Linkage with essential hypertension has been claimed for a microsatellite marker near the angiotensinogen gene (AGT; chromosome 1q42), as has association for the AGT variants M235T, G(-6)A and A(-20)C. To more rigorously evaluate AGT as a candidate gene for hypertension we performed sibpair analysis with multiple microsatellite markers surrounding this locus and using more sophisticated analysis programs. We also performed an association study of the AGT variants in unrelated subjects with a strong family history (two affected parents). For the linkage study, single and multiplex polymerase chain reaction (PCRs) and automated genescan analysis were conducted on DNA from 175 Australian Anglo-Celtic Caucasian hypertensives for the following markers: D1S2880-(2.1 cM)-D1S213-(2.8 cM)-D1S251-(6.5 cM)-AGT-(2.0 cM) -D1S235. Statistical evaluation of genotype data by nonparametric methods resulted in the following scores: Single-point analysis - SPLINK, P > 0.18; APM method, P > 0.25; ASPEX, MLOD < 0.28; SIB-PAIR, P > 0. 24; Multipoint analysis - MAPMAKER/SIBS, MLOD < 0.24; GENEHUNTER, P > 0.35. Exclusion scores of Lod -4.1 to -5.1 were obtained for these markers using MAPMAKER/SIBS for a lambda(s) of 1.6. The association study of G(-6)A, A(-20)C and M235T variants in 111 hypertensives with strong family history and 190 normotensives with no family history showed significant linkage disequilibrium between particular haplotypes, but we could find no association with hypertension. The present study therefore excludes AGT in the etiology of hypertension, at least in the population of Australian Anglo-Celtic Caucasians studied.

The research topic is the formation of nuclear family understanding and the politicization of nuclear family. Thus, the question is how did family historically become understood particularly as nuclear family and why did it become central in terms of politics and social? The research participates in discussions on the concept and phenomena of family. Central theme of analysis is to ask what is family? Family is seen as historically contingent and the discussions on the concept and phenomena are done via historical analysis. Center of attention is nuclear family, thus, a distinction between the concepts of family and nuclear family is made to be able to focus on historically specific phenomena of nuclear family. Family contrary to the concept of nuclear family -- in general is seen to be able to refer to families in all times and all cultures, as well as all types of families in our times and culture. The nuclear family understanding is examined through two separate themes, that of parent-child relationships and marital relations. Two simultaneous processes give nuclear family relations its current form: on the one hand the marital couple as the basis of family is eroding and losing its capacity to hold the family together; on the other, in Finland at least from 1950s on, the normal development of the child has became to be seen ontologically bound to the (biological) mother and (via her to) the father. In the nucleus of the family is the child: the biological, psychological and social processes of normal development are seen ontologically bound to the nuclear family relations. Thus, marriages can collapse, but nuclear family is unbreakable. What is interesting is the historical timing: as nuclear family relations had just been born, the marriage dived to a crisis. The concept and phenomena of nuclear family is analyzed in the context of social and politics (in Finnish these two collapses in the concept of yhteiskunnallinen , which refers both to a society as natural processes as well as to the state in terms of politics). Family is political and social in two senses. First, it is understood as the natural origin of the social and society. Human, by definition, is understood as a social being and the origin of social, in turn, is seen to be in the family. Family is seen as natural to species. Disturbances in family life lead to un-social behaviour. Second, family is also seen as a political actor of rights and obligations: family is obligated to control the life of its members. The state patronage is seen at the same time inevitable family life is way too precious to leave alone -- and problematic as it seems to disturb the natural processes of the family or to erode the autonomy of it. The rigueur of the nuclear family is in the role it seems to hold in the normal development of the child and the future of the society. The disturbances in the families first affect the child, then the society. In terms of possibility to re-think the family the natural and political collide: the nuclear family seems as natural, unchangeable, un- negotiable. Nuclear family is historically ontologised. The biological, psychological and social facts of family seem to be contrary to the idea of negotiation and politics the natural facts of family problematise the politics of family. The research material consists of administrational documents, memoranda, consultation documents, seminar reports, educational writings, guidebooks and newspaper articles in family politics between 1950s and 1990s.

