Describen las escalas de madurez gonadal macroscópicas, validadas mediante análisis microscópicos, de diez especies de peces: Engraulis ringens anchoveta peruana, Merluccius gayi peruanus merluza, Sarda chiliensis chiliensis bonito, Scomber japonicus peruanus caballa, Anchoa nasus anchoveta blanca, Paralabrax humeralis cabrilla, Paralichthys adspersus lenguado, Cynoscion analis cachema, Hippoglossina macrops lenguado de ojo grande y Vinciguerria lucetia. Todas las escalas tienen seis estadios de madurez para hembras y machos: 0 (virginal), I (reposo), II (en maduración), III (maduro), IV (desovante/expulsante), V (recuperación/post expulsante). Se describen características y criterios claros para diferenciación entre estadios de madurez por especie y se discute la importancia de la validación y sus múltiples aplicaciones

Taking into account the difficulties of taxonomic identification of larval anisakid nematodes based on morphological characters, genetic analyses were performed, together with those usually applied, in order to identify anisakid larvae found in the flounder Paralichthys isosceles from the littoral of the state of Rio de Janeiro, Brazil. The analysis of 1,820 larvae revealed a new species, similar to Hysterothylacium MD, Hysterothylacium 2, Hysterothylacium KB and Hysterothylacium sp regarding the absence of the larval tooth, an excretory pore situated below the nerve ring level, and slender lateral alae. Moreover, the new species differs from Hysterothylacium fortalezae and Hysterothylacium reliquens with regard to the number and size of spines present on the tail end and from Hysterothylacium patagonicus by the absence of interlabia. The maximum parsimony and neighbour joining tree topologies based on the 18S ribosomal DNA gene, complete internal transcribed spacer region and cytochrome oxidase 2 (COII) gene demonstrated that the Brazilian larvae belong to Raphidascarididae and represent a unique genetic entity, confirmed as a new Hysterothylacium species. Furthermore, the new species presents COII genetic signatures and shares polymorphisms with Raphidascarididae members. This is the first description of a new anisakid species from Brazil through the integration of morphological and molecular taxonomy data.

As micobacterioses afetam várias espécies de peixes, tanto comerciais como ornamentais. É uma enfermidade sistêmica com formação de granulomas, conhecida como micobacteriose de peixes. O desenvolvimento da aquicultura tem aumentado o número de casos com essa enfermidade que muitas vezes é de difícil diagnóstico. Neste trabalho apresentamos dois casos de micobacterioses em Paralichthys orbignyanus e Elacatinus figaro provenientes do Laboratório de Piscicultura Estuarina e Marinha da Universidade Federal do Rio Grande (FURG). As micobactérias de peixes possuem menos ácidos graxos que as de mamíferos, por isso foi utilizado o protocolo de coloração de Fite-Faraco, uma vez que o protocolo de coloração clássico de Ziehl Neelsen pode resultar em falsos negativos. Para determinar o tipo de micobactéria foi utilizada a imuno-histoquímica com a qual se pode diagnosticar a presença de Mycobacterium marinum. A enfermidade pode ser transmitida para o homem e com esse trabalho ressalta-se a necessidade de fazer o diagnóstico correto e implantar medidas de proteção para as pessoas que estão em contato com os peixes infectados.

A new species of a philometrid nematode, Margolisianum bulbosum, is described from the subcutaneous tissue in the mouth (larvigerous females), head (males, ovigerous, and larvigerous females), and eye (preovigerous females) of the southern flounder, Paralichthys lethostigma, from Mississippi Sound. It is placed in a new genus diagnosed by the combination of 8 large, paired but separate cephalic papillae; no inner cephalic papillae; an esophagus with a separate, muscular anterior bulb; a prominent mononuclear esophageal gland; and variable, irregularly distributed cuticular bosses in the females, as well as a vestigial rectum, particularly in larvigerous females. Some female specimens exhibit rows of lateral grooves and longitudinal ridges near the posterior end. Males have two small slightly subequal spicules, a barbed gubernaculum, 4 pairs of small cephalic papillae, and a bipartite hypodermal extension within a membranous cuticle on the posterior end. Males, ovigerous females, and larvigerous females appear to be present year round in this sporadic infection in Mississippi.

