Hydrophilic and lipophilic extracts of ten cultivars of Highbush and Rabbiteye Brazilian blueberries (Vaccinium corymbosum L. and Vacciniumashei Reade, respectively) that are used for commercial production were analysed for antioxidant activity by the FRAP, ORAC, ABTS and β-carotene-linoleate methods. Results were correlated to the amounts of carotenoids, total phenolics and anthocyanins. Brazilian blueberries had relatively high concentration of total phenolics (1,622-3,457 mg gallic acid equivalents per 100 g DW) and total anthocyanins (140-318 mg cyanidin-3-glucoside equivalents per 100 g DW), as well as being a good source of carotenoids. There was a higher positive correlation between the amounts of these compounds and the antioxidant activity of hydrophilic compared to lipophilic extracts. There were also significant differences in the level of bioactive compounds and antioxidant activities between different cultivars, production location and year of cultivation.

The present work reports the use of anthocyanins extracted from mulberry (Morus Alba L.), raspberry (Rubus Idaeus L.) and blueberry (Vaccinium myrtillus L.) as sensitizers in dye-sensitized solar cells. The conversion efficiency of these devices is dependent on the extracts employed and can be rationalized in terms of their composition and spectral properties. Solar cells sensitized by the mulberry extract showed the highest efficiency among the fruits investigated. Moreover, a 16 cm² active area solar cell with the mulberry extract has presented fair good efficiency of conversion for natural dye-based solar cells, besides stability over twenty weeks, showing perspectives for developing these low cost devices with a commercial viability.

In the NE part of the Aiguilles Rouges Massif near Martigny, at the eastern contact of the Variscan Vallorcine granite to adjacent gneisses, a series of pitchblende (UO2)-veins occur. This paper determines the level of enrichment and mobility of uranium in soils situated in the vicinity of such a UO2-vein 7 km west of Martigny. Within an area of 50 x 100 m, situated on a relatively steep slope and characterized by a strong gramma-ray anomaly, six soil profiles including their plant cover and a reference soil profile outside the influence of the UO2-vein have been examined. The soil shows pH-values between 4 and 5 and is colluvial. The applied analytical methods for the metal contents include extraction methods, common for soil studies, and bulk analysis performed with X-ray fluorescence and ICP-MS. Uranium contents found in the uppermost 20 cm of the soil profiles vary from 2,500 ppm close to the vein to 15 ppm at the lowermost point of the study area. The reference soil has around 3 ppm uranium. At greater depth (20 to 40 cm) the U-content decreases to about half of the surface values, indicating a vertical transport of uranium within the soil profile. No systematic dependance of uranium-contents to grain size (amount of clay) nor to the amount of organic matter has been found. However, the good correlation between uranium and free iron oxide concentration suggests adsorption of uranium on iron oxy-hydroxides. The ashes of grass and mosses contain up to 90 ppm U, the blueberry and redwood only up to 3 ppm. Our observations suggest that at the surface the uranium is transported by downhill creep (solifluxion) of uranium-rich rock fragments. Liberated by oxidation of the uppermost fragments in a given soil column, the uranium migrates vertically until the conditions are favourable to adsorption onto Fe-oxy-hydroxides. However, as high U-contents of local surface water show, this adsorption does not lead to a significant retention of the uranium.

The objective of this work was to evaluate the biochemical composition of six berry types belonging to Fragaria, Rubus, Vaccinium and Ribes genus. Fruit samples were collected in triplicate (50 fruit each) from 18 different species or cultivars of the mentioned genera, during three years (2008 to 2010). Content of individual sugars, organic acids, flavonols, and phenolic acids were determined by high performance liquid chromatography (HPLC) analysis, while total phenolics (TPC) and total antioxidant capacity (TAC), by using spectrophotometry. Principal component analysis (PCA) and hierarchical cluster analysis (CA) were performed to evaluate the differences in fruit biochemical profile. The highest contents of bioactive components were found in Ribes nigrum and in Fragaria vesca, Rubus plicatus, and Vaccinium myrtillus. PCA and CA were able to partially discriminate between berries on the basis of their biochemical composition. Individual and total sugars, myricetin, ellagic acid, TPC and TAC showed the highest impact on biochemical composition of the berry fruits. CA separated blackberry, raspberry, and blueberry as isolate groups, while classification of strawberry, black and red currant in a specific group has not occurred. There is a large variability both between and within the different types of berries. Metabolite fingerprinting of the evaluated berries showed unique biochemical profiles and specific combination of bioactive compound contents.

