Quality Management System has been implemented at the René Rachou Research Center since 2003. This study investigated its importance for collaborators (Cs) in laboratories. This was a quantitative and descriptive study performed in a group of 113 collaborators. It was based on the World Health Organization handbook: Quality Practices in Basic Biomedical Research. The questionnaires evaluated the parameters using the Likert scale. Biosafety, training and ethics were considered to be the most important parameters. Supervision and quality assurance, data recording, study plan, SOPs and file storage achieved intermediate evaluation. The lower frequency of responses was obtained for result report, result verification, personnel and publishing practices. Understanding the perception of the collaborators allows the development of improvement actions aiming the construction of a training program directing strategies for disseminating quality.

Este trabalho aborda o comportamento mecânico de elementos curvos em materiais compósitos aquando sujeitos a forças e momentos nas extremidades. Os modos de falha mais comuns são estudados em detalhe: falha transversal na matriz devido a tensões de flexão (tensões circunferenciais), ou falha por delaminagem devido as tensões interlaminares de tracção verificadas ao longo da espessura na região curva (tensões radiais). Inicialmente, faz-se uma abordagem sobre a influência de Z-Pins na resistência interlaminar, uma abordagem sobre os efeitos de bordo, e sobre tensões interlaminares com base em teorias Layerwise de ordem superior. Salienta-se, ainda, a importância do estudo feito com o World-Wide Failure Exercise (WWFE) (Hinton et al. 2004) na comunidade científica relacionada com o tema dos materiais compósitos. O presente trabalho, é essencialmente dividido em duas partes: modelos analíticos e modelos numéricos. São estudados provetes curvos, em forma de C (semicircular), geometria analisada por Ko e Jackson (1989); e em forma de L (cantoneira de abas iguais), analisados por Sun e Kelly (1988). Começa-se por fazer uma análise teórica, através da Teoria Multicamada, ao trabalho Ko e Jackson, autores da mesma. Analisou-se também, através desta teoria, a parte em quarto de círculo da viga estudada por Sun e Kelly. Após ter a solução analítica bem compreendida, essa teoria é aplicada ao principal estudo deste trabalho, um provete semicircular, usando os empilhamentos de Sun e Kelly. É ainda estudado, e aplicado a este último, o critério de falha em três dimensões (3D) proposto por Hashin (1980). Na segunda parte, é feita uma validação numérica dos três casos anteriores, com recurso ao software comercial de elementos finitos (EF), ANSYS. Estes estudos de EF têm a diferença, relativamente aos originais, de serem em 3D, podendo assim ser retiradas todas as tensões necessárias para se aplicar o critério de falha. No final, é feita uma comparação geral entre os resultados analíticos e numéricos.

O presente trabalho de final de Mestrado tem por objectivo estudar o desempenho actual da profissão de Relações Públicas em Portugal. Frequentemente associadas a actividades pouco relevantes ou vistas como um mero conjunto de técnicas executantes, assume-se neste trabalho uma visão de Relações Públicas como um elemento estratégico e fundamental à gestão organizacional. Assume-se aqui as RP como um acelerador de estratégia. As Relações Públicas, tal como as conhecemos hoje, são extremamente jovens e embora exista uma grande panóplia de definições e conceitos a si associados, estamos longe de chegar a um consenso e a uma definição institucionalizada sobre a profissão. Ao longo deste trabalho procurar-se-á reflectir um pouco sobre a evolução do conceito de Relações Públicas e perceber como estas passaram de um simples conjunto de técnicas para uma função estratégica, sendo hoje consideradas uma disciplina por excelência das Ciências da Comunicação. Analisar-se-á o desenvolvimento da disciplina e da profissão de Relações Públicas e, procurar-se-á fazer um enquadramento das RP em Portugal de modo a melhor compreendermos a realidade desta profissão no país.

