Analysis of the reactivity of IgM with self-antigens in tissues by a quantitative immunoblotting technique showed striking invariance among newborns in the human and in the mouse. The self-reactive repertoire of IgM of adults was also markedly conserved; it comprised most anti-self reactivities that prevailed among neonates. Multivariate analysis confirmed the homogeneity of IgM repertoires of neonates toward self- and non-self-antigens. Multivariate analysis discriminated between newborn and adult repertoires for reactivity with two of five sources of self-proteins and with non-self-antigens. Our observations support the concept that naturally activated B lymphocytes are selected early in development and throughout life for reactivity with a restricted set of self-antigens.

Esta pesquisa apresenta estudo de caso cujo objetivo foi analisar a aceitação do Portal Inovação, identificando os fatores preditivos da intenção comportamental de uso e do comportamento de uso direcionadores da adoção da tecnologia por seus usuários via extensão do Modelo Unificado de Aceitação de Tecnologia, denominado pela sigla UTAUT (Unified Theory of Acceptance and Use of Technololgy) de Venkatesh et al. (2003). O objeto da pesquisa o Portal Inovação foi desenvolvido pelo Ministério da Ciência, Tecnologia e Inovação (MCTI) em parceria com o Centro de Gestão e Estudos Estratégicos (CGEE), Associação Brasileira de Desenvolvimento Industrial (ABDI) e Instituto Stela, visando atender às demandas do Sistema Nacional de Ciência, Tecnologia e Inovação (SNCTI) do País. Para atingir os objetivos propostos, recorreu-se às abordagens qualitativa, que foi subsidiada pelo método estudo de caso (YIN, 2005) e quantitativa, apoiada pela metodologia UTAUT, aplicada a usuários do portal e que contemplou o resultado de 264 respondentes validados. Quanto ao material de análise, utilizou-se da pesquisa bibliográfica sobre governo eletrônico (e-Gov), Internet, Sistema Nacional de Inovação, modelos de aceitação de tecnologia, dados oficiais públicos e legislações atinentes ao setor de inovação tecnológica. A técnica de análise empregada quantitativamente consistiu no uso de modelagem por equações estruturais, com base no algoritmo PLS (Partial Least Square) com bootstrap de 1.000 reamostragens. Os principais resultados obtidos demonstraram alta magnitude e significância preditiva sobre a Intenção Comportamental de Uso do Portal pelos fatores: Expectativa de Desempenho e Influência Social. Além de evidenciarem que as condições facilitadoras impactam significativamente sobre o Comportamento de Uso dos usuários. A conclusão principal do presente estudo é a de que ao considerarmos a aceitação de um portal governamental em que a adoção é voluntária, o fator social é altamente influente na intenção de uso da tecnologia, bem como os aspectos relacionados à produtividade consequente do usuário e o senso de utilidade; além da facilidade de interação e domínio da ferramenta. Tais constatações ensejam em novas perspectivas de pesquisa e estudos no âmbito das ações de e-Gov, bem como no direcionamento adequado do planejamento, monitoramento e avaliação de projetos governamentais.

Implantada no Brasil na década de 50, a TV a mídia de maior abrangência e poder ideológico entre seus públicos encara sua segunda grande transformação: ela deixa de ser analógica e passa a ser digital. Com isso, traz à tona novas possibilidades de recepção e a possível convergência de meios. Nesse contexto, o objetivo deste estudo foi analisar o processo de instalação do Sistema Brasileiro de Televisão Digital Terrestre (SBTVD-t) a partir do clipping on-line do Fórum Nacional pela Democratização da Comunicação (FNDC). A análise desse material teve como foco averiguar se o FNDC foi tendencioso ou não na veiculação de matérias voltadas aos aspectos técnicos da nova tecnologia, em detrimento de seu potencial social. Para tanto, optou-se por uma pesquisa de base quantitativa em que as informações e os dados coletados levaram à constatação de que o FNDC se mostra pouco eficaz como aparato crítico-apreciativo da grande mídia, além de não cumprir alguns de seus objetivos ao reproduzir discursos e ideologias de outros veículos. Da mesma forma, verificou-se ainda que o Governo Federal também fugiu aos objetivos listados nos decretos presidenciais que instituem e dispõem sobre a implantação do SBTVD.(AU)

