The nature of the pre-morphotropic phase boundary (MPB) cubic-like state in the lead-free piezoelectric ceramics (1-x)Na1/2Bi1/2TiO3-(x)BaTiO3 at x similar to 0.06 has been examined in detail by electric field and temperature dependent neutron diffraction, x-ray diffraction, dielectric and ferroelectric characterization. The superlattice reflections in the neutron diffraction patterns cannot be explained with the tetragonal P4bm and the rhombohedral (R3c) phase coexistence model. The cubic like state is rather a result of long ranged modulated complex octahedral tilt. This modulated structure exhibits anomalously large dielectric dispersion. The modulated structure transforms to a MPB state on poling. The field-stabilized MPB state is destroyed and the modulated structure is restored on heating the poled specimen above the Vogel-Fulcher freezing temperature. The results show the predominant role of competing octahedral tilts in determining the nature of structural and polar states in Na1/2Bi1/2TiO3-based ferroelectrics. (C) 2013 AIP Publishing LLC.

The(1-x) BiFeO3-(x) PbTiO3 solid solution exhibiting a Morphotropic Phase Boundary (MPB) has attracted considerable attention recently because of its unique features such as multiferroic, high Curie point (T-C similar to 700 degrees C) and giant tetragonality (c/a -1 similar to 0.19). Different research groups have reported different composition range of MPB for this system. In this work we have conclusively proved that the wide composition range of MPB reported in the literature is due to kinetic arrest of the metastable rhombohedral phase and that if sufficient temperature and time is allowed the metastable phase disappears. The genuine MPB was found to be x=0.27 for which the tetragonal and the rhombohedral phases are in thermodynamic equilibrium. In-situ high temperature structural study of x=0.27 revealed the sluggish kinetics associated with the temperature induced structural transformation. Neutron powder diffraction study revealed that themagnetic ordering at room temperature occurs in the rhombohedral phase. The magnetic structure was found to be commensurate G-type antiferromagnetic with magnetic moments parallel to the c-direction (of the hexagonal cell). The present study suggests that the equilibrium properties in this solid solution series should be sought for x=0.27.

The nature of the stress and electric field driven structural and microstructural transformations in the morphotropic phase boundary (MPB) compositions of the high Curie point piezoelectric system BiScO3-PbTiO3 has been examined by ex situ based techniques. Using a powder poling technique, which is based on the concept of exploiting the irreversible structural change that occurs after the application of a strong electric field and stress independently, it was possible to ascertain that both moderate stress and electric field induce identical structural transformation-a fraction of the monoclinic phase transforms irreversibly to the tetragonal phase. Moreover, analysis of the dielectric response before and after poling revealed a counterintuitive phenomenon of poling induced decrease in the spatial coherence of polarization for compositions around the MPB and not so for compositions far away from the MPB range. Exploiting the greater sensitivity of this technique, we demonstrate that the criticality associated with the interferroelectric transition spans a wider composition range than what is conventionally reported in the literature based on bulk x-ray/neutron powder diffraction techniques.

A `powder-poling' technique was developed to study electric field induced structural transformations in ferroelectrics exhibiting a morphotropic phase boundary (MPB). The technique was employed on soft PZT exhibiting a large longitudinal piezoelectric response (d(33) similar to 650 pCN(-1)). It was found that electric poling brings about a considerable degree of irreversible tetragonal to monoclinic transformation. The same transformation was achieved after subjecting the specimen to mechanical stress, which suggests an equivalence of stress and electric field with regard to the structural mechanism in MPB compositions. The electric field induced structural transformation was also found to be accompanied by a decrease in the spatial coherence of polarization.

