This work was carried out to evaluate the functional response of Cryptolaemus montrouzieri Mulsant, 1850 (Coleoptera: Coccinellidae) fed with Planococcus citri Risso, 1813 (Hemiptera: Pseudococcidae) reared on a pumpkin hybrid (Cucurbita maxima x Cucurbita moscata) (Cucurbitaceae), seedlings of Rangpur lime (Citrus limonia) Rutaceae) and potato (Solanum tuberosum) (Solanaceae) at two temperatures. The predation rate of C. montrouzieri was measured using Petri dishes of 15 cm diameter with 1, 2, 4, 8, 16 and 24 adults of P. Citri. One third instar larva, one fourfh instar and one newly emerged adult (without differentiation of sex) of C. montrouzieri were added to each plate. The study was conducted in climatic chambers at temperatures of 25 and 30 degrees C and photophase of 12 hours. The predation rate was evaluated after 24 hours of prey exposition to the predator, by counting the number of preys trapped in the different treatments and control. The statistical design was completely randomized with four treatments x 6 subplots with 7 repetitions, the two temperatures. The values obtained were subjected to analysis of variance, to relate the number of scales preyed by larvae and adults of C. montrouzieri set up in different substrates. The amount of prey consumed by larvae and adults of the predator increased with increasing the prey density until it reaches a plateau, characterizing functional response type II. In general, the number of scales preyed by larvae and adults of C. montrouzieri was higher on potato and under temperature of 30 degrees C.

Salmonella enterica subsp. enterica serovar Gallinarum biovar Pullorum is a bird-restricted pathogen which causes pullorum disease. The strain FCAV198 was isolated from a pool of chicken ovaries in Brazil, and its genome may be helpful for studies involving molecular mechanisms related to pathogenesis and other related applications.

Huanglongbing (HLB) is associated with Candidatus Liberibacter spp., endogenous, sieve tube-restricted bacteria that are transmitted by citrus psyllid insect vectors. Transgenic expression in the phloem of specific genes that might affect Ca. Liberibacter spp. growth and development may be an adequate strategy to improve citrus resistance to HLB. To study specific phloem gene expression in citrus, we developed three different binary vector constructs with expression cassettes bearing the beta-glucuronidase (GUS) reporter gene (uidA) under the control of one of the three different promoters: Citrus phloem protein 2 (CsPP2), Arabidopsis thaliana phloem protein 2 (AtPP2), and Arabidopsis thaliana sucrose transporter 2 (AtSUC2). Transgenic lines of 'Hamlin', 'Pera', and 'Valencia' sweet oranges [Citrus sinensis (L.) Osbeck] were produced via Agrobacterium tumefaciens transformation. The epicotyl segments collected from in vitro germinated seedlings were used as explants. The gene nptII, which confers resistance to the antibiotic kanamycin, was used for selection. The transformation efficiency was expressed as the number of GUS-positive shoots over the total number of explants and varied from 1.54 to 6.08 % among the three cultivars and three constructs studied. Several lines of the three sweet orange cultivars analyzed using PCR and Southern blot analysis were genetically transformed with the three constructs evaluated. The histological GUS activity in the leaves indicates that the uidA gene was preferentially expressed in the phloem, which suggests that the use of the three promoters might be adequate for producing HLB-resistant transgenic sweet oranges. The results reported here conclusively demonstrate the preferential expression of GUS in the phloem driven by two heterologous and one homologous gene promoters. RpfG is a member of a class of wide spread bacterial two-component regulators with an HD-GYP cyclic di-GMP phosphodiesterase domain. In the plant pathogen Xanthomonas campestris, RpfG together with the sensor kinase RpfC regulates multiple factors as a response to the cell-to-cell Diffusible Signalling Factor (DSF). A dynamic physical interaction of RpfG with two diguanylate cyclase (GGDEF) domain proteins controls motility. Here we show that, contrary to expectation, regulation of motility by the GGDEF domain proteins does not depend upon their cyclic di-GMP synthetic activity. Furthermore we show that the complex of RpfG and GGDEF domain proteins recruits a specific PilZ domain adaptor protein, and this complex then interacts with the pilus motor proteins PilU and PiIT. The results support a model in which DSF signalling influences motility through the highly regulated dynamic interaction of proteins that affect pilus action. A specific motif that we identify to be required for HD-GYP domain interaction is conserved in a number of GGDEF domain proteins, suggesting that regulation via interdomain interactions is of broad relevance.
