to analyze the influence of environmental factors on the occurrence of this species and observe the recruitment pattern of its young. Crabs were collected monthly (July 2001 through June 2003) at depths of 5, 10, 15, 20, 25, 30 and 35 In, from a fishing boat equipped with two double-rig nets. Samples of water and sediment were collected for analysis of environmental factors. A total of 2112 spider crabs was obtained (701 juveniles and 1411 adults). The highest abundance was observed at depths of 20 and 25 m, in both regions. These localities were characterized by substrate composed of very fine sand and silt-clay. In regard to the temporal distribution, juveniles and adults predominated in the summer and winter months respectively. From these results, one can infer that the distribution of L. spinosa is related to environmental factors favorable for its life cycle; sediment type is the factor which most strongly determines its presence. © E. The objective of this study was to describe a new platelet-rich plasma (PRP) protocol with a reduced concentration of leukocytes and intact platelets. We collected 8 mL of venous blood (VB) from marginal ear veins of 10 male New Zealand white rabbits in acid dextrose citrate Vacutainer tubes. Tubes were centrifuged at 302g for 10 minutes. All plasma was collected in plastic tubes to avoid buffy-coat contamination and centrifuged at 2862g for 5 minutes. A 10% calcium chloride activator (10 PRP:2 CaCl2) was added to the lower third of this plasma (PRP), and the PRP gel was obtained. Mean platelet count was 317.7 x 10(3) +/- 39.9/microL in VB and 1344.9 x 10(3) +/- 347.5/microL in PRP. Leukocyte counts were 3.96 x 10(3) +/- 2.01/microL and 0.46 x 10(3) +/- 0.45/microL in VB and PRP, respectively. Mean platelet enrichment was 327.4 +/- 97.8%. All differences were statistically significant (P > .05). This protocol is practical and reproducible, resulting in a high concentration of intact platelets to help tissue repair and low levels of leukocytes.

Objective: The objective of this study was to investigate the mediators and the resident peritoneal cells involved in the neutrophil migration (NM) induced by mineral trioxide aggregate (MTA) in mice. Study design: MTA (25 mg/cavity) was injected into normal and pretreated peritoneal cavities (PC) with indomethacin (IND), dexamethasone (DEX), BWA4C, U75302, antimacrophage inflammatory protein-2 (MIP-2), and anti-interleukin-1β (IL-1β) antibodies and the NM was determined. The role of macrophage (MO) and mast cells (MAST) was determined by administration of thioglycollate 3% or 48/80 compound, respectively. The concentration of IL-1β and MIP-2 exudates was measured by ELISA. Results: MTA induced dose- and time-dependent NM into mice PC, with the participation of MO and MAST. NM was inhibited by DEX, BWA4C, and U75302, as well as anti-MIP-2 and anti-IL-1β antibodies. In the exudates, IL-1β and MIP-2 were detected. Conclusions: This study suggests that MTA induces NM via a mechanism dependent on MAST and MO mediated by IL-1β, MIP-2, and LTB4. © 2008 Mosby, Inc. All rights reserved.

This study aimed to compare the torque, torque ratio (Hamstrings:Quadriceps - H:Q), electromyographic (EMG) activity and EMG ratio (knee flexors:knee extensors EMG) in soccer players (SG, N=10) and active subjects (AG, N=10). Subjects performed three maximal voluntary isometric knee extensions and flexions at 45° and 90° to determine the peak torque and EMG activity. Torque and EMG activity of the knee flexor (biceps femoris [BF] and semitendinosus [ST]) were divided by the torque and EMG activity of the knee extensor (vastuls lateralis [VL] and rectus femoris [RF]) to calculate torque ratios (H:Q) and EMG ratios (BF:VL, BF:RF, ST:VL, ST:RF). The flexion torque was significantly higher for SG (p<0.05) in 45° and 90°. EMG activity for SG was significantly higher in agonist contractions for VL, RF and ST, and significantly lower in antagonist contractions for RF and ST when compared to AG Torque and EMG ratios were similar between groups and there were good correlations between torque ratio and BF:VL ratio (r=0.71, p=0.02) and BF:RF ratio (r=0.81, p=0.004) at 45. The EMG results could overestimate the joint balance calculated using torque ratios. Differences in recruitment pattern between soccer players and non-athletes can be related to the training routines and the EMG ratios presents applicable in trained populations.