The subject matter of this study is the cultural knowledge concerning romantic male-female relationships in autobiographies written by so called ordinary Finnish men and women born between 1901 and 1965. The research data (98 autobiographies) is selected from two collections by the Finnish Literature Society s folklore archives in the early 1990 s. Autobiographies are cultural representations where negotiation of shared cultural models and personal meanings given to hetero-relationship is evident in an interesting manner. In this research I analyze autobiographies as a written folklore genre. Information concerning male-female relationships is being analyzed using theoretically informed close readings thematic analysis, intertextual reading and reflexive reading. Theoretical implications stem from cognitive anthropology (the idea of cultural models) and an adaptation of discourse theory inspired by Michel Foucault. The structure of the analysis follows the structure of the shared knowledge concerning romantic male-female relationship: the first phase of analysis presents the script of a hetero-relationship and then moves into the actual structure, the cultural model of a relationship. The components of the model of relationship are, as mentioned in the title of the research, woman, man, love and sex. The research shows that all the writers share this basic knowledge concerning a heterosexual relationship despite their age, background or gender. Also the conflicts described and experienced in the relationships of the writers were similar throughout the timespan of the early 1900 s to 1990 s: lack of love, inability to reconcile sexual desires, housework, sharing the responsibility of childcare and financial problems. The research claims that the conflicts in relationships are a major cause for the binary view on gender. When relationships are harmonious, there seems to be no need to see men and women as opposites. The research names five important discourses present in the meaning giving processes of autobiographers. In doing so, the stabile cultural model of male-female relationship widens to show the complexity and variation in data. In this way it is possible to detect some age and gender specific shifts and emphasis. The discourses give meaning to the components of the cultural model and determine the contents of womanhood, manhood, sexuality and love. The way these discourses are spread and their authority are different: the romantic discourse evident in the autobiographies appeal to the authority of love supreme love is the purpose of male-female relationship and it justifies sexuality. In this discourse sex can be the place for confluence of genders. The ideas of romantic love are widely spread in popular culture. Popular scientific discourse defines a relationship as a site to become a man and a woman either from a psychological or a biological point of view. Genders are seen as opposites. These ideas are often presented in media and their authority in science which is seen as infallible. The Christian discourse defines men and women: both should work for the benefit of the nuclear family under the undisputed authority of God. Marital love is based on Christian virtues and within marriage sexuality is acceptable. The discourse I ve named folk tradition defines women and men as guardians of home and offspring. The authority of folk tradition comes from universal truth based in experience and truths known to the mediators of this discourse grandparents, parents and other elders or peers. Societal discourse defines the hetero relationship as the mainstay of society. The authority in societal discourse stems from the laws and regulations that control relationship practices.

The ownership of memories is sometimes disputed, particularly by twins. Examination of 77 disputed memories, 71 provided by twins, showed that most of the remembered events are negative and that the disputants appear to be self-serving. They claim for themselves memories for achievements and suffered misfortunes but are more likely to give away memories of personal wrongdoing. The research suggests that some of the memories in which we play a leading role might in fact have been the experiences of others.

Chromosome 5q22-33 is a region where studies have repeatedly found evidence for linkage to schizophrenia. In this report, we took a stepwise approach to systematically map this region in the Irish Study of High Density Schizophrenia Families (ISHDSF, 267 families, 1337 subjects) sample. We typed 289 SNPs in the critical interval of 8 million basepairs and found a 758 kb interval coding for the SPEC2/PDZ-GEF2/ACSL6 genes to be associated with the disease. Using sex and genotype-conditioned transmission disequilibrium test analyses, we found that 19 of the 24 typed markers were associated with the disease and the associations were sex-specific. We replicated these findings with an Irish case-control sample (657 cases and 414 controls), an Irish parent-proband trio sample (187 families, 564 subjects), a German nuclear family sample (211 families, 751 subjects) and a Pittsburgh nuclear family sample (247 families, 729 subjects). In all four samples, we replicated the sex-specific associations at the levels of both individual markers and haplotypes using sex- and genotype-conditioned analyses. Three risk haplotypes were identified in the five samples, and each haplotype was found in at least two samples. Consistent with the discovery of multiple estrogen-response elements in this region, our data showed that the impact of these haplotypes on risk for schizophrenia differed in males and females. From these data, we concluded that haplotypes underlying the SPEC2/PDZ-GEF2/ACSL6 region are associated with schizophrenia. However, due to the extended high LD in this region, we were unable to distinguish whether the association signals came from one or more of these genes.

