Background. Substantial evidence exists for HLA and other host genetic factors being determinants of susceptibility or resistance to infectious diseases. However, very little information is available on the role of host genetic factors in HIV-TB coinfection. Hence, a longitudinal study was undertaken to investigate HLA associations in a cohort of HIV seropositive individuals with and without TB in Bangalore, South India. Methods. A cohort of 238 HIV seropositive subjects were typed for HLA-A, B, and DR by PCR-SSP and followed up for 5 years or till manifestation of Tuberculosis. HLA data of 682 HIV Negative healthy renal donors was used as control. Results. The ratio of males and females in HIV cohort was comparable (50.4% and 49.6%). But the incidence of TB was markedly lower in females (12.6%,) than males (25.6%). Further, HLA-B* 57 frequency in HIV cohort was significantly higher among females without TB (21.6%, 19/88) than males (1.7%, 1/59); P = 0.0046; OR = 38. CD4 counts also were higher among females in this cohort. Conclusion. This study suggests that HIV positive women with HLA-B* 57 have less occurrence of TB as compared to males.

Theoretical and computational investigations of nucleation have been plagued by the sensitivity of the phase diagram to the range of the interaction potential. As the surface tension depends strongly on the range of interaction potential and as the classical nucleation theory (CNT) predicts the free energy barrier to be directly proportional to the cube of the surface tension, one expects a strong sensitivity of nucleation barrier to the range of the potential; however, CNT leaves many aspects unexplored. We find for gas-liquid nucleation in Lennard-Jones system that on increasing the range of interaction the kinetic spinodal (KS) (where the mechanism of nucleation changes from activated to barrierless) shifts deeper into the metastable region. Therefore the system remains metastable for larger value of supersaturation and this allows one to explore the high metastable region without encountering the KS. On increasing the range of interaction, both the critical cluster size and pre-critical minima in the free energy surface of kth largest cluster, at respective kinetic spinodals, shift towards smaller cluster size. In order to separate surface tension contribution to the increase in the barrier from other non-trivial factors, we introduce a new scaling form for surface tension and use it to capture both the temperature and the interaction range dependence of surface tension. Gene expression is the most fundamental biological process, which is essential for phenotypic variation. It is regulated by various external (environment and evolution) and internal (genetic) factors. The level of gene expression depends on promoter architecture, along with other external factors. Presence of sequence motifs, such as transcription factor binding sites (TFBSs) and TATA-box, or DNA methylation in vertebrates has been implicated in the regulation of expression of some genes in eukaryotes, but a large number of genes lack these sequences. On the other hand, several experimental and computational studies have shown that promoter sequences possess some special structural properties, such as low stability, less bendability, low nucleosome occupancy, and more curvature, which are prevalent across all organisms. These structural features may play role in transcription initiation and regulation of gene expression. We have studied the relationship between the structural features of promoter DNA, promoter directionality and gene expression variability in S. cerevisiae. This relationship has been analyzed for seven different measures of gene expression variability, along with two different regulatory effect measures. We find that a few of the variability measures of gene expression are linked to DNA structural properties, nucleosome occupancy, TATA-box presence, and bidirectionality of promoter regions. Interestingly, gene responsiveness is most intimately correlated with DNA structural features and promoter architecture.

