大豆是我国最重要的油料作物之一,在我国的农业乃至幽民经济中占有重要地位。但是,由于我国人豆品质较差、产量较低,严重地影响着我国的大豆牛产及其在国际市场上的竞争力。农杆菌介导的转基因方法是大豆改良的最简便和最经济的方法之一,然而大豆基因转化效率低一直是大豆基因工程的主要限制因素。本研究以含有GUS报告基因和NPT II筛选基因的农杆菌1301侵染大豆子叶节和下胚轴,并用组织化学定位法测定GUS基因在叶节和下胚轴上瞬时表达,研究了抗氧化剂对提高大豆基因转化效率的影响,为建立大豆高效基因转化体系奠定基础。 以子叶节为外植体,用农杆菌侵染和共培养后进行GUS染色,观察其瞬时表达情况。在培养基中不含抗氧化剂的情况下,子叶节上未发现染色点,仅在下胚轴侧面有极少染色点。而在培养基巾含有抗氧化剂的情况下,外植体上产生了人量GUS染色点,其中大部分位于下胚轴处,子叶节部位几乎很少被染色。这说明下胚轴比f叶节更容易接受外源基因。本论文分别以子叶节和下胚轴为外植体进行研究。 以子叶节为外植体,在抗氧化剂作用下用农杆菌侵染,诱导丛生芽,在含有潮霉素的筛选培养基上培养,以筛选抗性苗。实验只得到一株抗性幼苗。 为了进一步研究抗氧化剂对GUS暴凶在下胚轴中瞬时表达的影响,我们特别以下胚轴为外植体,测定了多种条件下的瞬时表达率情况。 我们研究了共培养时间对大豆下胚轴基因瞬时表达率的影响,通过确定合适的共培养时间以获得最佳转化率。在共培养2天后,GUS基因的表达率是8%,但是在3天后,GUS基因瞬时表达率大幅度上升,达到23.4%。随后两天GUS基因瞬时表达率没有太大变化,因此,大豆下胚轴的GUS基因瞬时表达的最适共培养时间为3天。 农杆菌再悬浮液稀释浓度对大豆下胚轴GUS基因瞬时表达也有较大影响:再悬浮培养基与农杆菌菌液等体积时,GUS基因瞬时表达率最高,随稀释浓度提高,转化效率降低。这说明随农杆菌菌液浓度提高,侵染几率增加,从而提高了GUS基因的瞬时表达率。 很多研究表明大豆基因转化存在很大的品种问差异,我们选择了四种基因型差异较大的品种在上述最佳条件下分别测定了GUS基因瞬时表达率,但没有发现品种之间存在显著差异,说明抗氧化剂在大豆下胚轴基因转化中具有品种普遍适用性。 上述研究结果表明抗氧化剂能够大大促进GUS基因在下胚轴的瞬时表达,这对于今后开展大豆基因组研究和品质改良工作具有一定意义。

