Tellurium tetrachloride adds to alkynes via two pathways: a concerted syn addition, that yields Z-tri- and tetra-substituted alkenes or by an anti addition that yields E-alkenes. The mechanistic aspects of these divergent pathways have been reevaluated at the light of crystallographic data. The molecules, of the title compound, in the crystal, are associated in a helical fashion with a Te...Te pitch of 6.3492(6) angstrom. As it exhibits inhibitory activity for cathepsin B and in order to gain more insight of the inhibition mechanism, a docking study was undertaken providing insight on why organic telluranes are more efficient inhibitors than inorganic ones as AS-101. TLC autographic assay revealed, in the EtOAc extract obtained from leaves and root bark of Maytenus aquifolium (Celastraceae), the presence of five compounds exhibiting antioxidant properties towards beta-carotene. They were isolated and identified as epigallocatechin (1), (+) ouratea-catechin (2), proanthocyanidin (3), kaempferol 3-O-alpha-L-rhamnopyranosyl (1-->6)-O-[beta-D-glucopyranosyl (1-->3)-O-alpha-L-rhamnopyranosyl-(1-->2)]-O-beta-D-glucopyranosyl (4) and quercetin 3-O-alpha-L-rhamnopyranosyl (1-->6)-O-beta-D-glucopyranosyl (1-->3)-O-alpha-L-rhamnopyranosyl-(1-->2)-O-beta-D-glucopyranosyl (5). The isolates were investigated for their redox properties using cyclic voltammetry and for their radical scavenging abilities through spectrophotometric assay on the reduction of 2,2-diphenyl-pycryl hydrazyl (DPPH). These results were correlated to the inhibition of beta-carotene bleaching on TLC autographic assay and to structural features of the flavonoids. Copyright (C) 2003 John Wiley Sons, Ltd.

The presence of tyrosine-phosphorylated proteins was studied in cultured rat pancreatic islets, Immunoblotting performed with total extracts of islets cultured in the presence of 1.8 or 5.6 mM glucose revealed at least three distinct tyrosine-phosphorylated bands (25 kDa, 95 kDa and 165-185 kDa). After 12 h incubation in medium containing 1.8 mM glucose, a pulse exposition to 11 or 22 mM glucose or to 10(-7) M insulin led to a substantial increase in the phosphorylation of all three bands, with no appearance of novel bands. Immunoprecipitation with specific antibodies demonstrated that the signal detected at 95 kDa corresponds to the beta subunit of the insulin receptor (IR) while the band at 165-185 kDa corresponds to the early substrates of the insulin receptor, IRS-1 and IRS-2. Immunoprecipitation with IRS-I or IRS-2 antisera detected their association with the lipid metabolizing enzyme phosphatidylinositol 3-kinase (PI 3-kinase), Thus, this is the first demonstration that elements involved in the insulin-signalling pathway of traditional target tissues are also present in pancreatic islets and are potentially involved in auto- and paracrine-signalling in this organ.

Atypical enteropathogenic Escherichia coli (aEPEC) strains are diarrheal pathogens that lack bundle-forming pilus production but possess the virulence-associated locus of enterocyte effacement. aEPEC strain 1551-2 produces localized adherence (LA) on HeLa cells; however, its isogenic intimin (eae) mutant produces a diffuse-adherence (DA) pattern. In this study, we aimed to identify the DA-associated adhesin of the 1551-2 eae mutant. Electron microscopy of 1551-2 identified rigid rod-like pili composed of an 18-kDa protein, which was identified as the major pilin subunit of type 1 pilus (T1P) by mass spectrometry analysis. Deletion of fimA in 1551-2 affected biofilm formation but had no effect on adherence properties. Analysis of secreted proteins in supernatants of this strain identified a 150-kDa protein corresponding to SslE, a type 2 secreted protein that was recently reported to be involved in biofilm formation of rabbit and human EPEC strains. However, neither adherence nor biofilm formation was affected in a 1551-2 sslE mutant. We then investigated the role of the EspA filament associated with the type 3 secretion system (T3SS) in DA by generating a double eae espA mutant. This strain was no longer adherent, strongly suggesting that the T3SS translocon is the DA adhesin. In agreement with these results, specific anti-EspA antibodies blocked adherence of the 1551-2 eae mutant. Our data support a role for intimin in LA, for the T3SS translocon in DA, and for T1P in biofilm formation, all of which may act in concert to facilitate host intestinal colonization by aEPEC strains. ©2013, American Society for Microbiology.

Os dados sísmicos terrestres são afetados pela existência de irregularidades na superfície de medição, e.g. a topografia. Neste sentido, para obter uma imagem sísmica de alta resolução, faz-se necessário corrigir estas irregularidades usando técnicas de processamento sísmico, e.g. correições estáticas residuais e de campo. O método de empilhamento Superfície de Reflexão Comum, CRS ("Common-Reflection-Surface", em inglês) é uma nova técnica de processamento para simular seções sísmicas com afastamento-nulo, ZO ("Zero-Offset", em inglês) a partir de dados sísmicos de cobertura múltipla. Este método baseia-se na aproximação hiperbólica de tempos de trânsito paraxiais de segunda ordem referido ao raio (central) normal. O operador de empilhamento CRS para uma superfície de medição planar depende de três parâmetros, denominados o ângulo de emergência do raio normal, a curvatura da onda Ponto de Incidência Normal, NIP ("Normal Incidence Point", em inglês) e a curvatura da onda Normal, N. Neste artigo o método de empilhamento CRS ZO 2-D é modificado com a finalidade de considerar uma superfície de medição com topografia suave também dependente desses parâmetros. Com este novo formalismo CRS, obtemos uma seção sísmica ZO de alta resolução, sem aplicar as correições estáticas, onde em cada ponto desta seção são estimados os três parâmetros relevantes do processo de empilhamento CRS.

Studies have shown a relationship between vitamin D and psoriasis. We compared serum levels of vitamin D of 20 psoriasis patients and 20 controls. The median vitamin D level was 22.80 +/- 4.60 ng/ml; the median in the cases was 23.55 +/- 7.6 ng/ml, and in controls 22.35 +/- 3.10 ng/ml (p = 0.73). Only 2 cases and 4 controls had sufficient levels of vitamin D, although without statistical significance between the groups (p = 0.608). Levels were lower in women with psoriasis compared with those in male patients (20.85 +/- 6.70 x 25.35 +/- 2.90 ng/ml, p = 0.03), a finding that was not observed among controls.

Pollen and geochemical data from Little Lake, western Oregon, suggest several patterns of millennial-scale environmental change during marine isotope stage (MIS) 2 (14,100–27,600 cal yr B.P.) and the latter part of MIS 3 (27,600–42,500 cal yr B.P.). During MIS 3, a series of transitions between warm- and cold-adapted taxa indicate that temperatures oscillated by ca. 2±–4±C every 1000–3000 yr. Highs and lows in summer insolation during MIS 3 are generally associated with the warmest and coldest intervals. Warm periods at Little Lake correlate with warm sea-surface temperatures in the Santa Barbara Basin. Changes in the strength of the subtropical high and the jet stream may account for synchronous changes at the two sites. During MIS 2, shifts between mesic and xeric subalpine forests suggest changes in precipitation every 1000–3000 yr. Increases in Tsuga heterophylla pollen at 25,000 and 22,000 cal yr B.P. imply brief warmings. Minimum summer insolation and maximum global ice-volumes during MIS 2 correspond to cold and dry conditions. Fluctuations in precipitation at Little Lake do not correlate with changes in the Santa Barbara Basin and may be explained by variations in the strength of the glacial anticyclone and the position of the jet stream.
