The control of parasitic diseases in small ruminants is mainly done with the use of synthetic anthelmintics. However, incorrect and indiscriminate use of these products has caused the emergence of parasite resistance. Plants with anthelmintic activity are used in folk veterinary medicine, but it is necessary to investigate and scientifically validate low-cost phytotherapeutic alternatives for future use to control gastrointestinal nematodes in small ruminants by family farmers. Thus, the aim of this study was to evaluate the in vitro anthelmintic effect of plant extracts from Melia azedarach and Trichilia claussenii by the egg hatch test (EHT) and larval development test (LDT) against sheep gastrointestinal nematodes. The hexane extract of M. azedarach fruits was extracted through cold percolation and the methanol extract of T. claussenii leaves was obtained by extraction at room temperature in solvents in order of increasing polarity. The efficacy results were analyzed using the Probit program of SAS. The M. azedarach extract showed a LC50 of 572.2 mu g/mL and LC99 of 1137.8 mu g/mL in the EHT, and LC50 of 0.7 mu g/mL and LC99 of 60.81 mu g/mL in the LDT. In turn, the T. claussenii extract presented a LC50 of 263.8 mu g/mL and LC99 of 522.5 mu g/mL in the EHTand LC50 of 1.11 mu g/mL and LC99 of 26.4 mu g/mL in the LDT. Comparing the extracts of the species from the Meliaceae family, T. claussenii showed greater anti-parasite potential in vitro than M. azedarach. However, studies on the isolated compounds, toxicity and administration forms to animals are also needed to validate low-cost alternative herbal remedies for use to control gastrointestinal nematodes by family farmers. The development of resistance to anthelmintics has prompted research into alternative methods of controlling intestinal nematodes in ruminants. This study aimed to assess the activity of Ananas comosus on Haemonchus contortus in Santa Inês sheep. The aqueous extract of pineapple skin (AEPS), bromelain from pineapple stems (B4882) and residue from pineapple processing was evaluated in in vitro and in vivo tests. The enzymatic activity of substances was analyzed by the azocasein method. The egg hatch test (EHT) and larval development test (LDT) were performed using the Embrapa2010 isolate of H. contortus. In the in vivo test, 36 sheep artificially infected with H. contortus were divided into six groups: G1: 2g/kg BW of the aqueous extract administered for three days; G2: 2g/kg BW of the industrial pineapple residue for 60 days; G3: 180mg/animal of bromelain in a single dose; G4: negative control I; G5: positive control (levamisole phosphate); and G6: negative control II. The eggs per gram (EPG) in the feces were counted till 28 days after treatment. LC50 and LC90 were obtained by the probit procedure, while the in vivo test results were analyzed by GLM. The aqueous extract in the in vitro and in vivo test, the bromelain and industrial residue presented 0.102, 0.157, 1.864 and 0.048 enzyme units/mL, respectively. In the egg hatch test, the LC50 and LC90 were respectively 31 and 81mg/mL for the aqueous extract and 0.50 and 2mg/mL for bromelain. In the larval development test, the LC50 and LC90 were respectively 1.7 and 7.3mg/mL for the aqueous extract and 0.019 and 0.086mg/mL for bromelain. In the in vivo test, the general efficacies of the treatments in relation to the negative control were 22.6%, 42.2%, 3.65% and 89% for the aqueous extract, industrial pineapple residue, bromelain and positive control respectively. The transformed EPG values were 3.19±0.59, 3.32±0.25, 2.85±0.66, 3.44±0.50, 2.28±0.93 and 2.75±0.94 for the aqueous extract, industrial residue, bromelain, negative control I, positive control and negative control II respectively. The results for all the treated groups differed significantly (p<0.05) from the positive control, and although the residue presented efficacy of 42.2%, there was no statistical difference (p>0.05) in relation to the negative control. Therefore, both the aqueous extract and bromelain were effective in vitro, but showed reduced anthelmintic efficacy in vivo. For the pineapple residue, the 42.2% in vivo efficacy in reducing the EPG and the possibility of reducing environmental contamination through reuse of industrial residue indicate it can also be useful for control of this parasite. © 2013 Elsevier B.V.

