O extrato enzimático foi preparado a partir da polpa e casca da maçã de cultivares Fuji e Gala utilizando tampão fosfato de sódio 100mM, pH 5,0 como solução extratora. Dentre as análises determinou-se a concentração de proteína nos extratos enzimáticos concentrados de polpa e casca, sendo que o cultivar Fuji apresentou teores mais elevados em comparação ao cultivar Gala. Os tratamentos térmicos foram realizados nas temperaturas de 60, 65, 70 e 75°C por períodos que variaram de 1 a 10 minutos, sendo observado diminuição da atividade de POD e PPO com o aumento da temperatura e tempo; no entanto a POD não chegou a ser inativada em nenhum dos tratamentos realizados. A PPO foi inativada totalmente após 10 minutos de tratamento a 75°C. A eletroforese mostrou uma composição diferente de isoenzimas aniônicas e catiônicas da peroxidase.

A peroxidase E.C. 1.11.1.7 (POD) e a polifenoloxidase E.C. 1.10.3.1 (PPO) foram extraídas da polpa de goiaba. Os extratos foram preparados utilizando-se a polpa da goiaba e solução tampão fosfato de sódio 100mM com pH variando de 6,0 a 7,0 em intervalos de 0,1. Foi determinada a atividade enzimática da peroxidase e da polifenoloxidase desses extratos, a fim de se observar o melhor pH para a extração de cada enzima. O pH 6,3 foi considerado o melhor para a extração da POD da polpa de goiaba, enquanto que para PPO, o pH foi 6,8. Os extratos brutos de POD e PPO foram submetidos a temperaturas de 60 °C, 65 °C, 70 °C, 75 °C e 80 °C por um período de 0 a 10 min. Os resultados demonstraram um decréscimo da atividade enzimática nos extratos com o aumento da temperatura e do tempo. No entanto, a total inativação não foi atingida o que sugere a presença de isoenzimas termoresistentes.

A peroxidase e a polifenoloxidase estão relacionadas com o escurecimento de frutas, por isso o controle das atividades destas enzimas é de grande importância para a tecnologia de alimentos. Neste trabalho estudaram-se as atividades dessas enzimas nas uvas frescas das cultivares Benitaka e Rubi, bem como as suas termoestabilidades e as suas atividades enzimáticas residuais no suco e nas geléias (extra e light). Foi também avaliada a qualidade microbiológica dos produtos elaborados. As atividades da enzima POD, tanto da fração solúvel quanto da ionicamente ligada, foram semelhantes nas uvas das duas variedades, Benitaka e Rubi. A atividade da enzima polifenoloxidase foi maior na variedade Rubi. As operações de cocção e pasteurização foram mais eficientes para baixar as atividades enzimáticas residuais da POD e PPO quando aplicadas às geléias de uva, em comparação com o suco. Embora não foram suficientes para a total inativação enzimática, essas operações reduziram-nas consideravelmente, e foram eficientes para garantir seguridade microbiológica dos produtos, geléias e suco.

Soil moisture and soil temperature affect pre-harvest infection with Aspergillus flavus and production of aflatoxin. The objectives of our field research in Niger, West Africa, were to: (i) examine the effects of sowing date and irrigation treatments on pod yield, infection with A. flavus and aflatoxin concentration; and (ii) to quantify relations between infection, aflatoxin concentration and soil moisture stress. Seed of an aflatoxin susceptible peanut cv. JL24 was sown at two to four different sowing dates under four irrigation treatments (rainfed and irrigation at 7, 14 and 21 days intervals) between 1991 and 1994, giving 40 different 'environments'. Average air and soil temperatures of 28-34 degrees C were favourable for aflatoxin contamination. CROPGRO-peanut model was used to simulate the occurrence of moisture stress. The model was able to simulate yields of peanut well over the 40 environments (r(2) = 0.67). In general, early sowing produced greater pod yields, as well as less infection and lower aflatoxin concentration. There were negative linear relations between infection (r(2) = 0.62) and the average simulated fraction of extractable soil water (FESW) between flowering and harvest, and between aflatoxin concentration (r(2) = 0.54) and FESW in the last 25 days of pod-filling. This field study confirms that infection and aflatoxin concentration in peanut can be related to the occurrence of soil moisture stress during pod-filling when soil temperatures are near optimal for A. flavus. These relations could form the basis of a decision-support system to predict the risk of aflatoxin contamination in peanuts in similar environments. (c) 2005 Elsevier B.V. All rights reserved.

