Enrofloxacin (ENR) is an antimicrobial used both in humans and in food producing species. Its control is required in farmed species and their surroundings in order to reduce the prevalence of antibiotic resistant bacteria. Thus, a new biomimetic sensor enrofloxacin is presented. An artificial host was imprinted in specific polymers. These were dispersed in 2-nitrophenyloctyl ether and entrapped in a poly(vinyl chloride) matrix. The potentiometric sensors exhibited a near-Nernstian response. Slopes expressing mV/Δlog([ENR]/M) varied within 48–63. The detection limits ranged from 0.28 to 1.01 µg mL−1. Sensors were independent from the pH of test solutions within 4–7. Good selectivity was observed toward potassium, calcium, barium, magnesium, glycine, ascorbic acid, creatinine, norfloxacin, ciprofloxacin, and tetracycline. In flowing media, the biomimetic sensors presented good reproducibility (RSD of ± 0.7%), fast response, good sensitivity (47 mV/Δlog([ENR]/M), wide linear range (1.0 × 10−5–1.0 × 10−3 M), low detection limit (0.9 µg mL−1), and a stable baseline for a 5 × 10−2 M acetate buffer (pH 4.7) carrier. The sensors were used to analyze fish samples. The method offered the advantages of simplicity, accuracy, and automation feasibility. The sensing membrane may contribute to the development of small devices allowing in vivo measurements of enrofloxacin or parent-drugs.

This work presents the development of a low cost sensor device for the diagnosis of breast cancer in point-of-care, made with new synthetic biomimetic materials inside plasticized poly(vinyl chloride), PVC, membranes, for subsequent potentiometric detection. This concept was applied to target a conventional biomarker in breast cancer: Breast Cancer Antigen (CA15-3). The new biomimetic material was obtained by molecularly-imprinted technology. In this, a plastic antibody was obtained by polymerizing around the biomarker that acted as an obstacle to the growth of the polymeric matrix. The imprinted polymer was specifically synthetized by electropolymerization on an FTO conductive glass, by using cyclic voltammetry, including 40 cycles within -0.2 and 1.0 V. The reaction used for the polymerization included monomer (pyrrol, 5.0×10-3 mol/L) and protein (CA15-3, 100U/mL), all prepared in phosphate buffer saline (PBS), with a pH of 7.2 and 1% of ethylene glycol. The biomarker was removed from the imprinted sites by proteolytic action of proteinase K. The biomimetic material was employed in the construction of potentiometric sensors and tested with regard to its affinity and selectivity for binding CA15-3, by checking the analytical performance of the obtained electrodes. For this purpose, the biomimetic material was dispersed in plasticized PVC membranes, including or not a lipophilic ionic additive, and applied on a solid conductive support of graphite. The analytical behaviour was evaluated in buffer and in synthetic serum, with regard to linear range, limit of detection, repeatability, and reproducibility. This antibody-like material was tested in synthetic serum, and good results were obtained. The best devices were able to detect 5 times less CA15-3 than that required in clinical use. Selectivity assays were also performed, showing that the various serum components did not interfere with this biomarker. Overall, the potentiometric-based methods showed several advantages compared to other methods reported in the literature. The analytical process was simple, providing fast responses for a reduced amount of analyte, with low cost and feasible miniaturization. It also allowed the detection of a wide range of concentrations, diminishing the required efforts in previous sample pre-treating stages.

A voltammetric biosensor for Ara h 6 (a peanut allergen) detection in food samples was developed. Gold nanoparticle-modified screen-printed carbon electrodes were used to develop a sandwich-type immunoassay using two-monoclonal antibodies. The antibody-antigen interaction was detected through the electrochemical detection of enzymatically deposited silver. The immunosensor presented a linear range between 1 and 100 ng/ml, as well as high precision (inter-day RSD ≤9.8 %) and accuracy (recoveries ≥96.7 %). The detection and quantification limits were 0.27 and 0.88 ng/ml, respectively. It was possible to detect small levels of Ara h 6 in complex food matrices.

This work aims to evaluate the feasibility of using image-based cytometry (IBC) in the analysis of algal cell quantification and viability, using Pseudokirchneriella subcapitata as a cell model. Cell concentration was determined by IBC to be in a linear range between 1 × 105 and 8 × 106 cells mL−1. Algal viability was defined on the basis that the intact membrane of viable cells excludes the SYTOX Green (SG) probe. The disruption of membrane integrity represents irreversible damage and consequently results in cell death. Using IBC, we were able to successfully discriminate between live (SG-negative cells) and dead algal cells (heat-treated at 65 °C for 60 min; SG-positive cells). The observed viability of algal populations containing different proportions of killed cells was well correlated (R 2 = 0.994) with the theoretical viability. The validation of the use of this technology was carried out by exposing algal cells of P. subcapitata to a copper stress test for 96 h. IBC allowed us to follow the evolution of cell concentration and the viability of copper-exposed algal populations. This technology overcomes several main drawbacks usually associated with microscopy counting, such as labour-intensive experiments, tedious work and lack of the representativeness of the cell counting. In conclusion, IBC allowed a fast and automated determination of the total number of algal cells and allowed us to analyse viability. This technology can provide a useful tool for a wide variety of fields that utilise microalgae, such as the aquatic toxicology and biotechnology fields.

