Il sistema viabile rappresenta una parte fondamentale nella dinamica dei trasporti al servizio della mobilità di trasporti e merci. La mobilità, ha fatto registrare ritmi di crescita sostenuti nel corso degli anni. Le trasformazioni sociali, del mercato del lavoro, lo sviluppo produttivo e commerciale, la posizione baricentrica tra le province jonca salentina e brindisina fanno di Manduria, una realtà dove è necessario prestare attenzione al sistema dei trasporti. Tuttavia la rete statale di vecchio stampo che risulta utilizzata, soprattutto da quote di mobilità interprovinciale per questioni di economicità del percorso e dal momento che manca una rete viaria ordinaria funzionale e scorrevole che colleghi in modo capillare il territorio fanno si che la Strada Statale 7ter risulti congestionata in maniera diffusa. La Superstrada di nuova costruzione sarebbe potuta essere un importante arteria stradale allacciandosi alla SS Taranto Brindisi Lecce collegando il sud della provincia Jonica in modo capillare ai comuni della provincia Taranto e Lecce, la quale non è stata ultimata ed avrebbe rappresentato una valida alternativa alla statale esistenti. Oltre a favorire i collegamenti tra il territorio jonico, brindisino e quello salentino, tale intervento sarebbe stato necessario per ridurre la componente di traffico di attraversamento di una serie di centri urbani, tra cui il Comune di Manduria. In questa tesi, sulla base dei dati raccolti dall’ultimo censimento nell’ambito provinciale e del Comune di Manduria, verrà implementato il funzionamento del Piano del Traffico stradale del Comune di Manduria e verranno condotte delle simulazioni, con l’ausilio del software di pianificazione di sistemi di trasporto stradale Omnitrans, versione demo, scaricabile gratuitamente dal sito www.omnitrans-international.com. Si ricercheranno stime sul funzionamento della rete, valutando la congestione del traffico ed eventualmente in termini di aumento della sicurezza e riduzione dell’inquinamento atmosferico.

Oxidative stress is considered to be of major relevance for a variety of pathological processes. Thus, it is valuable to identify compounds, which might act as antioxidants, i.e. compounds that antagonize the deleterious action of reactive oxygen species (ROS) on biomolecules. The mode of action of these compounds could be either to scavenge ROS directly or to trigger protective mechanisms inside the cell, thereby resulting in improved defense against ROS. Sulforaphane (SF) (1-isothiocyanato-(4R)-(methylsulfinyl)butane) is a naturally occurring cancer chemopreventive agent found as a precursor glucosinolate in Cruciferous vegetables like broccoli. Although SF is not a direct-acting antioxidant, there is substantial evidence that SF acts indirectly to increase the antioxidant capacity of animal cells and their abilities to cope with oxidative stress. Induction of phase 2 enzymes is one means by which SF enhances the cellular antioxidant capacity. Enzymes induced by SF include Glutathione S-transferases (GST) and NAD[P]H:quinone oxidoreductase (NQO1) which can function as protectors against oxidative stress. To protect themselves from oxidative stress, cells are equipped with reducing buffer systems including the GSH and thioredoxin (Trx) reductase. GSH is an important tripeptide thiol which in addition to being the substrate for GSTs maintains the cellular oxidation– reduction balance and protects cells against free radical species. Aim of the first part of this thesis was to investigate the ability of SF to induce the expression and the activity of different phase 2 and antioxidant enzymes (such as GST, GR, GPx, NQO1, TR, SOD, CAT) in an in vitro model of rat cardiomyocytes, and also to define if SF treatment supprts cells in counteracting oxidative stress induced by H2O2 It is well known that acute exhaustive exercise causes significant reactive oxygen species generation that results in oxidative stress, which can induce negative effects on health and well being. In fact, increased oxidative stress and biomarkers (e.g., protein carbonyls, MDA, and 8- hydroxyguanosine) as well as muscle damage biomarkers (e.g. plasmatic Creatine cinase and Lactate dehydrogenase) have been observed after supramaximal sprint exercises, exhaustive longdistance cycling or running as well as resistance-type exercises, both in trained and untrained humans. Markers of oxidative stress also increase in rodents following exhaustive exercise. Moreover, antioxidant enzyme activities and expressions of antioxidant enzymes are known to increase in response to exhaustive exercise in both animal and human tissues. Aim of this project was to evaluate the effect of SF supplementation in counteracting oxidative stress induced by physical activity through its ability to induce phase 2, and antioxidant enzymes in rat muscle. The results show that SF is a nutraceutical compound able to induce the activity of different phase 2 and antioxidant enzymes in both cardiac muscle and skeletal muscle. Thanks to its actions SF is becoming a promising molecule able to prevent cardiovascular damages induced by oxidative stress and muscle damages induced by acute exhaustive exercise.

