Cleome rosea é uma espécie nativa, de porte herbáceo, ocorrente em restingas brasileiras. Estudos recentes têm revelado o potencial medicinal da espécie para importantes propriedades farmacológicas, como por exemplo, as atividades anti-inflamatória, antigenotóxica, antiviral e antibacteriana. Porém, nos últimos anos, C. rosea não tem sido encontrada em várias regiões de seu ambiente natural, devido, principalmente, às ações antrópicas. Dessa forma, torna-se relevante o desenvolvimento de métodos de conservação que permitam o estudo e exploração das propriedades medicinais da espécie. O cultivo in vitro de raízes representa uma forma eficiente para produção de biomassa, devido ao rápido crescimento, produção estável de metabólitos, além de representar uma potencial fonte de explantes para a propagação em massa de diferentes espécies. O presente trabalho teve como objetivo a produção in vitro de culturas de raízes de C. rosea, associada à criopreservação, como forma de manutenção em longo prazo das culturas, monitorada através da análise de estabilidade genética. As culturas estabelecidas a partir de explantes radiculares de plantas propagadas in vitro de C. rosea demonstraram excelente capacidade de multiplicação de raízes em meio de cultura suplementado com o fitorregulador ANA, com manutenção dessa capacidade ao longo de sucessivas subculturas. Associado a esses resultados, o estabelecimento de protocolos de criopreservação pelo método de vitrificação resultou em elevados valores de frequência de recuperação do material após congelamento em nitrogênio líquido com as soluções de vitrificação PVS2 e PVS3. Os estudos de monitoramento da estabilidade genética, pela técnica de marcadores moleculares RAPD, revelaram a presença de polimorfismos significativos em uma das três culturas iniciadas a partir de raízes de C. rosea criopreservadas. Esses resultados demonstram as possibilidades de produção de raízes de C. rosea e conservação em longo prazo através da criopreservação, iniciando estudos inéditos para a espécie.

课题组在不断地创制新的同源四倍体材料的同时,连续多年以提高结实率为目的培育、筛选自交系材料,已获得自交繁殖十二年的高代自交系材料。相对于诱导创制初期,材料表现出的结实率低,同种系单株间的差异较大;高代材料已表现出较显著的结实率提升和较一致的农艺性状表型。 本实验选取课题组多年培育的同源四倍体水稻高代自交系材料,通过形态学、农艺性状和细胞遗传学比较,研究水稻同源四倍体与二倍体之间的异同。结果显示,所有同源四倍体材料的染色体组成均为2N=4X=48,花粉母细胞(PMC)减数分裂行为较正常,99%以上的染色体都能在减数分裂中期I(MI)发生联会、配对,形成四价体和二价体,这与理论染色体组构成相符。在减数分裂过程中,结实率较高的材料染色体异常现象较少而结实率较低的材料染色体异常现象较严重。统计分析表明,二价体和四价体的比例对结实率没有显著影响,但是三价体的数目对结实率有一定影响。这一结果表明了结实率和细胞减数分裂行为可能存在相关性,同源四倍体的减数分裂行为为筛选高结实率的同源四倍体种系提供了依据。 然后,对同源四倍体水稻高代自交系材料进行农艺性状和品质性状的统计与分析。主要针对结实率、每穗实粒数、有效分蘖和穗长等主要农艺性状,以及直链淀粉含量这一重要的品质性状进行统计。将统计结果与1996年诱导加倍的初代材料的数据相对比分析,结果显示所有材料经过多代选育培养,其农艺性状已经有了较显著的提高,同时同源四倍体材料的农艺性状稳定性也有了较显著的提升。如结实率的提高幅度较大,所有材料的平均结实率均显著高于加倍初代,而同种材料不同单株间的结实率差异也显著地减少,变异系数(CV)的平均值由1996年的41.15%减少到了2008年的28.81%。其他重要农艺性状也有不同程度的改良,种内变异系数也相应地降低。此外,实验测量了同源四倍体材料和来源二倍体材料的直链淀粉含量。分析结果显示,部分材料的直链淀粉含量与二倍体亲本产生了较显著的差异,这可能是诱导加倍过程中的遗传变异造成的;同源四倍体材料的种内变异系数(CV)平均值由1996年的6%下降到了2008年的3.88%,显示出在品质性状方面,同源四倍体材料的遗传稳定性也有较显著的增加。同源四倍体材料农艺性状经过多年的选育,表现出一定的提升,同时,经过多年自交纯化,所有材料种系内的性状差异逐渐缩小,说明同源四倍体水稻的遗传稳定性随着自交纯化而增强,这为同源四倍体水稻的进一步选育打下了良好的基础。 最后,通过测量连续两年的自交系材料的遗传多态性,分析材料间遗传差异和种群遗传结构,深入研究连续两代材料间的遗传差异,研究同源四倍体水稻与二倍体材料遗传稳定性之间的差异。实验采用18对SSR微卫星标记对连续两代15个材料,共94份样本进行差异分析。通过扩增条带长度多态性分析,计算不同材料以及同种材料不同世代间的遗传距离,构建同源四倍体和二倍体水稻的分子指纹库,并绘制聚类图。结果显示,同源四倍体和二倍体不同材料间的遗传差异比较大,遗传距离处于0.4757至0.2816之间;而相同品种不同世代材料间的遗传差异较小,但也表现出一定的遗传差异。同种同源四倍体材料不同世代间的遗传差异比二倍体材料更大,两代四倍体材料间遗传距离处于0.1359至0.0485之间;而两代二倍体材料间的遗传距离处于均小于0.0388。结果表明,同源四倍体水稻高代材料具有一定的遗传稳定性,但与来源二倍体材料相比,其世代间的遗传变异性仍然较强。这种结果说明,经过多代的自交纯化培育,同源四倍体水稻材料能够建立起相对稳定的遗传结构,同时,其强于二倍体亲本的变异性有能够为新品种的选育,农艺性状、品质性状的改良提供一定的遗传基础。此外,分析结果表明通过分子标记辅助检验,水稻材料间的遗传多态性能够有效地区分不同的品种,这为水稻品种的分子鉴定提供了一定的依据。 本研究从细胞学鉴定,农艺性状统计分析以及分子标记辅助聚类分析多方面地对同源四倍体水稻高代系进行了研究,对探究同源四倍体水稻的遗传规律,进一步揭示其遗传特性、农艺性状的遗传构成,为进一步选育优质的多倍体水稻提供了一定的理论依据。 