Työn tavoitteena oli kehittää nopeasti konvergoiva kuorielementti epälineaarisesti joustavien kappaleiden analysointiin. Kuorielementti perustuu absoluuttisten solmukoordinaattien menetelmään ja se hyödyntää kaarevuuden kuvausta elastisten voimien määrityksessä. Kehitettyä elementtiä verrattiin kontinuumimekaniikalla kehitettyyn kuorielementtiin ja kaupallisen elementtimenetelmän kuorielementtiin. Yksinkertaisimman kuormitustapauksen tuloksia verrattiin teknisen taivutusteorian mukaiseen analyyttiseen ratkaisuun. Staattisten testien tulokset tässä työssä kehitetyllä kuorielementillä vastasivat hyvin kaupallisella elementtimenetelmällä saatuja tuloksia. Deformaatioiden ollessa geometrisesti lineaarisella alueella, kehitetyllä kuorielementillä saadut tulokset vastasivat paremmin sekä analyyttistä ratkaisua että kaupallisella elementtimenetelmällä saatuja tuloksia kuin aiemman kontinuumimekaniikkaan perustuvan kuorielementin tulokset. Kehitetyn kuorielementin ongelmana verrattuna kontinuumimekaniikkaan perustuvaan elementtiin on monimutkaisempi kinematiikan kuvaus. Tästä on seurauksena laskenta-ajan huomattava kasvaminen. Jatkossa kannattaisi keskittyä numeeristen ratkaisumenetelmien kehittämiseen.

A simple flow cell for potentiometric detection is described. It was assembled by making use of two perspex pieces fixed together by means of four screws, and allow the connection of plane membrane conventional electrodes to flow system. Details about its construction are presented. The device performance was evaluated by making use of a cyanide ion-selective electrode. The relative standard deviation was about 0.5% with a detection limit of 8.0 x 10-6 mol CN- dm-3. Under experimental conditions, the linear range was 10-5 to10-2 mol dm-3.

The present work proposes the application of the 4-Hidroxy-3-(2-hydroxynaphtylazo)-benzenesulphonic acid (C.I. 15670), Alizarine Violet N (AVN), as a reagent for direct aluminium determination using molecular absorption spectrophotometry in the presence of tensoatives. Al(III) cation reacts with AVN in pH 9.4, forming a red complex, stable for at least 24 hours, with absorption minimum at 607nm and, against a reagent blank, (epsiloncomplex - epsilonreagent) = -2.71x10(4) L.mol-1.cm-1. The reaction occurs in the presence of a Triton-X100 and CTAB tensoatives mixture, in the presence of EDTA. Al(III) determination is possible in the linear range of 50 up to 400ng.mL-1, with a detection limit of 41 ng.mL-1.

In the present work four different analytical methodologies were studied for the determination of iron and titanium in Portland cement. The cement samples were dissolved with hot HCl and HF, being compared Fe and Ti concentrations through four analytical methods: molecular absorption spectrophotometry using the reagents 1,2-hydroxybenzene-3,5-disulfonic acid (Tiron) and the 5-chloro-salicylic acid (CSA), inductively coupled plasma atomic emission spectrometry (ICP-AES) and flame atomic absorption spectrophotometry (FAAS). In the spectrophotometric determinations were studied pH conditions, reagents addition order, interferences, amount of reagents, linear range and stability of the system. In the techniques of ICP-AES and FAAS were studied the best lines, interferences, sensibility and linear range. The obtained results were compared and the agreement was evaluated among the methods for the determination of the metals of interest.

This work presents an alternative method for determination of the herbicides tebuthiuron and hexazinone in ground water. The extraction was made with dichloromethane and the analyses by high performance liquid chromatography (HPLC), using reversed-phase column, C-18, mobile phase methanol/water 50:50, v/v, detection and quantification at 247 nm. The following validation parameters were obtained: limit of detection of method 0.02 and 0.03 µg L-1, limit of quantification of method 0.07 and 0.09 µg L-1; linear range limit of quantification of instrument - 300 µg L-1 (r² > 0.998); recoveries from 90.3 to 108.2% and 90.3 to 101.6%; intermediary precision (%RSD) < 8 and < 6%, for hexazinone and tebuthiuron, respectively. The method showed to be efficient and reliable for determination of the herbicides in ground water.