A randomized trial was carried out comparing cyclosporin A (CsA) and short-term methotrexate (MTX) versus CsA alone for graft versus host disease (GVHD) prophylaxis in patients with severe aplastic anemia (SAA) undergoing allogeneic bone marrow transplantation (BMT) from a compatible sibling. Seventy-one patients (median age, 19 years; range, 4-46 years) were randomized to receive either CsA and MTX or CsA alone for the first 3 weeks after BMT. Subsequently, both groups received CsA orally, with gradual drug reduction until discontinuation 8 to 12 months after BMT. Patients randomized in both arms had comparable characteristics and received the same preparative regimen (ie, cyclophosphamide 200 mg/kg over 4 days). The median time for neutrophil engraftment was 17 days (range, 11-31 days) and 12 days (range, 4-45 days) for patients in the CsA/MTX group and the CsA alone group, respectively (P =.01). No significant difference was observed in the probability of either grade 2, grade 3, or grade 4 acute GVHD or chronic GVHD developing in the 2 groups. The Kaplan-Meier estimates of 1-year transplantation-related mortality rates for patients given either CsA/MTX or CsA alone were 3% and 15%, respectively (P =.07). With a median follow-up of 48 months from BMT, the 5-year survival probability is 94% for patients in the CsA/MTX group and 78% for those in the CsA alone group (P =. 05). These data indicate that the use of CsA with MTX is associated with improved survival in patients with SAA who receive transplants from compatible siblings. (Blood. 2000;96:1690-1697)

Os sistemas familiares multigeracionais (três ou mais gerações vivas) passaram de raros no passado a uma realidade social cada vez mais comum na actualidade, devido ao fenómeno demográfico de envelhecimento populacional. Este estudo pretende contribuir para o conhecimento deste sistema social, situado entre a família nuclear e a comunidade, procurando descrever a sua estrutura, funções e padrões relacionais dos seus membros com outros familiares e com os sistemas sociais envolventes. Para tal, foi organizada uma amostra de 25 famílias multigeracionais (contendo quatro gerações), à qual foi aplicada uma entrevista de genograma a um elemento das gerações intermédias e quatro entrevistas de análise da rede social pessoal (uma por geração), perfazendo um total de 25 entrevistas de genograma e 92 de avaliação da rede social (menos 8 do que o esperado devido a mortalidade experimental). A análise dos dados possibilitou: 1) a aplicação do princípio de totalidade sistémica à família multigeracional, a sua definição como um sistema social de complexidade idiossincrática, a identificação dos subsistemas que compõem o objecto de estudo (indivíduo, núcleo familiar, composição familiar, geração, linhagem), e a definição de uma tipologia familiar multigeracional (família unificada, dispersa e fragmentada); 2) a identificação e análise de papéis sociais desempenhados pelos indivíduos no contexto familiar multigeracional especificamente relevantes neste nível sistémico (guardião das memórias familiares, elo de ligação familiar, pronto-socorro familiar); 3) a descrição das redes sociais pessoais e a identificação das diferenças estruturais e funcionais entre indivíduos pertencentes a diferentes subsistemas geracionais, que permitem reflectir sobre a evolução da rede social ao longo das várias fases de desenvolvimento individual e familiar; 4) um conhecimento mais aprofundado das redes sociais dos indivíduos mais idosos que pertencem a famílias multigeracionais, da composição da matriz relacional percepcionada como significativa e apoios disponibilizados. Em suma, os dois primeiros capítulos reflectem o esforço de desenvolvimento de um modelo heurístico para o sistema familiar multigeracional, numa perspectiva estruturalista, enquanto os dois capítulos subsequentes, apoiandose conceptualmente nos anteriores, facultam um quadro das relações entre subsistemas geracionais na actualidade, através de uma metodologia de análise das redes sociais. ABSTRACT: Multigenerational family systems comprising four generations are increasingly common nowadays due to the demographic phenomenon of population aging. This study aims at a deeper understanding of this social system, placed between the nuclear family and the community, in particular by describing its structure, functions and relational patterns of its members with other members in the family and surrounding social systems. A sample of 25 multigenerational families (comprising four generations) was selected. A genogram interview was applied to a member of the middle generations, and four personal social network questionnaires were administered to one element of each generation. A total of 25 genograms and 92 social network inventories were collected (minus 8 than expected due to experimental mortality). Main findings were: 1) the application of systemic principles to the multigenerational family, which allowed us to define it as a social system of idiosyncratic complexity, involving specific subsystems (individual, family nucleus, family composition, generation, lineage), and also to define a multigenerational family typology (unified, dispersed and fragmented family); 2) the identification and analysis of social roles assumed by individuals in the multigenerational family context, that are specifically relevant in this systemic level (keeper of family memories, connecting link in the family, family first aider); 3) the description of the personal social networks and the identification of structural and functional differences among individuals belonging to different generational subsystems, which allowed for a reflection on the evolution of social networks through the various stages of individual and family development; 4) a deeper knowledge of the eldest family members social networks, i.e. the relational matrix of significant individuals and supports in oldold age. In summary, the first two chapters reflect the development of a heuristic model directed at the multigenerational family, whereas the two subsequent chapters provide a view of contemporaneous relationships between generational subsystems, through a social network analysis methodology.

