Visible and near infrared (vis-NIR) spectroscopy is widely used to detect soil properties. The objective of this study is to evaluate the combined effect of moisture content (MC) and the modeling algorithm on prediction of soil organic carbon (SOC) and pH. Partial least squares (PLS) and the Artificial neural network (ANN) for modeling of SOC and pH at different MC levels were compared in terms of efficiency in prediction of regression. A total of 270 soil samples were used. Before spectral measurement, dry soil samples were weighed to determine the amount of water to be added by weight to achieve the specified gravimetric MC levels of 5, 10, 15, 20, and 25 %. A fiber-optic vis-NIR spectrophotometer (350-2500 nm) was used to measure spectra of soil samples in the diffuse reflectance mode. Spectra preprocessing and PLS regression were carried using Unscrambler® software. Statistica® software was used for ANN modeling. The best prediction result for SOC was obtained using the ANN (RMSEP = 0.82 % and RPD = 4.23) for soil samples with 25 % MC. The best prediction results for pH were obtained with PLS for dry soil samples (RMSEP = 0.65 % and RPD = 1.68) and soil samples with 10 % MC (RMSEP = 0.61 % and RPD = 1.71). Whereas the ANN showed better performance for SOC prediction at all MC levels, PLS showed better predictive accuracy of pH at all MC levels except for 25 % MC. Therefore, based on the data set used in the current study, the ANN is recommended for the analyses of SOC at all MC levels, whereas PLS is recommended for the analysis of pH at MC levels below 20 %.

RESUMO O conhecimento dos solos é cada vez mais importante para que o uso dele seja realizado corretamente na agropecuária, no crescimento urbano, na conservação dos recursos naturais, entre outros. Entretanto, verifica-se carência de profissionais qualificados para a caracterização e os mapeamentos pedológicos, particularmente em escalas de maior detalhamento. Essa carência, aliada aos avanços das ferramentas computacionais e do sensoriamento remoto, promoveu o surgimento do Mapeamento Digital de Solos (MDS), que busca auxiliar e agilizar as atividades de levantamento pedológico. Assim, este trabalho objetivou desenvolver uma metodologia de delimitaçao de unidades de solos em topossequências por meio do comportamento espectral dos solos no comprimento de onda do Visível-Infravermelho Próximo (Vis-NIR). A metodologia espectral consistiu na obtenção das curvas espectrais dos solos por meio do espectrorradiômetro FieldSpecPro e da redução do número de informações espectrais por meio da análise de Componentes Principais, seguida de agrupamento das amostras mediante método fuzzy k-médias. Foram selecionadas cinco topossequências com pontos equidistantes de 30 m para caracterizar as classes de solos e amostragens. Foram descritas oito classes de solos distintas, que tiveram caracterização detalhada e classificação em perfis pedológicos. No restante dos pontos, a caracterização das classes de solos foi feita com base na classificação dos solos realizada nos perfis pedológicos, com coleta de amostras por meio de tradagens nas profundidades de 0,00-0,20 e 0,80-1,00 m, perfazendo o total de 162 amostras ao longo das cinco topossequências. As amostras foram analisadas pelas metodologias convencional e espectral, para que os resultados pudessem ser comparados e avaliados. Dessa forma, foram realizadas análises morfológicas, físicas (textura) e químicas nas amostras de solo. Das cinco topossequências estudadas, os resultados foram satisfatoriamente semelhantes; alguns solos não foram perfeitamente individualizados pela metodologia espectral, em razão da grande semelhança em seus comportamentos espectrais, como demonstrado pelo Latossolo Vermelho Férrico e Nitossolo Vermelho Férrico. A metodologia espectral foi capaz de diferenciar solos com resposta espectral distinta e estabelecer limites nas topossequências, apresentando grande potencial para ser implementada em levantamentos pedológicos.

