Obesity is heritable and predisposes to many diseases. To understand the genetic basis of obesity better, here we conduct a genome-wide association study and Metabochip meta-analysis of body mass index (BMI), a measure commonly used to define obesity and assess adiposity, in up to 339,224 individuals. This analysis identifies 97 BMI-associated loci (P < 5 × 10(-8)), 56 of which are novel. Five loci demonstrate clear evidence of several independent association signals, and many loci have significant effects on other metabolic phenotypes. The 97 loci account for ∼2.7% of BMI variation, and genome-wide estimates suggest that common variation accounts for >20% of BMI variation. Pathway analyses provide strong support for a role of the central nervous system in obesity susceptibility and implicate new genes and pathways, including those related to synaptic function, glutamate signalling, insulin secretion/action, energy metabolism, lipid biology and adipogenesis.

This paper describes the port interconnection of two subsystems: a power electronics subsystem (a back-to-back AC/CA converter (B2B), coupled to a phase of the power grid), and an electromechanical subsystem (a doubly-fed induction machine (DFIM). The B2B is a variable structure system (VSS), due to presence of control-actuated switches: however, from a modelling simulation, as well as a control-design, point of view, it is sensible to consider modulated transformers (MTF in the bond graph language) instead of the pairs of complementary switches. The port-Hamiltonian models of both subsystems are presented and, using a power-preserving interconnection, the Hamiltonian description of the whole system is obtained; detailed bond graphs of all subsystems and the complete system are also provided. Using passivity-based controllers computed in the Hamiltonian formalism for both subsystems, the whole model is simulated; simulations are run to rest the correctness and efficiency of the Hamiltonian network modelling approach used in this work.

A espectroscopia na região do visível (VIS) e do infravermelho próximo (NIR) apresenta-se como uma promissora e rápida tecnologia de avaliação das características internas de várias espécies de frutas. A diferença de absorbância, provenientes dos frutos, ao serem submetidos a um espectro emitido entre dois comprimentos de onda próximos ao do pico de absorção da clorofila-a, foi utilizada para o desenvolvimento de um índice (Índice DA- IDA) que se correlaciona ao estádio de maturação dos frutos. Neste trabalho, objetivou-se avaliar as relações existentes entre o índice DA, obtidos com o equipamento DA-meter®, e os parâmetros físico-químicos de maturação utilizados em mangas da cultivar Tommy Atkins, na separação dos frutos em categorias distintas. Para isto, foram utilizados frutos pertencentes a um mesmo lote e adquiridos em um centro de distribuição local, na cidade de Pelotas-RS. Com os resultados obtidos, através do índice DA, foi possível separar os frutos em categorias de acordo com o grau de maturação, demonstrando a grande heterogeneidade com que os frutos são oferecidos ao consumidor, e as relações entre os valores dos índices DA e os parâmetros de qualidade, determinadas através dos coeficientes de determinação (R²), foram significativas para os atributos estudados.

En este artículo nos centraremos en el Atlántico y las relaciones en clave cultural (de ida y vuelta) entre América y Europa. Analizaremos los estudios que se han ocu­pado de la cultura escrita, la historiografía sobre la correspondencia, la circulación de los textos y las redes de distribución de li­bros en las relaciones atlánticas, donde la comunicación estaba sujeta a un ritmo pro­pio de contacto y transferencia. La mirada en torno a los fenómenos atlánticos supone romper ciertas barreras, admitir la mezcla y lo diverso, no centrarse únicamente en las interpretaciones institucionales y jurídicas, ya que la norma se fija, pero su transgresión se da de manera habitual en las relaciones atlánticas.

