Selecting a suitable place to install a new landfill is a hard work. Bauru is a Brazilian municipality where the local landfill currently in use has a life span that is almost over, and the selection of a new area for a future landfill is crucial and urgent. Here we use a geographic information system (GIS) approach to indicate possible suitable areas for installing the landfill. The considered criteria were: river network and the respective buffer zone, relief, urban areas and their respective buffer zone, existence of Areas for Environmental Protection (AEPs), occurrence of wells and their respective buffer zones, existence of airports and their buffer zones, wind direction, and the road network and its respective buffer zone. Due the facts that (1) Bauru has an urban area relatively large in relation to whole municipal area, (2) Bauru has two airports, and (3) this area encompasses parts of three AEPs, the model showed that there are few areas suitable and moderately suitable in Bauru, and the greater part of the municipality is unsuitable to install a new landfill. Due to this important finding reported here, the local policymakers should consider the suitable or even moderately suitable areas for analysis in situ or look for other creative solutions for destination of the solid waste. We highly encourage the use of GIS in studies that seek suitable areas for future landfills, having found that SIG was a tool that allowed fast and precise work and generated an outcome sufficiently clear of interpretation.Implications: Solid waste (SW) management is one of the main environmental concerns nowadays. Landfilling SW is still the main practice to disposal of such material. However, for many regions, suitable places for landfilling are getting scarce. This study proved this situation for a populous place in a southeastern Brazilian region. This study also showed how the decision makers should manage the problem in order to minimize the amount of SW generated and delivered for the landfill. Massive investment in education is a critical issue to reach the proposed aim.

The relationship between the microstructure and the magnetic properties of soft magnetic materials, have been studied by different researchers who seek to employ electrical systems, increasing their life span and reduce their energy consumption. Following this same line the Brazilian Synchrotron Light Laboratory developed a new synchrotron light source, the Sirius, where magnetic materials with high magnetic permeability values are being studied for use in accelerator dipoles. The low carbon steel is a ferromagnetic material that has a great relationship between cost and magnetic permeability. Aiming to raise the values of permeability of the material, heat treatments were done and evaluated the magnetic properties, microstructure and mechanical properties to correlate them. It was noted that the thermal annealing were the most effective, and the annealing performed with a small time threshold, which only phenomenon observed was the primary recrystallisation, was the most elevated values of magnetic permeability of the material, due to the average grain size ideal achieved. The heat treatments do not guide the magnetic domains of the material and not influence the mechanical properties of the material due to lack of carbon in the microstructure. The annealing treatments were shown to be an alternative to raising the values of the magnetic permeability of the material and facilitate the implementation of ultra low carbon steel in the dipoles of Sirius

The Brazilian Environmental Data Collection System (Sistema Brasileiro de Coleta de Dados Ambientais - SBCDA) has been providing reliable information regarding the monitoring of the Brazil environment to INPE (Instituto Nacional de Pesquisas Espaciais) and many Non-governmental organizations (NGO). This system is composed of a large number of Data Collection Plataforms (DCP), in charge of sending local data signals to satellites. Then the satellites automatically redirect each signal to reception centers located at INPE. Nowadays the SBCDA has been operating under the control of three satellites. It is important to mention that two of these satellites have already expired their life span around ten years ago. A strategy to keep the SBCDA operational is to develop a low cost satellite that involves Brazilian Universities and Public Institutions. This graduation work aims to design and simulate a signal conditioning module that enables the collection of a satellite status data, and then sending it to an onboard computer