The canvas support in easel paintings is composed mainly of cellulose. One of the maindegradation paths of cellulose is acid-catalysed hydrolysis, which means that in an acidic environment (low pH), its degradation proceeds at a faster rate (Strlič et al., 2005).The main effect of acid-catalysed hydrolysis is the breaking up of the polymer chains,measured by the “Degree of Polymerisation” (DP). The lowering of the DP value impliesa lower mechanical strength of the textile (Scicolone, 1993), and thus this parameter canbe used to monitor degradation. Knowing these two parameters can, therefore, be veryinformative regarding the condition of the canvas support.

The structural saturation and stability, the energy gap, and the density of states of a series of small, silicon-based clusters have been studied by means of the PM3 and some ab initio (HF/6-31G* and 6-311++G**, CIS/6-31G* and MP2/6-31G*) calculations. It is shown that in order to maintain a stable nanometric and tetrahedral silicon crystallite and remove the gap states, the saturation atom or species such as H, F, Cl, OH, O, or N is necessary, and that both the cluster size and the surface species affect the energetic distribution of the density of states. This research suggests that the visible luminescence in the silicon-based nanostructured material essentially arises from the nanometric and crystalline silicon domains but is affected and protected by the surface species, and we have thus linked most of the proposed mechanisms of luminescence for the porous silicon, e.g., the quantum confinement effect due to the cluster size and the effect of Si-based surface complexes.

O objetivo deste trabalho foi avaliar o potencial da espectroscopia de reflectância no VIS-NIR-SWIR, para a caracterização granulométrica de amostras de solos de diferentes classes texturais, e obter modelos de predição dos teores de argila, silte e areia no solo. Utilizou-se um conjunto de amostras representativas de Latossolos e Argissolo de cinco locais do Estado do Mato Grosso do Sul. Os espectros do visível e do infravermelho próximo ao infravermelho de ondas curtas (de 350 a 2.500 nm) das amostras foram obtidos e analisados. Empregaram-se a análise de componentes principais (ACP), agrupamento por "fuzzy c-means", regressão logística multinomial (RLM) e regressão por mínimos quadrados parciais. Espectros característicos para as diferentes classes texturais e a segregação de amostras de classes texturais e de locais de coleta com características distintas, por meio da ACP, "fuzzy c-means" e RLM, mostram o potencial semiquantitativo dos dados de reflectância no VIS-NIR-SWIR. Obteve-se quantificação satisfatória quanto à argila (R²=0,92, RPD=3,59), ao silte (R²=0,80, RPD=2,15) e à areia (R²=0,87, RPD=2,62). As técnicas de espectroscopia de reflectância podem auxiliar na determinação da textura e da variabilidade espacial do solo com metodologias semiquantitativas ou quantitativas.

Near-infrared spectroscopy (NIRS) was used to analyse the crude protein content of dried and milled samples of wheat and to discriminate samples according to their stage of growth. A calibration set of 72 samples from three growth stages of wheat (tillering, heading and harvest) and a validation set of 28 samples was collected for this purpose. Principal components analysis (PCA) of the calibration set discriminated groups of samples according to the growth stage of the wheat. Based on these differences, a classification procedure (SIMCA) showed a very accurate classification of the validation set samples : all of them were successfully classified in each group using this procedure when both the residual and the leverage were used in the classification criteria. Looking only at the residuals all the samples were also correctly classified except one of tillering stage that was assigned to both tillering and heading stages. Finally, the determination of the crude protein content of these samples was considered in two ways: building up a global model for all the growth stages, and building up local models for each stage, separately. The best prediction results for crude protein were obtained using a global model for samples in the two first growth stages (tillering and heading), and using a local model for the harvest stage samples.