Las relaciones entre actos festivos y espacios arquitectónicos, y las modalidades con que las ceremonias cortesanas llegaban a modificar los centros urbanos y las residen- cias palaciegas, han sido los temas principales del congreso internacional Making Space for Festival, 14001700. Interactions of Architecture and Performance in Late Medieval and Early Modern Festivals, que tuvo lugar del 21 al 24 de marzo de 2013 en el Palacio Pesaro-Papafava de Venecia, residencia de la Universidad de Warwick en la Laguna. La Society for European Festivals Research (SEFR), entidad vinculada a esta universidad, ha promovido el encuentro junto con la Red PALATIUM financia- da por el Research Networking Programme de la European Science Fundation (ESF) (2010-2015), dedicada al estudio de las residencias palaciegas de la Edad Moderna como espacios de intercambios políticos y culturales. El tema de las jornadas nacía de la unión de los intereses científicos de los grupos organizadores, dejando lugar a temáticas diferentes como: la articulación de un mensaje festivo a lo largo del recorri- do de procesiones y entradas públicas, la financiación y los aspectos más prácticos de la puesta en escena de un evento festivo, la tipificación de ceremonias y decoraciones en determinados contextos, el intercambio cultural a través de la fiesta, o la recepción de las modificaciones de los espacios por parte de los espectadores

Alanine aminotransferase (ALT) plays an important role in amino acid metabolism and gluconeogenesis. The preference of carnivorous fish for protein amino acids instead of carbohydrates as a source of energy lead us to study the transcriptional regulation of the mitochondrial ALT (mALT) gene and to characterize the enzyme kinetics and modulation of mALT expression in the kidney of gilthead sea bream (Sparus aurata) under different nutritional and hormonal conditions. 5′-Deletion analysis of mALT promoter in transiently transfected HEK293 cells, site-directed mutagenesis and electrophoretic mobility shift assays allowed us to identify HNF4α as a new factor involved in the transcriptional regulation of mALT expression. Quantitative RT-PCR assays showed that starvation and the administration of streptozotocin (STZ) decreased HNF4α levels in the kidney of S. aurata, leading to the downregulation of mALT transcription. Analysis of the tissue distribution showed that kidney, liver, and intestine were the tissues with higher mALT and HNF4α expression. Kinetic analysis indicates that mALT enzyme is more efficient in catalyzing the conversion of L-alanine to pyruvate than the reverse reaction. From these results, we conclude that HNF4α transactivates the mALT promoter and that the low levels of mALT expression found in the kidney of starved and STZ-treated fish result from a decreased expression of HNF4α. Our findings suggest that the mALT isoenzyme plays a major role in oxidazing dietary amino acids, and points to ALT as a target for a biotechnological action to spare protein and optimize the use of dietary nutrients for fish culture.

6-Phosphofructo-2-kinase/fructose-2,6-bisphosphatase (PFKFB)catalyzes the synthesis and degradation of fructose-2,6-bisphosphate, a key modulator of glycolysis-gluconeogenesis. To gain insight into the molecular mechanism behind hormonal and nutritional regulation of PFKFB expression, we have cloned and characterized the proximal promoter region of the liver isoform of PFKFB (PFKFB1) from gilthead sea bream (Sparus aurata). Transient transfection of HepG2 cells with deleted gene promoter constructs and electrophoretic mobility shift assays allowed us to identify a sterol regulatory element (SRE) to which SRE binding protein-1a (SREBP-1a)binds and transactivates PFKFB1 gene transcription. Mutating the SRE box abolished SREBP-1a binding and transactivation. The in vivo binding of SREBP-1a to the SRE box in the S. aurata PFKFB1 promoter was confirmed by chromatin immunoprecipitation assays. There is a great deal of evidence for a postprandial rise of PFKB1 mRNA levels in fish and rats. Consistently, starved-to-fed transition and treatment with glucose or insulin increased SREBP-1 immunodetectable levels, SREBP-1 association to PFKFB1 promoter, and PFKFB1 mRNA levels in the piscine liver. Our findings demonstrate involvement of SREBP-1a in the transcriptional activation of PFKFB1, and we conclude that SREBP-1a may exert a key role mediating postprandial activation of PFKFB1 transcription.

In the Arabidopsis thaliana genome, over 1000 putative genes encoding small, presumably secreted, signalling peptides can be recognized. However, a major obstacle in identifying the function of genes encoding small signalling peptides is the limited number of available loss-of-function mutants. To overcome this, a promising new tool, antagonistic peptide technology, was recently developed. Here, this antagonistic peptide technology was tested on selected CLE peptides and the related IDA peptide and its usefulness in the context of studies of peptide function discussed. Based on the analyses, it was concluded that the antagonistic peptide approach is not the ultimate means to overcome redundancy or lack of loss-of-function lines. However, information collected using antagonistic peptide approaches (in the broad sense) can be very useful, but these approaches do not work in all cases and require a deep insight on the interaction between the ligand and its receptor to be successful. This, as well as peptide ligand structure considerations, should be taken into account before ordering a wide range of synthetic peptide variants and/or generating transgenic plants.

