Extracts of the spice ginger (Zingiber officinale Roscoe) are rich in gingerols and shogaols, which exhibit antioxidant, anti-inflammatory, antifungal, anti mycobacterial, and anticarcinogenic proprieties. The present study evaluated the chemoprotective effects of a ginger extract on the DNA damage and the development of bladder cancer induced by N-butyl-N-(4-hydroxibutyl) nitrosamine (BBN)/N-methyl-N-nitrosourea (MNU) in male Swiss mice. Groups G1-G3 were given 0.05% BBN in drinking water for 18 weeks and four i.p. injections of 30 mg/kg body weight MNU at 1, 3, 10, and 18 weeks. Group G4 and G5 received only the BBN or MNU treatments, respectively, and groups G6 and G7 were not treated with BBN or MNU. Additionally, Groups G2, G3, and G6 were fed diets containing 1, 2, and 2% ginger extract, respectively, while Groups G1, G4, G5, and G7 were fed basal diet. Samples of peripheral blood were collected during the experiment for genotoxicity analysis; blood collected 4 hr after each MNU dose was used for the analysis of DNA damage with the Comet assay (assay performed on leukocytes from all groups), while reficulocytes collected 24 hr after the last MNU treatment of Groups G5-G7 were used for the micronucleus assay. At the end of the experiment, the urinary bladder was removed, fixed, and prepared for histopathological, cell proliferation, and apoptosis evaluations. Ginger by itself was not genotoxic, and it did not alter the DNA damage levels induced by the BBN/MNU treatment during the course of the exposure. The incidence and multiplicity of simple and nodular hyperplasia and transitional cell carcinoma (TCC) were increased by the BBN/MNU treatment, but dietary ginger had no significant effect on these responses. However, in Group G2 (BBN/MNU/2% ginger-treated group), there was an increased incidence of Grade 2 TCC. The results suggest that ginger extract does not inhibit the development of BBN-induced mouse bladder tumors.

An initiation-promotion medium-term bioassay for detection of chemical carcinogens, developed in the male F344 rat, uses 0.1% N-bis(2-hydroxypropyl)nitrosamine (DHPN) among five genotoxic chemicals for the initiation of carcinogenesis in multiple organs. To establish this bioassay in the Wistar strain, the effects of two dose levels of DHPN were evaluated on the main DHPN rat target organs: lung, thyroid gland, kidneys and liver. Four groups of male and female animals were studied: Control--untreated group; Multi-organ initiated group (also referred to as DMBDD, based on the initials of the five initiators)-treated sequentially with N-diethylnitrosamine (DEN, i.p.), N-methyl-N-nitrosourea (MNU, i.p.), N-butyl-N-(4-hydroxy butyl)nitrosamine (BBN, drinking water), N, N'-dimethylhydrazine (DMH, s.c.) and DHPN (drinking water) for 4 weeks; a third group treated with 0.1% DHPN in drinking water for 2 weeks and the last group treated with 0.2% DHPN in drinking water for 4 weeks. The animals were sacrificed after 30 weeks. DHPN at 0.2% induced preneoplasia in the liver and kidneys of rats of both sexes, the number and area of the putative preneoplastic liver glutathione S-transferase-positive hepatocyte foci being significantly increased in these animals. It also induced benign and malignant tumors in female and in male rats. However, there was no relationship between the increased incidence of preneoplastic lesions and tumor development in the 0.2% DHPN-exposed groups of both sexes. DHPN at 0.1% induced only a few preneoplastic lesions in the liver and kidney and no tumors in both male and female rats. A clear dose and sex-related carcinogenic activity of DHPN was registered, although Wistar rats of both sexes showed a relative resistance to the carcinogenic activity of this compound.

