Diesel spills contaminate aquatic and terrestrial environments. To prevent the environmental and health risks, the remediation needs to be advanced. Bioremediation, i.e., degradation by microbes, is one of the suitable methods for cleaning diesel contamination. In monitored natural attenuation technique are natural processes in situ combined, including bioremediation, volatilization, sorption, dilution and dispersion. Soil bacteria are capable of adapting to degrade environmental pollutants, but in addition, some soil types may have indigenous bacteria that are naturally suitable for degradation. The objectives for this work were (1) to find a feasible and economical technique to remediate oil spilled into Baltic Sea water and (2) to bioremediate soil contaminated by diesel oil. Moreover, the aim was (3) to study the potential for natural attenuation and the indigenous bacteria in soil, and possible adaptation to degrade diesel hydrocarbons. In the aquatic environment, the study concentrated on diesel oil sorption to cotton grass fiber, a natural by-product of peat harvesting. The impact of diesel pollution was followed in bacteria, phytoplankton and mussels. In a terrestrial environment, the focus was to compare the methods of enhanced biodegradation (biostimulation and bioaugmentation), and to study natural attenuation of oil hydrocarbons in different soil types and the effect that a history of previous contamination may have on the bioremediation potential. (1) In the aquatic environment, rapid removal of diesel oil was significant for survival of tested species and thereby diversity maintained. Cotton grass not only absorbed the diesel but also benefited the bacterial growth by providing a large colonizable surface area and hence oil-microbe contact area. Therefore use of this method would enhance bioremediation of diesel spills. (2) Biostimulation enhances bioremediation, and (3) indigenous diesel-degrading bacteria are present in boreal environments, so microbial inocula are not always needed. In the terrestrial environment experiments, the combination of aeration and addition of slowly released nitrogen advanced the oil hydrocarbon degradation. Previous contamination of soil gives the bacterial community the potential for rapid adaptation and efficient degradation of the same type of contaminant. When the freshly contaminated site needs addition of diesel degraders, previously contaminated and remediated soil could be used as a bacterial inoculum. Another choice of inoculum could be conifer forest soil, which provides a plentiful population of degraders, and based on the present results, could be considered as a safe non-polluted inoculum. According to the findings in this thesis, bioremediation (microbial degradation) and monitored natural attenuation (microbial, physical and chemical degradation) are both suitable techniques for remediation of diesel-contaminated sites in Finland.

Research on conducting polymers, organic light emitting diodes and organic solar cells has been an exciting field for the past decade. The challenge with these organic devices is the long term stability of the active material. Organic materials are susceptible to chemical degradation in the presence of oxygen and moisture. The sensitivity of these materials towards oxygen and moisture makes it imperative to protect them by encapsulation. Polymer nanocomposites can be used as encapsulation materials in order to prevent material degradation. In the present work, amine functionalized alumina was used as a cross-linking and reinforcing material for the polymer matrix in order to fabricate the composites to be used for encapsulation of devices. Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy and Raman spectroscopy were used to elucidate the surface chemistry. Thermogravimetric analysis techniques and CHN analysis were used to quantify grafting density of amine groups over the surface of the nanoparticles. Mechanical characterizations of the composites with various loadings were carried out with dynamic mechanical analyzer. It was observed that the composites have good thermal stability and mechanical flexibility, which are important for an encapsulant. The morphology of the composites was evaluated using scanning electron microscopy and atomic force microscopy.

A steel ball was slid on a steel flat lubricated by molybdenum disulfide (MoS2) particles suspended in hexadecane oil at 150 degrees C. The friction data is compared with that obtained when the ball was slid on the flat sprayed apriori with nominally dry MoS2 particles. The friction in the dry experiment was found to increase with temperature while the friction in wet condition was found to decrease with increasing temperature. Micro-Raman and Fourier transform IR spectroscopy are used to explore the roles of environmental moisture and chemical degradation of oil on the formation of antifriction film on the steel substrate.

