This work introduces two major changes to the conventional protocol for designing plastic antibodies: (i) the imprinted sites were created with charged monomers while the surrounding environment was tailored using neutral material; and (ii) the protein was removed from its imprinted site by means of a protease, aiming at preserving the polymeric network of the plastic antibody. To our knowledge, these approaches were never presented before and the resulting material was named here as smart plastic antibody material (SPAM). As proof of concept, SPAM was tailored on top of disposable gold-screen printed electrodes (Au-SPE), following a bottom-up approach, for targeting myoglobin (Myo) in a point-of-care context. The existence of imprinted sites was checked by comparing a SPAM modified surface to a negative control, consisting of similar material where the template was omitted from the procedure and called non-imprinted materials (NIMs). All stages of the creation of the SPAM and NIM on the Au layer were followed by both electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV). AFM imaging was also performed to characterize the topography of the surface. There are two major reasons supporting the fact that plastic antibodies were effectively designed by the above approach: (i) they were visualized for the first time by AFM, being present only in the SPAM network; and (ii) only the SPAM material was able to rebind to the target protein and produce a linear electrical response against EIS and square wave voltammetry (SWV) assays, with NIMs showing a similar-to-random behavior. The SPAM/Au-SPE devices displayed linear responses to Myo in EIS and SWV assays down to 3.5 μg/mL and 0.58 μg/mL, respectively, with detection limits of 1.5 and 0.28 μg/mL. SPAM materials also showed negligible interference from troponin T (TnT), bovine serum albumin (BSA) and urea under SWV assays, showing promising results for point-of-care applications when applied to spiked biological fluids.

This work presents the development of a low cost sensor device for the diagnosis of breast cancer in point-of-care, made with new synthetic biomimetic materials inside plasticized poly(vinyl chloride), PVC, membranes, for subsequent potentiometric detection. This concept was applied to target a conventional biomarker in breast cancer: Breast Cancer Antigen (CA15-3). The new biomimetic material was obtained by molecularly-imprinted technology. In this, a plastic antibody was obtained by polymerizing around the biomarker that acted as an obstacle to the growth of the polymeric matrix. The imprinted polymer was specifically synthetized by electropolymerization on an FTO conductive glass, by using cyclic voltammetry, including 40 cycles within -0.2 and 1.0 V. The reaction used for the polymerization included monomer (pyrrol, 5.0×10-3 mol/L) and protein (CA15-3, 100U/mL), all prepared in phosphate buffer saline (PBS), with a pH of 7.2 and 1% of ethylene glycol. The biomarker was removed from the imprinted sites by proteolytic action of proteinase K. The biomimetic material was employed in the construction of potentiometric sensors and tested with regard to its affinity and selectivity for binding CA15-3, by checking the analytical performance of the obtained electrodes. For this purpose, the biomimetic material was dispersed in plasticized PVC membranes, including or not a lipophilic ionic additive, and applied on a solid conductive support of graphite. The analytical behaviour was evaluated in buffer and in synthetic serum, with regard to linear range, limit of detection, repeatability, and reproducibility. This antibody-like material was tested in synthetic serum, and good results were obtained. The best devices were able to detect 5 times less CA15-3 than that required in clinical use. Selectivity assays were also performed, showing that the various serum components did not interfere with this biomarker. Overall, the potentiometric-based methods showed several advantages compared to other methods reported in the literature. The analytical process was simple, providing fast responses for a reduced amount of analyte, with low cost and feasible miniaturization. It also allowed the detection of a wide range of concentrations, diminishing the required efforts in previous sample pre-treating stages.