Os transtornos de ansiedade apresentam a maior incidência na população mundial dentre os transtornos psiquiátricos, e a eficácia clínica das drogas ansiolíticas é baixa, em parte devido ao desconhecimento acerca das bases neuroquímicas desses transtornos. Para uma compreensão mais ampla e evolutivamente substanciada desses fenômenos, a utilização de espécies filogeneticamente mais antigas pode ser uma aproximação interessante no campo da modelagem comportamental; assim, sugerimos o uso do paulistinha (Danio rerio Hamilton 1822) na tentativa de compreender a modulação de comportamentos tipo-ansiedade pelo sistema serotonérgico. Demonstramos que os níveis extracelulares de serotonina no encéfalo de paulistinhas adultos expostos ao teste de preferência claro/escuro [PCE] (mas não ao teste de distribuição vertical eliciada pela novidade [DVN]) apresentam-se elevados em relação a animais manipulados mas não expostos aos aparatos. Além disso, os níveis teciduais de serotonina no rombencéfalo e no prosencéfalo são elevados pela exposição ao PCE, enquanto no mesencéfalo são elevados pela exposição ao DVN. Os níveis extracelulares de serotonina estão correlacionados negativamente com a geotaxia no DVN, e positivamente com a escototaxia, tigmotaxia e a avaliação de risco no PCE. O tratamento agudo com uma dose baixa de fluoxetina (2,5 mg/kg) aumenta a escototaxia, a tigmotaxia e a avaliação de risco no PCE, diminui a geotaxia e o congelamento e facilita a habituação no DVN. O tratamento com buspirona diminui a escototaxia, a tigmotaxia e o congelamento nas doses de 25 e 50 mg/kg no PCE, e diminui a avaliação de risco na dose de 50 mg/kg; no DVN, ambas as doses diminuem a geotaxia, enquanto somente a maior dose diminui o congelamento e facilita a habituação. O tratamento com WAY 100635 diminui a escototaxia nas doses de 0,003 e 0,03 mg/kg, enquanto somente a dose de 0,03 mg/kg diminui a tigmotaxia e a avaliação de risco no PCE. No DVN, ambas as doses diminuem a geotaxia, enquanto somente a menor dose facilita a habituação e aumenta o tempo em uma “base” (“homebase”). O tratamento com SB 224289 não produziu efeitos sobre a escototaxia, mas aumentou a avaliação de risco na dose de 2,5 mg/kg; no DVN, essa droga diminuiu a geotaxia e o nado errático nas doses de 2,5 e 5 mg/kg, enquanto a dose de 2,5 mg/kg aumentou a formação de “bases”. O tratamento com DL-para-clorofenilalanina (2 injeções de 300 mg/kg, separadas por 24 horas) diminuiu a escototaxia, a tigmotaxia e a avaliação de risco no PCE, aumentou a geotaxia e a formação de bases e diminuiu a habituação no DVN. Quando os animais são pré-expostos a uma “substância de alarme” co-específica, observa-se um aumento nos níveis extracelulares de serotonina associados a um aumento na escototaxia, congelamento e nado errático no PCE; os efeitos comportamentais e neuroquímicos foram bloqueados pelo pré tratamento com fluoxetina (2,5 mg/kg), mas não pelo pré-tratamento com WAY 100,635 (0,003 mg/kg). Animais da linhagem leopard apresentam maior escototaxia e avaliação de risco no PCE, assim como níveis teciduais elevados de serotonina no encéfalo; o fenótipo comportamental é resgatado pelo tratamento com fluoxetina (5 mg/kg). Esses dados sugerem que o sistema serotonérgico dessa espécie modula o comportamento no DVN e no PCE de forma oposta; que a resposta de medo produzida pela substância de alarme também parece aumentar a atividade do sistema serotonérgico, um efeito possivelmente mediado pelos transportadores de serotonina, e ao menos um fenótipo mutante de alta ansiedade também está associado a esses transportadores. Sugere-se que, de um ponto de vista funcional, a serotonina aumenta a ansiedade e diminui o medo em paulistinhas.

Azimuthal decorrelations between the two central jets with the largest transverse momenta are sensitive to the dynamics of events with multiple jets. We present a measurement of the normalized differential cross section based on the full data set (∫Ldt=36  pb(-1)) acquired by the ATLAS detector during the 2010 sqrt(s)=7  TeV proton-proton run of the LHC. The measured distributions include jets with transverse momenta up to 1.3 TeV, probing perturbative QCD in a high-energy regime.

Donor-specific transfusions (DST) induce allograft tolerance in animals. Evidence is growing that FoxP3+ regulatory T cells are associated with tolerance in humans. Forty-four biopsies from 69 living donor kidney transplant recipients (LDT) after DST, 53 biopsies from 69 matched deceased donor transplant recipients (DDT), obtained for graft dysfunction, and 12 biopsies from LDT without DST were retrospectively analyzed. FoxP3 positivity was more frequent in LDT/DST than in DDT biopsies (67% vs. 44%, P=0.02). Considering only biopsies with acute rejection, FoxP3 positivity was observed in 92% (11/12) after LDT/DST, but only in 50% (6/12) after DDT (P=0.03). The number of FoxP3+ T cells per total infiltrating cells in rejection biopsies was higher (P<0.05) from LDT/DST (4.1%) than from DDT or LDT (2.6%) without DST (2.5%). Six-year graft survival was better in patients with LDT/DST than with DDT (87.5% vs. 79.7%, P=0.04). The present investigation demonstrates an association between DST and FoxP3+ T cells. The effect of DST on regulatory T cells deserves further analysis in transplantation.

