Tutkimus selvittää ajanmukaisien suunnittelujärjestelmien ominaisuuksia ja soveltuvuutta kartonkitehtaan kunnossapidon laitossuunnitteluun. Tutkimus on tehty keräämällä tietoja kirjallisista julkaisuista sekä tekemällä haastatteluja, empiirisiä kokeita ja kyselytutkimus. Saadut tutkimustulokset ovat suhteellisia ja niitä verrataan eräässä kartonkitehtaassa käytössä olevaan järjestelmään. Tutkimuksen tuloksina esitetään ajanmukaisien suunnittelujärjestelmien käytöllä saavutettavissa olevia hyötyjä ja käyttöön liittyviä ongelmia sekä näiden järjestelmien soveltuvuutta ja vertailukriteerejä laitoskäyttöön. Lisäksi käsitellään ajanmukaisiin järjestelmiin investoinnin kannattavuutta ja herkkyystekijöitä investointilaskelman (NPV) avulla. Tuloksena saatiin, etteivät tämänhetkiset suunnittelujärjestelmät sovi parhaalla mahdollisella tavalla yritysten suunnittelun ja tiedonhallinnan kokonaisuuteen. Keinona, jolla voidaan parantaa suunnittelujärjestelmien laitoskäyttöön sopivuutta, esitetään metsäteollisuuskonsernien välistä yhteistyötä.

Vakaviin reaktorionnettomuuksiin liittyviä ilmiöitä on tutkittu jo 1980-luvulta lähtien ja tutkitaan edelleen. Ilmiöt liittyvät reaktorisydämen ja muiden paineastian sisäisten materi-aalien sulamiseen sekä reagointiin veden ja höyryn kanssa. Ilmiöt on myös tärkeää tuntea ja niiden esiintymistä mallintaa käytössä olevilla laitoksilla, jotta voidaan varmistua turval-lisuusjärjestelmien riittävyydestä. Olkiluoto 1 ja 2 laitosten käyttölupa uusitaan vuoteen 2018 mennessä. Lupaprosessiin liit-tyy analyysejä, joissa mallinnetaan laitosten toimintaa vakavassa reaktorionnettomuudessa. Näiden analyysien tekoon Teollisuuden Voima Oyj on käyttänyt ohjelmaa nimeltä MEL-COR jo vuodesta 1994 lähtien. Käytössä on ollut useita eri ohjelmaversioita ja viimeisin niistä on 1.8.6, joka riittää vielä tulevan käyttöluvan uusintaprojektiin liittyvien analyysien tekoon. MELCOR:n vanhaa 1.8.6 ohjelmaversioita ei kuitenkaan enää päivitetä, joten siirtyminen uudempaan 2.1 versioon on tulevaisuudessa välttämätöntä. Uusimman versiopäivityksen yhteydessä on kuitenkin muuttunut koko ohjelman lähdekoodi ja vanhojen laitosmallien käyttö uudessa ohjelmaversiossa vaatii tiedostojen konvertoinnin. Tässä työssä esitellään MELCOR-version 2.1 ominaisuuksia ja selvitetään, mitä 1.8.6 versioon luotujen laitosmal-lien käyttöönotto versiossa 2.1 vaatii. Vaatimusten määrittelemiseksi laitosmalleilla tehdään ajoja molemmilla ohjelmaversioilla ja erilaisilla onnettomuuden alkutapahtuman määrittelyillä. Tulosten perusteella arvioidaan ohjelmaversioiden eroja ja pohditaan mitä puutteita laitosmalleihin konversion jälkeen jää. Näiden perusteella arvioidaan mitä jatkotoimenpiteitä konversio vaatii.

La peridermis és una estructura complexa que protegeix els òrgans vegetals madurs (secundaris) i les zones que han sofert ferides de la pèrdua d'aigua i dels patògens. Aquesta funció barrera és deguda al fel·lema o súber, un teixit format per cèl·lules suberificades. Tant el fel·lema com la suberina són crucials per la vida de les plantes terrestres, però pràcticament no es coneix res dels processos moleculars que regulen la seva formació, probablement degut a la manca de models adequats. En aquesta tesi s'han identificat i caracteritzat gens induïts al fel·lema mitjançant la combinació de dues plantes models. L'escorça d'alzina surera (Quercus suber) s'ha utilitzat per aïllar gens candidats de la formació del fel·lema i per investigar el comportament d'alguns d'aquests gens durant l'estació de creixement, mentre que la pela de la patata (Solanum tuberosum) s'ha utilitzat en estudis de genètica reversa per demostrar la funció d'alguns gens reguladors al fel·lema.

