The chemical investigation of the hexane and ethanol extracts from aerial parts of Vernonia chalybaea conducted to the isolation and characterization of a new aliphatic tetrahydroxyl ether, along with a series of known compounds such as 4 α,10 α-epoxyaromadendrane, friedelin, taraxasteryl acetate, pseudotaraxasteryl acetate, lupeyl acetate, lupeol, α-amiryn, β-amiryn, β-sitosterol, stigmasterol, 2,3-dimethyl-1,2,3-tri-hydroxybuthanol, angophorol, angophorol-7-O-glucoside, angophorol-7-O-rutinoside, 3,7-dimethoxy-5,3',4'-trihydroxyflavone and acacetin. The structures of all compounds were determined by spectroscopic analysis and comparison with published spectral data.

The chemical composition of the essential oils, obtained by hidrodistillation, from leaves of two Vernonia species (V. remotiflora and V. brasiliana), was determined by GC-FID and GC-MS. Both essential oils were predominantly constituted by sesquiterpenes (92.0 - 93.4%). The main constituents of the oil from V. brasilana were (E)-caryophyllene (36.7%), germacrene D (35.5%), and α-humulene (11.7%), while (E)-caryophyllene (42.2%) and bicyclogermacrene (20.0%) were the major ones in the oil from V. remotiflora. Although with moderate activity both oils showed acetylcholinesterase and antibacterial activities.

The chemical investigation of hexane and ethanol extracts from the aerial parts of Vernonia scorpioides resulted in the isolation and characterization of a new polyacetylene lactone, rel-4-dihydro-4β-hydroxy-5a-octa-2,4,6-triynyl-furan-2-(5H)-one, along with the new ethyl 3,4-dihydroxy-6,8,10-triynyl-dodecanoate, and seven known compounds: taraxasteryl acetate, lupeyl acetate, lupeol, lupenone, β-sitosterol, stigmasterol and luteolin. The structure of all compounds was determined by spectrometric techniques (HR-ESI-MS, ¹H and 13C NMR and IV) and comparison with published spectral data.

Descrevem-se surtos de intoxicação por Vernonia rubricaulis em bovinos em nove fazendas no Estado de Mato Grosso do Sul, entre setembro de 1999 e maio de 2001. No total morreram pelo menos 954 animais. Alguns fatores foram aparentemente importantes para a ocorrência da intoxicação: a utilização de técnicas que favorecem a brotação da planta incluindo a utilização da roçadeira ("trilho") para o controle de plantas invasoras, assim como a queima ou o desmatamento do campo; o transporte de animais procedentes de outras fazendas ou de outros piquetes da mesma fazenda, com ou sem manejo de animais no curral; e a superlotação de piquetes. O curso clínico variou de 12 a 48 horas. Os sinais clínicos foram, principalmente, agressividade ou depressão, salivação, cegueira, gemidos, incoordenação, retração de abdômen, fezes ressecadas com muco e estrias de sangue, focinho seco e decúbito. A letalidade foi de 100%. Os principais achados de necropsia foram observados no fígado. As lesões hepáticas variavam num mesmo animal e entre diferentes animais. Áreas focais vermelhas de hemorragias com tamanhos variando entre 1mm e 25cm, foram observadas em meio ao tecido hepático amarelado em alguns casos. Fígados inteiros ou áreas hepáticas com aspecto de noz-moscada e fígados difusamente amarelados com ou sem hemorragias petequiais, também foram observados. Ocorriam, também, hemorragias em diferentes tecidos e órgãos. As principais alterações histológicas caracterizaram-se por necrose centrolobular com vacuolização dos hepatócitos das áreas periportais e intermediárias ou necrose massiva e vacuolização difusa dos hepatócitos. A intoxicação foi reproduzida experimentalmente em três bovinos. A menor dose que causou morte foi de 3g/kg da planta verde fresca em fase de brotação. Os sinais clínicos e as lesões macro e microscópicas observadas foram semelhantes às apresentadas pelos animais intoxicados naturalmente.

A compreensão sobre a biologia de plantas daninhas pode contribuir significativamente no estabelecimento de estratégias adequadas para seu manejo, além de possibilitar o desenvolvimento de ferramentas de controle não químico. O objetivo deste trabalho foi estudar o efeito de profundidade de semeadura, tipo de solo, tipo e quantidade de palha na emergência de plântulas de Vernonia ferruginea (assa-peixe). No primeiro experimento, sementes da espécie foram colocadas em dois substratos (terra e areia) e em sete profundidades (0; 0,0025; 0,005; 0,01; 0,02; 0,04; e 0,08 m). No segundo experimento, determinou-se o efeito de palha de cana-de-açúcar, capim-braquiária e milho em cinco quantidades (0,0; 1,5; 3,0; 6,0; e 9,0 t ha-1) na emergência de V. ferruginea. Maior emergência foi obtida na superfície e em 0,0025 m em ambos os substratos. A palha de milho foi a que mais inibiu a emergência de V. ferruginea, em todas as quantidades testadas.