Antosyaanit ovat luonnossa esiintyvien flavonoidien suurin ryhmä. Niitä on tunnistettu yli 600 erilaista, mutta kuitenkin vain kuusi on luonnossa yleisesti esiintyviä. Eniten antosyaaneja on tummissa marjoissa kuten mustikassa, marja-aroniassa sekä variksenmarjassa. Antosyaaneilla on havaittu monia positiivisiaterveysvaikutuksia kuten antioksidatiivisia, antikarsinogeenisia, sekä antimikrobiaalisia vaikutuksia. Tässä työssä tutkittiin marjojen antosyaanien talteenottoa uuttamalla sekä niiden väkevöintiä. Tavoitteena oli saada mahdollisimman korkea antosyaanien saanto ja hyvä säilyvyys. Antosyaanien analysointi tehtiin nestekromatografisesti käyttäen ODS-2 kolonnia sekä nopeampaa spektrofotometristä pH-eromenetelmää. Tulokset laskettiin syanidiini-3-glukosidina. Antosyaanien väkevöintiä tutkittiin käyttäen erityyppisiä polymeerisiä makrohuokoisia adsorptiohartseja. Väkevöintiä kokeiltiin myös erottamalla epäpuhtauksia antosyaaniuutteesta sekä esikäsittelemällä puristejäännös eri olosuhteissa ennen antosyaanien uuttoa. Kuiva antosyaaniuute sisälsi ennen väkevöintiä noin 50 mg/g antosyaaneja ja väkevöinnin jälkeen yli 300 mg/g. Antosyaanien säilyvyyttä tutkittiin eri lämpötiloissa sekä kuivana että uutteena. Riippumatta säilytysolosuhteista kuivan puhdistetun antosyaanin havaittiin säilyvän hyvin.

A produção comercial de mirtilos na América do Sul tem passado por mudanças importantes nos últimos anos, em função do aumento da oferta de frutos frescos dos principais países produtores, causando forte queda de preços em plena safra, entre os meses de dezembro e fevereiro. Uma alternativa para esta situação é a produção de mirtileiros de maturação precoce ou muito tardia, fora da época de meia-estação, caracterizada pelos menores preços da fruta. Em 2010, foram introduzidas no Brasil novas variedades de mirtileiros de baixa exigência em frio desenvolvidas pela Universidade da Flórida, que podem ser cultivadas em regiões quentes e apresentam uma frutificação mais precoce, quando comparadas com as variedades tradicionalmente cultivadas no Brasil. Essas novas variedades apresentam grande potencial para a produção precoce de mirtilos no País em regiões sem frio hibernal, aumentando significativamente a rentabilidade da cultura. Para viabilizar esta produção, é necessário validar estratégias de manejo para estas variedades, devido à falta de informação técnica sobre seu cultivo nas condições edafoclimáticas brasileiras. Atualmente, estäo sendo conduzidas as primeiras pesquisas fitotécnicas no Estado de Säo Paulo para avaliar o crescimento e a produção destas variedades na ausência total de frio, situação de ocorrência particular em poucos países produtores do mundo.