In this work, we used sugarcane as a model due to its importance for sugar and ethanol production. Unlike the current plant models, sugarcane presents a complex genetics and an enormous allelic variation. Here, we report the analysis of SAGE libraries produced using the shoot apical meristem from contrasted genotypes by flowering induction (non-flowering vs. early-flowering varieties) grown under São Paulo state conditions. The expression pattern was analyzed using samples from São Paulo (SP) and Rio Grande do Norte (RN) states. These results showed that cDNAs identified by SAGE libraries had differential expression only in São Paulo state samples. Furthermore, the cDNA identified CYP (Citocrome P450) was chosen for in silico and genome characterization because it was found in SAGE libraries and subtractive libraries from samples from RN. Phylogenetic trees showed the relationship for these sequences. Furthermore, the qRT-PCR for CYP showed a potential role as flowering indutor for RN samples considering different isophorms. Considering the results present here, it can be consider that CYP gene may be used as molecular marker

The genome of all organisms is subject to injuries that can be caused by endogenous and environmental factors. If these lesions are not corrected, it can be fixed generating a mutation which can be lethal to the organisms. In order to prevent this, there are different DNA repair mechanisms. These mechanisms are well known in bacteria, yeast, human, but not in plants. Two plant models Oriza sativa and Arabidopsis thaliana had the genome sequenced and due to this some DNA repair genes have been characterized. The aim of this work is to characterized two sugarcane cDNAs that had homology to AP endonuclease: scARP1 and scARP3. In silico has been done with these two sequences and other from plants. It has been observed domain conservation on these sequences, but the cystein at 65 position that is a characteristic from the redox domain in APE1 protein was not so conservated in plants. Phylogenetic relationship showed two branches, one branch with dicots and monocots sequence and the other branch with only monocots sequences. Another approach in order to characterized these two cDNAs was to construct overexpression cassettes (sense and antisense orientation) using the 35S promoter. After that, these cassettes were transferred to the binary vector pPZP211. Furthermore, previously in the laboratory was obtained a plant from nicotiana tabacum containing the overexpression cassette in anti-sense orientation. It has been observed that this plant had a slow development and problems in setting seeds. After some manual crossing, some seeds were obtained (T2) and it was analyzed the T2 segregation. The third approach used in this work was to clone the promoter region from these two cDNAs by PCR walking. The sequences obtained were analyzed using the program PLANTCARE. It was observed in these sequences some motives that may be related to oxidative stress response