This group insists on creating new Autotetraploid Rice (Oryza sativa L.) materials, while improving the seed-setting of them for many years, cultivated and selected the inbred line materials, has obtained the high generation inbred lines after twelve years cultivation. Compared to the early induced materials, which shown the low seed setting, and the large difference between the different plants in the same germ-line; the high generation materials have shown significant improvement in seed setting and more uniform phenotype agronomic traits. The autotetraploid rice high generation inbred lines material, which has been cultivated for more than 12 years, was chose in this experiment. The similarities and differences between autotetraploid and diploid rice was studied through morphological, agronomic and cytogenetic ways. The results showed that all the chromosome of autotetraploid materials are composed of 2N=4X=48, the pollen mother cells (PMC) meiosis behavior is normal, more than 99% chromosomes in metaphase I(MI) were federated and paired to form tetravalents or bivalents, which constitutes a consistent theory of genome. In the meiosis process, the material with a higher seed setting showed less chromosome abnormal than the material whose seed setting is lower. However, statistical analysis showed that the bivalent and tetravalent rate had no significant impact on seed setting, but the number of trivalent had a certain impact on seed setting. The result shows that the seed setting may be related to the meiosis behavior, which provides a basis to cultivate new autotetraploid germ line with high seed setting through the meiotic behavior. Furthermore, the agronomic and quality traits of autotetraploid rice high generation inbred material were statistically analyzed. The statistically analysis was focused on major agronomic traits such as: seed setting, grains per panicle, effective tillers and panicle length, as well as the important quality trait amylose content. The statistic data was compared with the data in 1996, when the first induced generation of autotetraploid material, and the result shows that after a multi-generation breeding, the agronomic traits has been significantly improved in all the materials, while the stability of agronomic traits also significant upgraded. For instant, the seed setting increased significantly, the average seed setting of all materials was significantly higher than the first induced generation, and the differences between different plants in the same species also significantly reduced, the average of the coefficient of variation (CV) was reduced from 41.15% in 1996 to 28.81% in 2008. Other important agronomic traits had improved in different degrees; the coefficient of variation within species is also reduced accordingly. In addition, the amylose content of autotetraploid and diploid materials was measured in this experiment. The results shows that the amylose content of some of the material differed from diploid parents significantly, it may caused by the genetic change during the inducing, autotetraploid materials intra-specific coefficient of variation (CV) average reduced from 6% in 1996 to 3.88% in 2008, shows that this is a significant increase of quality traits stability in autotetraploid rice. Agronomic traits of autotetraploid material shows some improvement after years of breeding, at the same time, after years of purification, all material within the germ-line gradually narrow the differences in traits indicates that autotetraploid rice genetic stability was enhanced, which laid a good foundation for the further autotetraploid rice breeding. Finally, this experiment studied the genetic differences between materials of two generations and researched the difference of genetic stability between diploid and autotetraploid rice materials through investigating the genetic polymorphism, genetic