Specification of the centromere location in most eukaryotes is not solely dependent on the DNA sequence. However, the non-genetic determinants of centromere identity are not clearly defined. While multiple mechanisms, individually or in concert, may specify centromeres epigenetically, most studies in this area are focused on a universal factor, a centromere-specific histone H3 variant CENP-A, often considered as the epigenetic determinant of centromere identity. In spite of variable timing of its loading at centromeres across species, a replication coupled early S phase deposition of CENP-A is found in most yeast centromeres. Centromeres are the earliest replicating chromosomal regions in a pathogenic budding yeast Candida albicans. Using a 2-dimensional agarose gel electrophoresis assay, we identify replication origins (ORI7-LI and ORI7-RI) proximal to an early replicating centromere (CEN7) in C. albicans. We show that the replication forks stall at CEN7 in a kinetochore dependent manner and fork stalling is reduced in the absence of the homologous recombination (HR) proteins Rad51 and Rad52. Deletion of ORI7-RI causes a significant reduction in the stalled fork signal and an increased loss rate of the altered chromosome 7. The HR proteins, Rad51 and Rad52, have been shown to play a role in fork restart. Confocal microscopy shows declustered kinetochores in rad51 and rad52 mutants, which are evidence of kinetochore disintegrity. CENP-A(CaCse4) levels at centromeres, as determined by chromatin immunoprecipitation (ChIP) experiments, are reduced in absence of Rad51/Rad52 resulting in disruption of the kinetochore structure. Moreover, western blot analysis reveals that delocalized CENP-A molecules in HR mutants degrade in a similar fashion as in other kinetochore mutants described before. Finally, co-immunoprecipitation assays indicate that Rad51 and Rad52 physically interact with CENP-A(CaCse4) in vivo. Thus, the HR proteins Rad51 and Rad52 epigenetically maintain centromere functioning by regulating CENP-A(CaCse4) levels at the programmed stall sites of early replicating centromeres.

A obesidade é uma doença crônica, resultante do excesso de gordura no organismo. O aumento da obesidade no mundo, tem se revelado como um dos fenômenos clínicos e epidemiológicos da atualidade. Estudos populacionais e em modelos animais demonstram que a origem da epidemia da obesidade está relacionada a fatores genéticos, modificações de hábitos nutricionais, redução da atividade física, e alterações nutricionais durante a lactação, desempenhando um papel relevante no desenvolvimento da obesidade, DM2 e cardiomiopatias. As mitocôndrias são os coordenadores centrais do metabolismo energético, assim, alterações funcionais e estruturais dessa organela têm sido associadas à desordens metabólicas. Elas exercem um papel na sobrevivência e função dos cardiomiócitos devido à alta demanda energética do miocárdio. Desta forma, disfunções mitocondriais estão relacionadas com disfunções no miocárdio e conseqüente progressão de cardiomiopatias. Neste estudo, avaliamos a bioenergética e a ultraestrutura de cardiomiócitos de camundongos obesos e controle hiperalimentados durante a lactação. O consumo de oxigênio das fibras cardíacas foi avaliado por respirometria de alta-resolução, utilizando um oxígrafo-2K-Oroboros. A ultraestrutura dos cardiomiócitos foi analisada por microscopia eletrônica de transmissão e o conteúdo das proteínas Carnitina palmitoil transferase 1 (CPT1), Proteína desacopladora 2 (UCP2) , Transportador de glicose 1 e 4 (GLUT1) e (GLUT4), Proteína Kinase ativada por AMP (AMPK) e Proteína kinase ativada por AMP fosforilada p(AMPK) por Western blotting (WB). Além disso, o peso dos animais, a gordura retroperitoneal, epididimal e a glicemia em jejum foram determinadas. Nossos resultados confirmaram que os animais do grupo hiperalimentados (GH), aos 90 dias de vida, apresentaram aumento da massa corporal, de gordura epididimária e retroperitoneal comparado ao grupo controle (GC). As taxas respiratórias foram semelhantes nos dois grupos quando foram utilizados os substratos dos complexos I e II. Entretanto, quando o ácido graxo palmitoil-L-carnitina foi utilizado, a taxa respiratória máxima do GH foi significativamente menor. A análise ultraestrutural dos cardiomiócitos do GH demonstrou intenso dano na matriz mitocondrial e maior presença de gotículas de lipídios, caracterizando deposição ectópica. Os resultados do WB mostraram aumento significativo do conteúdo de CPT1 e UCP2 no GH comparado ao GC. Não foram encontradas diferenças significativas no conteúdo de GLUT1 entre os grupos, entretanto, observamos maior conteúdo do GLUT4 no GH. Além disso, encontramos maior conteúdo de AMPK no GH, ao passo que o conteúdo de pAMPK foi semelhante entre os grupos. Entretanto, a razão pAMPK/AMPK é significativamente menor no GH. Esses resultados sugerem que a hiperalimentação durante a lactação leva a obesidade na vida adulta com alterações na bioenergética e ultraestrutura dos cardiomiócitos.