The Kaposi sarcoma-associated herpesvirus (KSHV), or human herpesvirus 8, is a gammaherpesvirus etiologically linked to the development of Kaposi sarcoma, primary effusion lymphomas, and multicentric Castleman disease in humans. KSHV is unique among other human herpesviruses because of the elevated number of viral products that mimic human cellular proteins, such as a viral cyclin, a viral G protein-coupled receptor, anti-apoptotic proteins (e.g. v-bcl2 and v-FLIP), viral interferon regulatory factors, and CC chemokine viral homologues. Several KSHV products have oncogenic properties, including the transmembrane K1 glycoprotein. KSHV K1 is encoded in the viral ORFK1, which is the most variable portion of the viral genome, commonly used to discriminate among viral genotypes. The extracellular region of K1 has homology with the light chain of lambda immunoglobulin, and its cytoplasmic region contains an immunoreceptor tyrosine-based activation motif (ITAM). KSHV K1 ITAM activates several intracellular signaling pathways, notably PI3K/AKT. Consequently, K1 expression inhibits proapoptotic proteins and increases the life-span of KSHV-infected cells. Another remarkable effect of K1 activity is the production of inflammatory cytokines and proangiogenic factors, such as vascular endothelial growth factor. KSHV K1 immortalizes primary human endothelial cells and transforms rodent fibroblasts in vitro; moreover, K1 induces tumors in vivo in transgenic mice expressing this viral protein. This review aims to consolidate and discuss the current knowledge on this intriguing KSHV protein, focusing on activities of K1 that can contribute to the pathogenesis of KSHV-associated human cancers. Holocarboxylase synthetase (HCS) catalyzes the binding of biotin to lysine (K) residues in histones H3 and H4. Histone biotinylation marks play important roles in the repression of genes and retrotransposons. Preliminary studies suggested that K16 in histone H4 is a target for biotinylation by HCS. Here we demonstrated that H4K16bio is overrepresented in repeat regions {pericentromeric alpha satellite repeats; long terminal repeats (LTR)} compared with euchromatin promoters. H4K16bio was also enriched in the repressed interleukin-2 gene promoter. The enrichment at LTR22 and promoter 1 of the sodium-dependent multivitamin transporter (SMVT) depended on biotin supply; and was significantly lower in fibroblasts from an HCS-deficient patient compared with an HCS wild-type control. We conclude that H4K16bio is a real phenomenon and plays a role in the transcriptional repression of repeats and genes. HCS catalyzes the covalent binding of biotin to carboxylases, in addition to its role as a histone biotinyl ligase. HCS null individuals are not viable whereas HCS deficiency is linked to developmental delays and phenotypes such as short life span and low stress resistance. Here, we developed a 96-well plate assay for high-throughput analysis of HCS based on the detection of biotinylated p67 using IRDye-streptavidin and infrared spectroscopy. We demonstrated that the catalytic activity of rHCS depends on temperature and time, and proposed optimal substrate and enzyme concentrations to ensure ideal measurement of rHCS activity and its kinetics. Additionally, we demonstrated that this assay is sensitive enough to detect biotinylation of p67 by endogenous HCS from Jurkat lymphoid cells.

The reproductive biology and population dynamics of the cirolanid isopod Excirolana armata (Dana, 1853) were analysed through monthly samples from December 2003 to November 2005 on Una beach, Sao Paulo state (24 degrees S), in Southeastern Brazil. Sampling was performed along three transects established from the base of foredunes to the waterline. On Una beach, E. armata showed continuous reproduction with higher abundances of ovigerous females in winter and spring (July-November) with a higher peak of juveniles in spring (November 2004). The fecundity ranged from 2 to 18 eggs/embryos per female, depending on the female length. The incubation period was estimated as 2 months. The life span of males and females was nearly 1 year. The short life span and the high energetic expenditure inherent to reproduction with maternal care, probably kept females from producing more than one brood in their lifetime. When comparing the population of E. armata on Una beach (24 degrees S) with populations in Southern Brazil (32 degrees S), Uruguay (34 degrees S) and Argentina (36 degrees S), it was verified that several biological population traits (length of the smallest juvenile, length of the largest individual, length of the smallest and largest ovigerous females, range of fecundity and life span) tended to increase at higher latitudes, whereas other traits (instantaneous rate of mortality and the curvature parameter of von Bertalanffy growth function) tended to decrease. However, comparing E. armata on Una beach (24 degrees S) with a population situated at a close latitude (25 degrees S), unexpected differences in relation to population structure and to growth demonstrated and reinforced the importance of density-dependent factors over life history traits of E. armata on dissipative beaches.

