Balancing acts: Respiratory sensations, motor control and human posture

1. The present brief review covers some novel aspects of integration between respiration and movement of the body. 2. There are potent viscerosomatic reflexes in animals involving small-diameter pulmonary afferents that, when excited, would limit exercise. However, recent studies using lobeline injections to excite pulmonary afferents in awake humans suggest that there is no evoked reflex motoneuronal inhibition. Instead, the noxious respiratory sensations generated by the vagal afferents may be crucial in the decision to stop exercise. 3. While respiratory movements may affect limb movements, the control of the trunk and limbs can involve interaction (and even interference) with key respiratory muscles, such as the diaphragm. Recent studies have revealed that not only does the diaphragm receive feed-forward drive prior to some limb movements, but that it also contracts both phasically and tonically during repetitive limb movements. 4. Thus, challenges to posture can indirectly challenge ventilation, while coordinated diaphragm contraction may contribute to control of the trunk.

14-3-3 acts as an intramolecular bridge to regulate cdc25B localization and activity

One of the major regulators of mitosis in somatic cells is cdc25B. cdc25B is tightly regulated at multiple levels. The final activation step involves the regulated binding of 14-3-3 proteins. Previous studies have demonstrated that Ser-323 is a primary 14-3-3 binding site in cdc25B, which influences its activity and cellular localization. 14-3-3 binding to this site appeared to interact with the N-terminal domain of cdc25B to regulate its activity. The presence of consensus 14-3-3 binding sites in the N-terminal domain suggested that the interaction is through direct binding of the 14-3-3 dimer to sites in the N-terminal domain. We have identified Ser-151 and Ser-230 in the N-terminal domain as functional 14-3-3 binding sites utilized by cdc25B in vivo. These low affinity sites cooperate to bind the 14-3-3 dimer bound to the high affinity Ser-323 site, thus forming an intramolecular bridge that constrains cdc25B structure to prevent access of the catalytic site. Loss of 14-3-3 binding to either N-terminal site relaxes cdc25B structure sufficiently to permit access to the catalytic site, and the nuclear export sequence located in the N-terminal domain. Mutation of the Ser-323 site was functionally equivalent to the mutation of all three sites, resulting in the complete loss of 14-3-3 binding, increased access of the catalytic site, and access to nuclear localization sequence.

OxyR acts as a repressor of catalase expression in Neisseria gonorrhoeae

It has been reported that Neisseria gonorrhoeae possesses a very high level of catalase activity, but the regulation of catalase expression has not been investigated extensively. In Escherichia coli and Salmonella enterica serovar Typhimurium, it has been demonstrated that OxyR is a positive regulator of hydrogen peroxide-inducible genes, including the gene encoding catalase. The oxyR gene from N. gonorrhoeae was cloned and used to complement an E. coli oxyR mutant, confirming its identity and function. The gene was inactivated by inserting a kanamycin resistance cassette and used to make a knockout allele on the chromosome of N. gonorrhoeae strain 1291. In contrast to E. coli, the N. gonorrhoeae oxyR::kan mutant expressed ninefold-more catalase activity and was more resistant to hydrogen peroxide killing than the wild type. These data are consistent with OxyR in N. gonorrhoeae acting as a repressor of catalase expression.

Mitigated directness in Honglou meng: directive speech acts and politeness in eighteenth century Chinese

This paper is situated within a theoretical discussion that asserts a positive correlation between indirectness and politeness, and uses these claims as a springboard for an examination of polite ways of issuing directives in eighteenth century Chinese. On the basis of an analysis of directive speech acts from dialogue in the novel Honglou meng, it argues that the concept of indirectness, as it applies to the illocutionary transparency of individual speech acts, has no particular value in the culture and language of eighteenth century Chinese men. It is found that other linguistic devices such as particles, the reduplication of verbs, terms of address, and the presence and sequencing of supportive moves are far more significant to the communication of politeness. The findings suggest a need to rethink current theoretical positions on this subject and move beyond the analysis of individual speech acts to examine the role discourse structure and management play in the enactment of politeness. Mutating Identities: Clarice Lispector's um sopro de vida - pulsações and Virginia Woolf's between the acts

The questioning of fictional identities is central in both texts. Their structure illustrates the Bakhtinian notions of heteroglossia and dialogism and one of their main themes is the structuring and restructuring of social and personal history. Together with the use of irony and humour, the parody of social context, the use of intertextuality and metafiction, the exploration of fragment and of discontinuity, the development of self reflection and the interrogation of the author before the condition of his work, the focusing on the presence of a reader lost in the interpretation of the text, this problematisation of fictional identities places these two novels between the acts of modernism and postmodernism.

Aproximadamente una de cada cinco personas en el mundo mueren de cáncer. A pesar de los considerables avances en el tratamiento, incluyendo procedimientos de alta precisión en cirugía y protocolos detallados para rayo y quimioterapia, los tumores malignos son una de las causas de muerte más prominentes de la sociedad moderna. Las estrategias para el tratamiento dependen de un diagnostico preciso y temprano. No obstante algunos tumores son dificultosos para clasificarlos, desarrollando una incertidumbre en el pronóstico y tratamiento. Los tumores de tejidos blando y óseo son un grupo de neoplasias diverso y grande, que ocurren en todas las edades, normalmente sin un aparente factor etiológico. El diagnostico/ pronóstico puede ser complicado y el dilema del diagnóstico diferencial depende frecuentemente de los métodos histopatológicos y radiológicos. El presente proyecto intenta detectar las aberraciones cromosómicas en tumores sólidos utilizando las técnicas de biología molecular. Este método de detección es el más adecuado para diagnosticar y tratar eficientemente una gran cantidad de cánceres. Permite identificar alteraciones cromosómicas,tales como deleciones, translocaciones, amplificaciones y mutaciones que están asociadas con tipos específicos de tumores sólidos.

Small ubiquitin-like modifier conjugating enzyme with active site mutation acts as dominant negative inhibitor of SUMO conjugation in arabidopsis(F).

Small ubiquitin-like modifier (SUMO) conjugation affects a broad range of processes in plants, including growth, flower initiation, pathogen defense, and responses to abiotic stress. Here, we investigate in vivo and in vitro a SUMO conjugating enzyme with a Cys to Ser change in the active site, and show that it has a dominant negative effect. In planta expression significantly perturbs normal development, leading to growth retardation, early flowering and gene expression changes. We suggest that the mutant protein can serve as a probe to investigate sumoylation, also in plants for which poor genetic infrastructure precludes analysis via loss-of-function mutants.

The Acts of Paul and the Canonical Acts : A Phenomenon of Rereading

(Abstract only in french) Une approche littéraire de l'intertextualité, à l'aide des catégories posées par G. Genette, permet de renouveler l'approche du rapport entre Actes de Paul et Actes canoniques. A la différence de la critique littéraire classique, qui ne repère pas de dépendance littéraire hors de similitudes verbales ou narratives, l'« hypertextualité » désigne un phénomène de relecture dans lequel un texte-source est recomposé et réinterprété au sein d'un écrit second. Grâce à cette catégorie, cette contribution rend compte du jeu dialectique de parenté et de divergence que l'on observe entre les Actes de Paul et les Actes de Luc. La recomposition de la biographie de Paul dans les Actes de Paul a usé de créativité, elle a puisé dans l'imaginaire chrétien, mais elle s'est aussi servie de traditions préservées dans la suivance de l'apôtre.