Background: Fluctuations of estradiol and progesterone levels caused by the menstrual cycle worsen asthma symptoms. Conflicting data are reported in literature regarding pro and anti-inflammatory properties of estradiol and progesterone.Methods: Female Wistar rats were ovalbumin (OVA) sensitized 1 day after resection of the ovaries (OVx). Control group consisted of sensitized-rats with intact ovaries (Sham-OVx). Allergic challenge was performed by aerosol (OVA 1%, 15 min) two weeks later. Twenty four hours after challenge, BAL, bone marrow and total blood cells were counted. Lung tissues were used as explants, for expontaneous cytokine secretion in vitro or for immunostaining of E-selectin.Results: We observed an exacerbated cell recruitment into the lungs of OVx rats, reduced blood leukocytes counting and increased the number of bone marrow cells. Estradiol-treated OVx allergic rats reduced, and those treated with progesterone increased, respectively, the number of cells in the BAL and bone marrow. Lungs of OVx allergic rats significantly increased the E-selectin expression, an effect prevented by estradiol but not by progesterone treatment. Systemically, estradiol treatment increased the number of peripheral blood leukocytes in OVx allergic rats when compared to non treated-OVx allergic rats. Cultured-BAL cells of OVx allergic rats released elevated amounts of LTB4 and nitrites while bone marrow cells increased the release of TNF-α and nitrites. Estradiol treatment of OVx allergic rats was associated with a decreased release of TNF-α, IL-10, LTB4 and nitrites by bone marrow cells incubates. In contrast, estradiol caused an increase in IL-10 and NO release by cultured-BAL cells. Progesterone significantly increased TNF- α by cultured BAL cells and bone marrow cells.Conclusions: Data presented here suggest that upon hormonal oscillations the immune sensitization might trigger an allergic lung inflammation whose phenotype is under control of estradiol. Our data could contribute to the understanding of the protective role of estradiol in some cases of asthma symptoms in fertile ans post-menopausal women clinically observed. © 2010 de Oliveira et al; licensee BioMed Central Ltd.

The present study investigated the trail-following behavior of the subterranean termite Coptotermes gestroi (Wasmann Rhinotermitidae) under laboratory conditions. The results showed that workers were the first to initiate the exploration to the food source. When food was discovered they returned to the nest laying a trail for recruiting nestmates to the food source. In this situation, workers always traveled significantly faster when returning from the arenas. Both workers and soldiers were recruited to the food source; however, the soldier/worker proportion was higher during the first phase of the recruitment. When no food was available, the number of recruited nestmates and the speed on their way back to the nest were significantly lower. The results also showed that scout foragers always laid trail pheromones when entering into unknown territories, and that chemical signals found in the food could induce workers of C. gestroi to increase their travel speed. We evaluated the population dynamics of Acetes americanus Ortmann, 1893 focusing on sex ratio, individual growth, longevity, and the juvenile recruitment period. Samples were collected monthly from January 2006 to June 2007 in the bay of Ubatuba, Brazil. Specimen growth was identified for each gender, and the chosen cohorts were fitted in a von Bertalanffy Growth Model (VBGM); longevity was estimated by the von Bertalanffy inverse equation, considering 99% of the asymptotic length. A total of 6881 individuals (2343 males and 4538 females) were captured. On average the body size (total length) was greater in females (14.64 ± 3.34 mm) than in males (12.27 ± 1.86 mm). The mean growth curves (obtained by grouping the cohorts for each sex), provided estimates of TL∞ = 19.33 mm, k = 0.02 and t0 = -0.12 days for females and TL∞ = 15.13 mm, k = 0.03 and to = -0.07 days for males, where TL∞ is the asymptotic length, k is coefficient of growth and to is the theorical age when the size is equal to 0. Longevity was estimated at 0.61 years for females and 0.50 years for males. The sex ratio tended to favor females, which corroborates with others studies of sergestids. Our finding that males of A. americanus have higher values of k and therefore achieve a smaller size relative to females has been observed in other penaeids. We concluded that this differential growth pattern between the sexes is found across Dendrobranchiata. The life cycles of penaeids have an average duration of approximately 1-2 years, but our results corroborate other studies that estimate a shorter longevity for Acetes, as species of this genus are typically smaller in size. We found continuous recruitment with two main peaks observed during the study period, corroborating previous studies of Acetes. © The Crustacean Society, 2013. Published by Brill NV, Leiden.