Two growth patterns are recognized in shallow-water ophiuroids: (I) slow growth and early reproductive maturity over a long life span and (2) rapid growth with a short life span. For species with the first pattern, both growth and recruitment would reflect a reproductive pattern with long periods of resting and spawning concentrated in certain months of the year. To evaluate this hypothesis, the recruitment, population dynamics, and growth of the intertidal brittle star Ophionereis reticulata were analyzed from January 2001 to December 2002 at the Baleciro Isthmus on the southeast coast of Brazil. The species shows an annual gametogenic cycle with spawning taking place in summer. Densities varied from 0.46 to 9.46 individuals m(-2). Density variations and seawater temperature were not significantly correlated. The population structure of O. reticulata was polymodal, with at least four co-occurring cohorts. Recruitment events were recorded in March 2001, October 2001, January 2002, and September 2002. As indicated by the asymptote size (D(infinity)=11.47 mm +/- 1.46), growth constant (K=0.42 year(-1)+/- 0.12), and oscillation index (C=0.97 +/- 0.51), the growth pattern of O. reticulata seems to be based on high survivorship of juveniles and adults, where sexual maturity is reached at a small size with rapid growth in the first 2 years of life. A low level of settlement is to be expected based on these data; however, there must be a minimum successful survivorship and development for juveniles and adults. Another explanation for the lack of small individuals (disc diameter <1.0 mm) could be that recruitment is located in a different area and a post-settlement migration might be involved in the maintenance of the population.

Calorie restriction (CR) enhances animal life span and prevents age-related diseases, including neurological decline. Recent evidence suggests that a mechanism involved in CR-induced life-span extension is NO-stimulated mitochondrial biogenesis. We examine here the effects of CR on brain mitochondrial content. CR increased eNOS and nNOS and the content of mitochondria] proteins (cytochrome c oxidase, citrate synthase, and mitofusin) in the brain. Furthermore, we established an in vitro system to study the neurological effects of CR using serum extracted from animals on this diet. In cultured neurons, CR serum enhanced nNOS expression and increased levels of nitrite (a NO product). CR serum also enhanced the levels of cytochrome c oxidase and increased citrate synthase activity and respiratory rates in neurons. CR serum effects were inhibited by L-NAME and mimicked by the NO donor SNAP. Furthermore, both CR sera and SNAP were capable of improving neuronal survival. Overall, our results indicate that CR increases mitochondrial biogenesis in a NO-mediated manner, resulting in enhanced reserve respiratory capacity and improved survival in neurons. eNOS activation resulting in mitochondrial biogenesis is believed to play a central role in life span extension promoted by calorie restriction (CR). We investigated the mechanism of this activation by treating vascular cells with serum from CR rats and found increased Akt and eNOS phosphorylation, in addition to enhanced nitrite release. Inhibiting Akt phosphorylation or immunoprecipitating adiponectin (found in high quantities in CR serum) completely prevented the increment in nitrite release and eNOS activation. Overall, we demonstrate that adiponectin in the serum from CR animals increases NO center dot signaling by activating the insulin pathway. These results suggest this hormone may be a determinant regulator of the beneficial effects of CR.

Recent studies predict that several lineages of tropical animals are at particular risk given current estimates of global climate change. Yet, much uncertainty exists on the effects of climate shifts in ectothermic species from cool temperate regions such as Patagonia. In this study, we focus on the impact of environmental temperature on growth, age at sexual maturity, and life-span of the Patagonian gecko Homonota darwini. Skeletochronological methods were used to assess the bone growth rates Of individuals from three populations at different geographic and temporal scales: two populations from Chubut (warm site; 1941 and 2010) and one population from Rio Negro (cold site; 1997-1998). Populations displayed similar bone arrangement and the growth patterns fit a von Bertalanffy curve. Three populations attained reproductive size at a minimum age of 3 yr, but at the cold site two specimens were shown to mature in 4 yr. We found no differences in juvenile growth rates in body size or bone zone width between juveniles of 1 to 3 yr of age from the 1941 warm site and the 2010 warm site. However, these traits appeared to be higher at these two warm sites than at the cold site, which is consistent with the climatic differences among the three localities. Our results suggest that higher temperatures positively affect growth, denoting that global warming might benefit H. darwini, especially the southern populations.
