The objective of this work was to select the most informative morphoagronomic descriptors for cassava (Manihot esculenta) germplasm and to evaluate the ability of different methods to select the descriptors. Ninety-five accessions were characterized using 51 morphoagronomic descriptors. Data were subjected to a multiple correspondence analysis (MCA), whose information was used in the following four methods of descriptor selection: reverse order of the descriptor for the pth factorial axis of the MCA (Jolliffe); sequential, multiple correspondence analysis (SMCA); mean of the contribution orders of the descriptor in the first three factorial axes (C3PA); and C3PA method weighted by the respective eigenvalues of the full analysis (C3PAWeig). The correlations between the dissimilarity matrix with all descriptors and the most informative descriptors were high and significant (0.75, 0.77, 0.83, and 0.84 for C3PAWeig, C3PA, SMCA, and Jolliffe, respectively). The less informative descriptors were discarded, considering those common among the selection methods and relevant for the breeding interests. Therefore, 32 morphoagronomic descriptors with correlation between the dissimilarity matrices (r=0.81) were selected, due to their high capacity to discriminate cassava germplasm and to their ability to maintain some preliminary agronomic traits, useful for the initial characterization of the germplasm.

Ionotropic Receptors (IRs) are a recently characterized family of olfactory receptors in the fruit fly, Drosophila melanogaster. IRs are not related to insect Odorant Receptors (ORs), but rather have evolved from ionotropic glutamate receptors (iGluRs), a conserved family of synaptic ligand-gated ion channels. Here, we review the expression and function of IRs in Drosophila, highlighting similarities and differences with iGluRs. We also briefly describe the organization of the neuronal circuits in which IRs function, comparing and contrasting them with the sensory pathways expressing ORs. Finally, we summarize the bioinformatic identification and initial characterization of IRs in other species, which imply an evolutionarily conserved role for these receptors in chemosensation in insects and other protostomes.

Nitrofurazone (NF), 5-nitro-2-furaldehyde semicarbazone, a broad-spectrum antibiotic, has reported toxic effects and low solubility in water. It would be of great interest to form inclusion complexes between NF and a cyclodextrin, to develop more effective and safer antibiotic formulations. This paper focuses on the preparation of inclusion complexes of NF with 2-hydroxypropyl-β- cyclodextrin (HP-β-CD) and their initial characterization by evaluating rates of complex formation, photostability, solubility isotherms, release rate profiles, stoichiometry of the complexes and their morphology, as revealed by scanning electron microscopy. The kinetic tests of complex formation revealed that 17,3 h is enough for stabilization of the NF-cyclodextrin complex. The solubility isotherm studies showed that the isotherm changes from type A to type B, as a function of temperature. The photostability experiments showed that the insertion of the NF in the HP-β-CD cavity protects the drug from photodecomposition. The release kinetic tests showed that the profile of NF release from the complex is altered by the presence of HP-β-CD in the medium. A Job's plot indicated that the stoichiometry of the complex was 1:1 NF:HP-β-CD. The scanning electron micrographs showed changes in the crystal structure of NF in the complex. This study focused on the physicochemical properties of drug-delivery formulations that could potentially be developed into a novel type of therapy with NF.

Human development and population growth during the twentieth century increased the water demand, tripling its consumption between 1950 and 1990. As the water streams were polluted; and as water is the source of minerals and also regulates vital functions, it becomes the vehicle of transmission and consequently spreads many diseases. Probably, the industries are the major responsible for this pollution when they dump untreated effluents to water streams, saturating the already insufficient net of sanitation facilities polluting water and soil. An effective treatment has been established with low cost in Europe and the United States, through constructed systems on wetlands Constructed Wetland Systems - CWSs, gradually used in other countries in the last three decades. Lately, we observe a continuous growth in Brazilian poultry business, and poultry industry showed greatest dynamism in the country, following the global market. Pondering this information and the efficiency of such treatment, this work aimed to study prototypes, in a laboratory scale, simulating ascending and descending types of CWSs, vegetated with aquatic macrophytes Eichhornia crassipes and the uses of aggregates and soil, to treat industrial wastewater from slaughterhouses and aviary. We conducted the initial characterization of the effluent to have an idea of its constituents and to scale the system and the continuous flow. Furthermore, we characterized the soil to be used in this system. The collects are periodically made in the refrigeration industry FRICOCK FRIGORIFICAÇÃO AVICULTURA INDÚSTRIA E COMÉRCIO LTDA. for local treatment simulation. The effluent that was treated with 12 prototypes of CWSs are analyzed with some frequency. The results of these reviews were compared to the effluent coming from the industry... Children learn very early which groups are stigmatized and how they should be treated. They learn formally and informally, by listening and watching people and the world around them. The construction of children's representations about disabilities starts with access to information on the subject, which may come from their caregivers, media, school etc. This work is part of a monograph that examined the effects of an educational program involving strategies for non-disabled children on conceptions and attitudes concerning mental disability. This paper refers to the initial characterization of these children and their access to information on disability, and to identifying previous access, source and memory of such information. The study included 40 children from two classrooms of the first grade of a public school in Marília, SP. The children were interviewed using an individual semi structured questionnaire. Many children said they did not have access to information on the subject. Among those who had previous access to the information, the television was the most frequent source of information, and characteristics of the deficiencies, relations with diseases and encouragement of solidarity were among the contents reminded by them. The data emphasize the need for the media to expand concerns with the content presented to the public, for parents and teachers to become more involved with the issue, seeking to be informed and revise their own conceptions, in order to transform the subject into something that is part of daily school and family environment.