Night vision requires signaling from rod photoreceptors to adjacent bipolar cells in the retina. Mutations in the genes NYX and GRM6, expressed in ON bipolar cells, lead to a disruption of the ON bipolar cell response. This dysfunction is present in patients with complete X-linked and autosomal-recessive congenital stationary night blindness (CSNB) and can be assessed by standard full-field electroretinography (ERG), showing severely reduced rod b-wave amplitude and slightly altered cone responses. Although many cases of complete CSNB (cCSNB) are caused by mutations in NYX and GRM6, in approximately 60% of the patients the gene defect remains unknown. Animal models of human diseases are a good source for candidate genes, and we noted that a cCSNB phenotype present in homozygous Appaloosa horses is associated with downregulation of TRPM1. TRPM1, belonging to the family of transient receptor potential channels, is expressed in ON bipolar cells and therefore qualifies as an excellent candidate. Indeed, mutation analysis of 38 patients with CSNB identified ten unrelated cCSNB patients with 14 different mutations in this gene. The mutation spectrum comprises missense, splice-site, deletion, and nonsense mutations. We propose that the cCSNB phenotype in these patients is due to the absence of functional TRPM1 in retinal ON bipolar cells.

Dans le Cambodge angkorien, les souverains khmers administraient une paysannerie mouvante par le biais de temples-palais. Lorsque les Français prennent le contrôle, en 1845, ils se retrouvent devant une « masse paysanne inorganisée, inorganique même » (Delvert, 1961 : 201) et restent « confondus devant la mobilité des Cambodgiens » (Forest, 1980 : 30). À l’époque postcoloniale, les ethnologues feront essentiellement le même constat, pendant que John F. Embree (1950) proposera de catégoriser les sociétés indianisées du Sud-Est asiatique comme étant « loosely structured » : postulant une faible intégration individuelle des structures sociales donnant lieu à une prévalence de comportements individualistes ad hoc et à des communautés sans réelle organisation. La proposition fera école. Ces observations paraissent justes, mais l’analyse infructueuse. La structure dont parle Embree s’appuie sur une culture hautement syncrétique qui se reflétait aléatoirement dans les comportements. Mais l’organisation sociale khmère se trouve ailleurs : dans les solutions organisationnelles qui gouvernent les choix des individus lorsqu’ils doivent se regrouper afin d’effectuer des tâches récurrentes. À ce titre, les paysans khmers évoluaient dans une organisation sociale rigoureusement minimaliste et flexible. La maisonnée était l’élément essentiel, tandis que la communauté territoriale locale était contingente et fluctuante. Dans l’environnement naturel généreux du Cambodge, un petit groupe d’individus mobiles réunis sous un même toit pouvait aisément accomplir toutes les tâches nécessaires à sa survie. Alors on ne s’attachait jamais indéfiniment à une localité : seulement à des communautés sans cesse en évolution, centrées autour de pagodes agissant comme des ports d’ancrage.

Il est convenu de dire que la famille d’aujourd’hui se construit de multiples façons. Les familles recomposées, issues des nouvelles technologies de la reproduction, de l’adoption internationale ou dont les parents sont de même sexe font partie du paysage depuis de nombreuses années, au point où elles n’étonnent plus. Ces familles ont comme particularité de remettre en question l’adéquation entre la sexualité du couple et la reproduction tout en introduisant d’autres personnes dans la conception ou dans l’éducation des enfants. Malgré la polymorphie des familles contemporaines, perdure toutefois une certaine représentation de la famille basée sur le sang partagé entre les différents membres qui la composent. En ce sens, les « vrais » parents sont ceux qui sont à l’origine de la naissance de l’enfant (Déchaux, 2007; Deliège, 2005). Parallèlement à cela, on constate l’émergence d’un discours centré sur l’importance du père pour le développement et le bien-être de l’enfant, au point où l’absence du père au sein du foyer familial tend de plus en plus à être conceptualisée comme un problème social majeur (Kelly, 2009). De fait, les recherches démontrent que l’engagement paternel profite autant aux enfants qu’au père et à la mère. C’est dans ce contexte que s’inscrivent les familles lesboparentales dont les enfants sont nés d’un donneur connu. Cette thèse a comme objectif de comprendre le rôle du donneur au sein des familles lesboparentales, à savoir la place que lui font les mères dans leur projet parental. Nous cherchons également à comprendre comment ce rôle s’actualise. Pour ce faire, nous avons interrogé neuf (9) couples lesbiens et les neuf (9) donneurs à l’origine de leur projet parental. Les résultats présentent un niveau différencié d’implication des donneurs en fonction de la mise à distance ou de l’appropriation par les mères du schéma nucléaire traditionnel.