A avaliação da qualidade dos solos e a redução da Mata Atlântica tem sido alvo de diversas pesquisas no Brasil e no mundo, principalmente quando estes estão atrelados ao recorte espacial de Unidades de Conservação. No entanto, tem sido difícil para os gestores dessas unidades a avaliação dos impactos ambientais gerados ao longo dos anos nas bacias hidrográficas, principalmente pela falta de investimentos. Esta dissertação teve por objetivo geral avaliar o atual estágio de degradação da bacia hidrográfica do Córrego da Caçada pertencente à Área de Proteção Ambiental Federal de Cairuçu, no município de Paraty RJ, analisando quantitativamente e qualitativamente a redução dos fragmentos de Mata Atlântica e estabelecendo relações com a degradação física e química dos solos dentro e fora dos fragmentos florestais. A metodologia utilizada para a redução ou avanço dos fragmentos de Mata Atlântica baseou-se no uso de fotografias aéreas do ano de 1956 e imagens de satélite de 2012, onde possibilitou a avaliação espaço-temporal do uso e cobertura das terras, através da produção de um mapa temático final. Além disso, foram elaborados mapas temáticos de reconhecimento da área de estudo, como o de hipsometria, de declividade, de orientação e forma das encostas, além da geração de perfis topográficos. Para a avaliação da qualidade física, química e biológica dos solos foram determinadas as curvas de distribuição granulométrica, a densidade relativa dos grãos sólidos e a densidade aparente, porosidade total, os limites de liquidez e plasticidade, a estabilidade dos agregados em água, análises morfológicas, a saturação de bases, a capacidade de troca catiônica (CTC), a saturação por alumínio, fósforo, pH e o carbono orgânico. Para tal, foi realizada a abertura de três perfis, sendo um em área de fragmento florestal e dois em áreas de pasto. O resultado das análises permitiu, segundo a Sociedade Brasileira de Ciência do solo, a classificação de dois tipos de solos na bacia, sendo: Cambissolo Háplico Tb Distrófico Típico em área fragmento florestal e em área de pasto, e um Latossolo Amarelo Tb Distrófico Típico em área de pasto. Os resultados de laboratório mostraram que os solos avaliados têm baixa fertilidade e valores variados nos resultados de física do solo. No entanto, além do histórico de uso do solo caracterizado pelas práticas rudimentares do manejo empregado pelos Caiçaras, o clima predominante na região possibilita um regime pluviométrico anual que passa dos 2.000mm de chuva/ano, caracterizando solos muito lixiviados e pobres quimicamente. Portanto, conclui-se que a relação das propriedades físicas e químicas avaliadas junto ao manejo inadequado ao longo dos anos tem apresentado um cenário de grandes dificuldades para a recuperação florestal na bacia hidrográfica do Córrego da Caçada, o que mostra a importância da avaliação dos impactos ambientais não só pelo recorte de bacias hidrográficas, como contextualizar seu posicionamento dentro de Unidades de Conservação, com legislações e objetivos específicos.

该文通过大量实验,确定了制备单糖、寡糖及低聚糖的最佳生产工艺,并对几种制备方法做了比较研究.该文采用氧化降解和微波降解等不同技术对甲壳低聚糖的制备工艺进行了研究.H<,2>O<,2>作为氧化剂,在酸性条件下,采用4因素3水平进行正交实验,得到了制备分子量2000以下的寡糖的最佳条件.微波降解制备低聚糖目前尚未见报道,经过该实验证明此方法可降低能耗,减小污染,节省时间和原料,是一个具有产业化前景和市场潜力的好方法.通过以不同的壳聚糖为原料,探讨了在微波场作用下纯溶剂及盐效应对分子量变化的影响,最终产物的分子量均在1×10<'5>~10×10<'5>之间.根据IR光谱,壳聚糖降解前后主要峰的位置都无变化,只是随壳聚糖相对分子质量的降低各峰峰强有所变化,证明了壳聚糖氧化降解及微波降解制备寡糖、低聚糖是以开裂壳聚糖的β-1.4糖苷键来进行.该项研究对更好的开发利用甲壳质资源,促进中国海洋生物制品的发展具有十分重要的意义.

Climate change induced by anthropogenic warming of the earth's atmosphere is a daunting problem. This review examines one of the consequences of climate change that has only recently attracted attention: namely, the effects of climate change on the environmental distribution and toxicity of chemical pollutants. A review was undertaken of the scientific literature (original research articles, reviews, government and intergovernmental reports) focusing on the interactions of toxicants with the environmental parameters, temperature, precipitation, and salinity, as altered by climate change. Three broad classes of chemical toxicants of global significance were the focus: air pollutants, persistent organic pollutants (POPs), including some organochlorine pesticides, and other classes of pesticides. Generally, increases in temperature will enhance the toxicity of contaminants and increase concentrations of tropospheric ozone regionally, but will also likely increase rates of chemical degradation. While further research is needed, climate change coupled with air pollutant exposures may have potentially serious adverse consequences for human health in urban and polluted regions. Climate change producing alterations in: food webs, lipid dynamics, ice and snow melt, and organic carbon cycling could result in increased POP levels in water, soil, and biota. There is also compelling evidence that increasing temperatures could be deleterious to pollutant-exposed wildlife. For example, elevated water temperatures may alter the biotransformation of contaminants to more bioactive metabolites and impair homeostasis. The complex interactions between climate change and pollutants may be particularly problematic for species living at the edge of their physiological tolerance range where acclimation capacity may be limited. In addition to temperature increases, regional precipitation patterns are projected to be altered with climate change. Regions subject to decreases in precipitation may experience enhanced volatilization of POPs and pesticides to the atmosphere. Reduced precipitation will also increase air pollution in urbanized regions resulting in negative health effects, which may be exacerbated by temperature increases. Regions subject to increased precipitation will have lower levels of air pollution, but will likely experience enhanced surface deposition of airborne POPs and increased run-off of pesticides. Moreover, increases in the intensity and frequency of storm events linked to climate change could lead to more severe episodes of chemical contamination of water bodies and surrounding watersheds. Changes in salinity may affect aquatic organisms as an independent stressor as well as by altering the bioavailability and in some instances increasing the toxicity of chemicals. A paramount issue will be to identify species and populations especially vulnerable to climate-pollutant interactions, in the context of the many other physical, chemical, and biological stressors that will be altered with climate change. Moreover, it will be important to predict tipping points that might trigger or accelerate synergistic interactions between climate change and contaminant exposures.