Effects of tributyltin (TBT) which has been used for antifouling paint of ship's hulls and fishing nets on the immune system in Japanese flounder (Paralichthys olivaceus) were investigated. After short-term exposure to a high level of TBT, leucocytes in the head kidney from 1-year-old flounder were examined for the proportion of neutrophils in total leucocytes. Also examined were their respiratory burst activities using flow cytometry, the reduction of nitroblue tetrazolium (NBT) and lysozyme activities. Furthermore, long-term exposures to a relatively low level of TBT using young flounder were also carried out. The proportion of neutrophils in total leucocytes prepared from head kidney in each fish exposed to TBT at 20 microg/L for 5 days and the reduction of NBT by leucocytes prepared from the same experimental conditions increase compared to the control group. The contents were 42.0+/-6.8 and 52.5+/-6.3%, respectively. Significant differences of the NBT reduction were observed between 0 and 20 microg/L TBT exposure groups. On the other hand, the respiratory burst activity of cells in the exposure group clearly showed a tendency to decrease compared to the control group. Furthermore, high level of TBT also inhibited lysozyme activity which plays an important role for the bacteriocidal procedures. However, similar results were not obtained in the exposure group with a relatively low level of TBT. To determine the immunotoxic effects of TBT, infection experiments using pathogens which are naturally occurring should be further investigated.

El lenguado de California Paralichthys californicus es una especie con alto valor comercial debido a su gran tamaño y calidad de su carne. Esta especie presenta dimorfismo sexual en el crecimiento donde las hembras crecen más rápido que los machos, por lo tanto el cultivo monosexual de hembras resulta favorable para la producción. En el presente estudio se registró el proceso de diferenciación sexual mediante cortes histológicos de las larvas y gónadas del lenguado de California asimismo se probó el efecto de diferentes concentraciones de 17β-estradiol (E2) (2.5, 5 y 10 mg/kg) a través de la dieta para incrementar la proporción de hembras en el cultivo. A los 25 días después de la eclosión (DDE) se observó el primordio gonadal en ejemplares con una longitud total promedio de 6.96 ± 0.92 mm, hasta el día 75 DDE (37.58 ± 6.58 mm) se observó una gónada indiferenciada evidenciada por la presencia de células germinales primordiales. La primera evidencia de diferenciación se registró a los 115 DDE (55.93 ± 14.67 mm) donde se observó la cavidad ovárica y posteriormente a los 180 DDE (115.70 ± 17.02 mm) se evidencian ovarios con ovocitos en crecimiento y testículos con espermátidas. Por lo tanto, el periodo lábil para la diferenciación sexual se encuentra entre los días 75 y 115 después de la eclosión. Por otro lado, el suministro de E2 a través de la dieta a concentraciones de 2.5, 5 y 10 mg/kg incrementó el porcentaje de hembras de 26.67% (control, no adición de E2) a un 100% en todos los tratamientos. Se encontraron diferencias en la proporción de los tipos celulares (ovogonias, ovocitos primarios en fase I y II) entre el control y los tratamientos mientras que no se registraron alteraciones histológicas como atrofia gonadal en ninguno de los casos.

Improper dietary protein and energy levels and their ratio will lead to increased fish production cost. This work evaluated effects of dietary protein : energy ratio on growth and body composition of pacu, Piaractus mesopotamicus. Fingerling pacu (15.5 +/- 0.4 g) were fed twice a day for 10 weeks until apparent satiation with diets containing 220, 260, 300, 340 or 380 g kg-1 crude protein (CP) and 10.9, 11.7, 12.6, 13.4 or 14.2 MJ kg-1 digestible energy (DE) in a totally randomized experimental design, 5 x 5 factorial scheme (n = 3). Weight gain, specific growth rate increased and feed conversion ratio (FCR) decreased significantly (P < 0.05) when CP increased from 220 to 271, 268 and 281 g kg-1 respectively. Pacu was able to adjust feed consumption in a wide range of dietary DE concentration. Fish fed 260 CP diets showed best (P < 0.05) protein efficiency ratio and FCR with 11.7-12.6 MJ kg-1; but for the 380 CP-diets group, significant differences were observed only at 14.2 MJ kg-1 dietary energy level, suggesting that pacu favours protein as energy source. DE was the chief influence on whole body chemical composition. Minimum dietary protein requirement of pacu is 270 g kg-1, with an optimum CP : DE of 22.2 g MJ-1.