Feldspato é o nome dado a uma importante família de minerais, do grupo dos tectossilicatos, que compõem o material rochoso de mais de 60% da crusta terrestre. Estes minerais são constituintes essenciais da maior parte das rochas cristalinas e a sua importância é tal que o seu estudo constitui um capítulo fundamental da petrografia. Dada a sua grande incidência na crusta terrestre, os feldspatos ocorrem em diversos cenários geológicos, desde os depósitos pegmatitos, aos aplitos, granitos, sienitos nefelínicos e ainda areias feldspáticas. As suas características químicas, físicas e mineralógicas fazem com que este grupo de minerais esteja presente nas mais diversificadas indústrias, tais como a vidreira, cerâmica, tintas, borrachas, plásticos, abrasivos ligeiros, elétrodos de soldadura, entre outros. Em Portugal, as principais unidades produtoras de feldspatos encontram‐se distribuídas pelo Norte e Centro do país, mais concretamente pelos distritos de Viana do Castelo, Braga, Vila Real, Porto, Viseu e Guarda. Novos estudos geológicos acompanhados de prospeções, nomeadamente na região do maciço central, têm sido efetuados com vista à descoberta de novos depósitos com potencial para exploração deste importante mineral. O depósito aplítico da Argemela, no concelho do Fundão apresenta‐se como um depósito cujas dimensões e qualidade apresentadas indicam a possibilidade de exploração. Nesse sentido, o presente trabalho consiste em analisar e caracterizar os materiais presentes na quele depósito mineral, tendo em vista a respectiva aplicação industrial, e posteriormente propor uma instalação de processamento do material para que o mesmo seja comercializado. Também será objeto deste trabalho a análise da viabilidade económica do projeto, mais concretamente fazendo a previsão do investimento inicial de modo a somar os custos de produção correntes e avaliar o retorno com base na comercialização do material no modo “Ex Works”.

As ligações adesivas são frequentemente utilizadas na fabricação de estruturas complexas que não poderiam ou não seriam tão fáceis de ser fabricadas numa só peça, a fim de proporcionar uma união estrutural que, teoricamente, deve ser pelo menos tão resistente como o material de base. As juntas adesivas têm vindo a substituir métodos como a soldadura, e ligações parafusadas e rebitadas, devido à facilidade de fabricação, menor custo, facilidade em unir materiais diferentes, melhor resistência, entre outras características. Os materiais compósitos reforçados com fibra de carbono são amplamente utilizados em muitas indústrias, tais como de construção de barcos, automóvel e aeronáutica, sendo usados em estruturas que requerem elevada resistência e rigidez específicas, o que reduz o peso dos componentes, mantendo a resistência e rigidez necessárias para suportar as diversas cargas aplicadas. Embora estes métodos de fabricação reduzam ao máximo as ligações através de técnicas de fabrico avançadas, estas ainda são necessárias devido ao tamanho dos componentes, limitações de projecto tecnológicas e logísticas. Em muitas estruturas, a combinação de compósitos com metais tais como alumínio ou titânio traz vantagens de projecto. Este trabalho tem como objectivo estudar, experimentalmente e por modelos de dano coesivo (MDC), juntas adesivas em L entre componentes de alumínio e compósito de carbono epóxido quando solicitados a forças de arrancamento, considerando diferentes configurações de junta e adesivos de ductilidade distinta. Os parâmetros geométricos abordados são a espessura do aderente de alumínio (tP2) e comprimento de sobreposição (LO). A análise numérica permitiu o estudo da distribuição das tensões, evolução do dano, resistência e modos de rotura. Os testes experimentais validam os resultados numéricos e fornecem mecanismos de projecto para juntas em L. Foi mostrado que a geometria do aderente em L (alumínio) e o tipo de adesivo têm uma influência directa na resistência de junta.

Numa recensão às conferências sobre "Metafísica - Conceitos e Problemas" {Metaphysik. Begriffe und Probleme), proferidas por Adorno, na Universidade de Frankfurt, no segundo semestre de 1965 e publicadas postumamente, o filósofo dinamarquês Stâle R. S. Finke (1999) analisa os "esforços" de Adorno por "recuperar a concepção da experiência metafísica". Uma tal recuperação teria de ser compreendida num quadro de pensamento que é o da "dialéctica negativa", por sua vez tributária dos padrões kantianos de "consciência conceptual e autonomia moral" que Adorno articula naquilo a que Finke chama o "projecto" ou o "ideal de liberdade expressiva". O conceito de experiência metafísica, formulado nas referidas conferências, seria, assim, da maior importância para Adorno, pois que nele se daria expressão ao objectivo da "dialéctica negativa" (a Negative Dialektik foi publicada no ano seguinte, 1966): "a invocação do não-idêntico como condição de experiência e de expressão". "Relacionada intrinsecamente" com o "donnnio da experiência possível", histórica e cultural, isto é, com tudo aquilo que constrange a autonomia (por um lado permitindo, por outro lado limitando as possibilidades do eu), a concepção da experiência metafísica em Adorno convoca essas possibilidades do eu como inalcançadas