Ce mémoire analyse les articles et les publicités représentant les loisirs diffusés dans la Revue moderne et Châtelaine après la Seconde Guerre mondiale. Phénomène grandissant à l’ère de la consommation de masse, les loisirs sont de plus en plus populaires et l’abondance de leurs représentations illustre leur importance, notamment auprès des familles de la classe moyenne qui bénéficient de plus en plus de ressources pour en profiter. Ces représentations, utilisées pour faire la promotion de produits à usage quotidien, de produits du tourisme ou de conseils pour les vacances, semblent à première vue illustrer certains aspects de la vie quotidienne des Québécois, mais diffusent principalement certaines valeurs encouragées par la société d’après-guerre. En effet, plusieurs stéréotypes de genre peuvent être observés, dont celui de la mère ménagère et celui du père pourvoyeur. En plus d’être véhiculées dans les médias et par l’ensemble des acteurs sociaux de l’époque, les valeurs associées à la domesticité, la féminité, la masculinité et à la famille nucléaire se retrouvent jusque dans les loisirs, accessibles à de plus en plus de familles.

Les westerns de l’âge d’or étasunien (1948-1962) mettent en scène un "gender trouble" en créant des personnages de femmes et d’hommes qui empruntent les uns et les autres aux caractéristiques genrées associées par la société nord-américaine et occidentale en général aux genres binaires du féminin et du masculin. Ce trouble genré se développe entre autres par la volonté de trois hommes de recréer la cellule familiale nucléaire conventionnelle dans "Red River" (1948) de Howard Hawks, par le rapport de peur et d’oppression du groupe social sur les individus dans "High Noon" (1952) de Fred Zinnemann et "Johnny Guitar" (1954) de Nicholas Ray, ainsi que par la rencontre opposant l’homme de l’Est et l’homme de l’Ouest dont les idéologies et les valeurs divergent dans "The Man Who Shot Liberty Valance" (1962) de John Ford.

La búsqueda de alternativas que frenen eficazmente las consecuencias del conflicto armado en Colombia, es una labor que algunos ex mandatarios han liderado debido a los perjuicios que ocasionan no solo al núcleo familiar, sino en gran medida a la mujer. Ella es una de las víctimas de la violencia por ser líder comunitaria, por dirigir una fundación, ó por tener algún familiar vinculado a la Policía, el Ejército o a un grupo al margen de la ley. Todas estas condiciones, propias del conflicto armado, vulneran los derechos de la mujer pero, al mismo tiempo, impulsa a que ellas hagan uso de sus experiencias negativas con el fin de minimizar las problemáticas sociales y el impacto de la violencia en sus vidas. Este trabajo periodístico desarrolla cuatro reportajes mediante el uso de cuestionarios con preguntas a profundidad y es complementado con un video, con el propósito de destacar la lucha y la labor de cuatro gestoras de paz, quienes utilizan diferentes estrategias como la creación de movimientos sociales, la utilización de marchas o plantones, la divulgación de talleres sobre el perdón y reconciliación, o la construcción de un Banco del Trueque, para contrarrestar la violencia de la que son objeto. Se concluye que el papel de la mujer, como constructora de paz, no puede ser clasificado en un único objetivo, sino debe entenderse como la suma de múltiples prioridades debido a que el conflicto armado afecta de diferente manera a la mujer, a su familia, y a su comunidad. Su capacidad de liderazgo y de entrega hace que su labor sea de suma importancia pues su huella beneficia a toda la sociedad. Hay que destacar que el desafío de esta nueva gestora consiste en visibilizar su trabajo para ganar mayor reconocimiento político, promover su participación en acuerdos de paz, y minimizar las situaciones que ponen en riesgo su vida y la de sus organizaciones.

La doctrina cristiana protestante impone y promueve un modelo de familia nuclear (Padre-Madre e hijo)que genera un tipo de exclusión social a creyentes con familia monoparental como las madres cabeza de familia. Como consecuencia, las adeptas generan estrategias de negociación de empoderamiento o sumisión entre su doctrina y su realidad familiar.