Purpose: We compared and characterized the effects of intravesical bacillus Calmette-Guerin and/or staphylococcal enterotoxin B for nonmuscle invasive bladder cancer.Materials and Methods: A total of 75 female Fisher 344 rats were anesthetized. of the rats 15 received 0.3 ml saline (control) and 60 received 1.5 mg/kg MNU (N-methyl-n-nitrosourea) intravesically every other week for 6 weeks. The rats were divided into 5 groups. The MNU and control groups received 0.3 ml saline. The bacillus Calmette-Guerin group received 10(6) cfu bacillus Calmette-Guerin. The staphylococcal enterotoxin B group received 10 mu g/ml staphylococcal enterotoxin B. The bacillus Calmette-Guerin plus staphylococcal enterotoxin B group received the 2 treatments simultaneously. Each group was treated intravesically for 6 weeks. At 15 weeks all bladders were collected for histopathological and immunological evaluation, and Western blot.Results: Papillary carcinoma (pTa) and high grade intraepithelial neoplasia (carcinoma in situ) were more common in the MNU group. Papillary hyperplasia was more common in the bacillus Calmette-Guerin and enterotoxin groups. Flat hyperplasia was more common in the bacillus Calmette-Guerin plus enterotoxin group. No significant toxicity was observed. The apoptosis and cellular proliferation indexes decreased in the bacillus Calmette-Guerin, enterotoxin and bacillus Calmette-Guerin plus enterotoxin groups compared to the MNU group. Intensified vascular endothelial growth factor, matrix metalloproteinase-9, Ki-67 and insulin-like growth factor receptor-1 immunoreactivity was verified in the MNU group, moderate in the bacillus Calmette-Guerin and enterotoxin groups, and weak in the bacillus Calmette-Guerin plus enterotoxin and control groups. In contrast, intense endostatin immunoreactivity was verified in the control and bacillus Calmette-Guerin plus enterotoxin groups.Conclusions: Bacillus Calmette-Guerin and staphylococcal enterotoxin B showed similar anti-angiogenic effects. Bacillus Calmette-Guerin plus enterotoxin treatment had additional activity compared to that of monotherapy. It was more effective in restoring apoptosis and balancing cellular proliferation, and it correlated with increased endostatin, and decreased vascular endothelial growth factor, matrix metalloproteinase-9, Ki-67 and insulin-like growth factor receptor-1 reactivity.

Background. Characterization of novel rodent models for prostate cancer studies requires evaluation of either spontaneous and carcinogen-induced tumors as well as tumor incidence in different prostatic lobes. We propose a new short-term rodent model of chemically induced prostate carcinogenesis in which prostate cancer progression occurs differentially in the dorsolateral and ventral lobes. Methods. Adult gerbils were treated with MNU alone or associated with testosterone for 3 or 6 months of treatment. Tumor incidence, latency, localization, and immunohistochemistry (AR, PCNA, smooth muscle α-actin, p63, MGMT, and E-cadherin) were studied in both lobes. Results. Comparisons between both lobes revealed that lesions developed first in the DL while the VL presented longer tumor latency. However, after 6 months, there was a dramatic increase in tumor multiplicity in the VL, mainly in MNU-treated groups. Lesions clearly progressed from a premalignant to a malignant phenotype over time and tumor latency was decreased by MNU + testosterone administration. Three-dimensional reconstruction of the prostatic complex showed that the DL developed tumors exclusively in the periurethral area and showed intense AR, PCNA, and MGMT immunostaining. Moreover, VL lesions emerged throughout the entire lobe. MNU-induced lesions presented markers indicative of an aggressive phenotype: lack of basal cells, rupture of the smooth muscle cell layer, loss of E-cadherin, and high MGMT staining. Conclusions. There are distinct pathways involved in tumor progression in gerbil prostate lobes. This animal provides a good model for prostate cancer since it allows the investigation of advanced steps of carcinogenesis with shorter latency periods in both lobes. O câncer gástrico continua sendo uma importante causa de morte dentre os tipos de câncer no Brasil e no mundo. A origem do câncer de estomago provém, assim como nos demais, de acúmulo de alterações genéticas. Portanto, se faz necessário saber quais alterações genéticas são importantes para desencadear a patogênese do câncer gástrico. O MNU, conhecido carcinógeno, quando ingerido de forma oral em