BACKGROUND: Congenital intracranial dermoid tumors are very rare. The location of these dermoid lesions in the cavernous sinus and the complexity of the operative procedure for these lesions have been noted by several authors. Dermoid tumors originating in the cavernous sinus are usually interdural, and thus blurred vision is an uncommon presentation. CASE DESCRIPTION: Herein we report the first incidental case of a cavernous sinus dermoid cyst in a 21-year-old woman. CONCLUSIONS: A literature review was done and the possible treatments and approaches for this lesion are discussed. We consider that surgical treatment is indicated in most incidental cavernous sinus dermoid lesions due to the possible symptoms related to compression or rupture leading to chemical meningitis.

The aim of the present study is to evaluate cyst wall and protoscolex as an alternate source of antigen in serodiagnosis of cystic echinococcosis (CE). A total of 90 blood samples, 30 each of confirmed CE cases, disease controls and healthy controls were collected. Dot-ELISA using cyst wall, protoscolex and cyst fluid were used to demonstrate anti-hydatid antibodies. The sensitivity of Dot-ELISA using cyst wall, protoscolex and cyst fluid was 96.66%, 86.66% and 93.33% respectively and the specificity of the assay was 70% for Dot-ELISA using cyst fluid, protoscolex and cyst wall antigens. Results of the present study show that cyst wall and protoscolex can also be an useful source of antigen in detection of hydatid antibodies in the serodiagnosis of CE.

To turn wood into a construction material with enhanced properties, many methods of chemical modification have been developed in the last few decades. In this work, mechanical properties of pine wood were chemically modified, compared and evaluated. Maritime pine wood (Pinus pinaster) was modified with four chemical processes: 1,3-dimethylol-4,5- dihydroxyethyleneurea, N-methylol melamine formaldehyde, tetra-alkoxysilane and wax. The following mechanical properties were assessed experimentally: Modulus of elasticity measured statically, stiffness stabilization efficiency in different climates (30 and 87% of relative humidity), modulus of rupture, work maximum load, impact bending strength, compression, tensile and shear strength at indoor conditions (65% of relative humidity). In both types of active principle of modification, cell wall or lumen fill, no significant changes on the bending stiffness (modulus of elasticity) were found. In the remaining properties analysed significant changes in the modified wood-material took place compared to unmodified wood control: - Cell wall modification was the most effective method to achieve high stiffness stabilization efficiency (up to 60%) and also increased compression strength (up to 230%). However, modulus of rupture, tensile, shear and the impact bending strength were reduced by both resins, but in a varying extent, where the N-methylol melamine formaldehyde endured less reduction than 1,3-dimethylol-4,5-dihydroxyethyleneurea resin. In the latter, reduction up to 60% can take place. - In the lumen fill modification: tetra-alkoxysilane has no effect in the mechanical properties. Although, a slight increase in shear strength parallel to the grain was found. Wax specimens have shown a slight increase in bending strength, compression, tensile and shear strength as well as in the absorption energy capacity.

This study evaluated epidemiological factors for HCV infection associated with sharing perforating and cutting instruments among candidates for blood donation (CBD) in the city of Belém, Pará, Brazilian Amazon. Two definitions of HCV infection cases were used: anti-HCV positivity shown by EIA, and HCV-RNA detection by PCR. Infected and uninfected CBD completed a questionnaire about possible risk factors associated with sharing perforating and cutting instruments. The information was evaluated using simple and multiple logistic regressions. Between May and November 2010, 146 (1.1%) persons with anti-HCV antibodies and 106 (0.8%) with HCV-RNA were detected among 13,772 CBD in Belém. Risk factors associated with HCV infection based on the EIA (model 1) and PCR (model 2) results were: use of needles and syringes sterilized at home; shared use of razors at home, sharing of disposable razors in barbershops, beauty salons etc.; and sharing manicure and pedicure material. The models of HCV infection associated with sharing perforating and cutting instruments should be taken into account by local and regional health authorities and by those of other countries with similar cultural practices, in order to provide useful information to guide political and public strategies to control HCV transmission.