Chemical degradations often induce changes in protein conformation and thus influence protein activity and protein stability in solutions. One difficulty in studying of chemical degradations on protein aqueous properties is to obtain sufficient amount of chemically degraded protein which is well characterized. Chemical degradation protocols that are often used may induce also conformation changes and aggregation of the protein. In this article we studied the effect of methionine oxidation on the conformation of recombinant human growth hormone (r-hGH). In literature it is reported that oxidation of methionine residues induces conformation changes on r-hGH. In our study, oxidation of r-hGH was performed by incubation with hydrogen peroxide under mild conditions. Mass spectrometry and chromatographic analysis revealed that oxidation with hydrogen peroxide resulted in more than 90% of oxidized r-hGH. By extensive spectroscopic characterizations no detectable change in conformation and aggregation of r-hGH after oxidation was found. In conclusion, mild oxidation conditions led to selective oxidation of the two more accessible methionine residues of r-hGH (Met(14) and Met(125)) and did not results in any conformation change of the protein. These findings prove that oxidation of human growth hormone does not influence protein conformation and demonstrate the importance of employing mild conditions during production of oxidized protein. Os vidros bioativos constituem um material apropriado para o preenchimento de defeitos ósseos, como alternativa a enxertos autólogos, uma vez que, quando expostos a fluidos fisiológicos promovem a formação de uma ligação com o tecido ósseo sob a forma de uma camada de hidroxiapatite carbonatada. No presente trabalho caracterizaram-se vidros bioativos sem conteúdo alcalino, cuja composição incide no sistema binário de diópsido (CaMgSi2O6) e fosfato de tricálcio (3CaO·P2O5), em função da sua molhabilidade, carga superficial, perfil de degradação, carácter bioativo em fluido fisiológico simulado e do seu comportamento in vitro em contacto com células estaminais mesenquimais humanas (hMSCs). A medição do ângulo de contacto inicial de água sobre os vidros demonstrou o carácter hidrofílico dos vidros investigados. A determinação do potencial zeta mostrou que a carga superficial dos vidros é negativa, sendo mais negativa na composição Di-70. O estudo da biodegradação dos vidros, efetuado através da sua imersão em Tris-HCl, permitiu concluir que a perda de peso dos vidros foi reduzida. A caraterização in vitro em meio acelular foi efetuada através da imersão dos vidros numa solução de fluido fisiológico simulado (SBF) e verificou-se que estes possuem capacidade de formar uma camada de hidroxiapatite carbonatada à sua superfície após 7 dias, detetável por XRD, FTIR e SEM/EDS, sugerindo que este conjunto de vidros é potencialmente bioativo, e poderá estimular a proliferação e diferenciação celular. A resposta das hMSCs em cultura aos vidros bioativos foi avaliada em termos de atividade metabólica, morfologia, viabilidade, proliferação e diferenciação osteogénica e conclui-se que os biovidros Di-60 e Di-70 poderão constituir um suporte viável para a proliferação e diferenciação de hMSCs.