differences between materials and population genetic structure of inbred line materials of two consecutive years.18 pairs of SSR microsatellite markers for 15 materials of two generations were used in this experiment, and the total of 94 samples were analyzed. Through the amplification length polymorphism analysis of different materials and materials in different generations, the genetic distance between materials and generations was analyzed, a diploid and autotetraploid rice molecular fingerprint database and map rendering cluster were constructed. The result shows that the genetic distance is between 0.4757 to 0.2816 among different autotetraploid and diploid materials; the genetic distance between different generations of same species was less, but also shows a certain degree of genetic differences. The inter-generational genetic differences of autotetraploid materials were greater than of the diploid materials, which are 0.1359 to 0.0485 as the genetic distance; comparing with the 0.0388 of diploid materials. The result shows that high generation inbred autotetraploid rice material has a certain genetic stability, but the genetic variation between generations is still strong comparing with the source diploid materials. It indicates that, after many generations of purification cultivation, autotetraploid rice materials established a relatively stable genetic structure, at the same time, stronger variability than its diploid parents are useful in the breeding of new varieties, provides a genetic foundation to the agronomic and quality traits improvement. In addition, the analysis result shows that the through the molecular marker-assisted testing, rice genetic polymorphism between materials can effectively distinguish different species, provides a certain basis for molecular identification of varieties of rice. A series of investigation such as cytological identification, statistical analysis of agronomic traits, molecular marker-assisted cluster analysis was applied in this experiment to research genetic pattern of autotetraploid rice high generation inbred lines, revealed the genetic characteristics and the genetic composition of agronomic traits, provides a theoretical basis for the further selection of high quality autotetraploid rice.

The porcine circovirus type 2 (PCV2) genome encodes three major open reading frames (ORFs) encoding the replicase proteins (ORF1), the viral capsid protein (ORF2), and a protein with suggested apoptotic activity (ORF3). Previous phylogenetic analyses of complete genome sequences of PCV2 from GenBank have demonstrated 95-100% intra-group nucleotide sequence identity. However, although these isolates were readily grouped into clusters and clades, there was no correlation between the occurrence of specific PCV2 genotypes and the geographic origin or health status of the pig. In the present study, a unique dataset from a field study spanning the years pre and post the recognition of postweaning multisystemic wasting syndrome (PMWS) in Sweden was utilized. Using this dataset it was possible to discriminate three Swedish genogroups (SG1-3) of PCV2, of which SG1 was recovered from a pig on a healthy farm ten years before the first diagnosis of PMWS in Sweden. The SG1 PCV2/ORF2 gene sequence has been demonstrated to exhibit a high genetic stability over time and has subsequently only been demonstrated in samples from pigs on nondiseased farms. In contrast, SG2 was almost exclusively found on farms that had only recently broken down with PMWS whereas the SG3 genogroup predominated in pigs from PMWS-affected farms. These results further support the results obtained from earlier in vitro and in vivo experimental models and suggest the association of specific PCV2 genogroups with diseased and nondiseased pigs in the field.

