This work was focused to study the immobilization of enzymes on polymers. A large range of polymer matrices have been employed as supports for enzyme immobilization. Here polyaniline (PAN!) and poly(0~toluidine) (POT) were used as supports. PANI and POT provides an excellent support for enzyme immobilization by virtue of its facile synthesis, superior chemical and physical stabilities, and large retention capacity. We selected industrially important starch hydrolyzing enzymes a-amylase and glucoamylase for the study. In this work the selected enzymes were immobilized via adsorption and covalent bonding methods.To optimize the catalytic efficiency and stability of the resulting biocatalysts, the attempt was made to understand the immobilization effects on enzymatic properties. The effect of pH of the immobilization medium, time of immobilization on the immobilization efficiency was observed. The starch hydrolyzing activity of free 0:-amylase and glucoamylase were compared with immobilized forms. Immobilization on solid supports changes the microenvironment of the enzyme there by influences the pH and temperature relationship on the enzymatic activity. Hence these parameters also optimized. The reusability and storage stability of immobilized enzymes an important aspect from an application standpoint, especially in industrial applications. Taking in to consideration of this, the reusability and the long tenn storage stability of the immobilized enzyme investigated.

In the present paper, we studied the preparation of biomimetic triblock copolymer (ABA) membranes in aqueous solution and their deposition into solid supports. The self-assembly structures of the ABA in aqueous solution was investigated by using optical microscopy, dynamic light scattering, electron microscopy (EM) and SAXS. Spherical and tubular polymersomes were found at the highest concentrations investigated. The mechanism of deposition on solid supports (mica and glass) was elucidated by using atomic force microscopy (AFM). The deposition results in the formation of a uniform defect-free membrane at suitable polymer concentrations.

Poly(vinyl ether) gels SLURPS (Superior Liquid Uptake Resin for Polymer-supported synthesis) with low cross-linking levels have been synthesized for the first time in beaded form using a non-aqueous inverse suspension polymerisation approach. The synthetic protocol was optimized with regards to several parameters including reactions conditions, type and concentration of suspension stabilizer and controlled low temperature addition of co-initiator. Particle size measurements confirm the production of beads with average diameters of 700e950 mm. Optimization of the monomer composition of the poly (vinyl ether) gels resulted in a novel beaded polymer support with considerably improved as well as unique swelling characteristics in solvents ranging from hexane to water. The synthetic utility of the new gel was confirmed by carrying out a set of transformations with complete conversion leading to a useful amino and hydroxy terminated solid-phase precursor resin. Reaction progress could be monitored easily by 1H and 13C gel-phase NMR.

Solid polymer electrolytes based on amorphous polyether-urethane networks combined with lithium or sodium salts and a low molecular weight cosolvent (plasticizer) have been investigated in our laboratories for several years. Conductivity enhancements of up to two orders of magnitude can be obtained whilst still retaining solid elastomeric properties. In order to understand the effects of the plasticizers and their mechanism of conductivity enhancement, multinuclear NMR has been employed to investigate ionic structure in polymer electrolyte systems containing NaCF3SO3, LiCF3SO3 and LiClO3 salts.

With increasing dimethyl formamide (DMF) and propylene carbonate (PC) concentration the increasing cation chemical shift with fixed salt concentration indicates a decreasing anion-cation association consistent with an increased number of charge carriers. 13C chemical shift data for the same systems suggests that whilst DMF also decreases cation-polymer interactions, PC does the opposite, presumably by shielding cation-anion interactions. Temperature dependent 7Li spin-lattice relaxation times indicate the expected increase in ionic mobility upon plasticization with a shift of the T1 minimum to lower temperatures. The magnitude of T1 at the minimum increases upon addition of DMF whereas there is a slight decrease when PC is added. This also supports the suggestion that the DMF preferentially solvates the cation whereas the action of PC is limited to coulomb screening, hence freeing the anion.