OBJETIVO: Poucos estudos no Brasil investigaram a validação de testes cognitivos em amostras populacionais de idosos e nenhum deles analisou as propriedades psicométricas do Teste do Relógio de Tuokko. O objetivo do estudo foi traduzir e adaptar o modelo desse teste ao contexto brasileiro e avaliar sua validade de construto. MÉTODOS: Estudo transversal com uma amostra populacional constituída por 353 sujeitos de Juiz de Fora (MG), 2004-2005. Para avaliar as validades convergente e discriminante, utilizou-se a correlação de Pearson. Os subtestes do Teste do Relógio foram correlacionados com os instrumentos de referência: cubos e dígitos, e mini-exame do estado mental para avaliar a validade convergente. A validade discriminante foi investigada por meio da correlação dos subtestes com a Center for Epidemiologic Studies Depression Scale. RESULTADOS: Da amostra, 74,1% eram do sexo feminino, com idade entre 63 e 107 anos (73,8+8,5) e média de escolaridade foi 7,4 anos (DP=4,7). Obteve-se validade convergente, com correlações estatisticamente significativas entre todos os subtestes (p<0,01). Quanto à validade discriminante, o único subteste que teve correlação significativa com a escala de referência foi o "indicação das horas" (p<0,05). CONCLUSÕES: O Teste de Relógio traduzido e adaptado em uma amostra comunitária de idosos mostrou ser um instrumento de rastreio cognitivo breve, com boa validade de construto quando analisado com outros dados da literatura. Futuros trabalhos devem investigar outras propriedades psicométricas, como as validades de critério e de conteúdo.

OBJECTIVE: To analyze household risk factors associated with high lead levels in surface dental enamel. METHODS: A cross-sectional study was conducted with 160 Brazilian adolescents aged 14-18 years living in poor neighborhoods in the city of Bauru, southeastern Brazil, from August to December 2008. Body lead concentrations were assessed in surface dental enamel acid-etch microbiopsies. Dental enamel lead levels were measured by graphite furnace atomic absorption spectrometry and phosphorus levels were measured by inductively coupled plasma optical emission spectrometry. The parents answered a questionnaire about their children's potential early (05 years old) exposure to well-known lead sources. Logistic regression was used to identify associations between dental enamel lead levels and each environmental risk factor studied. Social and familial covariables were included in the models. RESULTS: The results suggest that the adolescents studied were exposed to lead sources during their first years of life. Risk factors associated with high dental enamel lead levels were living in or close to a contaminated area (OR = 4.49; 95% CI: 1.69;11.97); and member of the household worked in the manufacturing of paints, paint pigments, ceramics or batteries (OR = 3.43; 95% CI: 1.31;9.00). Home-based use of lead-glazed ceramics, low-quality pirated toys, anticorrosive paint on gates and/or sale of used car batteries (OR = 1.31; 95% CI: 0.56;3.03) and smoking (OR = 1.66; 95% CI: 0.52;5.28) were not found to be associated with high dental enamel lead levels. CONCLUSIONS: Surface dental enamel can be used as a marker of past environmental exposure to lead and lead concentrations detected are associated to well-known sources of lead contamination.

Estudo transversal visando estimar a prevalência das formas de deslocamento para o trabalho e identificar fatores associados em trabalhadores do Estado do Rio Grande do Sul. Um total de 2.265 trabalhadores responderam questionário sobre a forma de deslocamento utilizado para ir ao trabalho: caminhada/bicicleta, ônibus ou de carro/moto. Para estimar a associação entre o desfecho e variáveis sociodemográficas, ocupacionais e comportamental foi utilizada a regressão logística multinomial. O principal meio de deslocamento para o trabalho foi o ônibus (45,7%). Trabalhadores com maior condição socioeconômica tiveram maior probabilidade de deslocamento passivo.

E. faecium was the first reported VRE species, carrying the vanA gene in Brazil. In spite of this, vancomycin-resistant E. faecalis has become the predominant species in Brazilian hospitals. The aim of this study was to evaluate the genetic relatedness of VREs isolated in a Brazilian teaching hospital eight years apart from its first isolation. We analyzed 38 VRE strains obtained from 81 surveillance cultures of patients admitted to the four largest intensive care units in Hospital São Paulo in February, 2006. Presence of the vanA gene was assayed by PCR and PFGE analysis was used for molecular characterization. All VRE strains carried the vanA gene. Two distinct clonal groups were observed among vancomycin-resistant E. faecalis. Vancomycin-resistant E. faecium belonged to five distinct clones were demonstrated by molecular typing. All of these clones were different from the first vancomycin-resistant enterococci clone isolated eight years ago in our hospital.

Objective: To assess quantitative real-time polymerase chain reaction (q-PCR) for the sputum smear diagnosis of pulmonary tuberculosis (PTB) in patients living with HIV/AIDS with a clinical suspicion of PTB.Method: This is a prospective study to assess the accuracy of a diagnostic test, conducted on 140 sputum specimens from 140 patients living with HIV/AIDS with a clinical suspicion of PTB, attended at two referral hospitals for people living with HIV/AIDS in the city of Recife, Pernambuco, Brazil. A Löwenstein-Jensen medium culture and 7H9 broth were used as gold standard.Results: Of the 140 sputum samples, 47 (33.6%) were positive with the gold standard. q-PCR was positive in 42 (30%) of the 140 patients. Only one (0.71%) did not correspond to the culture. The sensitivity, specificity and accuracy of the q-PCR were 87.2%, 98.9% and 95% respectively. In 39 (93%) of the 42 q-PCR positive cases, the CT (threshold cycle) was equal to or less than 37.Conclusion: q-PCR performed on sputum smears from patients living with HIV/AIDS demonstrated satisfactory sensitivity, specificity and accuracy, and may therefore be recommended as a method for diagnosing PTB.