One of the most intriguing questions in ecology is how to identify which and how many species will be able to inhabit human-modified landscapes. Large-bodied mammals structure plant communities by trampling, herbivory, seed dispersal and predation, and their local extinction may have pervasive consequences in plant communities due to the breakdown of key interactions. Although much attention has been given to understanding the effects of defaunation on plant communities, information on the potential impacts on plant functional groups (seed dispersal, seed size and seedling leaves defense) inhabiting continuous forests after defaunation is scarce. We conducted mammal surveys (line transects and camera trapping) to determine the defaunation status of a continuous Atlantic forest in Brazil. Then, we evaluated the effects of defaunation on seedling diversity, richness and abundance of functional groups using 15 plot-pairs (each pair with one open and one exclusion plot) monitored over 36. months. We found that the studied area is partially defaunated because it exhibits high abundance of primates, while terrestrial mammals, such as large rodents and ungulates, are rare. We found no significant changes in either seedling richness and diversity or in the seedling composition of plant functional groups in response to mammal exclosure. Seedling mortality and recruitment were similar between plot types. Our findings suggest that at semi-defaunated areas, where arboreal species are still present, terrestrial mammals have low impacts on the plant community reassembly. © 2013 Elsevier Ltd.

Paracoccidioidomycosis is a human systemic mycosis caused by the fungus Paracoccidioides brasiliensis. The mechanisms involved in innate immune response to this fungus are not fully elucidated. Leukotrienes are known to be critical for the clearance of various microorganisms, mainly by mediating the microbicidal function of phagocytes. We investigated the involvement of leukotriene B4 in the early stages of experimental paracoccidioidomycosis, which was induced by intratracheal inoculation of the fungus in selected mouse lines. The mouse lines utilized were produced through bi-directional phenotypic selection, endowed with maximal or minimal acute inflammatory reactivity, and designated AIRmax and AIRmin, respectively. AIRmax mice were more resistant to the infection, which was demonstrated by reduced lung fungal loads. However, the two lines produced similar amounts of leukotriene B4, and pharmacological inhibition of this mediator provoked similar fungal load increases in the two lines. The lower fungal load in the AIRmax mice was associated with a more effective inflammatory response, which was characterized by enhanced recruitment and activation of phagocytic cells and an increased production of activator cytokines. This process resulted in an increased release of fungicidal molecules and a diminution of fungal load. In both lines, leukotriene production was associated with a protective response in the lung that was consequent to the effect of this eicosanoid on the influx and activation of phagocytes. © 2013 ISHAM.

Formigas são um dos mais importantes grupos animais nas florestas tropicais devido a sua abundância e seu número de espécies. Uma característica importante do grupo é a eusocialidade, que permite a ocorrência do comportamento de recrutamento quando um recurso alimentar é encontrado. Entretanto, existem duas questões principais acerca desse comportamento: (i) o recrutamento é um produto de pressões ambientais ou filogenéticas, e (ii) a velocidade de recrutamento é relacionada ao tamanho corpóreo das espécies de formigas. Neste trabalho nós analisamos essas duas questões em 17 espécies de formigas neotropicais, na floresta Amazônica densa de terras baixas. De acordo com os resultados, o recrutamento é fortemente relacionado com o tamanho da formiga, sendo que espécies menores exibem essa característica quando encontram uma fonte protéica. Entretanto, o tamanho das espécies não é importante na velocidade de recrutamento, o que sugere que a velocidade de recrutamento pode ser melhor explicado pelo tipo de recursos alimentares necessários à colônia.