There is a paradox between the remarkable genetic stability of measles virus (MV) in the field and the high mutation rates implied by the frequency of the appearance of monoclonal antibody escape mutants generated when the virus is pressured to revert in vitro (S. J. Schrag, P. A. Rota, and W. J. Bellini, J. Virol. 73: 51-54, 1999). We established a highly sensitive assay to determine frequencies of various categories of mutations in large populations of wild-type and laboratory-adapted MVs using recombinant viruses containing an additional transcription unit (ATU) encoding enhanced green fluorescent protein (EGFP). Single and double mutations were made in the fluorophore of EGFP to ablate fluorescence. The frequencies of reversion mutants in the population were determined by measuring the appearance of fluorescence indicating a revertant virus. This allows mutation rates to be measured under nonselective conditions, as phenotypic reversion to fluorescence requires only either a single-or a double-nucleotide change and amino acid substitution, which does not affect the length of the nonessential reporter protein expressed from the ATU. Mutation rates in MV are the same for wild-type and laboratory-adapted viruses, and they are an order of magnitude lower than the previous measurement assessed under selective conditions. Dada a extrema importância económica e ambiental que o montado de sobro tem em Portugal, e dado o declínio deste devido a várias razões (e.g. doença, idade das plantas) é premente desenvolver estratégias de preservação de sobreiros elite e optimizar técnicas para a propagação destes genótipos. No primeiro Capítulo expõe-se uma breve introdução sobre o montado actual e as técnicas actuais de regeneração/propagação do sobreiro. Descreve-se ainda as principais técnicas de preservação e avaliação de estabilidade genética referidas na literatura para sobreiro e outras lenhosas. No Capítulo II é apresentado um estudo de melhoramento das condições actuais de maturação de embriões somáticos de sobreiro com vista a aperfeiçoar o processo de conversão em plantas. Neste capítulo é apresentado um protocolo melhorado em relação ao actual que permite um desenvolvimento dos embriões somáticos dum modo semelhante aos embriões zigóticos em termos de substâncias de reserva. O Capítulo III mostra um estudo efectuado com o objectivo principal de avaliar estabilidade genética durante todo o processo de embriogénese somática. Neste capítulo são apresentados resultados duma análise feita por RAPD em fases distintas da embriogénese somática de sobreiro. Neste estudo mostra-se que não existem diferenças significativas entre plantas de campo, embriões somáticos e plantas regeneradas. No Capítulo VI, pretende-se complementar o estudo anterior. Neste Capítulo descreve-se a dinâmica do ciclo celular durante as primeiras fases de embriogénese somática na presença de reguladores de crescimento. Este trabalho permitiu concluir a importância dos reguladores de crescimento na indução e perceber o peso do factor genótipo durante o processo. Considerando os resultados anteriores, a necessidade de um processo eficiente de preservação de genótipos elite torna-se fundamental. No Capítulo V descreve-se um protocolo de criopreservação eficiente sem recursos a substâncias tóxicas. Nesta secção é ainda feita uma análise de variabilidade genética após criopreservação através de FCM, AFLP e SSR. Todos os resultados obtidos anteriormente são postos a prova no Capítulo VI onde se faz uma monitorização extensiva de 10 genótipos elite, tendo em conta a sua capacidade de produção de cortiça, através do processo de embriogénese somática. Durante esta secção são utilizados os protocolos desenvolvidos anteriormente e avaliados na sua eficiência. Neste capítulo é descrita a integração de vários segmentos deste estudo num só protocolo eficiente de regeneração e preservação de sobreiros através de embriogénese somática. Finalmente, no Capítulo VI são apresentadas as conclusões da presente Tese de Doutoramento, com especial incidência para linhas de investigação futuras a serem tomadas. Discute-se a importância deste novo protocolo na optimização da produção da cortiça e traçam-se possíveis aplicações alternativas.