doses determinadas desencadeia o desenvolvimento de adenocarcinoma gástrico do tipo intestinal, com aparecimento de estágios pré definidos característico deste tipo tumoral. Com base nestes conhecimentos, realizamos um experimento com 6 macacos da espécie Cebus apella induzidos à desenvolver câncer gástrico do tipo intestinal. Os animais ingeriam 16mg/kg de peso diariamente da droga, com desenvolvimento de lesões pré-neoplásicas em todos. Infelizmente, devido à toxicidade da droga, somente um animal sobreviveu ao tratamento e desenvolveu desenvolveu uma massa tumoral propriamente dita. Foram feitas avaliações periódicas dos animais, em dias pré-determinados (ao início, no 120º, 150º, 300º, 940º) com coletas de fragmentos da mucosa gástrica. Foram coletadas 20 amostras de tecido, distribuídas entre mucosa normal, gastrite, displasia, metaplasia e tumoral. Destas amostras extraímos DNA para as análises do gene CDH1. Não há na literatura sequencia deste gene para a espécie utilizada no estudo. Com este objetivo, utilizamos iniciadores construídos a partir de sequencias de CDH1 de Callithrix jacchus (espécie filogeneticamente) e seqüenciamos cerca de 342pb da região promotora de CDH1 de C. apella. As análises mostraram uma similaridade de 98% desta região com a dos humanos, com presença de vários sítios de ligação de fatores de transcrição (sp1, Ap2, NF-x, AREB6, Puf e CTF) além da presença do CAAT Box. Esta região ainda possui 30 sitios CpGs, o que indica que ela pode estar sofrendo regulação epigenética. Para se verificar como se encontrava o padrão de metilação desta região realizamos uma análise de MSP com iniciadores específicos e constatamos que houve predominância de alelos não metilados de CDH1 para todas as amostras pré neoplásicas e a amostra de adenocarcinoma encontra-se metilada. O que pode ser constatado com um ensaio de Imunohistoquímica, onde somente a amostra tumoral não expressava a proteína caderina. Desta maneira nossas análises sugerem que a metilação do gene CDH1 desempenha papel importante na tumorigênese gástrica em C. apella, e é um evento tardio, uma vez que não é observado nos outro tipos teciduais não tumorais E, partindo-se da similaridade entre as seqüências, podemos sugerir que o mesmo evento pode está ocorrendo em humanos. Esta metilação do promotor do CDH1 leva a um silenciamento deste gene o que desencadeia o estabelecimento de adenocarcinomas gástricos, fato apoiado pela literatura onde vários trabalhos apontaram que a hipermetilação do promotor da caderina está associada ao desenvolvimento do câncer gástrico.

Despite recent advances in understanding the biological basis ofprostate cancer (PCa), the management of this disease remains a challenge. Chemoprotective agents have been usedto protect against or eradicateprostatemalignancies. Here, we investigated the protective effect of -tocopherol on N-methyl-N-nitrosourea (MNU)-induced epithelial dysplasia in the rat ventral prostate (VP). Thirty-two male Wistar rats were divided into four groups (n=8): control (CT): healthy control animals fed a standard diet; control+-tocopherol (CT+T): healthy control animals without intervention fed a -tocopherol-enriched diet (20mg/kg); N-methyl-N-nitrosourea (MNU): rats that received a single dose of MNU (30mg/kg) plus testosterone propionate (100mg/kg) and were fed a standard diet; and MNU+-tocopherol (MNU+T): rats that received the same treatment of MNU plus testosterone and were fed with a -tocopherol-enriched diet (20mg/kg). After 4months, the VPs were excised to evaluate morphology, cell proliferation and apoptosis, as well as cyclooxygenase-2 (Cox-2), glutathione-S-transferase-pi (GST-pi) and androgen receptor (AR) protein expression, and matrix metalloproteinase-9 (MMP-9) activity. An increase in the incidence of epithelial dysplasias, such as stratified epithelial hyperplasia and squamous metaplasia, in the MNU group was accompanied by augmented cell proliferation, GST-pi and Cox-2 immunoexpression and pro-MMP-9 activity. Stromal thickening and inflammatory foci were also observed. The administration of a -tocopherol-enriched diet significantly attenuated the adverse effects of MNU in the VP. The incidence of epithelial dysplasia decreased, along with the cell proliferation index, GST-pi and Cox-2 immunoexpression. The gelatinolytic activity of pro-MMP-9 returned to the levels observed for the CT group. These results suggest that -tocopherol acts as a protective agent against MNU-induced prostatic disorders in the rat ventral prostate.