Relação entre fatores genéticos envolvidos em vias metabólicas mitocondriais e a doença de Parkinson

A doença de Parkinson (DP) é uma das desordens neurodegenerativas mais comuns associada ao envelhecimento, alcançando 2% aos 70 anos. É uma doença caracterizada pela degeneração progressiva de neurônios dopaminérgicos nigrais nos gânglios basais e pela presença de inclusões protéicas citoplasmáticas denominadas corpúsculos e neuritos de Lewy nos neurônios sobreviventes. A etiologia da DP é pouco conhecida, sendo considerada, na maioria dos casos, idiopática. Conhecimentos alcançados nos últimos 15 anos sobre a base genética da DP demonstram, claramente, que os fatores genéticos desempenham um importante papel na etiologia desta desordem. Neste trabalho, rastreamos mutações nos genes que codificam proteínas participantes de vias metabólicas mitocondriais (Parkin, PINK1 e DJ-1) em 136 pacientes brasileiros com manifestação precoce da DP, através do sequenciamento automático e da técnica de MLPA. Avaliamos a presença de variantes de sequência por meio do sequenciamento dos exons 1 a 12 do gene Parkin e dos exons 1 a 8 do gene PINK1. Em Parkin foram identificadas três mutações patogênicas ou potencialmente patogênicas, ambas em heterozigose: p.T240M, p.437L e p.S145N. Em PINK1 não encontramos variantes de ponto patogênicas. Através da técnica de MLPA investigamos alterações de dosagem nos genes Parkin, PINK1 e DJ-1. Identificamos cinco alterações no gene Parkin em quatro pacientes: uma duplicação heterozigota do exon 4 no paciente PAR2256, uma deleção heterozigota do exon 4 no probando PAR2099, uma deleção homozigota do exon 4 na paciente PAR3380 e um probando heterozigoto composto (PAR2396) com duas alterações, uma duplicação do exon 3 e uma deleção dos exons 5 e 6. No gene PINK1 identificamos uma deleção heterozigota do exon 1, que nunca foi descrita na literatura, em um paciente (PAR2083). Não encontramos alteração quantitativa no gene DJ-1. Neste estudo obtivemos uma frequência total de mutações patogênicas (pontuais e de dosagem) nos genes estudados de 7,3%, sendo 6,6% no gene Parkin e 0,7% no gene PINK1.

A tese busca identificar os elementos jurídicos e extrajurídicos que interferem sobre o comportamento judicial do Supremo Tribunal Federal. A análise é desenvolvida com base nos seguintes modelos decisórios: o modelo legalista, o modelo ideológico, o modelo institucional e o modelo estratégico de comportamento judicial. Ao longo do trabalho, examina-se a influência do direito, da ideologia, das normas que regem o Judiciário, das regras que regem as decisões colegiadas, do Poder Executivo, do Poder Legislativo, da opinião pública e da imprensa no processo decisório do Supremo Tribunal Federal.

Aquaculture production systems in developing countries are largely based on the use of unimproved species and strains. As knowledge and experience are accumulated in relation to the management, feeding and animal health issues of such production systems, the availability of genetically more productive stock becomes imperative in order to more effectively use resources. For instance, there is little point in providing ideal water conditions and optimum feed quality to fish that do not have the potential to grow faster and to be harvested on time, providing a product of the desired quality. Refinements in the production system and improvement of the stock used must progress hand in hand. In this paper we deal separately with genetic and non-genetic issues pertaining to the multiplication and dissemination of improved strains. The separation is somewhat arbitrary, and as will be evident from our discussion, there is frequent interaction between the two.