Bioactive glasses and glass–ceramics are a class of biomaterials which elicit special response on their surface when in contact with biological fluids, leading to strong bonding to living tissue. This particular trait along with good sintering ability and high mechanical strength make them ideal materials for scaffold fabrication. The work presented in this thesis is directed towards understanding the composition-structure-property relationships in potentially bioactive glasses designed in CaOMgOP2O5SiO2F system, in some cases with added Na2O. The main emphasis has been on unearthing the influence of glass composition on molecular structure, sintering ability and bioactivity of phosphosilicate glasses. The parent glass compositions have been designed in the primary crystallization field of the pseudo-ternary system of diopside (CaO•MgO•2SiO2) – fluorapatite (9CaO•3P2O5•CaF2) – wollastonite (CaO•SiO2), followed by studying the impact of compositional variations on the structure-property relationships and sintering ability of these glasses. All the glasses investigated in this work have been synthesized via melt-quenching route and have been characterized for their molecular structure, sintering ability, chemical degradation and bioactivity using wide array of experimental tools and techniques. It has been shown that in all investigated glass compositions the silicate network was mainly dominated by Q2 units while phosphate in all the glasses was found to be coordinated in orthophosphate environment. The glass compositions designed in alkali-free region of diopside – fluorapatite system demonstrated excellent sintering ability and good bioactivity in order to qualify them as potential materials for scaffold fabrication while alkali-rich bioactive glasses not only hinder the densification during sintering but also induce cytotoxicity in vitro, thus, are not ideal candidates for in vitro tissue engineering. One of our bioglass compositions with low sodium content has been tested successfully both in vivo and in preliminary clinical trials. But this work needs to be continued and deepened. The dispersing of fine glass particles in aqueous media or in other suitable solvents, and the study of the most important factors that affect the rheology of the suspensions are essential steps to enable the manufacture of porous structures with tailor-made hierarchical pores by advanced processing techniques such as Robocasting.

Un protocole inspiré du test de simulation 309 de l’Organisation de coopération et de développement économiques (OCDE) nous a permis de mesurer la dégradation chimique (excluant la photolyse) dans des eaux de surface de même que la dégradation chimique et biologique de neuf contaminants émergents dans l’effluent d’un décanteur primaire d’eau usée municipale. Les données étaient compatibles avec le modèle de cinétique de pseudo ordre un. Les résultats démontrant une persistance de plus d’un an dans les eaux de surface et de 71 jours dans l’effluent du décanteur primaire suggèrent que les dégradations chimique et biologique ne contribuent pas significativement à la diminution de: atrazine, déséthylatrazine, carbamazépine et diclofénac dans la phase aqueuse des systèmes testés. Les autres composés se sont dégradés à différents niveaux. Le 17ß-estradiol ainsi que l’éthinylestradiol, la noréthindrone, la caféine et le sulfaméthoxazole ont tous été sujet à la dégradation biologique dans les effluents du décanteur primaire d’eau usée avec des constantes de dégradation k et des demi-vies t1/2 mesurées allant respectivement de 0.0082 à 0.59 j-1 et de 1.2 à 85 jours. Les paramètres de cinétique mesurés peuvent être combinés aux concentrations typiques des composés à l’étude dans un décanteur primaire d’eau usée pour y calculer leur vitesse de dégradation. Cependant, puisque les décanteurs primaires dans les usines de traitement d’eaux usées ont généralement des temps de résidence de quelques heures seulement, il est improbable que les neufs contaminants émergents à l’étude diminuent significativement par ces processus durant leur passage dans le compartiment.

Biodegradation is the chemical degradation of materials brought about by the action of naturally occurring microorganisms. Biodegradation is a relatively rapid process under suitable conditions of moisture, temperature and oxygen availability. The logic behind blending biopolymers such as starch with inert polymers like polyethylene is that if the biopolymer component is present in sufficient amount, and if it is removed by microorganisms in the waste disposal environment, then the base inert plastic should slowly degrade and disappear. The present work focuses on the preparation of biodegradable and photodegradable blends based on low density polyethylene incorporating small quantities of ionomers as compatibilizers. The thesis consists of eight chapters. The first chapter presents an introduction to the present research work and literature survey. The details of the materials used and the experimental procedures undertaken for the study are described in the second chapter. Preparation and characterization of low density polyethylene (LDPE)-biopolymer (starch/dextrin) blends are described in the third chapter. The result of investigations on the effect of polyethylene-co-methacrylic acid ionomers on the compatibility of LDPE and starch are reported in chapter 4. Chapter 5 has been divided into two parts. The first part deals with the effect of metal oxides on the photodegradation of LDPE. The second part describes the function of metal stearates on the photodegradation of LDPE. The results of the investigations on the role of various metal oxides as pro-oxidants on the degradation of ionomer compatibilized LDPE-starch blends are reported in chapter 6. Chapter 7 deals with the results of investigations on the role of various metal stearates as pro-oxidants on the degradation of ionomer compatibilized LDPE-starch blends. The conclusion of the investigations is presented in the last chapter of the thesis.

Powder mixtures (1:1) of tibolone polymorphic forms I (monoclinic) and II (triclinic) and excipients have been prepared and compacted. The samples were stored at 50 degrees C and 90% RH for one month and subsequently were evaluated using differential scanning calorimetry (DSC) and high-performance liquid chromatography (HPLC). The results indicate that during the compaction, the applied pressure reduced the chemical stability of tibolone in both polymorph forms. The triclinic form was more chemically unstable, both pure and in contact with excipients, than the monoclinic form. Lactose monohydrate was shown to reduce chemical degradation for both forms. Ascorbyl palmitate was shown to affect the tibolone stability differently depending on the polymorphic form used.