Background: Microarray based comparative genomic hybridisation (CGH) experiments have been used to study numerous biological problems including understanding genome plasticity in pathogenic bacteria. Typically such experiments produce large data sets that are difficult for biologists to handle. Although there are some programmes available for interpretation of bacterial transcriptomics data and CGH microarray data for looking at genetic stability in oncogenes, there are none specifically to understand the mosaic nature of bacterial genomes. Consequently a bottle neck still persists in accurate processing and mathematical analysis of these data. To address this shortfall we have produced a simple and robust CGH microarray data analysis process that may be automated in the future to understand bacterial genomic diversity. Results: The process involves five steps: cleaning, normalisation, estimating gene presence and absence or divergence, validation, and analysis of data from test against three reference strains simultaneously. Each stage of the process is described and we have compared a number of methods available for characterising bacterial genomic diversity, for calculating the cut-off between gene presence and absence or divergence, and shown that a simple dynamic approach using a kernel density estimator performed better than both established, as well as a more sophisticated mixture modelling technique. We have also shown that current methods commonly used for CGH microarray analysis in tumour and cancer cell lines are not appropriate for analysing our data. Conclusion: After carrying out the analysis and validation for three sequenced Escherichia coli strains, CGH microarray data from 19 E. coli O157 pathogenic test strains were used to demonstrate the benefits of applying this simple and robust process to CGH microarray studies using bacterial genomes.

Due to their broad differentiation potential and their persistence into adulthood, human neural crest-derived stem cells (NCSCs) harbour great potential for autologous cellular therapies, which include the treatment of neurodegenerative diseases and replacement of complex tissues containing various cell types, as in the case of musculoskeletal injuries. The use of serum-free approaches often results in insufficient proliferation of stem cells and foetal calf serum implicates the use of xenogenic medium components. Thus, there is much need for alternative cultivation strategies. In this study we describe for the first time a novel, human blood plasma based semi-solid medium for cultivation of human NCSCs. We cultivated human neural crest-derived inferior turbinate stem cells (ITSCs) within a blood plasma matrix, where they revealed higher proliferation rates compared to a standard serum-free approach. Three-dimensionality of the matrix was investigated using helium ion microscopy. ITSCs grew within the matrix as revealed by laser scanning microscopy. Genetic stability and maintenance of stemness characteristics were assured in 3D cultivated ITSCs, as demonstrated by unchanged expression profile and the capability for self-renewal. ITSCs pre-cultivated in the 3D matrix differentiated efficiently into ectodermal and mesodermal cell types, particularly including osteogenic cell types. Furthermore, ITSCs cultivated as described here could be easily infected with lentiviruses directly in substrate for potential tracing or gene therapeutic approaches. Taken together, the use of human blood plasma as an additive for a completely defined medium points towards a personalisable and autologous cultivation of human neural crest-derived stem cells under clinical grade conditions.

Introduction Facing the challenging treatment of neurodegenerative diseases as well as complex craniofacial injuries such as those common after cancer therapy, the field of regenerative medicine increasingly relies on stem cell transplantation strategies. Here, neural crest-derived stem cells (NCSCs) offer many promising applications, although scale up of clinical-grade processes prior to potential transplantations is currently limiting. In this study, we aimed to establish a clinical-grade, cost-reducing cultivation system for NCSCs isolated from the adult human nose using cGMP-grade Afc-FEP bags. Methods We cultivated human neural crest-derived stem cells from inferior turbinate (ITSCs) in a cell culture bag system using Afc-FEP bags in human blood plasma-supplemented medium. Investigations of viability, proliferation and expression profile of bag-cultured ITSCs were followed by DNA-content and telomerase activity determination. Cultivated ITSCs were introduced to directed in vitro differentiation assays to assess their potential for mesodermal and ectodermal differentiation. Mesodermal differentiation was determined using an enzyme activity assay (alkaline phosphatase, ALP), respective stainings (Alizarin Red S, Von Kossa and Oil Red O), and RT-PCR, while immunocytochemistry and synaptic vesicle recycling were applied to assay neuroectodermal differentiation of ITSCs. Results When cultivated within Afc-FEP bags, ITSCs grew three-dimensionally in a human blood plasma-derived matrix, thereby showing unchanged morphology, proliferation capability, viability and expression profile in comparison to three dimensionally-cultured ITSCs growing in standard cell culture plastics. Genetic stability of bag-cultured ITSCs was further accompanied by unchanged telomerase activity. Importantly, ITSCs retained their potential to differentiate into mesodermal cell types, particularly including ALP-active, Alizarin Red S-, and Von Kossa-positive osteogenic cell types, as well as adipocytes positive in Oil Red O assays. Bag culture further did not affect the potential of ITSCs to undergo differentiation into neuroectodermal cell types coexpressing β-III-tubulin and MAP2 and exhibiting the capability for synaptic vesicle recycling. Conclusions Here, we report for the first time the successful cultivation of human NCSCs within cGMP-grade Afc-FEP bags using a human blood plasma-supplemented medium. Our findings particularly demonstrate the unchanged differentiation capability and genetic stability of the cultivated NCSCs, suggesting the great potential of this culture system for future medical applications in the field of regenerative medicine.