Theater findet in der Regel nicht ohne die gleichzeitige Anwesenheit von Zuschauenden und Agierenden statt: Eine begrenzte Öffentlichkeit wird zum räumlich definierten Publikum. Heute verlassen Theaterschaffende immer häufiger institutionelle Theaterräume und bewegen sich auf vielfältige Weise zwischen Theater und Öffentlichkeit. Theater erkundet seine Ränder und lädt unterschiedliche gesellschaftliche Akteure zur Mitgestaltung ein. Nichtprofessionelle Darsteller werden in die Stadttheater geholt und der alltägliche Raum wird zur Bühne gemacht. In vielen «sozialen Kunsttheaterprojekten» lösen sich die Grenzen zwischen Theaterkunst und Theaterpädagogik auf. Der erste Teil des Sammelbands fokussiert auf Phänomene, die in unterschiedlichen Zusammenhängen mit dem Terminus der Theatervermittlung in Verbindung gebracht werden. Im zweiten Teil wird die Theaterberichterstattung in Printmedien als Vermittlungskanal zwischen Theater und Öffentlichkeit untersucht.

In marmosets, it was observed that the synchrony among circadian activity profiles of animals that cohabite in family groups is stronger than those of the same sex and age of different families. Inside the group, it is stronger between the younger ones than between them and their parents. However, the mechanisms involved in the social synchrony are unknown. With the aim to investigate the synchronization mechanisms involved in the synchrony between the circadian activity profiles during cohabitation in pairs of marmosets, the motor activity was continuously registered by the use of actmeters on three dyads. The pairs were maintained in two different conditions of illumination: light-dark cycle LD 12:12 (LD cohabitation I – 21 days), and thereafter in LL (~350 lux). Under LL, the pairs were submitted to four experimental situations: 1. Cohabitation (LLJ I – 24 days), 2. Removal of one member of the pair to another room with similar conditions (LLS I – 20 days), 3. Reintroduction of the separated member in the cage of the first situation (LLJ II – 30 days) and 4. Removal of a member from each pair to another experimental room (LLS II – 7 days), to evaluate the mechanisms of synchronization. Ultimately, the members of each pair were reintroduced in the cage and were kept in LD cycle 12:12 (LDJ II – 11 days). The rhythms of pairs free-ran in LL, with identical periods between the members of each pair during the two stages of cohabitation. In the stages in which the animals were separated, only the rhythms of two females free-ran in the first stage and of three animals in the second one. In those conditions, the rhythms of animals of each pair showed different endogenous periods. Besides, during cohabitation in LD and LL, the members of each pair showed a stable phase relationship in the beginning of the active phase, while in the stages in which the animals were separated it was noticed a breaking in the stability in the phase relationships between the circadian activity profiles, with an increase in the difference in the phase angles between them. During cohabitation, at the transition between LD and LL, all animals showed free-running rhythms anticipating progressively the beginning and the end of the active phase in a phase similar to the previous condition, showing signs of entrainment to the previous LD. While in the posterior stages this was observed in only three animals between: LLT I and LLS I, and LLT II and LLS II, evidencing signs of entrainment to social cues between the members of each pair. On the other hand, one animal delayed progressively between LLT I and LLS I, three animals delayed between LLS I and LLT II, and three animals between LLT II and LLS II, perhaps by entrainment to the animals maintained outdoors in the colony. Similar process was observed in four animals between LLS II and LDT II, indicating entrainment to LD. In the transition between LLS I and LLT II, signs of masking was observed in the rhythm of a female in response to the male and in another pair in the rhythm of the male in regard to that of the female. The general and maximum correlations in the circadian activity profiles were stronger during cohabitation in LD and LL than in the absence of social contact in LL, evidencing the social effect. The cohabiting pairs had higher values of the maximum correlation in LD and LL than when the profiles were correlated to animals of different cages, with same or different sexes. Similar results were observed in the general correlation. Therefore, it is suggested that cohabitation induces a strong synchrony between circadian activity profiles in marmosets, which involves entrainment and masking. Nevertheless, additional studies are necessary to evaluate the effect of social cues on the synchronization of the circadian rhythm in pairs of marmosets in the absence of external social cues in order to confirm this hypothesis.

Visuelle Bildungsmedien erfahren wachsende Verbreitung in der universitären Lehre, und die Anzahl der verwendeten Medienformate steigt. Wir untersuchen empirisch den Lernerfolg dreier Formate - Lehrvideo plus Folien/Folien plus Audio/Audio - und stellen fest, dass der Lernerfolg die aufwändige Medienproduktion nicht in jedem Fall rechtfertigt. Das Untersuchungsdesign lädt zum Nachmachen und somit zum Ermitteln des geeignetsten Medienformats in einem spezifischen Lehrkontext ein. (DIPF/Orig.)