A doença de Parkinson (DP) é a segunda doença neurodegenerativa mais frequente no mundo, afetando 1-2% da população acima de 65 anos, caracterizada clinicamente por tremor em repouso, bradicinesia, instabilidade postural e rigidez muscular. Essas manifestações surgem devido à degeneração neuronal progressiva e à presença de inclusões proteicas ricas em α-sinucleína. A DP é decorrente da interação entre fatores ambientais e genéticos, e entre os fatores genéticos, variantes exônicas de transmissão dominante nos genes LRRK2 (leucine-rich repeat kinase 2), VPS35 (vacuolar protein sorting 35) e EIF4G1 (eukaryotic translation initiation factor 4-gamma 1) têm sido associadas à etiologia da doença. Entretanto, estudos sobre o efeito dessas variantes na população brasileira são raros ou inexistentes. Por essa razão, neste trabalho rastreamos mutações nos genes VPS35 (p.D620N; p.R524W), EIF4G1 (p.R1205H; p.A502V) e LRRK2 (p.G2019S) em uma amostra de 582 pacientes brasileiros com DP não aparentados e 329 indivíduos controles saudáveis. Além disso, conduzimos o primeiro estudo caso-controle para análise de variantes exônicas raras (p.Q1111H, p.T1410M, p.M1646T, p.S1761R, p.Y2189C) e comuns (p.N551K, p.R1398H, p.K1423K) no gene LRRK2 em um subgrupo de 329 pacientes brasileiros com DP, não aparentados, naturais da região sudeste. Esse subgrupo foi analisado e comparado com 222 indivíduos controles saudáveis a fim de verificar associações dessas variantes e a DP. Em relação às mutações dos genes VPS35 e EIF4G1, não foram encontradas alterações nos pacientes com DP. A mutação p.G2019S no gene LRRK2 foi encontrada em 15 probandos (2,6%), dos quais 9 são do sexo feminino (64,3%). O tremor em repouso foi observado em 47,36% dos pacientes com a mutação p.G2019S como primeiro sintoma motor. As análises das variantes raras no gene LRRK2 não revelaram qualquer associação estatisticamente significante. Entre as variantes comuns, a p.K1423K mostrou evidência de associação de risco com a DP (p<0,05) na estratificação contendo o grupo de indivíduos com história familiar da doença e para as variantes p.N551K e p.R1398H não foram observadas associações. A análise do haplótipo p.N551K-p.R1398H-p.K1423K revelou associação de proteção na amostra sudeste e na estratificação Rio de Janeiro (p<0,05). Esse haplótipo não está em desequilíbrio de ligação na amostra de 222 indivíduos controles brasileiros analisados (r2≤45). Os resultados obtidos neste estudo representam contribuições valiosas ao entendimento da relação entre as variantes genéticas estudadas e o risco de desenvolvimento da doença de Parkinson, principalmente no que se refere aos endofenótipos associados.

O Transtorno do Déficit de Atenção e Hiperatividade (TDAH) é uma desordem neurocomportamental caracterizada por graus variados de desatenção, atividade motora excessiva e impulsividade. Sua etiologia não é completamente conhecida, porém, um grande número de evidências sugere que, além de fatores de risco genéticos, a exposição gestacional ao etanol e a altas doses de cafeína contribuem para a manifestação deste transtorno. O etanol, presente na composição de bebidas alcoólicas, e a cafeína, presente na composição de diversas bebidas (refrigerantes, chás, café, e energéticos), alimentos derivados do cacau e medicamentos estão entre as substâncias neuroativas mais consumidas no mundo. Apesar das evidências epidemiológicas do uso combinado de cafeína e etanol por gestantes, as interações entre estas substâncias têm recebido pouca atenção em estudos experimentais. Este fato é particularmente importante visto que alguns estudos apontam que cafeína é capaz de ampliar alguns aspectos importantes da ação de outras substâncias com potencial neurotóxico. Nesse estudo avaliamos os efeitos da co-exposição à cafeína e ao etanol durante o desenvolvimento na atividade locomotora em camundongos adolescentes. Para tanto, camundongos Suíços foram expostos do primeiro dia gestacional até o vigésimo primeiro dia pós-natal (PN21), a solução de cafeína 0,1g/L (Grupo CAF1, 10 ninhadas), a solução de cafeína 0,3g/L (Grupo CAF3, 10 ninhadas) ou tiveram acesso à água potável (Grupo CAF0, 10 ninhadas). Em dias alternados de PN2 a PN8, os animais de cada ninhada receberam uma injeção intraperitoneal de 0,25 l/g de etanol (grupo ETOH25), 0,5 l/g de etanol (grupo ETOH50) ou de solução salina (grupo ETOH0). Em PN30, a atividade locomotora foi avaliada por 15 minutos no teste de Campo Aberto. A análise dos níveis de etanol sérico, realizada em uma amostra independente de animais em PN8, indicou que, para as duas doses de etanol, a alcoolemia dos animais do grupo CAF3 foi significativamente maior do que as dos grupos CAF0 e CAF1, que não diferiram entre si. No teste de campo aberto, apenas os animais expostos ao etanol apresentaram aumento da atividade locomotora. Tanto o grupo ETOH25 quanto o grupo ETOH50 tiveram a atividade maior que o grupo ETOH0. A exposição à cafeína, por si só, não afetou a atividade locomotora dos animais nem potencializou os efeitos do etanol. No grupo CAF3, a atividade locomotora não foi afetada pela exposição ao etanol. Nossos dados confirmam o papel da exposição precoce ao etanol na manifestação da hiperatividade locomotora. Além disso, a exposição à cafeína durante o desenvolvimento pode exercer um papel protetor para a manifestação da hiperatividade locomotora induzida pela exposição precoce ao etanol.