We have developed a digital holographic microscope (DHM), in a transmission mode, especially dedicated to the quantitative visualization of phase objects such as living cells. The method is based on an original numerical algorithm presented in detail elsewhere [Cuche et al., Appl. Opt. 38, 6994 (1999)]. DHM images of living cells in culture are shown for what is to our knowledge the first time. They represent the distribution of the optical path length over the cell, which has been measured with subwavelength accuracy. These DHM images are compared with those obtained by use of the widely used phase contrast and Nomarski differential interference contrast techniques.

This paper describes the importance of an innovative analytical technique for drugs and pharmaceuticals quantification, using Fouriertransform infrared (FTIR) transmission spectroscopy. This method does not use organic solvents, which is one great advantage over the most common analytical methods. This fact contributes to minimize the generation of organic solvent waste by the industry and thereby reduces the impact of its activities on the environment. The method involved absorbance measurements of the band corresponding to one of the chosen group in the molecule. Obviously, the method should be validated according to ICH guidelines, showing linearity, precision, accuracy and robustness, over a concentration range, using small amounts to prepare the analyte. The validated method is able to quantify drugs and pharmaceuticals and can be used as an environmentally friendly alternative for the routine analysis in pharmaceutical industry quality control.

This article contributes to an ongoing debate about how to measure sensitive topics in population surveys. We propose a novel technique that can be applied to the measurement of quantitative sensitive variables: the item sum technique (IST). This method is closely related to the item count technique, which was developed for the measurement of dichotomous sensitive items. First, we provide a description of our new technique and discuss how data collected by the IST can be analyzed. Second, we present the results of a CATI survey on undeclared work in Germany, in which the IST has been applied. Using an experimental design, we compare the IST to direct questioning. Our empirical results indicate that the IST is a promising data-collection technique for sensitive questions. We conclude by discussing the limitations of the new technique and outlining possible improvements for future studies.

A new surface analysis technique has been developed which has a number of benefits compared to conventional Low Energy Ion Scattering Spectrometry (LEISS). A major potential advantage arising from the absence of charge exchange complications is the possibility of quantification. The instrumentation that has been developed also offers the possibility of unique studies concerning the interaction between low energy ions and atoms and solid surfaces. From these studies it may also be possible, in principle, to generate sensitivity factors to quantify LEISS data. The instrumentation, which is referred to as a Time-of-Flight Fast Atom Scattering Spectrometer has been developed to investigate these conjecture in practice. The development, involved a number of modifications to an existing instrument, and allowed samples to be bombarded with a monoenergetic pulsed beam of either atoms or ions, and provided the capability to analyse the spectra of scattered atoms and ions separately. Further to this a system was designed and constructed to allow incident, exit and azimuthal angles of the particle beam to be varied independently. The key development was that of a pulsed, and mass filtered atom source; which was developed by a cyclic process of design, modelling and experimentation. Although it was possible to demonstrate the unique capabilities of the instrument, problems relating to surface contamination prevented the measurement of the neutralisation probabilities. However, these problems appear to be technical rather than scientific in nature, and could be readily resolved given the appropriate resources. Experimental spectra obtained from a number of samples demonstrate some fundamental differences between the scattered ion and neutral spectra. For practical non-ordered surfaces the ToF spectra are more complex than their LEISS counterparts. This is particularly true for helium scattering where it appears, in the absence of detailed computer simulation, that quantitative analysis is limited to ordered surfaces. Despite this limitation the ToFFASS instrument opens the way for quantitative analysis of the 'true' surface region to a wider range of surface materials.