Le bleuet démontre un potentiel thérapeutique dans le traitement du cancer et des maladies cardiovasculaires et neurodégénératives. Ces effets bénéfiques sont attribuables aux composés phénoliques abondants dans le bleuet, tels que les anthocyanines et les flavonoïdes. La biotransformation du jus de bleuet avec les bactéries Serratia vaccinii augmente sa teneur en composés phénoliques et son activité anti-oxydante, et modifie ses activités physiologiques. L’objectif de la présente étude est d’évaluer l’activité neuroprotectrice et le potentiel antidiabétique du jus de bleuet biostranformé (BJ). Le BJ est étudié dans différents tests dont : 1) La protection des neurones (N2a) contre le stress oxydatif (SO) induit par le peroxyde d’hydrogène; 2) La stimulation de la prise de glucose par les cellules musculaires (C2C12) et adipeuses (3T3-L1); 3) L’activité anti-hyperglycémique chez les souris obèses diabétiques KKAy. En effet, tandis que le jus de bleuet normal n’a aucun effet, le BJ augmente l’activité des enzymes anti-oxydantes, comme la catalase et la SOD (Superoxide Dimutase) et protège les neurones contre les changements de la signalisation des MAPKs et contre la toxicité induite par le peroxyde d’hydrogène. Le BJ augmente aussi la prise de glucose de 48% dans les cellules C2C12 et de 142% dans les cellules 3T3-L1. Cette augmentation n’est pas expliquée par une augmentation du calcium cytosolique mais plutôt par une stimulation de la phosphorylation de l’AMPK. De plus, le BJ inhibe l’adipogenèse chez les 3T3-L1. Le BJ diminue également l’hyperglycémie chez les souris obèses diabétiques KKAy et protège les jeunes souris pré-diabétiques contre le développement de l’obésité et du diabète. L’activité anti-hyperglycémique du BJ pourrait impliquer les adipokines puisque le BJ augmente le niveau d’adiponectine chez les souris diabétiques. Le BJ représente ainsi une approche prometteuse pour le traitement du diabète et les maladies neurodégénératives et une source de nouveaux agents thérapeutiques contre ces maladies.

Le diabète de type 2 est une maladie chronique dont l’incidence est en augmentation continuelle. Le risque de développer le diabète de type 2 chez les populations autochtones du Canada est de trois à cinq fois plus élevé que le reste de la population canadienne. La forêt boréale comporte plusieurs plantes médicinales ayant un potentiel pour le traitement ou la prévention du diabète. Certaines de ces plantes font partie de la médecine traditionnelle et alternative Crie. Des enquêtes ethnobotaniques ont amené notre équipe de recherche à identifier 17 extraits de plantes médicinales utilisées par les Cris d’Eeyou Istchee (Baie James, Québec) pour traiter les symptômes du diabète. Parmi ces extraits, certains ont montré des activités anti-diabétiques au niveau des cellules musculaires, des adipocytes et dans des études in vivo réalisées chez des animaux. Le but de cette thèse est d’élucider l’effet de ces 17 plantes sur l’homéostasie hépatique de glucose, d’identifier l’espèce la plus prometteuse et isoler ces constituants actifs. De même, le bleuet nain du genre Vaccinium angustifolium fait partie de la forêt boréale canadienne et est connu pour ses activités anti-diabétiques. Une biotransformation du jus de bleuet lui confère une activité antioxydante accrue et un profil biologique différent. Le deuxième but de cette thèse est d’élucider les mécanismes d’action par lesquels le jus de bleuet biotransformé (BJ) exerce son effet anti-diabétique et d’identifier ses principes actifs. Les résultats ont montré que trois extraits de plantes Cris se sont démarqués par leur effet sur l’homéostasie hépatique de glucose. Picea glauca exerce son effet en diminuant la production de glucose alors que Larix laricina agit en augmentant le stockage de glucose. Abies balsamea a montré le profil le plus prometteur, elle agit simultanément en diminuant l’activité de la Glucose-6-phosphatase (G6Pase) via la stimulation des voies insulino-dépendante et - indépendante et en augmentant l’activité de la Glycogène synthétase (GS) suite à la phosphorylation de la Glycogène synthase kinase-3. Le fractionnement de l’extrait d’Abies balsamea guidé par les deux bioessais a mené à l’isolation de trois composés actifs; l’acide abiétique (AA), l’acide déhydroabiétique (DAA) et le squalène (SQ). Les principes actifs ont montré le même mécanisme d’action que l’extrait brut en diminuant l’activité de la G6Pase et augmentant celle de la GS ainsi qu’en activant les voies de signalisation impliquées. Le DAA ii s’est démarqué par son effet le plus puissant et très comparable à celui de l’extrait d’Abies balsamea dans toutes les expériences. De son côté le BJ a montré un effet sur la diminution de la production hépatique de glucose, l’augmentation de son stockage ainsi que l’augmentation de son transport dans le muscle. Son fractionnement guidé par les bioessais a permis d’isoler sept fractions dont trois étaient les plus actives. L’identification des constituants de ces fractions actives a mené à isoler quatres composés phénoliques; l’acide chlorogénique, l’acide gallique, l’acide protocatéchique et le catéchol. Le catéchol s’est démarqué avec ses effets les plus puissants en diminuant l’activité de la G6Pase, augmentant celle de la GS et en stimulant le transport de glucose dans le muscle. Les résultats de cette thèse indiquent que la diminution de la production hépatique de glucose peut s’ajouter au profil anti-diabétique de certaines plantes médicinales Cries et surtout à celui d’A.balsamea dont les composés actifs peuvent aider dans le développement de nouvelles molécules anti-diabétiques. De plus, les résultats de cette thèse ont montré que l’activité antidiabétique du BJ implique le contrôle de l’homéostasie de glucose au niveau du foie et du muscle. L’identification du catéchol comme principe actif avec potentiel anti-diabétique prometteur pourra servir pour des fins thérapeutiques ultérieures.