A comparative study of field-induced domain switching and lattice strain was carried out by in situ electric-field-dependent high-energy synchrotron x-ray diffraction on a morphotropic phase boundary (MPB) and a near-MPB rhombohedral/pseudomonoclinic composition of a high-performance piezoelectric alloy (1-x) PbTiO3-(x)BiScO3. It is demonstrated that the MPB composition showing large d(33) similar to 425 pC/N exhibits significantly reduced propensity of field-induced domain switching as compared to the non-MPB rhombohedral composition (d(33) similar to 260 pC/N). These experimental observations contradict the basic premise of the martensitic-theory-based explanation which emphasizes on enhanced domain wall motion as the primary factor for the anomalous piezoelectric response in MPB piezoelectrics. Our results favor field-induced structural transformation to be the primary mechanism contributing to the large piezoresponse of the critical MPB composition of this system.

Purpose: Systemic exposure to parabens in the neonatal population, in particular propyl-parabens (PPB), remains a concern. Blood concentrations and kinetics of methyl-parabens (MPB) and PPB were therefore determined in neonates receiving medicines containing these excipients.
Methods: A multi-centre, non-interventional, observational study of excipient-kinetics in neonates. ‘Dried Blood Spot’ samples were collected opportunistically at the same time as routine samples and the observations modelled using a non-linear mixed effects approach.
Results: A total of 841 blood MPB and PPB concentration data were available for evaluation from 181 pre- and term-neonates. Quantifiable blood concentrations of MPB and PPB were observed in 99% and 49% of patients, and 55% and 25% of all concentrations were above limit of detection (10 ng/ml), respectively. Only MPB data was amenable to modelling. Oral bioavailability was influenced by type of formulation and disposition was best described by a two compartment model with clearance (CL) influenced by post natal age (PNA); CLPNA<21 days 0.57 versus CLPNA>21days 0.88 L/h.
Conclusions: Daily repeated administration of parabens containing medicines can result in prolonged systemic exposure to the parent compound in neonates. Animal toxicology studies of PPB that specifically address the neonatal period are required before a permitted daily exposure for this age group can be established.