The pantherine lineage of the cat family Felidae (order: Carnivora) includes five big cats of genus Panthera and a great many midsized cats known worldwide. Presumably because of their recent and rapid radiation, the evolutionary relationship among pantherines remains ambiguous. We provide an independent assessment of the evolutionary history of pantherine lineage using two complete mitochondrial (mt) genes (ND2 and ND4) and the nuclear beta-fibrmogen intron 7 gene, whose utility in carnivoran phylogeny was first explored. The available four mt (ND5, cytb, 12S, and 16SrRNA) and two nuclear (IRBP and TTR) sequence loci were also combined to reconstruct phylogeny of 14 closely related cat species. Our analyses of combined mt data (six genes; approximate to 3750 bp) and combined mt and nuclear data (nine genes; approximate to 6500 bp) obtained identical tree topologies, which were well-resolved and strongly supported for almost all nodes. Monophyly of Panthera genus in pantherine lineage was confirmed and interspecific affinities within this genus revealed a novel branching pattern, with P. tigris diverging first in Panthera genus, followed by P. onca, P. leo, and last two sister species P. pardus and P. uncia. In addition, close association of Neofelis nebulosa to Panthera, the phylogenetic redefinition of Otocolobus manil within the domestic cat group, and the relatedness of Acinonyx jubatus and Puma concolor were all important findings in the resulting phylogenies. The potential utilities of nine different genes for phylogenetic resolution of closely related pantherine species were also evaluated, with special interest in that of the novel nuclear beta-fibrinogen intron 7. The family Cyprinidae is the largest freshwater fish group in the world, including over 200 genera and 2100 species. The phylogenetic relationships of major clades within this family are simply poorly understood, largely because of the overwhelming diversity of the group; however, several investigators have advanced different hypotheses of relationships that pre- and post-date the use of shared-derived characters as advocated through phylogenetic systematics. As expected, most previous investigations used morphological characters. Recently, mitochondrial DNA (mtDNA) sequences and combined morphological and mtDNA investigations have been used to explore and advance our understanding of species relationships and test monophyletic groupings. Limitations of these studies include limited taxon sampling and a strict reliance upon maternally inherited mtDNA variation. The present study is the first endeavor to recover the phylogenetic relationships of the 12 previously recognized monophyletic subfamilies within the Cyprinidae using newly sequenced nuclear DNA (nDNA) for over 50 species representing members of the different previously hypothesized subfamily and family groupings within the Cyprinidae and from other cypriniform families as outgroup taxa. Hypothesized phylogenetic relationships are constructed using maximum parsimony and Basyesian analyses of 1042 sites, of which 971 sites were variable and 790 were phylogenetically informative. Using other appropriate cypriniform taxa of the families Catostomidae (Myxocyprinus asiaticus), Gyrinocheilidae (Gyrinocheilus aymonieri), and Balitoridae (Nemacheilus sp. and Beaufortia kweichotvensis) as outgroups, the Cyprinidae is resolved as a monophyletic group. Within the family the genera Raiamas, Barilius, Danio, and Rasbora, representing many of the tropical cyprinids, represent basal members of the family. All other species can be classified into variably supported and resolved monophyletic lineages, depending upon analysis, that are consistent with or correspond to Barbini and Leuciscini. The Barbini includes taxa traditionally aligned with the subfamily Cyprininae sensu previous morphological revisionary studies by Howes (Barbinae, Labeoninae, Cyprininae and Schizothoracinae). The Leuciscini includes six other subfamilies that are mainly divided into three separate lineages. The relationships among genera and subfamilies are discussed as well as the possible origins of major lineages. (c) 2008 Published by Elsevier Inc.

The family Cyprinidae is widely distributed in East Asia, and has the important phylogenetic significance in the fish evolution. In this study, the 5' end partial sequences (containing exon 1, exon 2 and indel 1) of S6K1 gene were obtained from 30 representative species in Cyprinidae and outgroup using PCR amplification and sequencing. The phylogenetic relationships of Cyprinidae were reconstructed with neighbor joining (NJ), maximum parsimony (MP), maximum likelihood (ML), and Bayesian methods. Myxocyprinus asiaticus (Catostomidae) was assigned to the outgroup taxon. Similar phylogenetic relationships within the family Cyprinidae were achieved with the four analyses. Leuciscini and Barbini were monophyletic lineages respectively with the high nodal supports. Leuciscini comprises Hypophthalmichthyinae, Xenocyprinae, Cultrinae, Gobioninae, Acheilognathinae and East Asian species of Leuciscinae and Danioninae. Monophyly of East Asian clade was supported with high nodal support. Barbini comprises Schizothoracinae, Barbinae, Cyprininae and Labeoninae. The monophyletic lineage consisting of Danio rerio, D. myersi, and Rasbora trilineata was basal in the tree. In addition, the large fragment indels in intron 1 were analyzed to improve the understanding of Cyprinidae relationships. The results showed that the large fragment indels were correlated with the relations among species. Some conserved regions in intron 1 were thought to be involved in the functional regulation. However, no correlation was found between sequence variations and species characteristic size.