The aim of this work is to provide a quantitative method for analysis of the concentration of superparamagnetic iron oxide nanoparticles (SPION), determined by means of ferromagnetic resonance (FMR), with the nanoparticles coupled to a specific antibody (AC133), and thus to express the antigenic labeling evidence for the stem cells C D133(+). The FMR efficiency and sensitivity were proven adequate for detecting and quantifying the low amounts of iron content in the C D133(+) cells (similar to 6.16 x 10(5) pg in the volume of 2 mu l containing 4.5 x 1011 SPION). The quantitative method led to the result of 1.70 x 10(-13) mol of Fe (9.5 pg), or 7.0 x 10(6) nanoparticles per cell. For the quantification analysis via the FMR technique it was necessary to carry out a preliminary quantitative visualization of iron oxide-labeled cells in order to ensure that the nanoparticles coupled to the antibodies are indeed tied to the antigen at the stem cell surface and that the cellular morphology was conserved, as proof of the validity of this method. The quantitative analysis by means of FMR is necessary for determining the signal intensity for the study of molecular imaging by means of magnetic resonance imaging (MRI).

Evaluate the distribution and variation of placental vascular indices according to gestational age and placental volume. From March to November 2007, three-dimensional (3D)-power Doppler ultrasound was performed in 295 normal pregnancies from 12 to 40 weeks of gestation. Using the same preestablished settings for all patients, power Doppler was applied to the placenta and placental Volume was obtained by the rotational technique (VOCAL(TM)). The 3D-power histogram was used to determine the placental vascular indices: vascularization index (VI), flow index (FI) and vascularization-flow index (VFI). The placental vascular indices were then plotted against gestational age and placental volume. All placental vascular indices showed constant distribution throughout gestation. A tendency for a reduction in placental vascular indices with increased placental volume was observed, but was only statistically significant when placental FI was considered (p < 0.05). All placental vascular indices estimated by 3D-power Doppler ultrasonography presented constant distribution throughout gestation, despite the increase in placental volume according to gestational age. Background: Although the influence of respiration on ventricular filling, as evaluated by Doppler technique, and the evaluation of diastolic velocities of mitral valve annulus (MVA), as measured by Doppler tissue imaging (DTI), can provide valuable information for the study of left ventricular (LV) diastolic function, the concomitant effects of aging, tidal volume (TV), and respiratory rate (RR) on these velocities have not been quantitatively investigated. Methods: We evaluated 12 normal male volunteers (Group I) aged 20-26 years (mean: 22.8) and 8 normal subjects aged 41 to 54 years old (mean: 45.9) (Group II). Using DTI we measured peak early (E-a) and late (A(a)) velocities of longitudinal axis expansion at lateral and medial MVA. Doppler mitral and tricuspid flow velocities were measured: peak early (E) and late (A) inflow velocity, early (E-i) and late (A(i)) flow integral, and deceleration time of peak early mitral flow velocity (DT). Respiratory cycles were simultaneously recorded at RR of 9, 12, 15, and 18 cycles/min and TV of 600 and 900 mL during respiration (RESP). Results and conclusions: (1) E, A, and A(i) in MV had negligible change during respiration, but E-i was significantly reduced during inspiration; (2) DT reduced slightly with inspiration, but the change was significant only with TV of 900 mL; (3) an important increase of E in right ventricular flow was observed during inspiration; (4) variations of RR and TV did not significantly influence right and left ventricular inflow in normal subjects, in the conditions of this investigation; (5) a significant increase of E-a at medial MVA was documented during inspiration only in young subjects; (6) a significant decrease of A(a) at medial MVA was observed during inspiration in both groups of volunteers; (7) RR and TV did not influence MVA velocities in young and adult subjects; (8) a consistent reduction in E-a and a significant increase in A(a) were observed with increasing age; (9) these changes were more conspicuous and consistent than those documented in ventricular filling when young and middle-age men are compared, suggesting that the DTI is more sensitive to detect changes in diastolic function; and (10) in addition, these data suggest that, for evaluation of diastolic function, in clinical context, it is not necessary to control rigorously RR or TV.