Electrical conductivity has been proposed as a rapid test to evaluate seed vigor; however, few researches have emphasized methodologies to its use in seeds of medicinal plants, such as chamomile. The objective of the research was to evaluate the electrical conductivity of chamomile seeds affected by different imbibition times and sample size. The evaluations consisted of moisture content, germination and vigor (first count of germination) to seed initial characterization. Then, it was evaluated the electrical conductivity, affected by imbibition time (6, 12, 24 e 48 hours) and seed amount per sample (25, 50, 75, 100). The completely randomized design was used with four replications, arranged as a 4 x 4 factorial. Means were compared by the Tukey test at 5% of probability. It was concluded that the electrical conductivity of chamomile seeds is affected by the number of seeds per sample and imbibition time isolately.

Limited information is available regarding the modulation of genes involved in the innate host response to Paracoccidioides brasiliensis, the etiologic agent of paracoccidioidomycosis. Therefore, we sought to characterize, for the first time, the transcriptional profile of murine bone marrow-derived dendritic cells (DCs) at an early stage following their initial interaction with P. brasiliensis. DCs connect innate and adaptive immunity by recognizing invading pathogens and determining the type of effector T-cell that mediates an immune response. Gene expression profiles were analyzed using microarray and validated using real-time RT-PCR and protein secretion studies. A total of 299 genes were differentially expressed, many of which are involved in immunity, signal transduction, transcription and apoptosis. Genes encoding the cytokines IL-12 and TNF-alpha, along with the chemokines CCL22, CCL27 and CXCL10, were up-regulated, suggesting that P. brasiliensis induces a potent proinflammatory response in DCs. In contrast, pattern recognition receptor (PRR)-encoding genes, particularly those related to Toll-like receptors, were down-regulated or unchanged. This result prompted us to evaluate the expression profiles of dectin-1 and mannose receptor, two other important fungal PRRs that were not included in the microarray target cDNA sequences. Unlike the mannose receptor, the dectin-1 receptor gene was significantly induced, suggesting that this beta-glucan receptor participates in the recognition of P. brasiliensis. We also used a receptor inhibition assay to evaluate the roles of these receptors in coordinating the expression of several immune-related genes in DCs upon fungal exposure. Altogether, our results provide an initial characterization of early host responses to P. brasiliensis and a basis for better understanding the infectious process of this important neglected pathogen.

A range of vectors were made in which the EYFP gene or the Cre gene were inserted in the start codon of the NG2 gene. The NG2-EYFP vectors were used to generate NG2-EYFP “knockin” mice by homologous recombination. The F1 generation showed lack of EYFP expression, due to NeoR cassette interference. Excision of the NeoR, by breeding the F1 generation to ELLA-Cre mice allowed proper expression of EYFP. NG2-EYFP heterozygous mice were characterized in detail for astrocytic, neurogenic and oligodendrocytic properties through antibody labeling. NG2-EYFP+ cells did not label for the astrocyte marker GFAP, but some cells did express S100 Beta. The cells did not label with any neuronal markers like Beta III tubulin, Neun, and double cortin, but many of the NG2-EYFP+ cells made intimate contacts to the neurons. These contacts are widespread throughout the grey and white matter of the brain. The NG2-EYFP+ cells did label for oligodendrocyte markers like PDGFα-R, NG2, Olig2, O4, and Sox 10. There were a few cells termed phantom cells that did label for NG2, but had no EYFP expression. This could have been caused by improper excision of the NeoR cassette in the F2 generation. The heterozygous mouse is a tool to allow the characterization of the in vivo properties of the NG2+ cells. Breeding of these mice to homozygosity yielded an NG2-knockout mouse, which was also subjected to initial characterization. The NG2-EYFP homozygous showed equivalent cell labeling results to the NG2-EYFP heterozygous mouse, but the phantom cells disappeared in the knockout. The results show that the NG2 cells are a heterogenous population that does not express astrocytic or neuronal markers. The homozygous mouse is an ideal tool to further dissect the properties of the cells, lacking NG2 in vivo.