Background. The last comprehensive review of experimental research on effects of homeopathic treatments on plants was published in 1984, and lacked formal predefined criteria to assess study quality. Since then several new studies with more advanced methods have been published.Objectives: To compile a review of the literature on basic research in homeopathy with healthy plants with particular reference to studies investigating specific effects of homeopathic remedies.Methods: The literature search included English, French, German, Italian, Portuguese and Spanish publications from 1920 to April 2009, using predefined selection criteria. We included experiments with healthy whole plants, seeds, plant parts and cells. The outcomes had to be measured by established procedures and statistically evaluated. We developed a Manuscript Information Score (MIS) and included only publications which provided enough information for proper interpretation (MIS >= 5). A formallised Study Methods Evaluation Procedure (SMEP) was used to evaluate these studies, and the subgroup of studies with adequate controls to identify specific effects.Results: A total of 86 studies in 79 publications was identified, 43 studies included statistics, 29 had MIS >= 5, and 15 studies investigated the specificity of homeopathic preparations. Specific effects of decimal, centesimal and fifty millesimal potencies were found including dilution levels far beyond the Avogadro number. In consecutive series of potencies only some of the tested potencies showed effects. There were many individual studies with diverse methods and very few reproduction trials.Conclusions: Healthy plant models seem an useful approach to investigate basic research questions about the specificity of homeopathic preparations. More investigations with more advanced methods are recommended, especially in the sectors of potentisation techniques, effective potency levels and conditions for reproducibility. Neste trabalho, são apresentados a metodologia de projeto e resultados de testes experimentais de um estabilizador de sistema de potência (ESP), implementado em um sistema de geração em escala reduzida de 10 kVA, localizado no Laboratório de Controle e Sistema de Potência (LACSPOT) da Universidade Federal do Pará (UFPA). O projeto do ESP é baseado em uma estratégia de controle robusto com ênfase em incertezas paramétricas estruturadas, as quais são tratadas com ferramentas da teoria de análise intervalar. Estas incertezas são decorrentes de mudanças do ponto de operação do sistema, que provocam variações nos parâmetros de um modelo matemático linearizado referente ao comportamento dinâmico do sistema elétrico de potência no referido ponto de operação. Para o projeto do ESP robusto intervalar, são realizados uma serie de ensaios experimentais com o propósito de estimar os parâmetros de modelos linearizados da planta, representando satisfatoriamente a dinâmica dos modos poucos amortecidos do sistema de geração interligado. O método de identificação é baseado em técnica de identificação paramétrica, baseado em mínimos quadrados. A partir de um conjunto de dados de entrada e saída, para cada ponto de operação, um modelo linear, do tipo auto-regressivo com entrada exógenos (ARX), estimado para fim de uso do projeto do ESP. Por fim, uma série de testes experimentais é realizada no sistema de geração interligado a rede elétrica local, com o propósito de verificar a efetividade da técnica de controle robusto intervalar proposta para a sintonia do ESP. A partir da análise da função custo do sinal de erro de desvio de potência elétrica na saída do gerador síncrono e a função custo do sinal de controle do ESP comprova-se experimentalmente o bom desempenho obtido pela técnica de controle proposta em comparação com uma técnica de controle clássica.

In this work particular attention was given to the study of secondary metabolites produced by some plants belonging to the Amaryllidaceae family, in the specific case isoquinoline alkaloids. At the first instance were characterized both qualitatively and quantitatively three different plants belonging to Amaryllidaceae family, such as: Crinum angustum Steud., Pancratium illyricum L., and Leucojum nicaeense Ard. The alkaloids extracts obtained were separately tested against enzymes involved in specific diseases or liable in multifactorial pathologies, like: MMPs, AChE,and PPO. From leaves extract of P.illyricum was isolated a new compound, 11α-hydroxy-O-methylleucotamine, with important role in AChE inbition. Considering the protection role against external bodies carried out by these metabolites in plant, extracts were also assayed against ATCC microorganisms and clinical isolates. Plants with promising pharmacological activities have been the basis for development of in vitro plant models.

The design of nuclear power plant has to follow a number of regulations aimed at limiting the risks inherent in this type of installation. The goal is to prevent and to limit the consequences of any possible incident that might threaten the public or the environment. To verify that the safety requirements are met a safety assessment process is followed. Safety analysis is as key component of a safety assessment, which incorporates both probabilistic and deterministic approaches. The deterministic approach attempts to ensure that the various situations, and in particular accidents, that are considered to be plausible, have been taken into account, and that the monitoring systems and engineered safety and safeguard systems will be capable of ensuring the safety goals. On the other hand, probabilistic safety analysis tries to demonstrate that the safety requirements are met for potential accidents both within and beyond the design basis, thus identifying vulnerabilities not necessarily accessible through deterministic safety analysis alone. Probabilistic safety assessment (PSA) methodology is widely used in the nuclear industry and is especially effective in comprehensive assessment of the measures needed to prevent accidents with small probability but severe consequences. Still, the trend towards a risk informed regulation (RIR) demanded a more extended use of risk assessment techniques with a significant need to further extend PSA’s scope and quality. Here is where the theory of stimulated dynamics (TSD) intervenes, as it is the mathematical foundation of the integrated safety assessment (ISA) methodology developed by the CSN(Consejo de Seguridad Nuclear) branch of Modelling and Simulation (MOSI). Such methodology attempts to extend classical PSA including accident dynamic analysis, an assessment of the damage associated to the transients and a computation of the damage frequency. The application of this ISA methodology requires a computational framework called SCAIS (Simulation Code System for Integrated Safety Assessment). SCAIS provides accident dynamic analysis support through simulation of nuclear accident sequences and operating procedures. Furthermore, it includes probabilistic quantification of fault trees and sequences; and integration and statistic treatment of risk metrics. SCAIS comprehensively implies an intensive use of code coupling techniques to join typical thermal hydraulic analysis, severe accident and probability calculation codes. The integration of accident simulation in the risk assessment process and thus requiring the use of complex nuclear plant models is what makes it so powerful, yet at the cost of an enormous increase in complexity. As the complexity of the process is primarily focused on such accident simulation codes, the question of whether it is possible to reduce the number of required simulation arises, which will be the focus of the present work. This document presents the work done on the investigation of more efficient techniques applied to the process of risk assessment inside the mentioned ISA methodology. Therefore such techniques will have the primary goal of decreasing the number of simulation needed for an adequate estimation of the damage probability. As the methodology and tools are relatively recent, there is not much work done inside this line of investigation, making it a quite difficult but necessary task, and because of time limitations the scope of the work had to be reduced. Therefore, some assumptions were made to work in simplified scenarios best suited for an initial approximation to the problem. The following section tries to explain in detail the process followed to design and test the developed techniques. Then, the next section introduces the general concepts and formulae of the TSD theory which are at the core of the risk assessment process. Afterwards a description of the simulation framework requirements and design is given. Followed by an introduction to the developed techniques, giving full detail of its mathematical background and its procedures. Later, the test case used is described and result from the application of the techniques is shown. Finally the conclusions are presented and future lines of work are exposed.