Si analizza il modello di Fister-Panetta per la crescita di cellule tumorali nella fase avascolare del tumore e si propone un percorso didattico di introduzione alla modellistica matematica in una quinta liceo. Si riporta poi una riflessione sulle motivazioni per cui l'introduzione degli aspetti modellistici e applicativi della matematica è importante anche nella scuola superiore.

The vertical profile of aerosol in the planetary boundary layer of the Milan urban area is studied in terms of its development and chemical composition in a high-resolution modelling framework. The period of study spans a week in summer of 2007 (12-18 July), when continuous LIDAR measurements and a limited set of balloon profiles were collected in the frame of the ASI/QUITSAT project. LIDAR observations show a diurnal development of an aerosol plume that lifts early morning surface emissions to the top of the boundary layer, reaching maximum concentration around midday. Mountain breeze from Alps clean the bottom of the aerosol layer, typically leaving a residual layer at around 1500-2000 m which may survive for several days. During the last two days under analysis, a dust layer transported from Sahara reaches the upper layers of Milan area and affects the aerosol vertical distribution in the boundary layer. Simulation from the MM5/CHIMERE modelling system, carried out at 1 km horizontal resolution, qualitatively reproduced the general features of the Milan aerosol layer observed with LIDAR, including the rise and fall of the aersol plume, the residual layer in altitude and the Saharan dust event. The simulation highlighted the importance of nitrates and secondary organics in its composition. Several sensitivity tests showed that main driving factors leading to the dominance of nitrates in the plume are temperature and gas absorption process. A modelling study turn to the analysis of the vertical aerosol profiles distribution and knowledge of the characterization of the PM at a site near the city of Milan is performed using a model system composed by a meteorological model MM5 (V3-6), the mesoscale model from PSU/NCAR and a Chemical Transport Model (CTM) CHIMERE to simulate the vertical aerosol profile. LiDAR continuous observations and balloon profiles collected during two intensive campaigns in summer 2007 and in winter 2008 in the frame of the ASI/QUITSAT project have been used to perform comparisons in order to evaluate the ability of the aerosol chemistry transport model CHIMERE to simulate the aerosols dynamics and compositions in this area. The comparisons of model aerosols with measurements are carried out over a full time period between 12 July 2007 and 18 July 2007. The comparisons demonstrate the ability of the model to reproduce correctly the aerosol vertical distributions and their temporal variability. As detected by the LiDAR, the model during the period considered, predicts a diurnal development of a plume during the morning and a clearing during the afternoon, typically the plume reaches the top of the boundary layer around mid day, in this time CHIMERE produces highest concentrations in the upper levels as detected by LiDAR. The model, moreover can reproduce LiDAR observes enhancement aerosols concentrations above the boundary layer, attributing the phenomena to dust out intrusion. Another important information from the model analysis regard the composition , it predicts that a large part of the plume is composed by nitrate, in particular during 13 and 16 July 2007 , pointing to the model tendency to overestimates the nitrous component in the particular matter vertical structure . Sensitivity study carried out in this work show that there are a combination of different factor which determine the major nitrous composition of the “plume” observed and in particular humidity temperature and the absorption phenomena are the mainly candidate to explain the principal difference in composition simulated in the period object of this study , in particular , the CHIMERE model seems to be mostly sensitive to the absorption process.

Il problema affrontato nel lavoro riguarda l'allocazione della spesa tra gruppi di beni alimentari (domestici ed extra-domestici) e le modificazioni che tale allocazione ha subito nell’arco dell’ultimo decennio. L’obiettivo principale dell'analisi proposta è, quindi, di spiegare come variazioni della quota di spesa destinata alle componenti del consumo alimentare siano attribuibili a fattori strettamente economici, oltre che alle caratteristiche struttura socio-demografiche dei consumatori. Allo scopo di valutare l’allocazione inter-temporale della spesa individuale viene proposto come schema di analisi il sistema di domanda Almost Ideal di Deaton e Muellbauer (AIDS).

The thesis is framed within the field of the stochastic approach to flow and transport themes of solutes in natural porous materials. The methodology used to characterise the uncertainty associated with the modular predictions is completely general and can be reproduced in various contexts. The theme of the research includes the following among its main objectives: (a) the development of a Global Sensitivity Analysis on contaminant transport models in the subsoil to research the effects of the uncertainty of the most important parameters; (b) the application of advanced techniques, such as Polynomial Chaos Expansion (PCE), for obtaining surrogate models starting from those which conduct traditionally developed analyses in the context of Monte Carlo simulations, characterised by an often not negligible computational burden; (c) the analyses and the understanding of the key processes at the basis of the transport of solutes in natural porous materials using the aforementioned technical and analysis resources. In the complete picture, the thesis looks at the application of a Continuous Injection transport model of contaminants, of the PCE technique which has already been developed and applied by the thesis supervisors, by way of numerical code, to a Slug Injection model. The methodology was applied to the aforementioned model with original contribution deriving from surrogate models with various degrees of approximation and developing a Global Sensitivity Analysis aimed at the determination of Sobol’ indices.
