O efeito do cultivo do mamoeiro em ambiente protegido foi estudado em três condições: sem cobertura e em dois telados construídos com tela de propileno branca, com malhas de 2 x 2 mm e 2 x 1 mm. Nessa área foram feitas avaliações na cultivar Baixinho de Santa Amália, contando-se o número de plantas com sintomas de ataque recente, para o ácaro branco Polyphagotarsonemus latus, com sintomas e presença de ácaros, para o ácaro rajado Tetranychus urticae e com presença de adultos ou ninfas nas folhas, no caso das moscas-brancas Trialeurodes sp., Bemisia tabaci biótipo B e uma terceira espécie não identificada. Para moscas-brancas, também foram realizadas contagens de ninfas e exúvias em laboratório. O cultivo em ambiente protegido favoreceu a sobrevivência e o desenvolvimento populacional das espécies estudadas, sendo que algumas possíveis causas são discutidas no texto. Considerando-se que o cultivo protegido pode ser uma boa alternativa para o controle de viroses, como o mosaico do mamoeiro, problema limitante para a cultura, estratégias de manejo de pragas nesses ambientes devem ser desenvolvidas, para viabilizar o seu uso.

Sequence diversity in the coat protein coding region of Australian strains of Johnsongrass mosaic virus (JGMV) was investigated. Field isolates were sampled during a seven year period from Johnsongrass, sorghum and corn across the northern grain growing region. The 23 isolates were found to have greater than 94% nucleotide and amino acid sequence identity. The Australian isolates and two strains from the U.S.A. had about 90% nucleotide sequence identity and were between 19 and 30% different in the N-terminus of the coat protein. Two amino acid residues were found in the core region of the coat protein in isolates obtained from sorghum having the Krish gene for JGMV resistance that differed from those found in isolates from other hosts which did not have this single dominant resistance gene. These amino acid changes may have been responsible for overcoming the resistance conferred by the Krish gene for JGMV resistance in sorghum. The identification of these variable regions was essential for the development of durable pathogen-derived resistance to JGMV in sorghum.

We report on the production and evaluation of passionflower transgenic lines for resistance to Cowpea aphid borne mosaic virus (CABMV). Genetic transformation was done using Agrobacterium tumefaciens and transgene integration was confirmed by Southern blot analyses, resulting in nine transgenic lines for `IAC 275` and three for `IAC 277`. Transgenic lines were clonally propagated and evaluated for resistance to CABMV After the third inoculation, under higher inoculum pressure, only propagated plants of the transgenic line T16 remained asymptomatic, indicating a high resistance to infection with CABMV. This transgenic line was self-pollinated and the RI generation was evaluated together with the RI generation of another resistant transgenic line (T2) identified previously. Plants were inoculated with CABMV by means of viruliferous Myzus nicotianae. All 524 T2R(1) plants became infected, whereas 13 of 279 T16R(1) remained asymptomatic after four successive inoculations. A TI6R(2) generation was obtained and plants were inoculated with CABMV mechanically or by aphids. After successive inoculations, 118 of 258 plants were symptomless, suggesting that the resistance to CABMV was maintained in the plant genome as the homozygous condition was achieved. Five selected resistant TI6R(2) plants which contained the capsid protein gene are being crossed for further analyses.

Neste trabalho avaliou-se o efeito da premunização com duas estirpes fracas do vírus-do-mosaico-do-mamoeiro - estirpe melancia ("papaya ringspot virus - type W", PRSV-W), combinada com a tolerância das plantas, no controle do mosaico e na produtividade da abóbora 'Menina Brasileira'. Testes realizados em casa de vegetação mostraram que as plantas dessa cultivar premunizadas com as estirpes fracas PRSV-W-1 e 2 ficaram totalmente protegidas contra a infecção por uma estirpe severa de Campinas. Em condições de campo avaliou-se comparativamente a produção de plantas premunizadas, infectadas com a estirpe severa de Campinas, e sadias e expostas à infecção natural (controle). A proteção foi medida com base na produção individual das plantas, cujos frutos foram classificados em comerciais e não-comerciais. As plantas premunizadas tiveram uma produção média de frutos comerciais (peso) 33% superior à daquelas naturalmente infectadas em campo. Quanto ao número de frutos comerciais, o aumento foi da ordem de 50%. A premunização combinada com a tolerância da abóbora 'Menina Brasileira' permitiu um melhor controle do mosaico, com ganhos na produção de frutos comerciais.