An increase of the reports involving mimetic systems has been observed. Briefly, these systems use biological phospholipids to exploit specific interactions between membrane-models and drugs. Here, the Layer-by-Layer (LbL) and Langmuir techniques were used to investigate the interaction between cardiolipin (CLP-negative phospholipid) and a cationic-like drug methylene blue (MB). Supported by a cationic polyelectrolyte (PAH), LbL films containing PAH/(CLP + MB) and PAH/(CLP + MB + AgNP) were grown up to 14 bilayers. The optical microscopy analysis revealed a decrease of the CLP vesicle sizes in the presence of MB as a possible consequence of the MB action onto the mechanical properties of the CLP membrane. From FTIR spectra, changes mainly related to peak position and band intensity and shape were observed in the spectra from PAH/CLP when in the presence of MB. The latter supports that the interactions between the phosphate and amine charged groups from CLP and PAH, respectively, established during the LbL film fabrication, besides the CLP hydrocarbon environment, are influenced by the presence of MB. Using the micro-Raman technique, a chemical mapping was build based on MB spectrum by resonance Raman scattering (RRS) and surface-enhanced resonance Raman scattering (SERRS). The later phenomenon was activated by Ag nanoparticles (AgNPs) trapped within the LbL film allowing collecting spectra for a single bilayer of PAH/(CLP + MB + AgNP). A rough estimation showed a SERRS amplification of 10(3) in comparison to RRS spectra. As a complementary approach, Langmuir films of CLP in the presence of co-spread MB were investigated through surface pressure vs mean molecular area (pi-A) isotherms. The results showed that for concentrations of MB below 100 mol%, the drug is expelled to water subphase for high values of surface pressure (condensed phase). Das Ziel der vorgelegten Arbeit war die Synthese von definierten, sphärischen Polystyrolpartikeln im Größenbreichen von Nanometern, die als Träger für die Immobilisierung von Metallocenkatalysatoren verwendet werden sollten. Ein wichtiger Anspruch an das System war dabei die Möglichkeit einer homogene Verteilung des Metallocenes auf dem Träger and eine homogene Fragmentierung des geträgerten Katalysators während der Polymerisation im Polymerprodukt. Für diese Zielsetzung wurden unterschiedliche Polystyrolnanopartikel hergestellt. Die Polystyrolnanopartikel waren mit unterschiedlichen funktionellen Gruppen wie Polyethylenoxid- und Polypropylenoxidketten oder Hydroxygruppen auf der Oberfläche versehen, um den Metallocenkatalysator und den Cokatalysator MAO immobilisieren zu können. In verschiedenen Experimenten wurde der Einfluss dieser Polystyrolnanopartikel als Träger auf die Katalysatoreigenschaften wie Aktivität oder Produktivität und die Eigenschaften des produzierten Polyolefins wie z.B. Molekulargewicht und Morphologie untersucht. Im Vergleich zu den PS- Nanopartikeln wurden außerdem PS-Mikropartikel, Silica und Dendrimere als Träger in der heterogenen Olefinpolymerisation eingesetzt. Von all diesen Trägersystemen wurde das Fragmentierungsverhalten durch konfocale Fluoreszenzmikroskopie untersucht. Aus den erhaltenen Ergebnissen kann geschlossen werden, dass die hergestellten Polystyrolnanopartikel neuartige und leistungsfähige Träger für heterogene Polymerisationsprozesse darstellen. Die hergestellten Polystyrolnanopartikel besaßen eine wohldefinierte sphärische Struktur, die eine homogene Verteilung des immobilisierten Metallocenkatalysators und somit auch eine vollständige Fragmentierung des geträgerten Katalysators im hergestellten Polyolefin ermöglichte. Die Katalysatorsysteme, die aus den PS- Nanopartikeln und dem Metallocenkatalysator zusammengesetzt waren, wurden in verschiedenen Polymerisationen wie der Ethylen- oder Propylenhomopolymersation und der Copolymerisation von Ethen mit α- Olefinen getestet. Die Oberflächen- funktionalisierten PS Nanopartikel immobilisierten den Metallocenkatalysator ausreichend gut, so dass kein „Leachen“ (Ablösen) des Katalysators von der Trägeroberfläche festgestellt werden konnte und deshalb Polymer von sehr guter Morphologie erhalten wurde. Um die Fragmentierung des Katalysators und den inneren Aufbau des Polymers näher untersuchen zu können, wurde die konfocale Fluoreszenzmikroskopie für das PS- Nanopartikelträgersystem angewendet. Durch farbstoffmarkierte Trägerpartikel konnte die Verteilung des fragmentierten Katalysators innerhalb des Polymers sichtbar gemacht und analysiert werden. Dabei wurde festgestellt, dass sich PS- Nanopartikel und auch Dendrimere als Träger ähnlich verhalten wie Ziegler- Natta- Katalysatoren, die auf MgCl2 immobilisiert für die heterogene Olefinpolymerisation verwendet werden. Das Fragmentierungsverhalten der Silica oder PS- Mirkopartikel geträgerten Systeme entsprach dagegen dem schichtweisen Fragmentierungsverhalten wie es bereits von Fink und Mitarbeitern beschrieben wurde.