A espectroscopia na região do visível (VIS) e do infravermelho próximo (NIR) apresenta-se como uma promissora e rápida tecnologia de avaliação das características internas de várias espécies de frutas. A diferença de absorbância, provenientes dos frutos, ao serem submetidos a um espectro emitido entre dois comprimentos de onda próximos ao do pico de absorção da clorofila-a, foi utilizada para o desenvolvimento de um índice (Índice DA- IDA) que se correlaciona ao estádio de maturação dos frutos. Neste trabalho, objetivou-se avaliar as relações existentes entre o índice DA, obtidos com o equipamento DA-meter®, e os parâmetros físico-químicos de maturação utilizados em mangas da cultivar Tommy Atkins, na separação dos frutos em categorias distintas. Para isto, foram utilizados frutos pertencentes a um mesmo lote e adquiridos em um centro de distribuição local, na cidade de Pelotas-RS. Com os resultados obtidos, através do índice DA, foi possível separar os frutos em categorias de acordo com o grau de maturação, demonstrando a grande heterogeneidade com que os frutos são oferecidos ao consumidor, e as relações entre os valores dos índices DA e os parâmetros de qualidade, determinadas através dos coeficientes de determinação (R²), foram significativas para os atributos estudados.

We report on a field-effect light emitting device based on silicon nanocrystals in silicon oxide deposited by plasma-enhanced chemical vapor deposition. The device shows high power efficiency and long lifetime. The power efficiency is enhanced up to 0.1 %25 by the presence of a silicon nitride control layer. The leakage current reduction induced by this nitride buffer effectively increases the power efficiency two orders of magnitude with regard to similarly processed devices with solely oxide. In addition, the nitride cools down the electrons that reach the polycrystalline silicon gate lowering the formation of defects, which significantly reduces the device degradation.

In this work we describe the processing of poly(styrene sulphonate) films (PSS) doped with neodymium (Nd). Optical density measurements in the UV-Vis-NIR region show the typical bands observed for neodymium chloride (NdCl3) in solution. In the case of films, the intensity ratio between the peaks at 800 nm (4I9/2 -> 4F5/2 + ²H7/2) and 580 nm (4I9/2 -> 4G5/2 + ²G7/2) is equal to 0.83. Infrared spectra present an enhancement in the absorption region of aromatic rings. Site selective luminescence spectroscopy shows that the incorporation of Nd introduces a hipsochromic shift and a line shape definition in UV luminescence compared to PSS film, decreasing the interaction between aromatic groups. In addition, the film exhibits an intense radiative transition at 1061 nm (4F3/2->4I11/2), comparable to the one present in crystalline materials doped with Nd.

A statistical mixture-design technique was used to study the effects of different solvents and their mixtures on the yield, total polyphenol content, and antioxidant capacity of the crude extracts from the bark of Schinus terebinthifolius Raddi (Anacardiaceae). The experimental results and their response-surface models showed that ternary mixtures with equal portions of all the three solvents (water, ethanol and acetone) were better than the binary mixtures in generating crude extracts with the highest yield (22.04 ± 0.48%), total polyphenol content (29.39 ± 0.39%), and antioxidant capacity (6.38 ± 0.21). An analytical method was developed and validated for the determination of total polyphenols in the extracts. Optimal conditions for the various parameters in this analytical method, namely, the time for the chromophoric reaction to stabilize, wavelength of the absorption maxima to be monitored, the reference standard and the concentration of sodium carbonate were determined to be 5 min, 780 nm, pyrogallol, and 14.06% w v-1, respectively. UV-Vis spectrophotometric monitoring of the reaction under these conditions proved the method to be linear, specific, precise, accurate, reproducible, robust, and easy to perform.

Electrode kinetics and complex formation of Zn(II) using doxycycline, chlortetracycline, oxytetracycline, tetracycline, minocycline, amoxicillin, chloramphenicol and cephaloglycin were reported at pH = 7.30 ± 0.01 in = 1.0 molL-1 NaClO4 used as supporting electrolyte at 25.0°C. Kinetic parameters viz. transfer coefficient (α), degree of irreversibility (λ) and rate constant (k) were determined. The study showed that 'Transition state' behaves between reactant (O) and product (R) response to applied potential. The stability constants varied from 2.14 to 10.31 showing that these drugs or their complexes could be used against Zn toxicity.