Lysine is the reference essential amino acid in fish feeds and usually the most limiting amino acid in feedstuffs. The dietary lysine requirement of juvenile pacu Piaractus mesopotamicus (4.3 g) was determined using five isonitrogenous (32% CP) test diets containing graded levels of lysine (0.9, 1.17, 1.44, 1.69 and 1.96% of dry diet) fed three times a day to four groups of 18 fish for 74 days. Growth, body composition, nutrient retention and hematological parameters of pacu were analyzed. Analysis of variance showed that all growth performance parameters were significantly affected by dietary treatments. The lysine requirements estimated using regression analysis for maximum weight gain and feed efficiency were 1.45 and 1.51% of dry diet, respectively. Nitrogen retention efficiency increased with increasing levels of dietary lysine up to 1.43% (p<0.05). Whole-body protein increased (p<0.05) and whole-body lipid decreased (p<0.05) with increasing dietary lysine level. Thus, the lysine requirement of juvenile pacu was estimated as being 1.4-1.5% of dry diet or 4.4-4.7% of dietary protein. (c) 2009 Elsevier B.V. All rights reserved.

Five species of monogeneans collected from marine fishes of the Pacific coast of Chile and Perú were studied. Three of them are new species: Interniloculus chilensis n. sp. (Capsalidae), Neoheterobothrium insalaris n. sp. (Diclidophoridae) and Loxura peruensis n. sp. (Axinidae) parasitic on Sebastes capensis (Scorpaenidae), Paralichthys sp. (Bothiidae) and Belone scapularis (Belonidae), respectively. Two new combinations are proposed, Intracotyle neghmei (Microcotylidae) for Neobivagina neghmei Villalba, 1987 and Hargicotlyle conceptionensis (Diclidophoridae) for Choricotyle conceptionensis Villalba, 1987.

A qualidade de 120 amostras de diferentes tipos de filé de peixe congelado foi avaliada com relação às características sensoriais, pH, teores de bases voláteis totais e histamina e presença de gás sulfídrico. Os resultados da análise sensorial indicaram boa qualidade dos peixes com valores médios de 5,52 (CV £ 13%) tanto para odor quanto para aspecto geral. Com relação a bases voláteis totais (BVT), teores médios de 62,71 mg N/100 g foram encontrados, com valores significativamente maiores (> 80 mg N/100 g) para namorado (Pseudopercis numida) e cação (famílias Carcharhinidae e Squalidae) e menores que 68 mg N/100 g para os demais tipos de peixe. Os valores médios de pH variaram de 6,11 em abrótea (Urophycis brasiliensis) a 7,49 em castanha (Umbrina sp.). Baseado nestes resultados, 79, 39 e 62% das amostras não atenderiam aos limites estipulados pela legislação vigente para BVT, pH e gás sulfídrico, respectivamente. Histamina foi encontrada em 37% das amostras com teores médios de 0,00 para linguado (Paralichthys sp. e Pleuronectes sp.) a 0,50 mg/100 g para namorado. A presença de histamina não foi detectada em abrótea. Foram observadas diferenças significativa entre os valores de BVT, pH e histamina das amostras de peixe agrupadas por famílias. Para uma melhor avaliação do potencial de utilização e estabelecimento de limites críticos para estes parâmetros como critério de qualidade, sugere-se que amostras sejam monitoradas desde o momento da captura. Os teores de histamina detectados nas amostras não seriam, por si só, capazes de causar intoxicação.