A exposição in utero a xenoestrógenos pode aumentar o risco de neoplasias de natureza endócrina na vida adulta. O Bisfenol A (BPA), componente de resinas e plástico, considerado xenoestrógeno e desregulador endócrino, tem sido investigado pelos seus potenciais efeitos adversos para a saúde humana. Como a Genisteína e o Indol-3-Carbinol possuem propriedades que podem inibir neoplasias de natureza endócrina, é possível que também atuem modulando/modificando os efeitos causados pela exposição gestacional ao BPA. O presente projeto teve como objetivos: (1) Avaliar os efeitos da exposição gestacional ao Bisfenol - A (BPA) sobre a morfogênese do útero e ovários na prole de fêmeas Sprague-Dawley (SD) da geração F1; (2) Avaliar se a exposição gestacional a genisteína e ao indol-3-carbinol altera os efeitos do BPA sobre sobre a morfogênese do útero e ovários na geração F1 e (3) avaliar os efeitos da exposição ao BPA, e às associações BPA e genisteína, BPA e indol-3-carbinol em relação à susceptibilidade a carcinogênese induzida pela N-Metil-N-Nitrosuréia (MNU). Portanto, fêmeas prenhas da linhagem SD foram divididas em 7 grupos experimentais e expostas ao Bisfenol A (BPA) (25 ou 250 ug/kg p.c.) DG 10 até o DG 21 (Moral et al. 2008), além de ração basal ou ração contendo genisteína (250 mg/kg) ou indol-3-carbinol (2000 mg/kg) durante toda a gestação. Parte da prole Fêmeas SD foi sacrificada parte no Dia Pós-Natal (DPN) 21 e parte ao final da 25ª semana após iniciação ou não com a MNU. Ao DPN 21 os ovários e útero foram removidos para contagem de folículos e morfometria, respectivamente. A prole restante de fêmeas recebeu uma única dose de MNU (50 mg/kg) ou solução de NaCl (1 ml/kg) no DPN 51 e foi sacrificada na 25ª semana após a aplicação de MNU ou de NaCl. Ovários e útero foi removidos para análises histológicas, incluindo a determinação de lesões proliferativas ...

Natural killer (NK) cell activity was evaluated after the initiation and promotion steps in a medium-term multi-organ bioassay for carcinogenesis. NK cell activity was assessed in vitro by Cr51 release assay at the 4th and 30th weeks of the experiment. Male Wistar rats were sequentially initiated with N-diethylnitrosamine (DEN i.p.), N-butyl-N-(4-hydroxybutyl)nitrosamine (BBN drinking water), N-methyl-N-nitrosourea (MNU i.p.), dihydroxy-di-N-propylnitrosamine (DHPN drinking water) and N,N'-dimethylhydrazine (DMH s.c.) at subcarcinogenic doses for 4 weeks (DMBDD initiation). One group was evaluated at the 4th week and the other was maintained without any further treatment until the 30th week. Two initiated groups were exposed through the diet to 2-acetylaminofluorene (2-AAF) or phenobarbital (PB), from the 6th until the 30th week. Five additional groups were studied to evaluate the effects of each initiator on NK activity. All groups submitted to initiation only, initiation plus promotion, or promotion only, developed significantly more preneoplastic lesions than the untreated control group. The main target organs for tumor development in the initiated animals were the liver and the colon, irrespective of treatment with 2-AAF or PB. NK cell activity was not affected by exposure to genotoxic carcinogens after initiation, at the 4th week. Treatments only with PB or 2-AAF did not change NK cell activity. However, decreased NK cell activity was registered in the group only initiated with DMBDD and in the group given DMBDD+2-AAF. This late depression of NK cell activity at the 30th week could be related to the production of suppressing molecules by the tumor cells.