Core-shell macromolecules with dendritic polyphenylene core and polymer shell Zusammenfassung / Abstract Core-shell macromolecular structures have become of great interest in materials science because they gave an opportunity to combine a large variety of chemical and physical properties in the single molecule, by combination of different (in terms of chemistry and physics) cores and shells. The interest in such complex structures was provoked by their potential applications in the coating and painting industry (latexes), as supports for catalysts in polymer industry, or as nano-containers and transporters for genes or drug delivery. The aim of this study was the synthesis, characterization and further application of core-shell macromolecules possessing a hydrophobic stiff core (polyphenylene dendrimers) surrounded with a hydrophilic, soft, covalently bonded polymer shell (poly(ethylene oxide) and its copolymers). The requirements for such complex substances were that they should be well-defined in terms of molecular weight (narrow molecular weight distribution) and in molecular structure. The preparation of core-shell molecules containing dendrimer as a core was possible via two synthetic routs: “grafting-onto” and “grafting-from”. The resulting core-shell macromolecules possessed narrow polydispersity as guaranteed by the excellent structural and functional definition of the dendrimer and the narrow polydispersity of the PEO, PS-b-PEO and PI-b-PEO attached to the dendrimer surface. Additional investigation of the size of the particles indicated a relation between both the length and the number of the polymer chains and the hydrodynamic radius determined by Dynamic Light Scattering and Fluorescent Correlation Spectroscopy. Core-shell nano-particles were applied as metallocene supports in heterogeneous olefin polymerizations. Our results indicate that such catalyst systems, that have a size of at least one order of magnitude smaller than the used by now organic supports, could be very useful as model compounds for investigations on catalyst fragmentation and its influence on the product parameters.

Studies have demonstrated that polymeric biomaterials have the potential to support osteoblast growth and development for bone tissue repair. Poly( beta- hydroxybutyrate- co- beta- hydroxyvalerate) ( PHBV), a bioabsorbable, biocompatible polyhydroxy acid polymer, is an excellent candidate that, as yet, has not been extensively investigated for this purpose. As such, we examined the attachment characteristics, self- renewal capacity, and osteogenic potential of osteoblast- like cells ( MC3T3- E1 S14) when cultured on PHBV films compared with tissue culture polystyrene ( TCP). Cells were assayed over 2 weeks and examined for changes in morphology, attachment, number and proliferation status, alkaline phosphatase ( ALP) activity, calcium accumulation, nodule formation, and the expression of osteogenic genes. We found that these spindle- shaped MC3T3- E1 S14 cells made cell - cell and cell - substrate contact. Time- dependent cell attachment was shown to be accelerated on PHBV compared with collagen and laminin, but delayed compared with TCP and fibronectin. Cell number and the expression of ALP, osteopontin, and pro- collagen alpha 1( I) mRNA were comparable for cells grown on PHBV and TCP, with all these markers increasing over time. This demonstrates the ability of PHBV to support osteoblast cell function. However, a lag was observed for cells on PHBV in comparison with those on TCP for proliferation, ALP activity, and cbfa- 1 mRNA expression. In addition, we observed a reduction in total calcium accumulation, nodule formation, and osteocalcin mRNA expression. It is possible that this cellular response is a consequence of the contrasting surface properties of PHBV and TCP. The PHBV substrate used was rougher and more hydrophobic than TCP. Although further substrate analysis is required, we conclude that this polymer is a suitable candidate for the continued development as a biomaterial for bone tissue engineering.

Fiber Bragg grating sensors recorded in poly(methyl methacrylate) fiber often exhibit hysteresis in the response of Bragg wavelength to strain, particularly when exposed to high levels of strain. We show that, when such a fiber grating sensor is bonded directly to a substrate, the hysteresis is reduced by more than 12 times, compared to the case where the sensor is suspended freely between two supports. © 2013 Optical Society of America.

When exposed to high levels of strain, polymer optical fibre grating sensors recorded in poly(methyl methacrylate) based fibre often exhibit hysteresis in the response of their Bragg wavelength to strain. We demonstrate that the application of pre-tension and annealing of the polymer fibre can reduce this hysteresis when the fibre is suspended freely between two supports, but much better performance is obtained when the sensor is attached directly to a substrate. In this case, the hysteresis can be lessened by more than a factor of 12. © 2014 IOP Publishing Ltd.

Porous 3D polymer scaffolds prepared by TIPS from PLGA (53:47) and PS are intrinsically hydrophobic which prohibits the wetting of such porous media by water. This limits the application of these materials for the fabrication of scaffolds as supports for cell adhesion/spreading. Here we demonstrate that the interior surfaces of polymer scaffolds can be effectively modified using atmospheric air plasma (AP). Polymer films (2D) were also modified as control. The surface properties of wet 2D and 3D scaffolds were characterised using zeta-potential and wettability measurements. These techniques were used as the primary screening methods to assess surface chemistry and the wettability of wet polymer constructs prior and after the surface treatment. The surfaces of the original polymers are rather hydrophobic as highlighted but contain acidic functional groups. Increased exposure to AP improved the water wetting of the treated surfaces because of the formation of a variety of oxygen and nitrogen containing functions. The morphology and pore structure was assessed using SEM and a liquid displacement test. The PLGA and PS foam samples have central regions which are open porous interconnected networks with maximum pore diameters of 49 μm for PLGA and 73 μm for PS foams.