The karyotypes and cytogenetic characteristics of flatfishes species Paralichthys orbignyanus, Paralichthys patagonicus, Citarichthys spilopterus and Etropus crossotus (Paralichthyidae), Bothus ocellatus (Bothidae) and Symphurus tessellatus (Cynoglossidae) were investigated by conventional [Giemsa staining, C-banding, Ag- and chromomycin (CMA(3))-stainings] and molecular [in situ hybridization (ISH)] cytogenetic techniques. The results showed 2n = 46 and FN = 48 (2msm + 46sta) in P. orbignyanus, 2n = 46 and FN = 46 (46sta) in P. patagonicus, 2n = 26 and FN = 44 (18msm + 8sta) in C. spilopterus, 2n = 38 and FN = 64 (26msm + 12sta) in E. crossotus, 2n = 32 and FN = 50 (18msm + 14sta) in B. ocellatus, and 2n = 46 and FN = 62 (46msm + 62sta) in S. tessellatus. All species exhibited weak C-band positive segments in terminal and centromeric positions of some chromosome pairs. Silver staining of the nucleolus organizer regions (Ag-NOR) technique showed a single Ag-NOR-bearing chromosome pair in all species except E. crossotus. All these sites were CMA(3) positive and showed clear ISH signals after probing with a 18S rRNA probe. Etropus crossotus presented until seven chromosomes with Ag-NORs and CMA(3) positively stained segments in five chromosome pairs. Conversely only one chromosome pair was identified with the ISH experiments in this species. The available results show that the fishes of the order Pleuronectiformes experienced a marked chromosome evolution that included reduction in diploid number, mainly due to Robertsonian rearrangements, and several chromosome inversions. Specific dietary contents from six fishes collected in Mississippi Sound are recorded. In order of their importance, primary components grouped in major taxonomic categories were fishes, penaeid shrimps, and other crustaceans for Cynoscion nebulosus; Crustaceans and fishes for C. arenarius; fishes and crustaceans for C. nothus; crustaceans, pelecypods, and polychaetes for Pogonias cromis; crustaceans, molluscs, polychaetes, and fishes for Archosargus probatocephalus; and fishes and penaeid shrimps for Paralichthys lethostigma. Principal items in the diets of most of the fishes included Anchoa mitchilli, Penaeus aztecus, P. setiferus, and Callinectes sapidus. Those crustaceans show that competition exists for commercial shellfishes in Mississippi Sound. Ratios among the different dietary items vary, according at least to species of fish, length of fish, season, specific location, and abundance of available prey. Some of these variations are documented and are additionally related to selected findings by other authors sampling different localities. We suggest that examination of food items in Archosargus probatocephalus can serve as a practical means to sample and assess seasonal prevalence and abundance of a wide range of invertebrates throughout different habitats in Mississippi Sound and elsewhere.

In gene-banking, primordial germ cells (PGCs), which are embryonic precursor cells of germ cells, are useful for cryopreservation because PGCs have a potential to differentiate into both eggs and sperm via germ-line chimera. Here, we have established vitrification methods for PGCs cryopreservation using 12- to 17-somite stage embryos in loach, Misgurnus anguillicaudatus, which were dechorionated, removed their yolk and injected with green fluorescent protein (GFP) -nos1 3'UTR mRNA to visualize their PGCs. In order to optimize cryopreservation medium for vitrification, the toxicity of cryoprotectants was analyzed. Different concentrations (2, 3, 4, 5 m) of dimethyl sulfoxide (DMSO), methanol (MeOH), ethylene glycol (EG) and propylene glycol (PG) as cryoprotectants were tested. Then, 5 m DMSO showed significantly-high toxicity. Based on this information, combinations called DMP (2 m (14.2% [v/v]) DMSO, 2 m (8.1% [v/v]) MeOH and 2 m (14.4% [v/v]) PG), DP (2 m (14.2% [v/v]) DMSO and 4 m (28.7% [v/v]) PG) and DE (2.1 m (15% [v/v]) DMSO and 2.7 m (15% [v/v]) EG) were evaluated for their toxicities and efficacy of PGCs cryopreservation using two types of equilibration step: direct immersion of cryopreservation media (one-step) and serial exposure to half and full concentration of cryopreservation media (two-step). Viable PGCs were obtained from post-thaw embryos which were cryopreserved by DP and DE with both 1- and 2-step equilibrations. Despite DP showing the highest toxicity, it gave the highest survival rate of embryonic cells after cryopreservation. When PGCs recovered from vitrified embryos were transplanted into host embryos at the blastula stage, the transplanted PGCs were able to migrate to a host genital ridge similarly as endogenous PGCs. It suggests that our methods could be useful to create a germ-line chimera for the production of gametes from PGCs of cryopreserved embryos.