We have proposed a novel robust inversion-based neurocontroller that searches for the optimal control law by sampling from the estimated Gaussian distribution of the inverse plant model. However, for problems involving the prediction of continuous variables, a Gaussian model approximation provides only a very limited description of the properties of the inverse model. This is usually the case for problems in which the mapping to be learned is multi-valued or involves hysteritic transfer characteristics. This often arises in the solution of inverse plant models. In order to obtain a complete description of the inverse model, a more general multicomponent distributions must be modeled. In this paper we test whether our proposed sampling approach can be used when considering an arbitrary conditional probability distributions. These arbitrary distributions will be modeled by a mixture density network. Importance sampling provides a structured and principled approach to constrain the complexity of the search space for the ideal control law. The effectiveness of the importance sampling from an arbitrary conditional probability distribution will be demonstrated using a simple single input single output static nonlinear system with hysteretic characteristics in the inverse plant model.

The approach uses recent developments in neural networks, especially in the context of modelling statistical distributions, which are applied to forward and inverse plant models. Provided that certain conditions are met, an estimate of the intrinsic uncertainty for the outputs of neural networks can be obtained using the statistical properties of networks. More generally, multicomponent distributions can be modelled by the mixture density network. In this work a novel robust inverse control approach is obtained based on importance sampling from these distributions. This importance sampling provides a structured and principled approach to constrain the complexity of the search space for the ideal control law. The performance of the new algorithm is illustrated through simulations with example systems.

The developed methodology circumvents the dynamic programming problem by using the predicted neural network uncertainty to localise the possible control solutions to consider. Convergence of the output error for the proposed control method is verified by using a Lyapunov function. Several simulation examples are provided to demonstrate the efficiency of the developed control method. The manner in which such a method is extended to nonlinear multi-variable systems with different delays between the input-output pairs is considered and demonstrated through simulation examples.

In this paper a new framework has been applied to the design of controllers which encompasses nonlinearity, hysteresis and arbitrary density functions of forward models and inverse controllers. Using mixture density networks, the probabilistic models of both the forward and inverse dynamics are estimated such that they are dependent on the state and the control input. The optimal control strategy is then derived which minimizes uncertainty of the closed loop system. In the absence of reliable plant models, the proposed control algorithm incorporates uncertainties in model parameters, observations, and latent processes. The local stability of the closed loop system has been established. The efficacy of the control algorithm is demonstrated on two nonlinear stochastic control examples with additive and multiplicative noise.

In nonlinear and stochastic control problems, learning an efficient feed-forward controller is not amenable to conventional neurocontrol methods. Provided that certain conditions are met, an estimate of the intrinsic uncertainty for the outputs of neural networks can be obtained using the statistical properties of networks. More generally, multicomponent distributions can be modelled by the mixture density network. Based on importance sampling from these distributions a novel robust inverse control approach is obtained. This importance sampling provides a structured and principled approach to constrain the complexity of the search space for the ideal control law.