Os vírus representam sérios obstáculos para o sucesso da olericultura no mundo inteiro, constituindo a identificação daqueles de maior incidência numa região, papel fundamental para o estabelecimento de estratégias de controle. Visitas de campo foram realizadas a plantios de espécies de cucurbitáceas em áreas produtoras do Maranhão e amostras foliares foram coletadas de 118 plantas com sintomas ou suspeita de sintomas de vírus, sendo 46 de abóbora (Cucurbita moschata), 30 de melancia (Citrullus lanatus), 23 de maxixe (Cucumis anguria), 13 de pepino (C. sativus) e seis de melão (C. melo). Todas as amostras foram testadas contra anti-soros específicos para os principais vírus das famílias Bromoviridae, Comoviridae e Potyviridae que infetam cucurbitáceas no Nordeste, mediante "enzyme-linked immunosorbent assay" (ELISA) indireto e dupla difusão em agar. Os resultados revelaram a identificação sorológica de Papaya ringspot vírus (PRSV) em 64,4% das amostras analisadas, seguido de Watermelon mosaic virus-2 (WMV-2) em 15,2%, Cucumber mosaic virus (CMV) em 6,8%, Squash mosaic virus (SqMV) em 3,4% e Zucchini yellow mosaic virus (ZYMV) em 3,4%. Este levantamento confirma a predominância do PRSV em espécies de cucurbitáceas cultivadas no estado do Maranhão.

As viroses constituem o principal grupo de doenças do mamoeiro (Carica papaya), ocasionando grandes perdas na produção, podendo chegar à destruição total das plantações afetadas. Embora mais de dez vírus tenham sido constatados infetando naturalmente o mamoeiro, em todo o mundo, no Brasil, até o presente, foram assinaladas apenas as ocorrências do vírus da mancha anelar do mamoeiro (Papaya ringspot virus, PRSV), do vírus do amarelo letal do mamoeiro (Papaya lethal yellowing virus, PLYV) e do vírus da meleira que se encontra em fase de caracterização. A mancha anelar causada pelo PRSV é, inquestionavelmente, o mais importante problema sanitário do mamoeiro. O controle do PRSV mostra-se imprescindível, apesar de bastante difícil, em razão da sua forma de disseminação rápida e eficiente por diversas espécies de afídeos e ausência de resistência genética em C. papaya. Na tentativa de controlar o PRSV, várias medidas já foram testadas, não existindo, até o momento, nenhuma estratégia eficiente e duradoura para seu controle no Brasil. O desenvolvimento de plantas transgênicas de mamoeiro expressando o gene da capa protéica (cp) do PRSV, imunes ao mesmo, abriu nova possibilidade para solução do problema.

The main objective of this work was to investigate the ability of Aphis gossypii and Myzus persicae to transmit Cucumber mosaic virus (CMV) singly and mixed with two potyviruses (Papaya ringspot virus - type W, PRSV-W and Zucchini yellow mosaic virus, ZYMV), to zucchini squash plants (Cucurbita pepo). The results showed that the potyviruses in general were more efficiently transmitted by both species of aphids as compared to CMV. The transmission of PRSV-W, ZYMV and CMV separately was more efficient than in mixture.

O ZLCV é um tospovírus encontrado com freqüência causando severos danos em cucurbitáceas. Nesse trabalho avaliaram-se os danos causados pelo ZLCV em abobrinha de moita 'Caserta', em campo na ESALQ/USP, Piracicaba-SP, onde esse vírus é freqüente. Plantas obtidas pela semeadura direta foram monitoradas periodicamente quanto à infecção pelo ZLCV por meio dos sintomas e por PTA-ELISA. Monitorou-se ainda a contaminação com Papaya ringspot virus - type W e Zucchini yellow mosaic virus, desconsiderando a produção dessas plantas. As plantas foram agrupadas em função da época de aparecimento dos sintomas do ZLCV, avaliando a produção de frutos comerciais (FC) e não comerciais (FNC) de cada grupo e comparando com a de plantas que permaneceram sem sintomas até o final do experimento. As plantas que apresentaram sintomas até os 23 dias após a emergência (DAE) não produziram qualquer tipo de frutos. FC foram colhidos de plantas que apresentaram sintomas a partir dos 42 DAE. Mesmo assim, houve redução de 78,5 % na produção de FC. Plantas que mostraram sintomas por ocasião da última colheita (55 DAE) apresentaram redução na produção de FC de 9,6 %. A infecção com o ZLCV até o início da frutificação inviabiliza a produção de FC de abobrinha de moita 'Caserta'.

Cucurbits species grown in 38 of 40 agricultural regions in the state of Sao Paulo, Brazil, were surveyed for the relative incidence of Cucumber mosaic virus (CMV), Papaya ringspot virus-type W (PRSV-W), Watermelon mosaic virus-2 (WMV-2), Zucchini lethal chlorosis virus (ZLCV), and Zucchini yellow mosaic virus (ZYMV) during May 1997 and June 1999. Samples from 621 plants, representing eight cultivated species, six wild species, and one commercial hybrid (Cucurbita moschata x C. maxima), were analyzed by plate trapped antigen enzyme-linked immunosorbent assay (PTA-ELISA). PRSV-W and ZYMV were the most frequently found viruses, accounting for 49.1 and 24.8%, respectively, of 605 samples tested. ZLCV, CMV, and WMV-2 were detected in 7.8, 6.0, and 4.5% of 612, 497, and 423 samples tested, respectively. Double infection was found in 97 samples, and triple infection was found in 10 samples. Quadruple infection was detected in one C. pepo sample. Plants that were symptomatic but negative by PTA-ELISA might be due to abiotic agents, infection by virus for which antiserum was not available, such as Squash mosaic virus, or infection with an as yet uncharacterized virus.