Several recent molecular phylogenetic studies of the sisorid catfishes (Sisoridae) have challenged some aspects of their traditional taxonomy and cladistic hypotheses of their phylogeny. However, disagreement with respect to relationships within this family in these studies highlights the need for additional data and analyses. Here we subjected 15 taxa representing 12 sisorids genera to comprehensive phylogenetic analyses using the second intron of low-copy nuclear S7 ribosomal protein (rpS7) gene and the mitochondrial 16S rRNA gene segments both individually and in combination. The competing sisorid topologies were then tested by using the approximately unbiased (AU) test and the Shimodaira-Hasegawa (SH) test. Our results support previously suggested polyphyly of Pareuchiloglanis. The genus Pseudecheneis is likely to be nested in the glyptosternoids and Glaridoglanis might be basal to the tribe Glyptosternini. However, justified by AU and SH test, the sister-group relationship between Pseudecheneis and the monophyletic glyptosternoids cannot be rejected based on the second intron of rpS7 gene and combined data analyses. It follows that both gene segments are not suitable for resolving the phylogenetic relationships within the sisorid catfishes. Overall, the second intron of rpS7 gene yielded poor phylogenetic performance when compared to 16S rRNA gene, the evolutionary hypothesis of which virtually agreed with the combined data analyses tree. This phenomenon can be explained by the insufficient length and fast saturation of substitutions in the second intron of rpS7 gene, due to substitution patterns such as frequent indels (insertion/deletion events) of bases in the sequences during the evolution.

Intron loss and its evolutionary significance have been noted in Drosophila. The current study provides another example of intron loss within a single-copy Dfak gene in Drosophila. By using polymerase chain reaction (PCR), we amplified about 1.3 kb fragment spanning intron 5-10, located in the position of Tyr kinase (TyK) domain of Dfak gene from Drosophila melanogaster species group, and observed size difference among the amplified DNA fragments from different species. Further sequencing analysis revealed that D. melanogaster and D. simulans deleted an about 60 bp of DNA fragment relative to other 7 Drosophila species, such as D. elegans, D. ficusphila, D. biarmipes, D. takahashii, D. jambulina, D. prostipennis and D. pseudoobscura, and the deleted fragment located precisely in the position of one intron. The data suggested that intron loss might have occurred in the Dfak gene evolutionary process of D. melanogaster and D. simulans of Drosophila melanogaster species group. In addition, the constructed phylogenetic tree based on the Dfak TyK domains clearly revealed the evolutionary relationships between subgroups of Drosophila melanogaster species group, and the intron loss identified from D. melanogaster and D. simulans provides a unique diagnostic tool for taxonomic classification of the melanogaster subgroup from other group of genus Drosophila.