沙棘广泛分布于亚欧大陆的温带地区和亚洲亚热带的高海拔地区。沙棘能适应多种生态环境,能耐受多种逆境(如干旱、低温、高温和盐害等)。在中国,沙棘常常被用作植被恢复中的先锋树种而大量栽培。本文以中国沙棘为试验材料,探索沙棘适应干旱机制,以及沙棘对干旱胁迫的适应机制是否存在种群间的差异,同时试图通过分析干旱胁迫下沙棘叶片蛋白质表达变化探索沙棘适应干旱胁迫的分子机理。 对三个分别来自低海拔湿润地区、低海拔干旱地区和高海拔湿润地区的中国沙棘种群进行干旱胁迫处理。干旱胁迫能提高根冠比,比叶面积,降低平均叶面积和总生物量,提高沙棘的抗氧化性酶活性、脯氨酸含量、脱落酸(ABA)含量、降低光合作用,提高长期用水效率。实验中的这两个低海拔种群比高海拔种群抵抗干旱的能力更强,不同的种群采用了不同的策略来耐受干旱胁迫和过氧化胁迫。 在2004 年度的实验中,干旱胁迫处理下,高海拔湿润种群(道孚种群)严重失水,生长也受到更大的抑制,非气孔因素在抑制光合作用方面占支配地位,抗坏血酸含量下降,ABA和脯氨酸含量增加幅度比九寨沟种群的要高,这可能是因为道孚种群严重失水而引起的;而低海拔湿润种群(九寨沟种群)的体内水分状况几乎不受干旱的影响,生长情况也较道孚种群要好。 在2005 年度的试验中,和高海拔湿润地区种群(道孚)相比较,低海拔干旱地区种群(定西)在叶片相对水含量、根冠比、抗氧化酶活性(过氧化氢酶、抗坏血酸过氧化物酶和谷胱甘肽过氧化物酶)、保护性物质(脯氨酸,脱落酸)含量等方面都要高,光能热耗散能力也更强,而且气体交换参数(气孔扩散阻力和胞间CO2浓度等)对干旱也更不敏感。 分析了干旱胁迫下沙棘叶片蛋白质表达的变化。共发现319 个蛋白质,有4 个蛋白在干旱胁迫下消失(Putative ABCtransporter ATP-binding protein 、Hypothetical proteinXP-515578,热激蛋白Hslu219 和一个没得到鉴定的蛋白),4 个只在干旱胁迫下出现(没命名的蛋白质产物,对甲基苯-丙酮酸双加氧酶,NTrX 和一个没得到鉴定的蛋白),46 个蛋白质的表达丰度变化显著,包括32 个干旱负调蛋白,14 个干旱正调蛋白(3 个Rubisco 的大亚基、J-type–co-chaperone Hsc20、putative protein DSM3645-2335、putative acyl-COA 脱氢酶、nesprin-2 和两个没有得到鉴定的蛋白质)。这些蛋白质参与了氮代谢调控、抗氧化行物质的合成、脂肪酸β-氧化、核骨架构造、[Fe-S]基团组装、物质跨膜运输、细胞分裂或作为分子伴侣和蛋白质酶起作用。putative ABC transporter ATP-binging protein、NtrX、nesprin-2 和Hslu 是本试验新发现的高等植物蛋白,我们主要从它们的保守结构域或在其他生物中的同源物来猜测它们的功能。实验结果为我们研究植物抗干旱机制提供了新线索和新视野。 Seabuckthorn (Hippophae rhamnoides L.) is widly distributed throughtout the temperatureresiogn of Europe and Asia and sub-tropical plateau zone of Asia. H. rhamnoides can adapatseveral different environments, and can tolerant several envioronmental stresses (e.g, lowtemperature, high temperature, drought and salty). It has been widely used in forest restoration asthe pioneer species in China. In present study, we applied H.rhamnoides subsp. Sinensis asexperimental materials to study its drought-tolerant mechanism, and expected to findpopulational difference in drought-tolerant mechanism that may exist among populations, and tryto get some insight in drought-tolerant mechanism of it at morecular level through analyzing thechange of leaf protein expression. Three populations from high altitude wet zone, low altitude wet zone and low altitude arid znoe,respectively, were applied in our experiment, and were subjected to drought. Drought increasedthe root/shoot ratio(RS), special leaf area, long-term water use efficinency, activity of antioxidantenzymes, proline content and abscisic acid (ABA) content, declined the net photosynthesis rate(A), average leaf area (ALA), total biomass (TB). Both two low altitude populations were moredrought-tolerant than the high altitude population, and different population applied differentstratedgies to tolerant oxidant stress