Os estuários são ambientes complexos, biologicamente diversos e muito importantes no que respeita à produtividade primária. As zonas intertidais destes ecossistemas são ocupadas por organismos que possuem uma elevada capacidade de sobrevivência e adaptação face às variadas e rápidas alterações nos factores ambientais (tais como temperatura, salinidade, conteúdo hídrico, etc.). As cadeias tróficas com origem no ecossistema estuarino bentónico são essencialmente herbívoras, regulando o fluxo de energia desde o fundo sedimentar e através do ecossistema. Nas áreas estuarinas intertidais a produção primária é essencialmente suportada pelo microfitobentos (MPB). Estas comunidades de microalgas bênticas constituem uma importante fonte de matéria orgânica e são por si só a principal fonte alimentar para as populações de Hydrobia. Neste contexto, a interacção MPB - Hydrobia é um modelo-chave na investigação da cadeia trófica estuarina de origem bentónica, actuando como um importante canal de transporte de energia para os níveis tróficos superiores, especialmente se considerarmos que Hydrobia é uma importante presa para peixes, aves e caranguejos. O presente estudo tem por objectivos gerais: i) a investigação do controlo ambiental (particularmente da luz e do teor em água do sedimento) e endógeno na migração vertical do MPB e ii) a identificação e potencial utilização de marcadores tróficos (pigmentos e ácidos gordos) úteis à investigação da interacção MPB – Hydrobia em laboratório e em condições naturais, considerando a existência de uma elevada plasticidade trófica por parte da Hydrobia e a elevada densidade populacional que estes organismos podem apresentar. A primeira fase de investigação resultou na comparação do papel dos estímulos ambientais e do controlo endógeno nos padrões de comportamento migratório vertical do microfitobentos, demonstrando a existência de um controlo essencialmente endógeno na formação e desintegração do biofilme superficial. A regulação e manutenção da biomassa à superfície do sedimento são claramente controladas pela variação dos factores ambientais, em especial da luz, cuja presença é essencial à formação total do biofilme microalgal à superfície do sedimento intertidal. Foi proposta uma nova abordagem metodológica com vista à estimativa nãodestrutiva do teor de água de sedimentos intertidais vasosos , possibilitando o estudo da influência da acção do vento no conteúdo hídrico dos sedimentos e o consequente impacto da dessecação na comunidade microfitobêntica. Observou-se que a dessecação provoca efeitos limitantes não só na biomassa superficial mas também na actividade fotossintética dos biofilmes microfitobênticos, conduzindo à diminuição da produtividade primária. No que respeita à dinâmica trófica da interacção MPB - Hydrobia foi estabelecido o uso do pigmento feoforbide a, quantificado nas partículas fecais da fauna, como marcador trófico que permite estimar a quantidade de biomassa de microalgas (clorofila a) incorporada pelos organismos animais.Para tal foi investigada e comprovada a existência de uma relação significativa entre a concentração de feopigmentos excretados e a concentração de clorofila a ingerida. Estes estudos foram desenvolvidos numa primeira fase à escala diária, considerando os efeitos dos ciclos sazonais, dia-noite e maré, e depois com a validação em condições naturais, numa escala mensal. A taxa de ingestão média de indivíduos de H. ulvae varia ao longo do dia, com o máximo em torno dos períodos diurnos de maré baixa, o que pode estar relacionado com a disponibilidade de MPB. As taxas de ingestão (TI) de H. ulvae variam ainda em função da estação do ano (TI verão > TI primavera) e em função da densidade de indivíduos (> densidade, < ingestão). Verificou-se um efeito negativo na concentração de clorofila disponível após herbívoria independentemente da densidade de indivíduos. Finalmente, a comparação dos perfis de ácidos gordos de H. ulvae provenientes de diferentes habitats com os perfis de potenciais fontes alimentares permitiu demonstrar que os ácidos gordos são ferramentas úteis na identificação do habitat ocupado por estes organismos. No entanto, apesar da ocupação de diferentes habitats e da integração de múltiplas fontes de produção primária na sua dieta foram sempre observados significativos níveis de ácidos gordos específicos de microalgas (em particular diatomáceas), reforçando o papel importante das comunidades de microalgas bênticas na dieta das populações de H. ulvae.