Vernonia scorpioides has been widely used in Brazil to treat skin problems and chronic wounds, such as ulcers of the lower limbs and diabetic lesions. In the present study, we investigated the effect of a dichloromethane (DCM) fraction of V. scorpioides leaf extract on Ehrlich ascitic and solid tumor-bearing mice. The animals were treated once a day with the DCM fraction at a concentration of 5 mg/kg, administered ip during and after the development of the tumor. The lifespan, weight, number and type of leukocytes, number of tumor cells, volume of solid and ascitic tumors were measured. The development of the tumor with pre-treated tumor cells in vitro with the DCM fraction (5 mg/kg) was analyzed and the animals were sacrificed after 7 days. The DCM fraction (5 mg/kg) totally inhibited tumor development when in direct contact with tumor cells, and also ascitic tumor development with in vitro treatment or when administered ip, in loco (after 7 days). Animals treated with the DCM fraction increased their lifespan ca. 2 weeks and maintained their body weight for 30 days. When applied immediately after the inoculation of the tumor cells in vivo, it totally abolished tumor development, with tumor development only decreasing when treatment was started 3 days after the tumor challenge. These data suggest an antineoplastic activity of the fraction. Oral or ip administration of DCM fraction (5 mg/kg) for 7 days did not reduce the solid tumor volume. The cytotoxic activity described here differs from the conventional immune suppressing profile of standard chemotherapy because it increases neutrophil influx to the peritoneal cavity. These results show that, besides exhibiting a tumoricidal activity, the DCM fraction also exhibits inflammatory activity.

The volatile compounds were isolated from the headspace fraction of "assa-peixe" honeys by adsorptive column chromatography, eluted with acetone and analysed by GC-FID and GC-MS. Volatile compounds were separated using a polar phase column. Low- and medium-boiling point volatile compounds predominated in the headspace. A large proportion of 3-penten-2-one (80.5 ± 13.9 µg.kg-1) and benzaldehyde (25.9 ± 4.2 µg.kg-1) was found in the headspace fraction, while 2-penten-1-ol, 3-hexenyl butanoate, octadecane and hexanoic acid (<0.01 µg.kg-1) were by far the less abundant volatile compounds. A total of 12 volatile compounds were identified and, among them, 5 compounds were reported as "assa-peixe" honey constituents for the first time. Of the 5 new volatile compounds reported, 3-penten-2-one, dodecane, tridecane and benzaldehyde were definitively identified in the headspace fraction of Brazilian "assa-peixe" honeys.

Methanolic (VPME) and chloroformic (VPCL) extracts, obtained from the aerial parts of Vernonia polyanthes, were investigated for its antiulcerogenic properties. Administration of VPME (250 mg/kg) and VPCL (50 mg/kg) significantly inhibited the gastric mucosa damage (64% and 90%, respectively) caused by absolute ethanol (p.o.). Otherwise, in NSAID-induced gastric damage, their gastroprotective effects have decreased. Since the VPCL extract resulted to be more effective than the VPME we focused our efforts over VPCL action mechanism of action. © 2007 Elsevier B.V. All rights reserved.

Background: Cholera, a severe acute watery diarrhea caused by Vibrio cholerae is endemic in Nigeria with most cases occurring in the rural areas. In South West Nigeria, some individuals resort to alternative treatments such as Ogi-tutu, Psidium guajava and Vernonia amygdalina during infections. The effectiveness of these alternatives in the prevention and treatment of V. cholerae infection requires experimental investigation. Objective: This study was designed to investigate the ameliorative effects of Ogi-tutu, Vernonia amygdalina and Psidium guajava on intestinal histopathology of experimental mice infected with V. cholerae. Methods: Preliminary investigation of in vitro vibriocidal activities of these alternatives were carried out using agar cup diffusion assay. For ameliorative effects, adult mice were inoculated with 100 μl (106 cells) of Vibrio cholerae and dosed at 0 h (immediate prevention) and 4 h (treatment of infection) and their intestines were histopathologically evaluated. Results: The histopathological changes were the same irrespective of the treated groups, but the lesions varied in extent and severity. The ameliorative effects in decreasing order were V. amygdalina > P. guajava > Ogi-tutu. Conclusion: V. amygdalina gave the best ameliorative effects in the prevention and treatment of V. cholerae infection.