There is considerable interest in the potential of a group of dietary-derived phytochemicals known as flavonoids in modulating neuronal function and thereby influencing memory, learning and cognitive function. The present review begins by detailing the molecular events that underlie the acquisition and consolidation of new memories in the brain in order to provide a critical background to understanding the impact of flavonoid-rich diets or pure flavonoids on memory. Data suggests that despite limited brain bioavailability, dietary supplementation with flavonoid-rich foods, such as blueberry, green tea and Ginkgo biloba lead to significant reversals of age-related deficits on spatial memory and learning. Furthermore, animal and cellular studies suggest that the mechanisms underpinning their ability to induce improvements in memory are linked to the potential of absorbed flavonoids and their metabolites to interact with and modulate critical signalling pathways, transcription factors and gene and/or protein expression which control memory and learning processes in the hippocampus; the brain structure where spatial learning occurs. Overall, current evidence suggests that human translation of these animal investigations are warranted, as are further studies, to better understand the precise cause-and-effect relationship between flavonoid intake and cognitive outputs.

Evidence suggests that flavonoid-rich foods are capable of inducing improvements in memory and cognition in animals and humans. However, there is a lack of clarity concerning whether flavonoids are the causal agents in inducing such behavioral responses. Here we show that supplementation with pure anthocyanins or pure flavanols for 6 weeks, at levels similar to that found in blueberry (2% w/w), results in an enhancement of spatial memory in 18 month old rats. Pure flavanols and pure anthocyanins were observed to induce significant improvements in spatial working memory (p = 0.002 and p = 0.006 respectively), to a similar extent to that following blueberry supplementation (p = 0.002). These behavioral changes were paralleled by increases in hippocampal brain-derived neurotrophic factor (R = 0.46, p<0.01), suggesting a common mechanism for the enhancement of memory. However, unlike protein levels of BDNF, the regional enhancement of BDNF mRNA expression in the hippocampus appeared to be predominantly enhanced by anthocyanins. Our data support the claim that flavonoids are likely causal agents in mediating the cognitive effects of flavonoid-rich foods.