Anthelmintics used for intestinal helminthiasis treatment are generally effective; however, their effectiveness in tissue parasitosis (i.e. visceral toxocariasis) is moderate. The aim of this study was to evaluate the in vitroactivity of lapachol, β-lapachone and phenazines in relation to the viability of Toxocara canis larvae. A concentration of 2 mg/mL (in duplicate) of the compounds was tested using microculture plates containing Toxocara canis larvae in an RPMI-1640 environment, incubated at 37 °C in 5% CO2 tension for 48 hours. In the 2 mg/mL concentration, four phenazines, lapachol and three of its derivatives presented a larvicide/larvistatic activity of 100%. Then, the minimum larvicide/larvistatic concentration (MLC) test was conducted. The compounds that presented the best results were nor-lapachol (MLC, 1 mg/mL), lapachol (MLC 0.5 mg/mL), β-lapachone, and β-C-allyl-lawsone (MLC, 0.25 mg/mL). The larvae exposed to the compounds, at best MLC with 100% in vitro activity larvicide, were inoculated into healthy BALB/c mice and were not capable of causing infection, confirming the larvicide potential in vitro of these compounds.

Only a small percentage of individuals living in endemic areas develop severe malaria suggesting that host genetic factors may play a key role. This study has determined the frequency of single nucleotide polymorphisms (SNPs) in some pro and anti-inflammatory cytokine gene sequences: IL6 (-174; rs1800795), IL12p40 (+1188; rs3212227), IL4 (+33; rs2070874), IL10 (-3575; rs1800890) and TGFb1 (+869; rs1800470), by means of PCR-RFLP. Blood samples were collected from 104 symptomatic and 37 asymptomatic subjects. Laboratory diagnosis was assessed by the thick blood smear test and nested-PCR. No association was found between IL6 (-174), IL12p40 (+1188), IL4 (+33), IL10 (- 3575), TGFb1 (+869) SNPs and malaria symptoms. However, regarding the IL10 -3575 T/A SNP, there were significantly more AA and AT subjects, carrying the polymorphic allele A, in the symptomatic group (c2 = 4.54, p = 0.01, OR = 0.40 [95% CI - 0.17- 0.94]). When the analysis was performed by allele, the frequency of the polymorphic allele A was also significantly higher in the symptomatic group (c2 = 4.50, p = 0.01, OR = 0.45 [95% CI - 0.21-0.95]). In conclusion, this study has suggested the possibility that the IL10 - 3575 T/A SNP might be associated with the presence and maintenance of malaria symptoms in individuals living in endemic areas. Taking into account that this polymorphism is related to decreased IL10 production, a possible role of this SNP in the pathophysiology of malaria is also suggested, but replication studies with a higher number of patients and evaluation of IL10 levels are needed for confirmation.

Although the protease inhibitors have revolutionized the therapy of chronic hepatitis C (CHC), the concomitant use of pegylated-interferon (PEG-IFN) and ribavirin (RBV) is associated to a high rate of adverse effects. In this study, we evaluated the consequences of PEG-IFN and RBV and their relationship with mortality in patients with cirrhosis. METHODS: Medical records of CHC who underwent treatment with PEG-IFN and RBV in a public hospital in Brazil were evaluated. All the patients with cirrhosis were selected, and their clinical and laboratory characteristics, response to treatment, side effects and mortality were evaluated. RESULTS: From the 1,059 patients with CHC, 257 cirrhotic patients were evaluated. Of these, 45 (17.5%) achieved sustained viral response (SVR). Early discontinuation of therapy occurred in 105 (40.8%) patients, of which 39 (15.2%) were due to serious adverse effects. The mortality rate among the 257 cirrhotic patients was 4.3%, occurring in 06/242 (2.4%) of the Child-A, and in 05/15 (33.3%) of the Child-B patients. In conclusion, the treatment of patients with cirrhosis due to HCV with PEG-IFN and RBV shows a low SVR rate and a high mortality, especially in patients with liver dysfunction.