A method for determining copper by solid phase spectrophotometry (SPS) was optimized using the Doehlert design. Copper(II) was sorbed on a styrene-divinylbenzene anion-exchange resin as a Cu(II)-1-(2-pyridylazo)-2-naphthol (PAN) complex, at pH 7.0. Resin phase absorbances at 560 and 800 nm were measured directly. The detection limit was found to be 2.5 µg L-1. The relative standard deviation on ten replicate determinations of 10 µg Cu(II) in 1000 mL samples was 1.1%. The linear range of the determination was 5.0-100 µg L-1. The method was applied successfully to the determination of Cu(II) in natural water and vegetable samples.

Two spectrophotometric methods are described for the simultaneous determination of ezetimibe (EZE) and simvastatin (SIM) in pharmaceutical preparations. The obtained data was evaluated by using two different chemometric techniques, Principal Component Regression (PCR) and Partial Least-Squares (PLS-1). In these techniques, the concentration data matrix was prepared by using the mixtures containing these drugs in methanol. The absorbance data matrix corresponding to the concentration data matrix was obtained by the measurements of absorbances in the range of 240 - 300 nm in the intervals with Δλ = 1 nm at 61 wavelengths in their zero order spectra, then, calibration or regression was obtained by using the absorbance data matrix and concentration data matrix for the prediction of the unknown concentrations of EZE and SIM in their mixture. The procedure did not require any separation step. The linear range was found to be 5 - 20 µg mL-1 for EZE and SIM in both methods. The accuracy and precision of the methods were assessed. These methods were successfully applied to a pharmaceutical preparation, tablet; and the results were compared with each other.

In the present work, a method was developed and validated for the quantification of benzyl isothiocyanate (BITC) in the fruits of Carica papaya. The quantification of this compound was carried out by gas chromatography (GC) with selective detectors - nitrogen phosphorus detector (NPD) and flame photometric detector (FPD). The performance of these detectors showed a higher sensitivity of the NPD with a broader linear range of detection. The LOD/LOQ were 0.038/0.100 µg/mL for NPD and 5.78/19.29 µg/mL for FPD. The recovery of the method for BITC was 90,64%. An average value of BITC concentration in all the analyzed samples was 16,23 µg BITC/g.

This paper reports an analytical method for the determination of ambroxol in micellar medium by spot test-diffuse reflectance spectroscopy. The reflectance measurements were performed analyzing the colored compound (λ= 520 nm) produced from the reaction between ambroxol and p-dimethylaminocinnamaldehyde on the surface filter paper. The linear range was from 1.21 × 10"3 to 9.65 × 10"3 mol L-1 (500 - 4000 μg mL-1). The limit of detection and quantification were 3.50 x 10-4 mol L-1 (145 μg mL-1) and 1.16 x 10-3 mol L-1 (481 μg mL-1), respectively. Five commercial samples were analysed and the results obtained by the proposed method were in good agreement with those obtained by the literature method at 95% confidence level.

The present work purposes the preparation of a silica gel sorbent organically modified with 2-aminoethyl-3-aminobutylmethyldimethoxysilane (AAMDMS) and imprinted with Cu2+ ions by means surface imprinting technique and its use for selective on-line sorbent preconcentration of Cu2+ ions with further UV-VIS spectrophotometric determination by flow injection analysis. The Cu2+-imprinted silica gel, when compared with non imprinted silica gel and silica gel, showed from the binary mixture of Cu2+/Ni2+ relative selectivity coefficient (k') of 6.84 and 5.43 and 6.64 and 19.83 for the mixture Cu2+/Pb2+, thus demonstrating higher selectivity of Cu2+-imprinted silica gel towards Cu2+ ions. Under optimized condition, the on-line preconcentration method provided detection limit of 3.4 μg L-1 and linear range ranging from 30.0 up to 300.0 μg L-1 (r = 0.995). The accuracy of method was successfully assessed by analyzing different kind of spiked water samples with recovery values ranging from 92.2 up to 103.0%.

A simple and more environmentally friendly method by combined spot test-diffuse reflectance spectroscopy for determining metoclopramide in pharmaceutical formulations is described. The method is based on the reaction between metoclopramide and p-dimethylaminocinnamaldehyde, in the presence of HCl, producing a colored compound (λmáx = 580 nm) on the filter paper. The linear range was from 5.65 x 10-4-6.21x10-3 mol L-1 (r = 0.999). The limit of detection was 1.27 x 10-4 mol L-1. The proposed reflectometric method was applied successfully to the determination of metoclopramide in pharmaceuticals and it was favorably compared with the Brazilian or British Pharmacopoeia methods at 95% confidence level.