Measuring protein biomarkers from sample matrix, such as plasma, is one of the basic tasks in clinical diagnostics. Bioanalytical assays used for the measuring should be able to measure proteins with high sensitivity and specificity. Furthermore, multiplexing capability would also be advantageous. To ensure the utility of the diagnostic test in point-of-care setting, additional requirements such as short turn-around times, ease-ofuse and low costs need to be met. On the other hand, enhancement of assay sensitivity could enable exploiting novel biomarkers, which are present in very low concentrations and which the current immunoassays are unable to measure. Furthermore, highly sensitive assays could enable the use of minimally invasive sampling. In the development of high-sensitivity assays the label technology and affinity binders are in pivotal role. Additionally, innovative assay designs contribute to the obtained sensitivity and other characteristics of the assay as well as its applicability. The aim of this thesis was to study the impact of assay components on the performance of both homogeneous and heterogeneous assays. Applicability of two different lanthanide-based label technologies, upconverting nanoparticles and switchable lanthanide luminescence, to protein detection was explored. Moreover, the potential of recombinant antibodies and aptamers as alternative affinity binders were evaluated. Additionally, alternative conjugation chemistries for production of the labeled binders were studied. Different assay concepts were also evaluated with respect to their applicability to point-of-care testing, which requires simple yet sensitive methods. The applicability of upconverting nanoparticles to the simultaneous quantitative measurement of multiple analytes using imaging-based detection was demonstrated. Additionally, the required instrumentation was relatively simple and inexpensive compared to other luminescent lanthanide-based labels requiring time-resolved measurement. The developed homogeneous assays exploiting switchable lanthanide luminescence were rapid and simple to perform and thus applicable even to point-ofcare testing. The sensitivities of the homogeneous assays were in the picomolar range, which are still inadequate for some analytes, such as cardiac troponins, requiring ultralow limits of detection. For most analytes, however, the obtained limits of detection were sufficient. The use of recombinant antibody fragments and aptamers as binders allowed site-specific and controlled covalent conjugation to construct labeled binders reproducibly either by using chemical modification or recombinant technology. Luminescent lanthanide labels were shown to be widely applicable for protein detection in various assay setups and to contribute assay sensitivity.

Escherichia coli K-12 (pEGFPluxABCDEAmp) (E. coli-lux), constitutively emitting bioluminescence (BL), was constructed and its BL emitting properties tested in different growth and killing conditions. The BL emission directly correlated with the number of viable E. coli-lux cells, and when subjected to the antimicrobial agent, the diminishment of the BL signal was linked directly to the number of killed bacterial cells. The method provided a very convenient application, especially when compared to conventional plate counting assays. This novel real-time based method was utilized in both immunological and toxicological assessments. The parameters such as the activation phase, the lytic phase and the capacity of the killing of the serum complement system were specified not only in humans but also in other species. E. coli-lux was also successfully used to study the antimicrobial activities of insect haemolymph. The mechanisms of neutrophil activity, like that of a myeloperoxidase (MPO)-H2O2-halide system, were studied using the E. coli-lux approach. The fundamental role of MPO was challenged, since during the actual killing in described circumstances in phagolysosome the MPO system was inactivated and chlorination halted. The toxicological test system, assessing indoor air total toxicity, particularly suitable for suspected mold damages, was designed based on the E. coli-lux method. Susceptibility to the vast number of various toxins, both pure chemicals and dust samples from the buildings and extracts from molds, were investigated. The E. coli-lux application was found to possess high sensitivity and specificity attributes. Alongside the analysis system, the sampling kit for indoor dust was engineered based on the swipe stick and the container. The combination of practical specimen collector and convenient analysis system provided accurate toxic data from the dust sample within hours. Neutrophils are good indicators of the pathophysiological state of the individual, and they can be utilized as a toxicological probe due to their ability to emit chemiluminescence (CL). Neutrophils can either be used as probe cells, directly exposed to the agent studied, or they can act as indicators of the whole biological system exposed to the agent. Human neutrophils were exposed to the same toxins as tested with the E. coli-lux system and measured as luminol amplified CL emission. The influence of the toxins on the individuals was investigated by exposing rats with moniliniformin, the mycotoxin commonly present in Finnish grains. The activity of the rat neutrophils was found to decrease significantly during the 28 days of exposure.