The purpose of this work was to evaluate the effects of ethylene action blockade and cold storage on the ripening of `Golden` papaya fruit. Papayas harvested at maturity stage 1 (up to 15% yellow skin) were evaluated. Half of the fruits, whether treated or not treated with 100 nL L(-1) of 1-methylcyclopropene (1-MCP), were stored at 23A degrees C, while the other half were stored at 11A degrees C for 20 days prior to being stored at 23A degrees C. Non-refrigerated fruits receiving 1-MCP application presented a reduction in respiratory activity, ethylene production, skin color development and pectinmethylesterase activity. Even with a gradual increase in ethylene production at 23A degrees C, fruits treated with 1-MCP maintained a high firmness, but presented a loss of green skin color. Cold storage caused a decrease in ethylene production when fruits were transferred to 23A degrees C. The results suggest that pulp softening is more dependent on ethylene than skin color development, and that some processes responsible for loss of firmness do not depend on ethylene.

The carotenoid composition was evaluated during ripening of papaya cv. `Golden` under untreated (control) conditions and treated with ethylene and 1-methylcyclopropene (1-MCP). At the end of the experiments, the total carotenoid content in the control group (2194.4 mu g/100 g) was twice as high as that found in ethylene (1018.1 mu g/100 g) and 1-MCP (654.5 mu g/100 g) gas-treated samples. Separation of 21 carotenoids by HPLC connected to photodiode array and mass spectrometry detectors showed that no minor carotenoids seemed to be particularly favoured by the treatments. Lycopene was the major carotenoid in all untreated and gas-treated samples, ranging from 461.5 to 1321.6 mu g/100 g at the end of the experiments. According to the proposed biosynthetic pathway, lycopene is the central compound, since it is the most abundant carotenoid indicating a high stimulation of its upstream steps during ripening, and it is the source for the synthesis of other derivative compounds, such as beta-cryptoxanthin. Papaya (Carica papaya) is a relevant tropical crop and physico-chemical changes take place very quickly, as a consequence of activation of biochemical pathways by de nova synthesis of several proteins. Thus, in order to have information on the changes in gene expression in ripening papaya, transcripts from the pulp of unripe and ripe fruit were profiled by differential-display RT-PCR (DDRT-PCR). Seventy transcript derived fragments (TDFs) isolated from gels were re-amplified by PCR and differential expression of 40 papaya genes was confirmed by reverse northern blotting. Twenty-nine positively cloned TDFs were sequenced, and 17 were putatively identified by homology search. Ten of these genes were downregulated during ripening and UDP-glucose glucosyltransferase, alpha-2 importin, RNase L inhibitor-like protein, and a syntaxin protein were identified. Among the up-regulated genes there was a carboxylesterase, an integral membrane Yip1 family protein, a glycosyl hydrolase family-like protein and an endopolygalacturonase. Considering their relatedness to papaya quality, the fragments of genes potentially implicated in carbohydrate metabolism and pulp softening may be considered of interest for further studies. According to the results, differential display was a feasible approach to investigate differences in gene expression during fruit ripening, and can provide interesting information about those fruits whose genomic data is scarce, as is the case of papayas. Papaya (Carica papaya L) fruit has a short shelf life due to fast ripening induced by ethylene, but little is known about the genetic control of ripening and attributes of fruit quality. Therefore, we identified ripening-related genes affected by ethylene using cDNA-AFLP (Amplified Fragment Length Polymorphism of cDNA). Transcript profiling of non-induced and ethylene-induced fruit samples was performed, and 71 differentially expressed genes were identified. Among those genes some involved in ethylene biosynthesis, regulation of transcription, and stress responses or plant defence were found (heat shock proteins, polygalacturonase-inhibiting protein, and acyl-CoA oxidases). Several transcription factors were isolated, and except for a 14-3-3 protein, an AP2 domain-containing factor, a salt-tolerant zinc finger protein, and a suppressor of PhyA-105 1, most of them were negatively affected by ethylene, including fragments of transcripts similar to VRN1, and ethylene responsive factors (ERF). With respect to fruit quality, genes related to cell wall structure or metabolism, volatiles or pigment precursors, and vitamin biosynthesis were also found. (C) 2010 Elsevier Ireland Ltd.