Induced pluripotent stem cells (iPSc) have great potential for applications in regenerative medicine, disease modeling and basic research. Several methods have been developed for their derivation. The original method of Takahashi and Yamanaka involved the use of retroviral vectors which result in insertional mutagenesis, presence in the genome of potential oncogenes and effects of residual transgene expression on differentiation bias of each particular iPSc line. Other methods have been developed, using different viral vectors (adenovirus and Sendai virus), transient plasmid transfection, mRNA transduction, protein transduction and use of small molecules. However, these methods suffer from low efficiencies; can be extremely labor intensive, or both. An additional method makes use of the piggybac transposon, which has the advantage of inserting its payload into the host genome and being perfectly excised upon re-expression of the transposon transposase. Briefly, a policistronic cassette expressing Oct4, Sox2, Klf4 and C-Myc flanked by piggybac terminal repeats is delivered to the cells along with a plasmid transiently expressing piggybac transposase. Once reprogramming occurs, the cells are re-transfected with transposase and subclones free of tranposon integrations screened for. The procedure is therefore very labor intensive, requiring multiple manipulations and successive rounds of cloning and screening. The original method for reprogramming with the the PiggyBac transposon was created by Woltjen et al in 2009 (schematized here) and describes a process with which it is possible to obtain insert-free iPSc. Insert-free iPSc enables the establishment of better cellular models of iPS and adds a new level of security to the use of these cells in regenerative medicine. Due to the fact that it was based on several low efficiency steps, the overall efficiency of the method is very low (<1%). Moreover, the stochastic transfection, integration, excision and the inexistence of an active way of selection leaves this method in need of extensive characterization and screening of the final clones. In this work we aime to develop a non-integrative iPSc derivation system in which integration and excision of the transgenes can be controlled by simple media manipulations, avoiding labor intensive and potentially mutagenic procedures. To reach our goal we developed a two vector system which is simultaneously delivered to original population of fibroblasts. The first vector, Remo I, carries the reprogramming cassette and GFP under the regulation of a constitutive promoter (CAG). The second vector, Eneas, carries the piggybac transposase associated with an estrogen receptor fragment (ERT2), regulated in a TET-OFF fashion, and its equivalent reverse trans-activator associated with a positive-negative selection cassette under a constitutive promoter. We tested its functionality in HEK 293T cells. The protocol is divided in two the following steps: 1) Obtaining acceptable transfection efficiency into human fibroblasts. 2) Testing the functionality of the construct 3) Determining the ideal concentration of DOX for repressing mPB-ERT2 expression 4) Determining the ideal concentration of TM for transposition into the genome 5) Determining the ideal Windows of no DOX/TM pulse for transposition into the genome 6) 3, 4 and 5) for transposition out of the genome 7) Determination of the ideal concentration of GCV for negative selection We successfully demonstrated that ENEAS behaved as expected in terms of DOX regulation of the expression of mPB-ERT2. We also demonstrated that by delivering the plasmid into 293T HEK cells and manipulating the levels of DOX and TM in the medium, we could obtain puromycin resistant lines. The number of puromycin resistant colonies obtained was significantly higher when DOX as absent, suggesting that the colonies resulted from transposition events. Presence of TM added an extra layer of regulation, albeit weaker. Our PCR analysis, while not a clean as would be desired, suggested that transposition was indeed occurring, although a background level of random integration could not be ruled out. Finally, our attempt to determine whether we could use GVC to select clones that had successfully mobilized PB out of the genome was unsuccessful. Unexpectedly, 293T HEK cells that had been transfected with ENEAS and selected for puromycin resistance were insensitive to GCV.

The purpose of the present work is to report studies on structural phase transition for PMN-xPT ferroelectric, with melt PbTiO3 composition around the MPB (x = 0.35 mol %), using infrared spectroscopy technique. The study was centered on monitoring the behavior of the 1-(NbO), 1-(TiO) and 1-(MgO) stretching modes as a function of temperature. The increasing as a function of temperature for 1-(TiO) and 1-(MgO) modes, observed between 230 and 300 K, can be related to the monoclinic (MC) + tetragonal (T) phase coexistence in the PMN-PT.

Este artigo analisa as características de experimentação e criatividade em composições de Walter Franco, nos discos Ou não (1973) e Revolver (1975). Quatro aspectos serão observados: a letra, os elementos musicais (instrumentação e arranjos), as relações entre letra, arranjo e performance, e o design das capas dos discos em diálogo com a natureza conceitual das canções.