As mutações no gene de supressão tumoral p53 estão entre as anormalidades genéticas mais comuns encontradas numa ampla variedade de tumores. Embora a função do gene p53 ainda não esteja completamente esclarecida, ele parece ser um fator de transcrição nuclear que controla a proliferação celular, a apoptose e a manutenção da estabilidade genética. A angiogênese é essencial para o crescimento e a metastatização de tumores sólidos. O Fator de Crescimento do Endotélio Vascular (VEGF, Vascular Endothelial Growth Factor), um fator de crescimento identificado recentemente com propriedades angiogênicas significativas, pode ser um importante regulador da angiogênese tumoral. A associação entre as expressões da proteína p53 e do VEGF e o prognóstico tem sido pouco estudada. Foram estudadas peças cirúrgicas de 47 pacientes com carcinoma epidermóide de esôfago (CEE) submetidos à esofagectomia em estágios II e III, utilizando-se coloração imuno-histoquímica. As expressões da proteína p53 e do VEGF foram observadas em 53% e 40% dos tumores, respectivamente. As expressões da proteína p53 e do VEGF coincidiram em somente 21% dos casos, e não foi encontrada correlação entre elas. Nenhum dos fatores clinicopatológicos se correlacionaram significativamente com as expressões da proteína p53 ou do VEGF. Em relação ao prognóstico, não havia associação significativa entre as expressões da proteína p53 e do VEGF e pior prognóstico. Os autores concluem que tanto a expressão da proteína p53 como a expressão do VEGF não se correlacionaram com o prognóstico em pacientes com CEE em estágios II e III.

Human multipotent mesenchymal stromal cells (MSCs), also known as mesenchymal stem cells, have become an important and attractive therapeutic tool since they are easily isolated and cultured, have in vitro expansion potential, substantial plasticity and secrete bioactive molecules that exert trophic effects. The human umbilical cord as a cell source for cell therapy will help to avoid several ethical, political, religious and technical issues. One of the main issues with SC lines from different sources, mainly those of embryonic origin, is the possibility of chromosomal alterations and genomic instability during in vitro expansion. Cells isolated from one umbilical cord exhibited a rare balanced paracentric inversion, likely a cytogenetic constitutional alteration, karyotype: 46,XY,inv(3)(p13p25~26). Important genes related to cancer predisposition and others involved in DNA repair are located in 3p25~26. Titanium is an excellent biomaterial for bone-implant integration; however, the use can result in the generation of particulate debris that can accumulate in the tissues adjacent to the prosthesis, in the local bone marrow, in the lymph nodes, liver and spleen. Subsequently may elicit important biological responses that aren´t well studied. In this work, we have studied the genetic stability of MSC isolated from the umbilical cord vein during in vitro expansion, after the cryopreservation, and under different concentrations and time of exposition to titanium microparticles. Cells were isolated, in vitro expanded, demonstrated capacity for osteogenic, adipogenic and chondrogenic differentiation and were evaluated using flow cytometry, so they met the minimum requirements for characterization as MSCs. The cells were expanded under different concentrations and time of exposition to titanium microparticles. The genetic stability of MSCs was assessed by cytogenetic analysis, fluorescence in situ hybridization (FISH) and analysis of micronucleus and other nuclear alterations (CBMN). The cells were able to internalize the titanium microparticles, but MSCs preserve their morphology, differentiation capacity and surface marker expression profiles. Furthermore, there was an increase in the genomic instability after long time of in vitro expansion, and this instability was greater when cells were exposed to high doses of titanium microparticles that induced oxidative stress. It is necessary always assess the risks/ benefits of using titanium in tissue therapy involving MSCs, considering the biosafety of the use of bone regeneration using titanium and MSCs. Even without using titanium, it is important that the therapeutic use of such cells is based on analyzes that ensure quality, security and cellular stability, with the standardization of quality control programs appropriate. In conclusion, it is suggested that cytogenetic analysis, FISH analysis and the micronucleus and other nuclear alterations are carried out in CTMH before implanting in a patient