and drought stress. The results of the exprement in 2004 showed that Daofu population was more drought-sensitivethan Jiuzhai population. Under drought conditions, leaf relative water content (RWC) greatlydecreased in Daofu population, but not in Jiuzhai population. The large loss of water in Daofupopulation resulted in a limitation on A mainly caused by non-stomatal factors, severer suppression in growth rate and a significant reduction in ascorbic acid (AsA) content, comparedwith Jiuzhai population. The greater increase in content of ABA and proline in Daofu populationmay be also induced by large loss in water, so that enable plants to cope with sever drought. In the exprement of 2005, drought significantly increased RS, activities of catalase (CAT),peroxidase (POD), glutathione peroxidase (GPX) and ascorbate peroxidase (APX), and alsosignificantly increased ABA and proline contents. On the other hand, compared with Daofupopulation, drought induced larger RS and activities of CAT, GPX and APX, and higher ABAcontent in Dingxi population, whereas gas exchange traits, e.g., stomatal limitation value (LS) andintercellular CO2 concentration (Ci), were less responsive to drought in Dingxi population thanthose in Daofu population. All these factors enable Dingxi population to tolerant drought betterthan Daofu population. The leaf protein profile of seabuchthorn subjected to drought was analyzed. Altogether 319proteins were detected in well-watered sample, four proteins disappeard by drought (putativeABCtransporter ATP-binding protein, hypothetical protein XP-515578, Hslu219and aunidentified protein), four only appeared under drought (a probable nitrogen regulation protein(NtrX), a 4-hydroxyphenylpyruvate dioxygenase , an unnamed protein product and an identified protein), 32 drought down-regulated proteins, and 14 drought up-regulated proteins (nine wereidentified: three large subunits of Rubisco, a hypothetical protein DSM3645-23351, a putativeacyl-COA dehydrogenase, a nesprin-2, a J-type-co-chaperone HSC20 and two unmatchedproteins). These proteins may involve in β-oxidation, cross-membrane transport, cell division,cytoskeleton stabilization, iron-sulfur cluster assembly, nitrogen metabolism regulation andantioxidant substance biosynthesis or function as molecular chaperone or protease. Four proteins(a putative ABC transporter ATP-binging protein, NtrX, nesprin-2, Hslu) were new found in highplants, and their functions were estimated from their conserved domain or their homologues inother organism. Our results provided new clue and new insight for us to study thedrought-tolerant mechanism in plants.