A teoria Neo-Institucional define Campos Organizacionais como espaços onde atores compartilham regras, esquemas interpretativos e consentem com as instituições estabelecidas. A partir dessa perspectiva, espera-se que as formas sociais sejam adotadas e difundidas pelos membros do campo. Essa visão de campo tem sido expandida para dar conta dos conflitos e inovação. A partir da investigação do campo de Jazz e da MPB, aborda-se a tranformação em campos de produção musical. Em primeiro lugar, investiga-se as relações teóricas entre o conceito de campo e redes sociais, atavés da aproximação das teorias desenvolvidas principalmente por Bourdieu e White. Evitando-se a síntese desses dois corpos teóricos, sugere-se o potencial de complementaridade entre as duas abordagens. Assim, sugere-se que mudanças sociais podem ser acompanhadas pelo descasamento entre a estrutura de redes sociais e a concentração de oportunidades. A partir do mapeamento das redes sociais do campo de Jazz, de 1930 a 1969, obtem-se uma topografia dos líderes de banda e seus respectivos estilos. Verifica-se que o lugar de emersão de novos estilos pode ser explicado pela evolução da estrutura do campo. As tranformações internas a um campo impactam as carreiras dos músicos. Constroi-se uma carreira “típica” de um músico de jazz, que sugere uma trilha a ser percorrida para que se alcance o sucesso. Em contrapartida, a mudança na estrutura e lógica do campo de Jazz levou ao favorecimento de músicos que não seguiram a carreira típica. Essa contraposição é importante para aproximar a análise à ação dos individuos co-responsáveis pela mudança na lógica do campo. A institucionalização de um campo possibilita a definição daqueles estilos que são legítimos, em contraposição com aqueles que são segregados. Dessa forma, a permeabilidade de um campo conjuga a a geração autônoma de normas com elementos externos. Novamente, essa permeabilidade é reflexo da transformação da estrutura e lógica do campo. A partir da penetração da Bossa Nova no campo de Jazz americano, analisou-se a participação dos críticos na avaliação, legitimação e tradução da Bossa Nova. Essa influência cruzada foi acompanhada pela redifinição das fronteiras entre os campos. A comunicação entre campos distintos surge como um fator crucial na explicação da emersão de novos campos. A comunicação entre os campos de Música Popular Brasileira e Música Pop (principalmente Jovem Guarda) foi realizada pelos músicos do movimento Tropicalista. A criação de uma nova posição possibilitou a articulação entre dois espaços sociais que encontravam-se polarizados. Percebe-se o papel do conflito nessas dinâmicas: o conflito dentro do campo do jazz permitiu a sua abertura e emersão de uma pluralidade de estilos que articulavam de formas distintas a tradição do jazz e as influências externas. A polarização dentro do campo da MPB criou a possibilidade de criação de novas posições internas e articulações externas. As mudanças nessas lógicas foram concomitantes com o desenho de novos contornos do campo e novas trajetórias das carreiras. Em contrapartida, esses conflitos revelaram elementos compartilhados que se tornaram institucionalizados e aceitos por grupos opostos.

In a previous study, we showed that purified commercial esterase activity can be detected in a chemiluminescent assay based on the hydrolysis of 2-methyl-1-propenylbenzoate (MPB) to 2-methyl-1-propenol, which is subsequently oxidized by the horseradish peroxidase (HRP)-H2O2 system. The purpose of this study was to verify the applicability of this assay to human serum. The existence of an esterase activity capable of hydrolysing MPB is indicated by the fact that the MPB-scruin-HRP-H2O2 System consumes oxygen and emits light. Both signals were abolished by prior serum heat inactivation and were preserved when serum was stored at less than or equal to4 degreesC. Addition of aliesterase inhibitors, such as fluoride ion and trichlorfon or the cholinesterase inhibitor eserine, totally prevents light emission. The butyrylcholinesterase-specific substrate benzoylcholine causes a delay in both O-2 uptake and light emission, while the specific acetylcholinesterase substrate, acetyl-beta -methylcholine, had practically no effect. Purified butyrylcholinesterase, but not acetylcholinesterase, triggered light emission. The finding that butyryleholinesterase is responsible for the hydrolysis of MPB in serum should serve as the basis for the development of a specific chemiluminescent assay for this enzyme. The discovery of a new monoclinic phase in the PbZr1-xTixO3 (PZT) system in the vicinity of the morphotropic phase boundary (MPB), previously considered as a region where the rhombohedral and tetragonal phases of PZT coexist, was recently reported. Investigations of this new phase were reported using different techniques such as high-resolution synchrotron x-ray powder diffraction and Raman spectroscopy. The main objective has been to define a new phase diagram of PZT. In this context, infrared spectroscopic studies were performed in the vicinity of the MPB and studies were initially centred on a PZT sample with x = 0.49 mol% Ti content. Results suggested that the monoclinic --> tetragonal phase transition occurs at 237 K, confirming the use of IR as a useful technique to investigate this phase transition.