The phylogeny is one of the main activities of the modern taxonomists and a way to reconstruct the history of the life through comparative analysis of these sequences stored in their genomes aimed find any justification for the origin or evolution of them. Among the sequences with a high level of conservation are the genes of repair because it is important for the conservation and maintenance of genetic stability. Hence, variations in repair genes, as the genes of the nucleotide excision repair (NER), may indicate a possible gene transfer between species. This study aimed to examine the evolutionary history of the components of the NER. For this, sequences of UVRA, UVRB, UVRC and XPB were obtained from GenBank by Blast-p, considering 10-15 as cutoff to create a database. Phylogenetic studies were done using algorithms in PAUP programs, BAYES and PHYLIP package. Phylogenetic trees were build with protein sequences and with sequences of 16S ribosomal RNA for comparative analysis by the methods of parsimony, likelihood and Bayesian. The XPB tree shows that archaeal´s XPB helicases are similar to eukaryotic helicases. According to this data, we infer that the eukaryote nucleotide excision repair system had appeared in Archaea. At UVRA, UVRB and UVRC trees was found a monophyletic group formed by three species of epsilonproteobacterias class, three species of mollicutes class and archaeabacterias of Methanobacteria and Methanococci classes. This information is supported by a tree obtained with the proteins, UVRA, UVRB and UVRC concatenated. Thus, although there are arguments in the literature defending the horizontal transfer of the system uvrABC of bacteria to archaeabacterias, the analysis made in this study suggests that occurred a vertical transfer, from archaeabacteria, of both the NER genes: uvrABC and XPs. According the parsimony, this is the best way because of the occurrence of monophyletic groups, the time of divergence of classes and number of archaeabacterias species with uvrABC system

The use and inadequate exploitation of natural resources is restricting the occurrence of aroeira (Myracrodruon urundeuva F.F. & M.F. Allemao), which now is on the FAO list of endangered species. This exploitation causes a decrease in the genetic base of M. urundeuva populations, which makes it difficult to find genotypes with stability and adaptability to different growing conditions. This study aimed at estimating the genetic variation and productivity, stability and adaptability of progenies of a M. urundeuva natural population, from the Ecological Station of Paulo de Faria - SP, under different planting systems. DBH (diameter at breast height) was evaluated in four progeny tests of M. urundeuva: i) planted with Anandenanthera falcata and Guazuma ulmifolia (TP-AMA); ii) single (TP-ASO); iii) planted with annual crops (TP-SAF) and iv) planted with Corymbia citriodora (TP-EUCA), installed in Selviria-MS. The experimental design consisted of complete randomized blocks with three replications and a variable number of plants per plot in each of the four planting systems. From the joint analysis of the planting systems studied, it was found that: i) there were variations among planting systems particularly in TP-SAF; ii) only in TP-EUCA it was possible to detect variations among the progenies; iii) the effects of the genotype x environment interaction were not significant. Thereby, the harmonic mean of genotypic values (MHVG), the relative performance of genotypic values from the mean of each site (PRVG) and the harmonic mean of the relative performance of genotypic values (MHPRVG) for DBH showed, respectively: progenies with greater stability, adaptability, and stability and simultaneous adaptability within different planting systems. The use of these selection criteria provided a more refined selection of the best progenies of M. urundeuva under the different planting systems studied.