It was a study of the effect of non-cognitive factors on cognitive aging. The purpose of the study was to research the effect of attitude towards aging on memory performance of the elderly, and trying to explore the mechanism of the attitude towards aging on memory performance. The entire study was divided into three phases and included six experiments. The questionnaire of attitude towards aging was compiled in phase 1. Then the validity and reliability of it was tested in the elderly. The self-esteem scale was also tested as above. These prepare the instruments for the study. It was a static study in phase 2. Through comparing the old groups in different environment, analyzing relationship between different non-cognitive factors and memory performance in the old subjects in identical environment, the paths of the effect of the attitude towards aging on memory performance was explored. It was a dynamic study in phase 3. By changing attitude towards aging of the elderly with direct thinking (positive, negative), the effect on memory performance was observed and the mechanism was explored. Main results: 1. The validity and reliability of the questionnaire towards aging were good. Its construct had two factors (positive, negative). Both reliability of retest and split test were over 0.90. 2. Direct thinking task was able to change the attitude towards aging effectively in old or young adults. The scores of the attitudes towards aging between positive and negative thinking subjects were different significantly. 3. The effect of attitude towards aging on memory performance was significant. The results in static or dynamic condition showed that the elderly with higher score of the questionnaire of attitude towards aging (more positive), their memory self-efficacy and memory performance were higher. In the opposite (negative), they were lower. 4. Mechanism of the effect of the attitude towards aging on memory performance: in static condition there were two paths i.e. direct path and through memory self-efficacy; in dynamic condition, besides the two paths, emotion change constituted another path. 5. Different non-cognitive factors have different effect on psychological function. The results of this study found that self-esteem has no significant correlation with memory performance, but correlates with depression; The factor of education has effect on many non-cognitive factors and memory performance.

A newly developed experimental model called simulation of real mission was used to explore law of time perception and user endurance for feedback delay under Network-Supported Co-operative Work. Some non-technological factors influencing time perception and user endurance (mission type、difficulty level、feedback method、partner type、gender and A type behavior pattern) were also examined. Results of the study showed that: (1) Under condition of waiting without feedback, mission type and difficulty level demonstrated significant main effects on judgment of waiting duration. People will wait more time to receive partner's feedback if he or she perceives that partner's task is difficult, and the longest waiting duration (LWD) in the mission of computation is longer than the LWD in the mission of proof searching. (2) Under condition of waiting with feedback, experimental data perfectly supported Vierordt's Law: short duration is underestimated, long duration is overestimated, only proper duration (2-6 second) can be estimated correctly. The proper duration will vary with the changing of difficulty levels of mission. More long the waiting duration is, more estimation error will be occurred. The type difference of partner has no significant effect on the law of time perception. (3) Under condition of waiting with feedback, non-technology factors can significantly effect user's endurance. When subjects were told their partner was human, mission type and difficulty level of mission could significantly effect user's endurance. When subjects were told their partner was computer, A type behavior pattern and difficulty level of mission could significantly effect user's endurance. The two-way interaction effect between A type behavior pattern and gender was detected.

This research, mainly by using perceptual experiment and relating its results to the acoustic correlates, investigated the acoustic correlates of mandarin prominences of prosodic word、prosodic phrase、sentence and their mutual influences. The results are as follow: (1) for the prominence of prosodic word, according the order of the extents to which these acoustic correlates contribute to the perception of the prominence, its acoustic correlates are duration、pitch、spectral tilt、energy and pitch-change, the results also have showed that the perception of the prominence is influenced by some non-acoustic factors; (2) for the prominence of prosodic phrase, baseing on the experimental results, the existing of prosodic phrase prominence in mandarian has been proposed, and the experiments proved that the prosodic phrase prominence fall on the semantic focus of the phrase typically; (3) for the prominence of sentence, the experimental results show that, the lengthening of duration, similar with the pitch-change, mainly localised on the prosodic head of the domain on which the sentence prominence emphisize, and this lengthening would, in different levels, spread on other syllables in the phonetic constituents which contain the prosodic head, and this pattern of lengthening of duration does not vary with changing of domain of sentence prominence, that means, in mandarin sentence, there is generally no acoustic cues which can been used in the perception of the domain of sentnece prominence; (4) for the acoustic cues of mutual influences of the prominences of different levels, the experiment have proved that, not only the pitch-change but also the lengthening of duration are subjective to melodic grid law. In the last part of the dissertation, I have tried to given an explantion about the above experimental results from psylogical view point.