人LMBR1(Limb region 1 homolog (mouse)) 基因位于染色体7q36区域,全长约210.2 kb,含17个外显子,编码一个由490个氨基酸构成的跨膜蛋白。研究表明,LMBR1 基因的表达活性与脊椎动物四肢的手指或脚趾数目变化有关;另外,发生在其重要元件——intron 5 内的许多变异与多种表型的轴前多指症((PPD, Preaxial polydactyly)存在相关性,这主要是因为LMBR1 intron 5 内含有一个与骨骼系统发育有关的基因(SHH(Sonic hedgehog)基因)的远程顺式调控元件。本研究旨在探究LMBR1基因 intron 5 内的遗传多样性,进而评估HapMap计划的样本选择策略,并检测该区域是否受自然选择的作用。 国际人类基因组单体型图计划(HapMap Project,The International Haplotype Map Project) 于2002年10月正式启动,该计划旨在构建人类基因组中常见变异的遗传图式。自其数据发布以来,广泛应用于生物医学、群体遗传学等领域,在复杂疾病的遗传机理研究、自然选择的检测等方面做出了前所未有的贡献;但是HapMap计划中样本的代表性有待评估。 本研究中,我们综合考虑地理来源信息及线粒体单倍型类群 (Haplogroup)信息选择了41个东亚人作为样本(以保证样本的代表性),测定位于LMBR1 基因intron 5 内的目的片段中存在的单核苷酸多态性(SNP, Singe nucleotide polymorphism)位点,通过所得数据与HapMap数据的比较,发现二者之间差异显著且HapMap数据不能覆盖所有我们得到的常见变异,因而我们认为:HapMap计划中国部分的样本选择策略有待进一步完善。 关于自然选择的研究不仅可以使我们了解生物的进化机制,同时还对复杂疾病的遗传机理研究具有重要的提示作用,因而,对于自然选择的检测,一直以来都是生物学研究的重点。平衡选择是一种维持遗传多态性的自然选择方式,现已发现很多与特定疾病或性状相关的基因或调控序列受平衡选择的作用,如 G6PD 基因、PTC 基因、FMO3 基因、FSHB 基因及 CCR5 基因5’端顺式调控区等我们对41个东亚样本中LMBR1 intron 5 内一段长为9256 bp (Chr7: 156280954-156271699 (Build36))的序列进行以 Tajima’s D 检验为主的群体遗传学分析,发现该区域在进化历程中受到平衡选择的作用。LMBR1 intron 5 内的多态位点与多种表型的多指症存在相关性,受其调控的 SHH 基因在骨骼系统发育中具有重要作用,人类骨骼系统的适应性进化等三方面的因素为该区域受平衡选择的作用提供了进一步的佐证。 总之,本研究对HapMap计划的样本选择策略和数据应用提供了一定的参考;同时还发现一个与骨骼系统发育有关的基因调控元件受平衡选择的作用。

应用GUS(葡萄糖苷酶 )基因标记技术将标记基因GUS导入受体菌S .fredii8855,标记菌株形成的根瘤可被GUS染色缓冲液染成蓝色 ,而土著菌形成的根瘤不能着色 .由此即可十分简便地确定土著菌的影响程度 .盆栽试验表明 ,S .fredii 8855的结瘤抗酸碱能力高于土著菌 ,能在土壤中较大范围内迁移 .当它的根瘤占有率不小于 43%时 ,接种能显著提高大豆产量 ,大豆产量与根瘤占有率呈正相关 (r=0 .98) ,而与总瘤数关系不大 (r=0 .1 3) .土壤N素显著抑制其结瘤 ,补加P能缓解这种抑制作用 .

In this study, the background activity of beta-glucuronidase (GUS) was analyzed histochemically and fluorometrically in the negative control of Laminaria japonica (Phaeophyta) thalli, showing low level of activity. GUS gene transformation without selectable gene in L. japonica was performed using four different promoters, i.e., Cauliflower mosaic virus 35S promoter (CaMV35S) from cauliflower mosaic virus, ubiquitin promoter (UBI) from maize, adenine-methyl transfer enzyme gene promoter (AMT) from virus in green alga Chlorella, and fucoxanthin chlorophyll a/c-binding protein gene promoter (FCP) from diatom Phaeodactylum tricornutum. The GUS transient activity was determined fluorometrically after bombarding sliced parthenogenetic sporophytes explants, and it was found that the activity resulting from CaMV35S and FCP promoters (in 114.3 and 80.6 pmol MU min(-1) (mg protein)(-1), respectively) was higher than for the other two promoters. The female gametophytes were bombarded and regenerated parthenogenetic sporophytes. FCP was the only promoter that resulted in detectable GUS chimeric expression activity during histochemical staining and polymerase chain reaction. Results of Southern blot showed that GUS gene was integrated with the L. japonica genome.
