以黄土高原地区典型草本(白羊草)、灌木(沙棘)和乔木(辽东栎)为对象,研究了3种植物细根比根长在不同土层的分布状况以及与其它细根参数和土壤物理因子之间的相关性。结果表明,3种植物细根比根长的变化范围为6~55 mm/mg。在0~80cm土层,白羊草、沙棘和辽东栎细根比根长变化范围分别为18~55 mm/mg,14~40 mm/mg,6~33 mm/mg。3种植物0~80cm土层平均细根比根长从大到小依次为白羊草>沙棘>辽东栎。3种植物0~10cm土层细根比根长依次为沙棘>辽东栎>白羊草,10~80cm依次为白羊草>辽东栎>沙棘,表明3种植物细根比根长不仅在这两土层中的分布不具一致性,而且与0~80cm土层平均比根长也不具有一致性,进一步说明3种植物沿土壤剖面的生物量分配策略不同。相关分析表明,3种植物细根比根长与其它细根参数之间的相互关系各不相同,制约程度存在差异。与土壤物理因子的相关分析表明,3种植物细根比根长均随土壤含水量的增加而减少。土壤各级水稳性团聚体和土壤颗粒对3种植物细根比根长并无一致的影响。

A field and market basket study (similar to 1300 samples) of locally grown fruits and vegetables from historically mined regions of southwest (SW) England (Cornwall and Devon), and as reference, a market basket study of similarly locally grown produce from the northeast (NE) of Scotland (Aberdeenshire) was conducted to determine the concentration of total and inorganic arsenic present in produce from these two geogenically different areas of the U.K. On average 98.5% of the total arsenic found was present in the inorganic form. For both the market basket and the field survey, the highest total arsenic was present in open leaf structure produce (i.e., kale, chard, lettuce, greens, and spinach) being most likely to soil/dust contamination of the open leaf structure. The concentration of total arsenic in potatoes, swedes, and carrots was lower in peeled produce compared to unpeeled produce. For baked potatoes, the concentration of total arsenic in the skin was higher compared to the total arsenic concentration of the potato flesh, this difference in localization being confirmed by laser ablation inductively coupled plasma mass spectroscopy (LA-ICP-MS). For all above ground produce (e.g., apples), peeling did not have a significant effect on the concentration of total arsenic present.

A plasticidade ecológica observada em plantas que ocupam diferentes formações vegetais é frequentemente acompanhada por variações morfológicas, sendo a folha o órgão vegetativo com a maior variação estrutural. Diante disso, cinco espécies de Polygala L. ocorrentes em restinga e cerrado do Estado de São Paulo tiveram a anatomia da folha analisada com o objetivo de efetuar um inventário dos seus principais caracteres e descrever sua estrutura, com o propósito de compará-las, e de avaliar se é possível evidenciar padrões estruturais comuns às condições ecológicas de ambas as formações vegetacionais. As espécies pesquisadas foram: P. cyparissias A.St. –Hil. & Moq., P. laureola A.St.-Hil. & Moq., P. paniculata L., encontradas em restinga; P. angulata DC. e P. violacea Aubl. emend. Marques, que ocorrem em campo cerrado e em margem de cerradão, respectivamente. A região mediana da lâmina foliar e a região proximal do pecíolo de cada folha foram processadas segundo as técnicas usuais para microscopia eletrônica de varredura e de luz. Os resultados obtidos evidenciam dois padrões estruturais comuns às espécies procedentes das formações de restinga e cerrado: características mesomórficas são observadas em P. laureola, P. paniculata e P. violacea, e xeromórficas em P. cyparissias e P. angulata.