Opinnäytetyön tarkoituksena oli selvittää millaisia menetelmiä on käytetty nuorten sukupuolitautitartuntojen ehkäisyssä kansainvälisesti. Opinnäytetyö on osa Seksuaaliterveyden osaamisen kehittäminen metropolialueella ja hoitotyön koulutuksessa -hanketta. Opinnäytetyö toteutettiin systemaattista kirjallisuuskatsausta soveltaen. Käytetty aineisto rajattiin koskemaan länsimaalaisia vuoden 2000 jälkeen julkaistuja artikkeleita, jotka koskivat 15-25 -vuotiaita nuoria. Aineiston muodostivat kymmenen artikkelia, jotka käsittelivät sukupuolitautien seulontaa, kampanjointia sukupuolitautien ehkäisemiseksi, sukupuolitautien ehkäisyvälineitä, partnerin jäljitystä ja seksuaalineuvontaa terveydenhuollon ammattilaisen toteuttamana. Kirjallisuuskatsauksen tulosten mukaan tehokkaita nuorten sukupuolitautien ehkäisymenetelmiä ovat systemaattinen sukupuolitautien seulonta, näkyvä kampanjointi, uusien sukupuolitautien ehkäisyvälineiden kehittäminen naisille, partnerin jäljityksen tehostaminen sekä terveydenhuollon ammattilaisen antama asiantunteva seksuaalineuvonta. Partnerin jäljitys tehostuu kotinäytteenoton sekä erilaisten partnerin tiedotusmenetelmien käyttöönoton avulla. Naisille suunnatut sukupuolitautien ehkäisymenetelmät ovat myös tärkeitä nuorten sukupuolitautien ehkäisyssä. Tutkimustuloksissa esille tulleiden menetelmien hyödyntäminen Suomessa nuorten sukupuolitautitartuntojen ehkäisyssä vähentäisi sukupuolitautitartuntoja ja edistäisi täten kansanterveyttä. Kansainvälisten tutkimustulosten tarkastelu ja niiden esille tuominen palvelee yhteiskuntaamme nuorten sukupuolitautitartuntojen ehkäisyn kehittämisessä.

OBJETIVO: Avaliar a eficácia da ultrassonografia transvaginal (USTV) e da histerossonografia (HSG) e compará-las na avaliação de alterações endometriais em portadoras de sangramento uterino anormal. MATERIAIS E MÉTODOS: Estudo transversal com 30 pacientes, idade entre 29 e 71 anos, 21 delas (70%) na pré-menopausa e 9 (30%) na pós-menopausa. Utilizou-se solução salina a 0,9% para contraste na HSG. Foi considerado o achado histeroscópico e/ou histopatológico como método padrão. Utilizou-se o teste de MacNemar para comparação dos testes diagnósticos. RESULTADOS: A histeroscopia diagnosticou 18 casos (60%) de alterações intracavitárias, sendo 10 pólipos (33,3%). A USTV apresentou sensibilidade e especificidade de 83,3% e a HSG mostrou sensibilidade de 94,4% e especificidade de 91,6%. O teste de MacNemar evidenciou sensibilidade (p = 0,500) e especificidade (p = 1,000) semelhantes entre a USTV e a HSG para detecção de alterações endometriais. No diagnóstico de pólipo, a HSG apresentou maior sensibilidade (90,9% × 27,3%; p = 0,016), com especificidade semelhante (89,5% × 94,7%; p = 1,000). CONCLUSÃO: A HSG e a USTV apresentam boas taxas de predição para doenças endometriais em pacientes com sangramento uterino anormal. A HSG apresenta sensibilidade e especificidade semelhantes às da USTV na detecção dessas doenças, porém apresenta sensibilidade maior para pólipos.