Pods play a key role in encapsulating the developing seeds and protecting them from pests and pathogens. In addition to this protective function, it has been shown that the photosynthetically active pod wall contributes assimilates and nutrients to fuel seed growth. Recent work has revealed that signals originating from the pod may also act to coordinate grain filling and regulate the reallocation of reserves from damaged seeds to those that have retained viability. In this review we consider the evidence that pods can regulate seed growth and maturation, particularly in members of the Brassicaceae family, and explore how the timing and duration of pod development might be manipulated to enhance either the quantity of crop yield or its nutritional properties.

Based on pollen analysis of a sediment core from the Atlantic Ocean off Liberia the West African vegetation history for the last 400 ka is reconstructed. During the cold oxygen isotope stages 12, 10, 8, 6, 4, 3 and 2 an arid climate is indicated, resulting in a southward shifting of the southern border of the savanna. Late Pleistocene glacial stages were more arid than during the Middle Pleistocene. A persistence of the rain forest in the area, even during the glacial stages, is recorded. This suggests a glacial refuge of rain forest situated in the Guinean mountains. Afromontane forests with Podocarpus occurred in the Guinean mountains from the stages 12 to 2 and disappeared after. The tree expanded from higher to lower elevations twice in the warm oxygen isotope stage 11 (pollen subzones 11d, 11b) and at least twice during the warm stage 5 (pollen subzones 5d, 5a), indicating a relative cool but humid climate for these periods.

In this work we propose a method to accelerate time dependent numerical solvers of systems of PDEs that require a high cost in computational time and memory. The method is based on the combined use of such numerical solver with a proper orthogonal decomposition, from which we identify modes, a Galerkin projection (that provides a reduced system of equations) and the integration of the reduced system, studying the evolution of the modal amplitudes. We integrate the reduced model until our a priori error estimator indicates that our approximation in not accurate. At this point we use again our original numerical code in a short time interval to adapt the POD manifold and continue then with the integration of the reduced model. Application will be made to two model problems: the Ginzburg-Landau equation in transient chaos conditions and the two-dimensional pulsating cavity problem, which describes the motion of liquid in a box whose upper wall is moving back and forth in a quasi-periodic fashion. Finally, we will discuss a way of improving the performance of the method using experimental data or information from numerical simulations

Se desarrollan varias técnicas basadas en descomposición ortogonal propia (DOP) local y proyección de tipo Galerkin para acelerar la integración numérica de problemas de evolución, de tipo parabólico, no lineales. Las ideas y métodos que se presentan conllevan un nuevo enfoque para la modelización de tipo DOP, que combina intervalos temporales cortos en que se usa un esquema numérico estándard con otros intervalos temporales en que se utilizan los sistemas de tipo Galerkin que resultan de proyectar las ecuaciones de evolución sobre la variedad lineal generada por los modos DOP, obtenidos a partir de instantáneas calculadas en los intervalos donde actúa el código numérico. La variedad DOP se construye completamente en el primer intervalo, pero solamente se actualiza en los demás intervalos según las dinámicas de la solución, aumentando de este modo la eficiencia del modelo de orden reducido resultante. Además, se aprovechan algunas propiedades asociadas a la dependencia débil de los modos DOP tanto en la variable temporal como en los posibles parámetros de que pueda depender el problema. De esta forma, se aumentan la flexibilidad y la eficiencia computacional del proceso. La aplicación de los métodos resultantes es muy prometedora, tanto en la simulación de transitorios en flujos laminares como en la construcción de diagramas de bifurcación en sistemas dependientes de parámetros. Las ideas y los algoritmos desarrollados en la tesis se ilustran en dos problemas test, la ecuación unidimensional compleja de Ginzburg-Landau y el problema bidimensional no estacionario de la cavidad. Abstract Various ideas and methods involving local proper orthogonal decomposition (POD) and Galerkin projection are presented aiming at accelerating the numerical integration of nonlinear time dependent parabolic problems. The proposed methods come from a new approach to the POD-based model reduction procedures, which combines short runs with a given numerical solver and a reduced order model constructed by expanding the solution of the problem into appropriate POD modes, which span a POD manifold, and Galerkin projecting some evolution equations onto that linear manifold. The POD manifold is completely constructed from the outset, but only updated as time proceeds according to the dynamics, which yields an adaptive and flexible procedure. In addition, some properties concerning the weak dependence of the POD modes on time and possible parameters in the problem are exploited in order to increase the flexibility and efficiency of the low dimensional model computation. Application of the developed techniques to the approximation of transients in laminar fluid flows and the simulation of attractors in bifurcation problems shows very promising results. The test problems considered to illustrate the various ideas and check the performance of the algorithms are the onedimensional complex Ginzburg-Landau equation and the two-dimensional unsteady liddriven cavity problem.