Focal adhesion kinase (FAK) regulates cellular processes that affect several aspects of development and disease. The FAK N-terminal FERM (4.1 protein-ezrin-radixin-moesin homology) domain, a compact clover-leaf structure, binds partner proteins and mediates intramolecular regulatory interactions. Combined chemical cross-linking coupled to MS, small-angle X-ray scattering, computational docking and mutational analyses showed that the FAK FERM domain has a molecular cleft (similar to 998 angstrom(2)) that interacts with sarcomeric myosin, resulting in FAK inhibition. Accordingly, mutations in a unique short amino acid sequence of the FERM myosin cleft, FP-1, impaired the interaction with myosin and enhanced FAK activity in cardiomyocytes. An FP-1 decoy peptide selectively inhibited myosin interaction and increased FAK activity, promoting cardiomyocyte hypertrophy through activation of the AKT-mammalian target of rapamycin pathway. Our findings uncover an inhibitory interaction between the FAK FERM domain and sarcomeric myosin that presents potential opportunities to modulate the cardiac hypertrophic response through changes in FAK activity.

Despite striking differences in climate, soils, and evolutionary history among diverse biomes ranging from tropical and temperate forests to alpine tundra and desert, we found similar interspecific relationships among leaf structure and function and plant growth in all biomes. Our results thus demonstrate convergent evolution and global generality in plant functioning, despite the enormous diversity of plant species and biomes. For 280 plant species from two global data sets, we found that potential carbon gain (photosynthesis) and carbon loss (respiration) increase in similar proportion with decreasing leaf life-span, increasing leaf nitrogen concentration, and increasing leaf surface area-to-mass ratio. Productivity of individual plants and of leaves in vegetation canopies also changes in constant proportion to leaf life-span and surface area-to-mass ratio. These global plant functional relationships have significant implications for global scale modeling of vegetation–atmosphere CO2 exchange.

A number of aberrant morphological phenotypes were noted during propagation of the Arabidopsis thaliana DNA hypomethylation mutant, ddm1, by repeated self-pollination. Onset of a spectrum of morphological abnormalities, including defects in leaf structure, flowering time, and flower structure, was strictly associated with the ddm1 mutations. The morphological phenotypes arose at a high frequency in selfed ddm1 mutant lines and some phenotypes became progressively more severe in advancing generations. The transmission of two common morphological trait syndromes in genetic crosses demonstrated that the phenotypes are caused by heritable lesions that develop in ddm1 mutant backgrounds. Loss of cytosine methylation in specific genomic sequences during the selfing regime was noted in the ddm1 mutants. Potential mechanisms for formation of the lesions underlying the morphological abnormalities are discussed.

This thesis provides new knowledge on an understudied group of grasses, some of which are resurrection grasses (i.e. able to withstand extreme drought). The sole Australian species (Tripogon loliiformis) is morphologically diverse and could be more than one species. This study sought to determine how many species of Tripogon occur in Australia, their relationships to other species in the genus and to two other genera of resurrection grasses (Eragrostiella and Oropetium). Results of the research indicate there is not enough evidence, from DNA sequence data, to warrant splitting up T. loliiformis into multiple species. The extensive morphological diversity seems to be influenced by environmental conditions. The three genera are so closely related that they could be grouped into a single genus. This new knowledge opens up pathways for future investigations, including studying genes responsible for desiccation tolerance and the conservation of native grasses that occur in rocky habitats.

Host feeding selection by the female pea leafminer, Liriomyza huidobrensis, on 47 species of plants was studied. The leaves were sectioned by microtome, and 15 characteristics of the leaf tissue structure were measured under a microscope. Correlation analysis between host feeding selection and leaf tissue structure indicated that the preference of host feeding selection was positively correlated with the percentage of moisture content of leaves and negatively with thickness of the epidermis wall, and densities of the palisade and spongy tissues of leaves. Leaf tissue structure was influential in feeding and probing behavior of female L. huidobrensis. So, thickness of epidermis wall, densities of the palisade and spongy tissues can act as a physical barrier to female oviposition. Furthermore, higher densities of palisade and spongy tissues can be considered a resistant trait which affects mining of leaf miner larvae as well. As a result, plants with lower leaf moisture content may not be suitable for the development of L. huidobrensis.
